Biodegradation and detoxification of neonicotinoid insecticide thiamethoxam by white-rot fungus Phanerochaete chrysosporium

Anwei Chen,Wenjie Li,Xiaoxiao Zhang,Cui Shang,Si Luo,Ruoyu Cao,Doudou Jin

The extensive use of neonicotinoid pesticides in the past two decades caused serious impacts on many kinds of living beings. Therefore, it has been strongly suggested to detoxify and eliminate neonicotinoids’ residual levels in environment. Here, the degradation and detoxification of thiamethoxam (THX) by white-rot fungus Phanerochaete chrysosporium was conducted. Results shown that P. chrysosporium can tolerate THX and degraded 49% of THX after incubation for 15 days, and then 98% for 25 days at the initial concentration of 10 mg/L, which indicates the excellent degradation ability of this fungus to THX. Based on the by-products identified, THX underwent dechlorination , nitrate reduction , and C–N cleavage between the 2-chlorothiazole ring and oxadiazine. ( Z )-N-(3-methyl-1,3,5-oxadiazinan-4-ylidene)nitramide and 3-methyl-1,3,5-oxadiazinan-4-imine were identified as the main metabolites. The impacts of THX and its corresponding degradation intermediates on the growth of E. coil and Microcystis aeruginosa as well as the germination of rape and cabbage demonstrated that P . chrysosporium effectively degrades THX into metabolites and reduces its biotoxicity. The present work demonstrates that P . chrysosporium can be effectively used for degradation and detoxification of THX.

Excavation, expression, and functional analysis of a novel zearalenone-degrading enzyme

Gao Huihui,Lu Dan,Xing Mingyan,Xu Qing,Xue Feng

Zearalenone (ZEN) is a toxic secondary metabolite of Fusarium sp. commonly found in wheat, corn, and other crops. In addition to economic losses, ZEN can seriously endanger the health of both humans and livestock, thus presenting an urgent need for ZEN-detoxifying enzymes that function in the extreme heat or pH conditions of industrial fermenters. Here, we identify and characterize the activity of the ZEN-degrading enzyme from Exophiala spinifera , ZHD_LD, which shares 60.15% amino acid identity and a conserved catalytic triad with the well-characterized ZEN-detoxifying protein ZHD101 from Clonostachys rosea . Biochemical activity and stability assays indicated that purified recombinant ZHD_LD exhibited high activity against ZEN with optimal reaction conditions of 50 ℃ and pH 7.0–10.0. Structural modeling of the ZHD_LD active site and comparison with ZHD101 revealed its likely mechanism of ZEN degradation. This research provides an industrially valuable candidate enzyme for ZEN detoxification in food and livestock feed.

Dual-enzyme cascade colorimetric biosensor for safe, rapid on-site detection and detoxification of ochratoxins

Xiao Liang,Zhuobing Zhang,Yilei Du,Ziliang Li,Nana Xu,Jian Gao,Huimin Qin,Liping Du,Song You,Jinshan Li,Weidong Liu,Yanhe Ma

Ochratoxins, toxic mycotoxins present in food and feed, pose a threat to human health due to their various toxic effects. Developing a safe, convenient, and rapid on-site detection tool for monitoring ochratoxins is essential for food and environmental safety, especially in developing countries. Although numerous effective ochratoxin sensing and detection systems are available, most do not combine detoxification with detection capabilities. In this work, an ochratoxin-detoxifying enzyme was cascaded with phenylalanine dehydrogenase to construct a colorimetric biosensor capable of simultaneously detecting and removing Ochratoxin A (OTA) and Ochratoxin B (OTB) contamination. Once the ochratoxins are detoxified into non-toxic OTα/OTβ and phenylalanine, 1-methoxy-5-methylphenazinium methyl sulfate (MPMS) functions as a signal reporter, converting the ochratoxin concentration into a colorimetric signal that gradually shifts from red to green. This allows for real-time visual observation with the naked eye. Additionally, OTA/OTB within a concentration range of 0–50 μg/mL can be quantitatively analyzed using a microplate reader or a smartphone. Under the optimized conditions, the colorimetric biosensor effectively eliminated and detected OTA/OTB at a spiked concentration of 5 μg/L in Chinese liquor samples within 1 h. Hence, the dual-enzyme cascade colorimetric biosensor has been demonstrated to be a safe, portable, rapid, and accurate tool for the detection and elimination of OTA/OTB in food.

Detoxification of three toxins in castor meal by a novel continuous phase-transition extraction in a pilot-scale

Guo Liu,Mingjuan Liao,Baoyan Guo,Qixin Kan,Shuangde Zhou,Konglong Feng,Wanhuang Lin,Yahui Huang,Jianyin Miao,Yong Cao

Castor ( Ricinus communis L.) meal, a by-product of castor oil production, is a high-quality protein source. Unfortunately, the toxic substance limits its application. Herein, a novel continuous phase transition extraction (CPTE) technology at pilot scale was explored for castor meal detoxification. The detoxification conditions were optimized using response surface methodology (RSM) as: treatment time of 3 h, extraction pressure of 0.16 MPa and temperature of 84.8°C. And the removal rate of allergens, hemagglutinin, and ricin reached 79.2 %, 99.8 % and 99.0 %, respectively. The protein content of castor meal increased from 38.4% to 39.0%. The nontoxicity nature of the treated caster meal was verified via an acute toxicity tests where the mean lethal dose was >5000 mg/kg body weight. Overall, CPTE offers a novel and efficient tool for castor meal detoxification, with the superiority of non-nutrient loss, it improved the application value of castor meal and reduced the risk of environmental pollution.

Measurement and control of containing-fluorine particulate matter emission during spent pot lining combustion detoxification process

Funian Han,Liang Yu,Xinyun Mo,Lu Zhang,Jiaqi Jia,Jingmeng Dong,Shaopeng Gu

The particle size distribution (PSD), composition, morphology, and formation mechanism of particulate matter (PM) released from the combustion of spent pot lining with and without CaSiO 3 were investigated. The results showed that NaF and Na 3 AlF 6 were found to be the main compositions of PM, and the particle size distribution of PM shows a bimodal distribution. CaSiO 3 substantially inhibited the emission of PM by transforming NaF, Na 3 AlF 6 , and CaF 2 into stable Ca 4 Si 2 O 7 F 2 . Moreover, CaSiO 3 also limited the formation of high hazardous PM 0.2 by providing SiO 2 , Al 2 O 3 , and NaAlSiO 4 with high melting points as the core of promoting the growth of PM in particle size.

Visible-light-driven simultaneous decontamination of multi-antibiotics by facile synthesized BiOCl loaded food wastes biochar

Xiaoqian Zhang,Zhipeng Wu,Yixiao Wu,Abdulmoseen Segun Giwa,Shaobin Huang,Lishan Niu

Environmental dissemination caused by widespread use of antibiotics has been regarded as a possible hazard to aquatic ecosystem and human health. The increasing misgivings make it imperative to develop a novel catalyst with remarkable visible-light-driven activity to remove antibiotics, especially for their simultaneous decontamination . Herein, C/BiOCl composites were successfully prepared by decorating BiOCl nanosheets on food wastes biochar (C) by a simple hydrolysis strategy. Not only the binary system of tetracycline antibiotics, but also the ternary mixture could be simultaneously photodegraded over 25% C/BiOCl within 15 min irradiation. The improved photocatalytic activities could be ascribed to the introduction of biochar, endowing increased surface area, enhanced separation of photo-generated charge carriers, and better light absorption . The as-prepared 25% C/BiOCl also demonstrated satisfactory stability and positive removal effect in actual water samples. The present work provides new insights into the development of biochar-based photocatalysts for simultaneous degradation of multiple antibiotics.

Solvent-free one-step mechanochemical synthesis of magnetic graphene composite adsorbent for efficient removal of multiple aflatoxins in vegetable oils

Libin Wan,Fanghui Zhao,Ming Lv,Shucai Gao,Haidong Gao,Mantang Chen,Lianjun Song,Wei Liu,Hongwei Liu,Fayun Wang

Aflatoxin-contaminated vegetable oils pose severe threats to human health. Although magnetic graphene-based materials can be used for aflatoxins (AFs) removal, their current preparation methods involve complex, multi-step procedures and are time-consuming. Herein, we proposed a simple, efficient and solvent-free mechanochemical approach for one-step synthesis of magnetic graphene composite (MGC) directly from graphite and Fe 3 O 4 . The MGC was used as an adsorbent to remove four AFs from vegetable oils, and the adsorption mechanism was further explored by conducting batch adsorption experiments. The results demonstrated that MGC could simultaneously remove 85.8–100 % of AFs from 13 types of oils within 60 min at 30 °C. The detoxification process exhibited low cytotoxicity towards HepG2 cells, and has little effect on the quality of oils. Thus, MGC exhibited promising application potential for vegetable oil detoxification in food industry due to its facile scalable production, efficient detoxification capability, good biosafety, and easy recovery and regeneration.
