Comparison of glutaraldehyde and carbodiimides to crosslink tissue engineering scaffolds fabricated by decellularized porcine menisci

Shuang Gao,Zhiguo Yuan,Weimin Guo,Mingxue Chen,Shuyun Liu,Tingfei Xi,Quanyi Guo

The objectives of this study were to fabricate porous scaffolds using decellularized meniscus, and to explore a preferable crosslinking condition to enhance mechanical properties of scaffolds. Moreover, the microstructure, porosity, biodegradation and cytotoxicity were also evaluated. EDAC or GTA in different concentration was used to crosslink scaffolds. FTIR demonstrated functional groups change in crosslinking process. SEM photography showed that crosslinked scaffolds had blurry edges, which resulted scaffolds crosslinked by 1.2   mol/l EDAC had smaller porosity than other groups. The structure change enhanced antidegradation property. After immersing in enzyme solution for 96   h, scaffolds crosslinked by GTA and EDAC could maintain their mass >   70% and 80%. Most importantly, mechanical properties of crosslinked scaffolds were also improved. Uncrosslinked Scaffolds had only 0.49   kPa in compression modulus and 12.81   kPa in tensile modulus. The compression and tensile modulus of scaffolds crosslinked by 1.0% GTA were 1.42 and 567.44   kPa respectively. The same value of scaffolds crosslinked by 1.2   mol/l EDAC were 1.49 and 532.50   kPa. Scaffolds crosslinked by 1.0% and 2.5% GTA were toxic to cells, while EDAC groups showed no cytotoxicity. Chondrocytes could proliferate and infiltrate within scaffolds after seeding. Overall, 1.2   mol/l EDAC was a preferable crosslinking condition.

A Double Cross-Linked Injectable Hydrogel Derived from Muscular Decellularized Matrix Promotes Myoblast Proliferation and Myogenic Differentiation

Zhao Huang,Jianwen Cheng,Wei Su

Injectable hydrogels possess tremendous merits for use in muscle regeneration; however, they still lack intrinsic biological cues (such as the proliferation and differentiation of myogenic cells), thus considerably restricting their potential for therapeutic use. Herein, we developed a double cross-linked injectable hydrogel composed of methacrylamidated oxidized hyaluronic acid (MOHA) and muscular decellularized matrix (MDM). The chemical composition of the hydrogel was confirmed using1H NMR and Fourier transform infrared spectroscopy. To achieve cross-linking, the aldehyde groups in MOHA were initially reacted with the amino groups in MDM through a Schiff-based reaction. This relatively weak cross-linking provided the MOHA/MDM hydrogel with satisfactory injectability. Furthermore, the methacrylation of MOHA facilitated a second cross-linking mechanism via UV irradiation, resulting in improved gelation ability, biomechanical properties, and swelling performance. When C2C12 myogenic cells were loaded into the hydrogel, our results showed that the addition of MDM significantly enhanced myoblast proliferation compared to the MOHA hydrogel, as demonstrated by live/dead staining and Cell Counting Kit-8 assay after seven days of in vitro cultivation. In addition, gene expression analysis using quantitative polymerase chain reaction indicated that the MOHA/MDM hydrogel promoted myogenic differentiation of C2C12 cells more effectively than the MOHA hydrogel, as evidenced by elevated expression levels of myogenin, troponin T, and MHC in the MOHA/MDM hydrogel group. Moreover, after four to eight weeks of implantation in a full-thickness abdominal wall-defect model, the MOHA/MDM hydrogel could promote the reconstruction and repair of functional skeletal muscle tissue with enhanced tetanic force and tensile strength. This study provides a new double cross-linked injectable hydrogel for use in muscular tissue engineering. Keywords:hydrogels;biomaterials;muscular decellularized matrix;proliferation;myogenic differentiation

The study of a new detergent (octyl-glucopyranoside) for decellularizing porcine pericardium as tissue engineering scaffold

Jiaoming Dong,Yu Li,Xiumei Mo

Background The use of extracellular matrix (ECM) derived from decellularized tissue is increasingly frequent in regenerative medicine and tissue engineering . However, it is recognized that currently used decellularization procedures have negative effects on ECM integrity. The objective of this study was to investigate the impact of a decellularization protocol with a new detergent on the ECM integrity of porcine pericardium (PP) compared with other traditional detergents. Materials and methods Fresh PP were decellularized by sodium deoxycholate in combination with Triton X-100 (SDT), sodium dodecyl sulfate (SDS), and octyl-glucopyranoside (OGP), respectively. Histologic analysis and scanning electron microscopy were performed to confirm the removal of cells and to examine the structure of ECM. DNA content was examined by the method of DNA extraction . Mechanical properties and biochemical compositions of ECM were also studied. Results Histologic analysis and DNA determination demonstrated that SDS and OGP completely removed the cells, and the major ECM structure was preserved well for PP treated with 1% (wt/vol) OGP but disrupted for PP treated with SDS; whereas treatment with SDT was insufficient to remove cells from PP. Uniaxial tensile tests showed that PP decellularized by OGP had similar mechanical properties to native PP, whereas the mechanical properties of PP decellularized by SDS and SDT decreased. The biochemical compositions of PP decellularized by OGP were also well conserved, except that glycosaminoglycans markedly decreased. Moreover, the results obtained in the MTT study further indicated that the cytotoxicity of PP decellularized by OGP was significantly lower than that decellularized by SDS and SDT. Conclusion It is suggested that the environmentally friendly and nontoxic OGP can be used as a decellularizing agent. The OGP method could achieve both complete removal of cells from native PP and preservation of the matrix structure; thus, it might be a suitable approach to preparation of tissue engineering heart valve scaffold.

A Sponge-Like Double-Layer Wound Dressing with Chitosan and Decellularized Bovine Amniotic Membrane for Promoting Diabetic Wound Healing

Yang Yang,Yanyan Zhang,Yishu Yan,Qian Ji,Yutong Dai,Suyuan Jin,Yanxian Liu,Jinghua Chen,Liping Teng

The diabetic wounds do not heal easily in part because they are susceptible to infection due to environmental influences. Wound dressing is crucial to wound healing, as it can basically protect the wound from external damages and provide a suitable microenvironment for tissue regeneration. In this study, a double-layer membrane that consists of chitosan sponge and decellularized bovine amniotic membrane (dBAM) has been developed by freeze-casting method. The results showed that the porous structure of the sponge layer improved the performances of blood coagulation and swelling. The dense dBAM can optimize the mechanical property of wound dressing. In vitro studies revealed that the bilayer membrane had favorable biocompatible, especially for human foreskin fibroblast cells (HFF-1) cell adhesion and proliferation. Moreover, the full-thickness skin defects of diabetic model mice that treated with bilayer membrane showed over 80% closure in 8 days. Our findings imply that the double-layer dressing has great potentials to be used in diabetic patients. Keywords:chitosan;decellularized bovine amniotic membrane;diabetic wound healing;double-layer dressing

3D printing electrospinning fiber-reinforced decellularized extracellular matrix for cartilage regeneration

Weiming Chen,Yong Xu,Yaqiang Li,Litao Jia,Xiumei Mo,Gening Jiang,Guangdong Zhou

Cartilage decellularized matrix (CDM) is considered a promising biomaterial for fabricating cartilage tissue engineering scaffolds. An ideal CDM-based scaffold should possess customizable 3D shape for complex tissue regeneration and proper pore size for cell infiltration , as well as provide mechanical support for cell growth. 3D printing is an efficiently technique for preparing customizable 3D scaffolds, however, fabricating CDM-based 3D-printed scaffolds with customizable shapes, proper pore structure and satisfactory mechanical properties remains a challenge. In the current study, to achieve customizable CDM-based 3D scaffolds, CDM was successfully processed into inks suitable for 3D printing. Further, the poor mechanics of CDM-based scaffolds were significantly improved by adding electrospinning fiber into the CDM-based inks for 3D printing. Importantly, the 3D-printed electrospinning fiber-reinforced CDM-based scaffold presented good biocompatibility and can enhance repair articular cartilage defects in rabbits. The current study provides a novel strategy for printing electrospinning fiber-reinforced CDM-based scaffolds for tissue regeneration.

An electrospun fiber reinforced scaffold promotes total meniscus regeneration in rabbit meniscectomy model

Shuang Gao,Mingxue Chen,Pei Wang,Yan Li,Zhiguo Yuan,Weimin Guo,Zengzeng Zhang,Xueliang Zhang,Xiaoguang Jing,Xu Li,Shuyun Liu,Xiang Sui,Tingfei Xi,Quanyi Guo

Low vascularization in meniscus limits its regeneration ability after injury, and tissue engineering is the most promising method to achieve meniscus regeneration. In this study, we fabricated a kind of composite scaffold by decellularized meniscus extracellular matrix/polycaprolactone (DMECM/PCL) electrospinning fibers and porous DMECM, in which DMECM/PCL fibers were used as reinforcing component. The tensile modulus of the composite scaffold in longitudinal and crosswise directions were 8.5 ± 1.9 and 2.3 ± 0.3 MPa, respectively. Besides that, the DMECM/PCL electrospinning fibers enhanced suture resistance of the composite scaffold more than 5 times than DMECM scaffold effectively. In vitro cytocompatibility showed that the porous structure provided by DMECM component facilitated meniscus cells’ proliferation. DMECM was also the main component to regulate cell behaviors, which promoted meniscus cells expressing extracellular matrix related genes such as COL I, COL II, SOX9 and AGG . Rabbits with total meniscectomy were used as animal model to evaluated the composited scaffolds performance in vivo at 3 and 6 months. Results showed that rabbits with scaffold implanting could regenerate neo-menisci in both time points. The neo-menisci had similar histology structure and biochemical content with native menisci. Although neo-menisci had inferior tensile modulus than native ones, its modulus was improved with implanting time prolonging. MRI imaging showed the signal of neo-meniscus in the body is clear, and X-ray imaging of knee joints demonstrated the implantation of scaffolds could relief joint space narrowing. Moreover, rabbits with neo-menisci had better cartilage condition in femoral condyle and tibial plateau compared than meniscectomy group. Statement of Significance We fabricated the meniscus scaffold by combining porous decellularized meniscus extracellular matrix (DMECM) and DMECM/PCL electrospinning fibers together, which used the porous structure of DMECM, and the good tensile property of electrospinning fibers. We believe single material cannot satisfy increasing needs of scaffold. Therefore, we combined not only materials but also fabrication methods together to develop scaffold to make good use of each part. DMECM in electrospinning fibers also made these two components possible to be integrated through crosslinking. Compared to existing meniscus scaffold, the composite scaffold had (1) soft structure and extrusion would not happen after implantation, (2) ability to be trimmed to suitable shape during surgery, and (3) good resistance to suture.

A 3D bioprinted potential colorectal tumor model based on decellularized matrix/gelatin methacryloyl/nanoclay/sodium alginate hydrogel

Xinyue Liu,Yan Shu,Jingjing Zhu,Huan Fang,Ya Su,Hailin Ma,Bing Li,Jie Xu,Yuen Yee Cheng,Bo Pan,Kedong Song

Colorectal cancer (CRC) is now the third most common cancer worldwide. However, the development cycle for anticancer drugs is lengthy and the failure rate is high, highlighting the urgent need for new tumor models for CRC-related research. The decellular matrix (dECM) offers numerous cell adhesion sites, proteoglycan and cytokines. Notably, porcine small intestine is rich in capillaries and lymphatic capillaries, which facilitates nutrient absorption. This study, we utilized dECM, along with methylacryloyl gelatin (GelMA), sodium alginate (SA) and nanoclay (NC) to create a hydrogel scaffold through 3D extrusion bioprinting. Human CRC cells (HCT8) were seeded onto the scaffold and their drug resistance was tested using 5-fluorouracil (5-FU). Our findings indicate that dECM enhances the hydrophilic properties, mechanical strength and biocompatibility of the scaffold. Furthermore, compared to traditional two-dimensional (2D) models, the three-dimensional (3D) scaffold supports the long-term growth of tumor spheres. After 2 days of 5-FU treatment, the cell survival rate reaches 88.06 ± 0.51 %. This suggests that our scaffold provides a promising alternative platform for in vitro research on cancer mechanisms, anti-cancer drug screening and new drug development.

Designing injectable dermal matrix hydrogel combined with silver nanoparticles for methicillin-resistant Staphylococcus aureus infected wounds healing

Chen Sunfang,Yao Jun,Huo Shicheng,Xu Chennan,Yang Ruting,Tao Danhua,Fang Bin,Ma Gaoxiang,Zhu Zaihua,Zhang Ye,Guo JingJing

Hydrogel-based delivery systems have now emerged as a pivotal platform for addressing chronic tissue defects, leveraging their innate capacity to suppress pathogenic infections and facilitate expedited tissue regeneration. In this work, an injectable hydrogel dressing, termed AgNPs-dermal matrix hydrogel (Ag@ADMH), has been designed to expedite the healing process of wounds afflicted with methicillin-resistant Staphylococcus aureus (MRSA), featuring sustained antibacterial efficacy. The synthesis of the hydrogel dressing entailed a self-assembly process of collagen fibers within an acellular dermal matrix to construct a three-dimensional scaffold, encapsulated with plant polyphenol-functionalized silver nanoparticles (AgNPs). The Ag@ADMH demonstrated exceptional biocompatibility, and enables a sustained release of AgNPs, ensuring prolonged antimicrobial activity. Moreover, the in vitro RT-qPCR analysis revealed that compared with ADMH, Ag@ADMH diminish the expression of iNOS while augmenting CD206 expression, thereby mitigating the inflammatory response and fostering wound healing. Especially, the Ag@ADMH facilitated a reduction in M1 macrophage polarization, as evidenced by a significant decrement in the M1 polarization trend and an enhanced M2/M1 ratio in dermal matrix hydrogels laden with AgNPs, corroborated by confocal microscopy and flow cytometry analyses of macrophage phenotypes. The in vivo assessments indicated that Ag@ADMH minimized fibrous capsule formation. In a full-thickness skin defect model of MRSA infection, the formulation significantly attenuated the inflammatory response by reducing MPO and CD68 expression levels, concurrently promoting collagen synthesis and CD34 expression, pivotal for vasculogenesis, thereby accelerating the resolution of MRSA-infected wounds. These attributes underscore the injectable extracellular matrix hydrogel as a formidable strategy for the remediation and regeneration of infected wounds. Graphical Abstract

Caprine dermal scaffolds for repair of full-thickness skin wounds in rabbits

Parvez Ahmad,Sangeeta Devi Khangembam,Anil Kumar Gangwar,Vipin Kumar Yadav,Prafull Kumar Singh,Yogendra Singh,Ravi Prakash Goyal,Surendra Pratap Chakraverty,Rajesh Kumar Verma

Biological scaffolds prepared after decellularization are used for the restoration of damaged tissues. A number of chemicals are used for bioscaffold preparation, and some of them damage their composition and architecture. Herein, we investigated the Sapindus mukorossi fruit pericarp extract (SPE) (5%) for decellularization of the caprine dermis. The dermal samples were processed in 5% SPE over magnetic stirrer for 96h at room temperature. The decellularization efficiency of SPE was analyzed by histological examination, DAPI staining, scanning electron microscopy (SEM), quantification of DNA hydroxyproline and hemocompatibility determination. Further, these acellular caprine dermal scaffolds were transplanted on full thickness skin wounds of group III New Zealand white rabbits. The wounds were left open in group I (Sham) and reconstructed by autograft in group II (n = 6 in each group). Continuous agitation of native caprine dermal tissues in 5% SPE for 96 hours leads to complete decellularization without affecting the extracellular matrix architecture. Microscopic observation of decellularized samples did not show any nuclei. DNA quantity was reduced (p < .05) in decellularized samples and scaffolds were found to be hemocompatible. Complete healing was observed on day 28 in groups II and III. No significant difference was noted in IgG in all the groups. Quantitative assessment of MDA showed a significant increase in groups I and II. Our results suggested that the 5% SPE solution effectively decellularized the native caprine dermis and the scaffolds were well tolerated by the animals.

Effect of human heart valve-derived ECM and NP/PCL electrospun nanofibrous sheet on mice bone marrow mononuclear cells and cardiac repair

Yao Chen,Zhanghao Huang,Cheng Ji,Jia-Hai Shi

Background Biomaterials can improve cardiac repair combined with transplantation of bone marrow mononuclear cells (BMMNCs). In this study, we compared the phenotype and cardiac repair between human heart valve-derived scaffold (hHVS) and natural protein/polycaprolactone (NP/PCL) anchored BMNNCs. Methods and results BMMNCs were obtained from mice five days following myocardial infarction. Subsequently, BMMNCs were separately cultured on hHVS and PCL. Proliferation and cardiomyogenic differentiation were detected in vitro. Cardiac function was measured after transplantation of cell-seeded cardiac patch on MI mice. After that, the BMMNCs were collected for mRNA sequencing after culturing on the scaffolds. Upon anchoring onto hHVS or PCL, BMMNCs exhibited an increased capacity for proliferation in vitro, however, the cells on hHVS exhibited superior cardiomyogenic differentiation ability. Moreover, both BMMNCs-seeded biomaterials effectively improved cardiac function after 4 weeks of transplantation, with reduced infarction area and restricted LV remodeling. Cell-seeded hHVS was superior to cell-seeded PCL. Conclusion BMMNCs on hHVS showed better capacity in both cell cardiac repairing and improvement for cardiac function than on PCL. Compared with seeded onto PCL, BMMNCs on hHVS had 253 genes up regulated and 189 genes down regulated. The reason for hHVS’ better performance than PCL as a scaffold for BMMNCs might be due to the fact that optimized method of decellularization let more cytokines in ECM retained.

A novel approach for engineering DHCM/GelMA microgels: application in hepatocellular carcinoma cell encapsulation and chemoresistance research

Dandan Zhou,Xiaoxiao Li,Wencun Liu,Mingjun Zhang,Ying Cheng,Zhousong Xu,Jian Gao,Yiyang Wang

Liver cancer, a highly aggressive malignancy, continues to present significant challenges in therapeutic management due to its pronounced chemoresistance. This resistance, which undermines the efficacy of conventional chemotherapy and targeted therapies, is driven by multifaceted mechanisms, with increasing emphasis placed on the protective role of the tumor microenvironment (TME). The hepatocellular carcinoma extracellular matrix (ECM), a primary non-cellular component of the TME, has emerged as a critical regulator in cancer progression and drug resistance, particularly in hepatocellular carcinoma cell (HCC). In this study, a hybrid biomimetic hydrogel was engineered by integrating decellularized hepatocellular carcinoma matrix (DHCM) with gelatin methacrylate (GelMA) precursors. This composite DHCM/GelMA hydrogel was designed to replicate the physicochemical and functional properties of the hepatocellular carcinoma ECM, thereby offering a biomimetic platform to explore the interactions between HCCs and their microenvironment. Leveraging a custom-designed microfluidic 3D printing platform, we achieved high-throughput fabrication of HCC-encapsulated DHCM/GelMA microgels, characterized by enhanced uniformity, biocompatibility, and scalability. These microgels facilitated the construction of hepatocellular carcinoma microtissues, which were subsequently employed for chemoresistance studies. Our findings revealed that DHCM/GelMA microgels closely mimic the hepatocellular carcinoma tumor microenvironment, effectively recapitulating key features of ECM-mediated drug resistance. Mechanistic studies further demonstrated that DHCM significantly upregulates the expression of Aquaporin 3 (AQP3) in the encapsulated HCCs. This upregulation potentially activates mTOR signaling-associated autophagy pathways, thereby enhancing chemoresistance in HCCs. These biomimetic models provide a robust and versatile platform for studying the underlying mechanisms of drug resistance and evaluating therapeutic interventions. This innovative approach highlights the potential of DHCM/GelMA microgels as a transformative tool in cancer-associated tissue engineering and anticancer drug screening. By enabling detailed investigations into the role of ECM in chemoresistance, this study contributes to advancing therapeutic research and offers promising strategies to overcome drug resistance, ultimately improving clinical outcomes in liver cancer treatment.

Xiaofei Gao,Xinxiang Chai,Yanzhen Lou,Liming Ruan,Zhengfei He,Yunjie Ni,Linggang Sun,Yanfang Sun,Guohua Jiang

Electrical stimulation (ES) of skin wounds can promote cell proliferation, protein synthesis, inflammatory response, and improve neurological function. In this study, the dynamical cross-linked conductive hydrogels have been developed by integration of poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS) into porcine acellular dermal matrix (PADM) and oxidized hyaluronic acid (OHA) using dopamine-Fe complex (DA@Fe) as cross-linking agent. The as-obtained conductive composite hydrogels have the characteristics of rapid gelation, excellent deformation ability, high water absorption, and suitable degradation rate. Four-point probes resistivity measurement system has been used to test the electrical properties of the as-obtained hydrogels, and showing their conductivity at 0.207–0.322 S/m. In addition, the developed dynamical cross-linked conductive hydrogels exhibit excellent biocompatibility, thereby inducing cells proliferation and migration. In vivo results show that the resultant composite hydrogel can accelerate wound healing with combination of electrical stimulation (ES) by promoting expression of vascular factor (CD31) and decreasing the expression of tumor necrosis factor-α (TNF-α).
