Chitin is the second most abundant biopolymer in the world after cellulose and commonly found in crustaceans. It is also the most abundant natural nitrogen-containing polysaccharide. The challenge in separation of chitin directly from snow crab ( Chionoecetes opilio ) shells is that the solvent used must be versatile in removing calcium carbonate and protein while also being efficient, environmentally friendly, and cost-effective. In this study, chitin was extracted from snow crab shells using a DES consisting of TEBAC and lactic acid. DES successfully removed minerals and proteins, with a deproteinization rate of 95.51% and a decalcification rate of 96.41%, while the purity of chitin was 91.15% and the yield was 21.31%. Furthermore, the extracted chitin has large pores on its surface, making it an ideal matrix for biomedical materials. This study provides an environmentally friendly and cost-effective solution for the utilization of crustacean waste using multifunctional DES.

Natural rubber (NR) latex was initially deproteinized, then, the interactions between chitin nanocrystals (ChNCs) and deproteinized natural rubber (DNR) latex particles in an aqueous system and the properties of DNR/ChNCs composites were investigated. Composite films were obtained by casting and evaporating off the prevulcanized DNR/ChNCs complexation rather than simply mixing of DNR latex and ChNCs suspension. The results of DLS and TEM confirmed that this kind of complexation was formed between negative charged DNR and positively charged ChNCs by self-assembly. The tensile strength of composite films containing 4 wt% ChNCs was 25.00 ± 1.42 MPa, which was 3.11 multiples of the pure pre-vulcanized DNR. DNR/ChNCs composite films could provide excellent mechanical properties and avoid allergies caused by proteins in NR latex.

Existing methods for chitin extraction usually produce substantial waste, which poses ecological hazards. Natural deep eutectic solvent (NADES) offers a promising one-step pretreatment alternative, replacing the resource-intensive demineralization (DM) and deproteinization (DP) process. Hence, in this study, the influence of various acidic NADES, on achieving a simplified one-step DM and DP in the chitin extraction process was investigated. The study yielded chitin with 87.73 % purity, and microstructural analysis showed that NADES pretreatment minimally affected chitin quality without deacetylation. In addition, chitin extracted using choline chloride-oxalic acid as a carrier displayed excellent performance in the immobilization of Geobacillus thermocatenulatus lipase 2 (GTL2) because of obvious Ca 2+ activation effect. This process contributed to enhancement of immobilized enzyme activity. The immobilized GTL2 showed excellent hydrolytic capabilities, with its highest activity reaching 547.80 ± 20.62 U/mg, significantly better than the five commercial lipases that exhibited <40 % of the enzyme activity. Furthermore, the hydrolytic capacity of immobilized GTL2 was notably high for 4-nitrophenyl butyrate, measuring 935.47 ± 51.60 U/mg. This study provided a constructive approach for the one-step pretreatment of shrimp shells with organic acid-based NADES to isolate and purify chitin and its potential application as an immobilized carrier to enhance enzyme activity.

Three-dimensional (3D) printing provides a novel approach to repair bone defects using customized biomimetic tissue scaffolds. To make a bone substitute closest to natural bone structure and composition, two different types of hydroxyapatite, Nano hydroxyapatite (nHA) and deproteinized bovine bone (DBB), were dispersed into collagen (CoL) to prepare the bioink for 3D printing. In doing so, a porous architecture was manufactured with 3D printing technology. The physical and chemical properties of the materials were evaluated, including biocompatibility and effect on the osteogenic differentiation of the human bone marrow-derived mesenchymal stem cells (hBMSCs). The XPS, XRD, FTIR, and the mechanical analysis of the material indicated that the two HA were consistent in their elements, but different in their chemical bonds and crystal phases. The SEM results showed the different surface morphologies of the HA crystals as well as the scaffolds, which would be the main factors affecting the internal porous structure of the scaffold. There were no differences between the two composite scaffolds in cell proliferations. FITC-phalloidin/vinculin/DAPI staining indicated that hBMSCs can adhere well to the 3D-printed surfaces. Alkaline phosphatase (ALP) staining reflected ALP expressed on both of the osteogenic-induction medium (OM) group, but not on proliferation medium (PM) group. The real-time polymerase chain reaction results showed the expression levels of osteogenesis-related genes RUNX2, SOX9, OCN, and COL1A1 in OM group were significantly increased after 7 days compared with the PM group (p < 0.01). The expression of SOX9, OCN, and COL1A1 in nHA/CoL scaffolds was higher than that in CoL scaffolds (p < 0.05). The expression of OCN and COLIA1 in DBB/CoL scaffolds was higher than that in CoL scaffolds (p < 0.05). In conclusion, the physicochemical and biological properties of 3D bioprinted scaffolds consisting of nHA/CoL or DBB/CoL would be well suited for the scaffolds to being a porous customized bone substitute, 3D printing scaffolds would be a prospective candidate for clinical application in future.Impact StatementBone loss due to trauma, inflammation, and surgical processes has posed great difficulty in the aesthetic reconstruction of a functional alveolar bone. Tissue engineering and biomaterials, which can promote alveolar bone regeneration, have become a popular focus of current studies. Three-dimensional (3D) printing provides a novel approach to repair bone defects using customized biomimetic tissue scaffolds. Nano hydroxyapatite (nHA) and deproteinized bovine bone (DBB) are two materials mainly used in clinical practice, particularly DBB are widely used in dentistry and craniomaxillofacial orthosis because of the porosity characteristic. To make a bone substitute closest to natural bone structure and composition, nHA and DBB were dispersed into collagen (CoL) to prepare the bioink for 3D printing. The physicochemical and biological properties between the two 3D printing scaffolds were compared. Both nHA/CoL and DBB/CoL 3D printing scaffold would be promising candidate for the clinical applications in the future.

Bone grafts are widely used in bone regeneration to increase the speed and quality of new bone formation. While they are routinely characterized based on their biocompatible and bioactive properties, they also exert a profound impact on host immune responses, which in turn can display a significant effect on the healing and repair process. In this study, we investigated the role of macrophage behavior on deproteinized bovine bone matrix (DBBM, BioOss) to investigate their impact on creating either a pro- or anti-inflammatory microenvironment for tissue integration. RT-PCR and immunofluorescence staining results demonstrated the ability for RAW 264.7 cells to polarize toward M2 wound-healing macrophages in response to DBBM and positive control (IL-4). Interestingly, significantly higher expression of interleukin-10 and higher number of multinucleated giant cells (MNGCs) was observed in the DBBM group. Thereafter, conditioned media (CM) from macrophages cultured with DBBM seeded with MC3T3-E1 cells demonstrated a marked increase in osteoblast differentiation. Noteworthy, this effect was reversed by blocking IL10 with addition of IL10 antibody to CM from the DBBM macrophages. Furthermore, the use of dendritic cell specific transmembrane protein (DC-STAMP)-knockout to inhibit MNGC formation in the DBBM group resulted in a significant reduction in osteoblast differentiation, indication a pivotal role for MNGCs in biomaterials-induced osteogenesis. The results from this study indicate convincingly that the immune response of macrophages towards DBBM has a potent effect on osteoblast differentiation. Furthermore, DBBM promoted macrophage fusion and polarization towards an M2 wound-healing phenotype, further created a microenvironment favoring biomaterial-induced osteogenesis. © 2018 Wiley Periodicals, Inc.
