Harnessing Spatiotemporal-Specific Tumorous Exosome Dynamics: Ultra-Sensitive Lanthanide Luminescence Detection Strategy Enabled by Exosomal Membrane Engineering for Melanoma Immunotherapy Monitoring

Nanhang Zhu,Yue Yu,Di An,Yating Zeng,Ke Kang,Qiangying Yi,Yao Wu

Focused on the newly secreted tumorous exosomes during melanoma immunotherapy, this work has pioneered an ultra-sensitive spatiotemporal-specific exosome detection strategy, leveraging advanced exosomal membrane engineering techniques. The proposed strategy harnesses the power of amplified lanthanide luminescence signals on these exosomes, enabling precise and real-time monitoring of the efficacy of melanoma immunotherapy. The methodology comprises two pivotal steps. Initially, Ac4ManNAz-associated metabolic labeling is employed to evolve azide groups onto the membranes of newly secreted exosomes with remarkable selectivity. These azide groups serve as versatile clickable artificial tags, enabling the precise identification of melanoma exosomes emerging during immunotherapy. Subsequently, lanthanide-nanoparticle-functionalized polymer chains are controllably grafted onto the exosome surfaces through click chemistry and in situ Fenton–RAFT polymerization, serving as robust signal amplifiers. When integrated with time-resolved fluorescence detection, this strategy yields detection signals with an exceptionally high signal-to-noise ratio, enabling ultra-sensitive detection of PD-L1 antigen expression levels on the spatiotemporal-specific exosomes. The detection strategy boasts a wide linear concentration range spanning from 1.7 × 104 to 1.7 × 109 particles/mL, with a remarkable theoretical detection limit of 1.28 × 103 particles/mL. The remarkable enhancements in detection sensitivity and accuracy facilitate the evaluation of the efficacy of immunotherapeutic interventions in the mouse B16 melanoma model, notably revealing a substantial disparity in PD-L1 levels between immunotherapy-treated and untreated groups (P < 0.01) and further emphasizing the cumulative therapeutic effect that intensifies with repeated treatments (P < 0.001).

Innovative Separator Engineering: Hydrogen Bond-Driven Layer-By-Layer Assembly for Enhanced Stability and Efficiency in Lithium Metal Batteries

Zhuqing Huang,Xingtao Qi,Hai Zhang,Liequan Liu,Ze Zhang,Zhenyu Yang,Junchao Wei

Lithium metal batteries (LMBs) face critical challenges due to uncontrolled lithium dendrite growth and inhomogeneous Li+ flux, largely attributed to conventional separators’ poor interfacial compatibility. To address this, we propose a hydrogen bond-driven layer-by-layer (LbL) assembly strategy for engineering functional separators using poly(vinyl alcohol) (PVA) and tannic acid (TA). The optimized PP/(TA/PVA)15 separator leverages the synergistic interplay between PVA’s hydroxyl groups and TA’s carbonyl moieties, forming a robust hydrogen-bonded network that simultaneously enhances lithiophilicity, regulates Li+ flux uniformity, and immobilizes anions. The interfacial design achieves exceptional electrochemical performance: Li//Li symmetric cells maintain stable operation for 800 h at 0.5 mA cm–2/0.5 mAh cm–2, while Li//LiFePO4 half cells retain 73.8% capacity after 1000 cycles at 5C (decay rate: 0.026% per cycle). The separator further exhibits high ionic conductivity (0.94 mS cm–1) and Li+ transference number (0.63), outperforming conventional polyolefin counterparts. By integrating simplicity, scalability, and eco-friendliness, this work pioneers a universal interface chemistry paradigm for next-generation LMBs, offering transformative insights into separator engineering through molecular-level hydrogen-bonding control.

Cell Membrane Engineered Polypeptide Nanonets Mimicking Macrophage Aggregates for Enhanced Antibacterial Treatment

Jiang Xiao,Zhongquan Song,Tengfei Liu,Zengchao Guo,Xiaohui Liu,Hui Jiang,Xuemei Wang

Drug-resistant bacterial infections and their lipopolysaccharide-related inflammatory complications continue to pose significant challenges in traditional treatments. Inspired by the rapid initiation of resident macrophages to form aggregates for efficient antibacterial action, this study proposes a multifunctional and enhanced antibacterial strategy through the construction of novel biomimetic cell membrane polypeptide nanonets (R-DPB-TA-Ce). The design involves the fusion of end-terminal lipidated polypeptides containing side-chain cationic boronic acid groups (DNPLBA) with cell membrane intercalation engineering (R-DPB), followed by coordination with the tannic acid-cerium complex (TA-Ce) to assemble into a biomimetic nanonet through boronic acid-polyphenol-metal ion interactions. In addition to the ability of RAW 264.7 macrophages cell membrane components' (R) ability to neutralize lipopolysaccharide (LPS), R-DPB-TA-Ce demonstrated enhanced capture of bacteria and its LPS, leveraging nanoconfinement-enhanced multiple interactions based on the boronic acid–polyphenol nanonets skeleton combined with polysaccharide. Utilizing these advantages, indocyanine green (ICG) is further employed as a model drug for delivery, showcasing the exceptional treatment effect of R-DPB-TA-Ce as a new biomimetic assembled drug delivery system in antibacterial, anti-inflammatory, and wound healing promotion. Thus, this strategy of mimicking macrophage aggregates is anticipated to be further applicable to various types of cell membrane engineering for enhanced antibacterial treatment.

A Versatile Plasma Membrane Engineered Cell Vehicle for Contact-Cell-Enhanced Photodynamic Therapy

Shi-Ying Li,Wen-Xiu Qiu,Hong Cheng,Fan Gao,Feng-Yi Cao,Xian-Zheng Zhang

In this paper, a plasma membrane engineering approach is reported for tumor targeting drug delivery and contact-cell-enhanced photodynamic therapy (“CONCEPT”) by anchoring functionalized conjugates to cell vehicles. The membrane anchoring conjugates are comprised of a positively charged tetra-arginine peptide sequence, a palmitic-acid-based membrane insertion moiety, and a lysine linker whose ε-amine is modified with camptothecin (CPT), protoporphyrin IX (PpIX), or fluorescein (FAM). The amphipathic CPT, PpIX, or FAM conjugates (short as aCPT, aPpIX, or aFAM, respectively) can easily and steadily anchor or coanchor on the cell membrane of RAW264.7 cells (short as RCs), red blood cells, or mesenchymal stem cells. After anchoring aPpIX in RC cells, the tumor targeting ability and therapeutic effect of aPpIX-anchored RC cells (short as aPRCs) is demonstrated in vitro and in vivo. Importantly, aPRCs exhibit the “CONCEPT” effect, which can enhance the therapeutic efficacy and reduce side effects at the single cell level. Due to the good tumor-targeting ability, aPRCs can efficiently inhibit the tumor growth with no systemic toxicity after photoirradiation by photodynamic therapy.

Enzyme-activated anchoring of peptide probes onto plasma membranes for selectively lighting up target cells

Julan Xia,Sujuan Sun,Xianhua Wu,Yan Huang,Chunyang Lei,Zhou Nie

In a cellular microenvironment, numerous biomolecules are involved in various physiological and pathological processes. However, for the in-depth and comprehensive understanding of their roles at the molecular level, there is still a lack of detection techniques for the in situ tracking of these biomolecules in a local environment. Herein, we engineered a membrane insertion peptide (MIP) as an enzyme-activated membrane insertion peptide probe (eaMIP) that allowed the in situ tracking of the activity of target enzymes in living cells. In this strategy, the membrane insertion capacity of the MIP motif in each eaMIP was caged by appending a chemical moiety. In the presence of target enzymes, the caging moiety in each eaMIP was removed by enzymatic decaging, leading to the generation of active MIPs. The versatility of this design was demonstrated by lighting up different tumor cells with distinct fluorescence signal patterns, affording an alternative tool for clinical diagnostics, biochemical research and membrane engineering.

MOF-derived pearl-like nanospheres enable tailored regulation of interface ion transport in dendrite-free lithium metal batteries

Xiaoxin Wu,Si Chen,Huasen Shen,Yunan Tian,Guoning Wu,Tingting Ma,Mengjun Li,Yuyu Li,Zhaohuai Li

Separators constitute vital components in high-energy-density lithium metal batteries (LMBs), yet their propensity to fail under lithium (Li) dendrite growth remains a critical limitation. In this work, we report a commercial Celgard separator engineered with pearl-like nanospheres (Celgard@PNS) to enable electrochemically stable and dendrite-free lithium metal anodes (LMAs). The PNS-functionalized layer in the Celgard@PNS ensures rapid and homogeneous Li ion flux, which stabilizes LMAs and effectively mitigates dendritic Li deposition. Moreover, the PNS modification fosters the development of a robust solid-electrolyte interphase (SEI) layer. The Celgard@PNS-based Li||Cu half-cell exhibits stable Coulombic efficiency for 110 cycles, and the corresponding Li||Li symmetric cell sustains long cycling stability (>900 h) at a high current density of 10 mA cm −2 with an areal capacity of 1 mAh cm −2 . Impressively, when paired with a high-mass-loading NCM811 cathode (22.5 mg cm −2 ), the Li||NCM811 full cell retains 81.4 % of its initial capacity after 140 cycles at 0.5 C. These results underscore the potential of the Celgard@PNS separator for practical implementation in high-capacity LMBs.

Triazine and hydroxyl covalent organic framework modified separator for Zn-ion fast and Selective transport and dendrite-free deposition in zinc–iodine battery

Tiao Huang,Shenglin Wang,Jingyi Wu,Hui Hu,Jianyi Wang,Xiaosong Zhang,Yanan Gao

Aqueous zinc–iodine (Zn–I) batteries show a great development potential in the field of energy storage due to their advantages of low cost, high safety and ecological friendliness. However, Zn dendrite growth and shuttle effect of polyiodide seriously aggravate the self-discharge behavior and thus decrease the cycle life of batteries . Therefore, controlling the diffusion of zinc ion and inhibiting soluble polyiodide in the separator are the most effective strategy to extend the lifespan of batteries. Herein, we design and prepare a multifunctional Janus separator composed of a covalent organic framework (COF) layer, a commercial glass fiber (GF) substrate and a graphene (Gr) layer to achieve durable Zn–I batteries. The COF layer features abundant polar groups and uniform open-channel mesopores enabling homogeneous Zn 2+ flux and simultaneously inhibiting polyiodide species migration. The highly conductive Gr layer can not only load iodide but also effectively activate the redox kinetics of iodine species. The full cell assembled with the TTA-DHTPA-COF@GF@Gr Janus separator demonstrated a recorded energy density of up to 6.4 mA h·cm −2  at a high current density of 20 mA cm −2 in aqueous Zn–I batteries. The design of the multifunctional Janus separator provides a new insight into separator engineering for durable Zn-ion batteries.

Subnanometer Two-Dimensional Graphene Oxide Channels for Ultrafast Gas Sieving

Jie Shen,Gongping Liu,Kang Huang,Zhenyu Chu,Wanqin Jin,Nanping Xu

Two-dimensional (2D) materials with atomic thickness and extraordinary physicochemical properties exhibit unique mass transport behaviors, enabling them as emerging nanobuilding blocks for separation membranes. Engineering 2D materials into membrane with subnanometer apertures for precise molecular sieving remains a great challenge. Here, we report rational-designing external forces to precisely manipulate nanoarchitecture of graphene oxide (GO)-assembled 2D channels with interlayer height of ∼0.4 nm for fast transporting and selective sieving gases. The external forces are synergistic to direct the GO nanosheets stacking so as to realize delicate size-tailoring of in-plane slit-like pores and plane-to-plane interlayer-galleries. The 2D channels endow GO membrane with excellent molecular-sieving characteristics that offer 2–3 orders of magnitude higher H2 permeability and 3-fold enhancement in H2/CO2 selectivity compared with commercial membranes. Formation mechanism of 2D channels is proposed on the basis of the driving forces, nanostructures, and transport behaviors.

Separator with high ionic conductivity enables electrochemical capacitors to line-filter at high power

Hu Yajie,Li Puying,Lai Guobin,Lu Bing,Wang Haiyan,Cheng Huhu,Wu Mingmao,Liu Feng,Dang Zhi-Min,Qu Liangti

Line-filtering electrochemical capacitors (LFECs) are demonstrating advantages in line filtering over traditional electrolytic capacitors. However, they can only function at no-load or low-power conditions due to the limited high-frequency capacitance resulting from the excessive ionic resistance, despite much progress in electrode materials. Here, we show separators dominate both ion migration and capacitance in LFECs. A 3 μm-thick thread-anchor structured separator is developed, featuring both accelerated ionic transport and reliability, leading to a low ionic resistance of 25 mΩ cm 2 . With a phase angle of −80° at 120 Hz, the assembled device has an areal capacitance of 6.6 mF cm −2 . Furthermore, stack integration in parallel breaks the trade-off between capacitance and frequency response, boosting the areal capacitance by two orders of magnitude without decay of frequency characteristics. The On-board field test demonstrates that voltage ripples are steadily suppressed below 5% even for practical high-power line filtering with a load power density of 2.5 W cm −2 , three orders of magnitude higher than previous instances. This work opens up a perspective of separator engineering for the development of high-performance line-filtering electrochemical capacitors and promotes their applications in practical high-power scenarios.

Metabolic Engineering for Improved Curcumin Biosynthesis in Escherichia coli

Jieyuan Wu,Wei Chen,Yutong Zhang,Xuanxuan Zhang,Jian-Ming Jin,Shuang-Yan Tang

The biosynthetic efficiency of curcumin, a highly bioactive compound from the plant Curcuma longa, needs to be improved. In this study, we performed host cell and biosynthetic pathway engineering to improve curcumin biosynthesis. Using in vivo-directed evolution, the expression level of curcuminoid synthase (CUS), the rate-limiting enzyme in the curcumin biosynthetic pathway, was significantly improved. Furthermore, as curcumin is a highly hydrophobic compound, two cell membrane engineering strategies were applied to optimize the biosynthetic efficiency. Curcumin storage was increased by overexpression of monoglucosyldiacylglycerol synthase from Acholeplasma laidlawii, which optimized the cell membrane morphology. Furthermore, unsaturated fatty acid supplementation was used to enhance membrane fluidity, which greatly ameliorated the damaging effect of curcumin on the cell membrane. These two strategies enhanced curcumin biosynthesis and demonstrated an additive effect.

Cellular Membrane-Engineered Nanovesicles as Three-Stage Booster to Target Lesion Core

Fanyi Huang,Yawei Du,Yanjia Chen,Zeping Qiu,Zhiyan Wang,Yingze Fan,Yunjing Shi,Zhuojin Li,Ke Yang,Wenguo Cui,Wei Jin

Lesion core is the area with the most serious injury and vigorous repair. Existing nanocarriers are difficult to break through the targeted delivery to lesion core and the precise treatment for intracellular and extracellular microenvironment. Herein, a cellular membrane-engineered nanovesicle (CMEV) with hierarchical structure is constructed using double emulsion-extrusion method, by integrating neutrophil membrane, functional antibody and gelled drug-loaded core, as ‘three-stage booster’ to target the lesion core and deliver catestatin (CST), a small therapeutic peptide, for ischemic cardiomyopathy therapy. By coating neutrophil membrane outside the shell, CMEV is endowed with the function of neutrophil-like migration to achieve the first stage of tissue targeting. Based on the specific anchoring to injured myocardium, myosin light chain 3 (MLC3) antibody was embedded to fulfill the second stage of CMEV accumulation in lesion core. The gelled core containing CST-sodium alginate (NaAlg) with pH-responsive shell is prepared by ionic cross-linking to accomplish the third stage of precise CST administration. Triggered by the microenvironment, NaAlg electrostatically adheres to the lesion core for sustained release, enhancing the efficacy of CST in improving cardiomyocyte apoptosis, excessive fibrosis, macrophage polarization and angiogenesis. Thus, the “three-stage booster” nanovesicle significantly ameliorates cardiac function and adverse remodeling to treat ischemic cardiomyopathy. This article is protected by copyright. Thermally triggered polyrotaxane translational motion helps proton transfer

Ge Xiaolin,He Yubin,Liang Xian,Wu Liang,Zhu Yuan,Yang Zhengjin,Hu Min,Xu Tongwen

Synthetic polyelectrolytes, capable of fast transporting protons, represent a challenging target for membrane engineering in so many fields, for example, fuel cells, redox flow batteries, etc. Inspired by the fast advance in molecular machines, here we report a rotaxane based polymer entity assembled via host–guest interaction and prove that by exploiting the thermally triggered translational motion (although not in a controlled manner) of mechanically bonded rotaxane, exceptionally fast proton transfer can be fulfilled at an external thermal input. The relative motion of the sulfonated axle to the ring in rotaxane happens at ~60 °C in our cases and because of that a proton conductivity (indicating proton transfer rate) of 260.2 mS cm −1 , which is much higher than that in the state-of-the-art Nafion, is obtained at a relatively low ion-exchange capacity (representing the amount of proton transfer groups) of 0.73 mmol g −1 .
