A highly selective “turn-on” fluorescent sensor for zinc ion based on a cinnamyl pyrazoline derivative and its imaging in live cells

Yun-Shang Yang,Chun-Mei Ma,Ying-Peng Zhang,Qing-Hua Xue,Jia-Xi Ru,Xiao-Yu Liu,Hui-Chen Guo

A fluorescence probe based on cinnamyl pyrazoline was developed and synthesized. The probe detected Zn2+ ion that was mixed with a series of cations in aqueous ethanol with high selectivity and high sensitivity. After adding Zn2+, the fluorescence intensity was significantly enhanced, with a 56-fold increase. The detection limits and binding constants of 2.95 × 10−8 M and 1.468 × 104 M−1 were calculated by a fluorescence titration experiment. The 1 : 1 binding chemometrics of the 1–Zn2+ complex was analyzed by Job's plot. The probe showed good reversibility when adding PPi. Additionally, the “turn-on” fluorescent probe with excellent membrane permeability was satisfactorily applied to microscopic imaging for the detection of Zn2+ in baby hamster kidney (BHK)-21 cells.

COVID-19-inspired “artificial virus” to combat drug-resistant bacteria by membrane-intercalation- photothermal-photodynamic multistage effects

Yongsheng Ni,Jingyao Wang,Mengyi Wang,Lizhi Liu,Hongqing Nie,Qiaoling Wang,Jing Sun,Tianli Yue,Ming-Qiang Zhu,Jianlong Wang

COVID-19 threatens human life because of the super destructiveness produced from its coronal morphology and strong transmembrane infection based on spike glycoprotein. Inspired by the coronal morphology of COVID-19 and its means of infecting, we designed an “artificial virus” with coronal morphology based on the concept of “defeating superbacteria with superviruses” by self-assembling a transacting activator of transduction peptide with triple-shell porous graphitic carbon nitride (g-C 3 N 4 ) embedded with cobalt nanoparticles to forcefully infect methicillin-resistant Staphylococcus aureus (MRSA). The results confirmed that this “artificial virus” had unique properties of crossing the bacterial cell membrane barrier, heating the internal bacterial microenvironment and triggering ROS outbreak, based on its coronal morphology, membrane penetration, temperature-rising and heat insulation, oxidase-like activity and excellent visible-light harvesting properties. It had a high sterilization efficiency of 99.99% at 20 min, which was 18.6 times that of g-C 3 N 4 , and the efficiency remained at 99.99% after 3 rounds of recycling and reuse. Additionally, it can rapidly inactivate bacteria in river water and accelerate wound healing.

Improving the Membrane Permeability of 5-Fluorouracil via Cocrystallization

Xia-Lin Dai,Song Li,Jia-Mei Chen,Tong-Bu Lu

Pharmaceutical cocrystallization is proposed as a new method to enhance membrane permeability of a BCS class III model drug, 5-fluorouracil (5FU). Three cocrystals of 5FU, 5FU/3-hydroxybenzoic acid (1), 5FU/4-aminobenzoic acid (2), and 5FU/cinnamic acid (3), were successfully synthesized by a slurry method or a liquid-assisted grinding process. Spectroscopic methods, thermal analysis, and X-ray diffraction were used to characterize these new forms. The permeability was studied using a Franz diffusion cell and silicone membrane. All of the cocrystals showed improved membrane permeability compared to free 5FU. The cumulative amount per unit area of permeated 5FU in the first 10 h for 1–3 was increased by 41, 70, and 83%, and the steady penetration rates of 1–3 were increased by 38, 66, and 79%, respectively, as compared to the pure drug. Structure–permeability correlation study finds a link between intermolecular interactions and molecular packing in cocrystals and their permeability behavior and has important implications for use of a cocrystallization approach to improve drugs’ permeability in the pharmaceutical field.

Unveiling the anti-listerial effect of Citrus bergamia essential oil: Mechanism of membrane disruption and anti-hemolytic activity

Hafiz Abdul Rasheed,Abdur Rehman,Xiaochen Chen,Tariq Aziz,Fahad Al-Asmari,Majid Alhomrani,Abdulhakeem S. Alamri,Haiying Cui,Lin Lin

Listeria monocytogenes is a prominent cause of foodborne illnesses and public health concerns worldwide. Naturally active compounds have been extensively explored for their potential to reduce foodborne pathogenic contamination. This work is designed to study the chemical composition, antibacterial effect , and inhibition mechanism of Citrus bergamia essential oil (CB-EO) against L. monocytogenes . The CB-EO composition revealed the major components of the total account are d -limonene (23.2%), linalyl acetate (14.01%), and linalool (9.96%). The inhibitory effect was examined at different concentrations of CB-EO, and the MIC value was 1.5 mg/mL against L. monocytogenes . The CB-EO concentration of 1.5 mg/mL significantly disturbed the membrane permeability and fluidity, leading to leakage of intracellular protein and DNA constituents, as revealed by the BCA protein assay and UV–Vis spectrophotometric analysis, respectively. However, a decrease in concentration to 0.75 mg/mL indicated a significant decline in intracellular macromolecule leakage as well as membrane disruption. Moreover, SEM and CLSM observations provided extensive insight into the inhibition mechanism of CB-EO against L. monocytogenes . Finally, while investigating the anti-hemolysis activity, CB-EO and its major compounds, limonene , linalool, and linalyle acetate, have been found to be effective against the virulence agent listeriolysin O (LLO). Thus, it was proposed that CB-EO could act on L. monocytogenes cells through multiple mechanisms of action, with EO concentration providing a crucial role in determining the mode of action. This work offers deep insight into the inhibition mechanisms of natural substances, expanding the potential uses of CB-EO as a natural antimicrobial.

Octa-arginine modified lipid emulsions as a potential ocular delivery system for disulfiram: A study of the corneal permeation, transcorneal mechanism and anti-cataract effect

Chang Liu,Qi Lan,Wei He,Changlu Nie,Chunjuan Zhang,Tonghua Xu,Tongying Jiang,Siling Wang

The purpose of the study was to design a novel octa-arginine (R8) modified lipid emulsion (LE) system for the ocular delivery of the lipophilic drug disulfiram (DSF). The influence of the particle size of the lipid emulsions and the presence of R8 on corneal permeation was studied. DSF-loaded lipid emulsions with different particle sizes (DSF-LE1, DSF-LE2, DSF-LE3) and DSF-loaded lipid emulsions modified with R8 (DSF-LE1-R8 and DSF-LE2-R8) were prepared. The Zeta potential of the lipid emulsions was changed from negative to a positive value after modification of R8. The mucoadhesion of different preparations was investigated, and DSF-LE1-R8 was found to produce the strongest mucoadhesion. The in vitro corneal penetration study and in vivo ocular distribution study showed that the R8 modified lipid emulsion (DSF-LE1-R8) with a nano particle size, exhibited the highest permeability and the largest amount of DDC distributed in ocular issues. Coumarin-6 labelled LE1-R8 displayed more homogeneous fluorescence with the deeper penetration into the cornea compared with other preparations at various times. Confocal laser scanning microscopy showed that, in addition to paracellular routes, LE-R8 could also transport across the corneal epithelium by transcellular routes as a result of increased uptake due to the R8 modification. Furthermore, the anti-cataract effect was evaluated and it was found that DSF-LE1-R8 exhibited a marked anti-cataract effect. Therefore, the lipid emulsions with nano-sized particles and modification of R8 were proposed as a potential ocular delivery system to improve the corneal penetration and ocular delivery of DSF.

The role of NhaA protein in modulating antibiotic tolerance in Escherichia coli

Zhen Tao,Chuanjun Tian,Chunyan Zhong,Bingjie Ji,Wenwen Li,Yongshan Zhao

As microbial resistance and recurrent bacterial infections escalate, the growing understanding of the interplay between antibiotic resistance and tolerance has sparked significant interest in the latter. Previous studies have demonstrated that the deletion of cation/proton antiporters (CPAs) induces bacterial phenotypes, such as slow growth and prolonged lag phases, which contribute to the development of tolerance. This study investigates the role of the NhaA protein in antibiotic tolerance in Escherichia coli using CRISPR/Cas9 gene editing to delete the NhaA protein. Our results suggest that the NhaA protein plays a key role in modulating antibiotic tolerance. In response to NhaA deletion, E. coli adapts through multiple mechanisms, including changes in membrane permeability, enhanced efflux activity, increased membrane fluidity, disruption of the proton motive force (PMF), and a reduction in intracellular ATP levels. These adaptive changes collectively promote the development of antibiotic tolerance. Understanding these tolerance mechanisms could uncover new therapeutic targets, help prevent the emergence of tolerance, or sustain bacteria cells in a tolerant state, providing crucial strategies to combat the rise of antibiotic-resistant bacteria.

Green Synthesis of an Alginate-Coated Silver Nanoparticle Shows High Antifungal Activity by Enhancing Its Cell Membrane Penetrating Ability

Shunyu Xiang,Xiaozhou Ma,Huan Shi,Ting Ma,Cuiling Tian,Ya Chen,Haitao Chen,Xue Chen,Ke Luo,Lin Cai,Daibin Wang,Yang Xue,Jin Huang,Xianchao Sun

As a kind of promising nanopesticide, in contrast to traditional synthesis strategies, the application of a polysaccharide in silver nanoparticle preparation can improve its stability and avoid the usage of harmful substances. In this work, a two-step strategy for synthesizing silver nanoparticles (A-AgNPs) from aldehyde-modified sodium alginate (ASA) was introduced. The size of the A-AgNPs synthesized can be controlled from 6 to 40 nm with a high dispersibility in water. Furthermore, compared to naked AgNPs (n-AgNPs), the A-AgNPs showed improved broad-spectrum antimicroorganism activity. We found that the A-AgNPs mainly exerted their antifungal activity through the changing of cell membrane permeability and affecting the soluble protein synthesis, destruction of DNA structure, and inhibition of DNA replication. Meanwhile, the A-AgNPs showed no inhibition of rice and N. benthamiana seed germination. Considering its high biocompatibility and the highly efficient antimicroorganism activity, A-AgNPs can be potentially applied in plant protection science research.

Hyaluronic Acid Modified Lutein Nanoparticles for Effective Application in Eye Drops: Development, Characterization, and Evaluation

Peng Liu,Dong Ling,Cai Tianqi,Liu Kai,Gao Xingtong,Sun Wenlong,Li Aixiang,Song Xinhua

In this study, the possibility of using hyaluronic acid (HA) modified lutein as the main component of eye drops for ophthalmic treatment was discussed. Firstly, hyaluronic acid grafted lutein (HA- g -lutein) was synthesized by hydrophilic modification of lutein by Steglich esterification. The grafted copolymer was self-assembled in water to form micelles and then loading unreacted lutein to prepare composite nanoparticles (denoted HA-lutein NPs). The findings from the FTIR, UV–Vis, and XRD analyses indicate that the graft copolymers were synthesized while preserving the structural integrity of lutein. Furthermore, the modification demonstrated efficient inhibition of lutein crystallization. TEM showed that HA-lutein NPs self-assembled into spherical micelles in water, the particle size of which was on the nanometer scale. The saturation solubility of HA-lutein NPs exhibits a 48-fold increase compared to that of free lutein. Then the cytotoxicity of HA-lutein NPs and the effect on in-vitro transcorneal permeation of excised-rabbit cornea, corneal hydration level, and in-vivo ocular irritation after topical administration of HA-lutein NPs in rabbit eyes were studied. The results indicated the ocular safety of HA-lutein NPs. The quantitative analysis of permeability parameters by HPLC showed that the apparent permeability coefficient and the cumulative penetration quantity within 360 minutes of the HA-lutein NPs group were 3.71 and 7.04 times higher compared to the free lutein group. In summary, the graft modification of lutein with HA to form nanoparticles is an effective method to improve the availability of lutein in the eye and has potential application in ocular therapy.

Ultrahigh permeability of graphene-based membranes by adjusting D-spacing with poly (ethylene imine) for the separation of dye wastewater

Jia-Jie Lu,Yi-Hang Gu,Yan Chen,Xi Yan,Ya-Jun Guo,Wan-Zhong Lang

Graphene oxide (GO) as a unique 2D material attracts much attention. It can be employed to prepare graphene-based membranes by vacuum filtration on microporous substrates. However, the permeability of graphene-based membranes is mainly limited by its interlamellar spacing. In this work, the interlamellar spacing of graphene-based membranes is expanded by grafting poly (ethylene imine) (PEI) on GO sheets. The novel graphene-based membranes present ultrahigh permeability without sacrificing the rejection for the filtration of dye wastewater. The original GO membrane (GOM) only has a pure water permeation flux (PWP) value of 27.0 L·m −2 ·h −1  bar −1 . However, the novel graphene-based membranes prepared with the modified GO by PEI-600 and PEI-10000 (recorded as PGOM-600 and PGOM-10000) markedly increase to 208.1 and 450.2 L·m −2 ·h −1 ·bar −1 respectively. In the separation of dye wastewater, GOM, PGOM-600 and PGOM-10000 exhibit much higher rejections for direct red and methyl blue. However, the modified graphene-based membranes (PGOM-600 and PGOM-10000) present more than 7–20 times higher permeation flux than that of original GOM without sacrificing dye rejection. Thus, this work provides graphene-based membranes with ultrahigh permeation flux for dye removal from wastewater.

A potential carrier based on liquid crystal nanoparticles for ophthalmic delivery of pilocarpine nitrate

Jing Li,Lin Wu,Weijun Wu,Baoyan Wang,Zhongyuan Wang,Hongliang Xin,Qunwei Xu

Poor corneal penetration and short preocular retention of a clinical hydrophilic drug, pilocarpine nitrate (PN), for the treatment of open-angle glaucoma and acute angle-closure glaucoma, limit its ocular application. The purpose of this study was to investigate the potential of liquid crystal nanoparticles (LCNPs) for ocular delivery of PN. LCNPs were developed by a top-down method using glyceryl monoolein (GMO) and water in the presence of stabilizer Poloxamer 407 . They were characterized by transmission electron microscopy (TEM) and small angle X-ray diffraction (SAXS). The size of LCNP is 202.28   ±   19.32   nm and the encapsulation efficiency reached 61.03%. The in vitro release profiles indicated that PN could keep sustained release from PN-loaded LCNPs for 8   h. An ex vivo corneal permeation study revealed that the apparent permeability coefficient of PN-loaded LCNPs was 2.05-fold higher than that of commercial eye drops. In addition, the topical administration test showed that PN-loaded LCNPs had a prolonged effect on decreasing intraocular pressure (IOP) of rabbits compared with commercial drug and physiological saline. In conclusion, LCNPs had been demonstrated to be potential for controlled-release ocular drug delivery.

Essence of the Enhanced Osmotic Energy Conversion in a Covalent Organic Framework Monolayer

Zhiwei Huang,Munan Fang,Bin Tu,Jinlei Yang,Zhuang Yan,Haftu Gebrekiros Alemayehu,Zhiyong Tang,Lianshan Li

Low membrane conductivity originated from a high membrane thickness has long been the “Achilles heel” of the conventional polymeric membrane, greatly hampering the improvement of the output power density in osmotic power generation. Herein, we demonstrate a molecularly-thin two-dimensional (2D) covalent organic framework (COF) monolayer membrane, featured with ultimate thickness, high pore density, and tight pore size distribution, which performs as a highly efficient osmotic power generator. Despite the large pore size up to 3.8 nm and relatively low surface charge density of 2.2 mC m–2, the monolayer COF membrane exhibits a high osmotic current density of 16.7 kA m–2 and an output power density of 102 W m–2 under 50 times the NaCl salinity gradient (0.5 M/0.01 M). This superior power density could be further improved to 170 W m–2 in the real seawater/river water gradient system. When the large pore size and low surface charge density are considered, this superior performance is not expected. Computational studies further reveal that the ultimate membrane permeability originated from the high membrane porosity, rather than ion selectivity, plays a dominant role in the production of high current density, especially under high salinity. This work provides an alternative strategy to realize improved output power density in ultrapermeable membranes.

Adjusting the Ion Permeability of Polyelectrolyte Multilayers through Layer-by-Layer Assembly under a High Gravity Field

Chao Jiang,Caijun Luo,Xiaolin Liu,Lei Shao,Youqing Dong,Yingwei Zhang,Feng Shi

The layer-by-layer (LbL) assembled multilayer has been widely used as good barrier film or capsule due to the advantages of its flexible tailoring of film permeability and compactness. Although many specific systems have been proposed for film design, developing a versatile strategy to control film compactness remains a challenge. We introduced the simple mechanical energy of a high gravity field to the LbL assembly process to tailor the multilayer permeability through adjusting film compactness. By taking poly(diallyldimethylammonium chloride) (PDDA) and poly{1–4[4-(3-carboxy-4-hydroxyphenylazo)benzenesulfonamido]-1,2-ethanediyl sodium salt} (PAzo) as a model system, we investigated the LbL assembly process under a high gravity field. The results showed that the high gravity field introduced effectively accelerated the multilayer deposition process by 20-fold compared with conventional dipping assembly; the adsorption rate was positively dependent on the rotating speed of the high gravity equipment and the concentration of the building block solutions. More interestingly, the film compactness of the PDDA/PAzo multilayer prepared under the high gravity field increased remarkably with the growing rotational speed of the high gravity equipment, as demonstrated through comparisons of surface morphology, cyclic voltammetry curves, and photoisomerization kinetics of PDDA/PAzo multilayers fabricated through the conventional dipping method and through LbL assembly under a high gravity field, respectively. In this way, we have introduced a simple and versatile external form of mechanical energy into the LbL assembling process to improve film compactness, which should be useful for further applications in controlled ion permeability, anticorrosion, and drug loading.
