Spatiotemporally Monitoring Cell Viability through Programmable Mitochondrial Membrane Potential Transformation by Using Fluorescent Carbon Dots

Xiaohui Yin,Yuanqiang Sun,Xin Geng,Jianjun Li,Ran Yang,Ke Zhang,Lingbo Qu,Zhaohui Li

Mitochondria are the powerhouse of the cell, which maintain the metabolism of living cells. Mitochondrial membrane potential (MMP) is the key parameter of mitochondria and represents cellular activity. The programmable MMP transformations, i.e., normal, decrease, and vanishing, are indicative mitochondria healthy/damaged/dead states. Therefore, methods for monitoring MMP are of great importance. In this work, carbon dots responsive to mitochondrial membrane potential (MMP-CDs) are prepared by a simple microwave method. The intracellular distribution of the MMP-CDs is dependent on the mitochondrial membrane potential. Three different spatiotemporally organelle distributions (mitochondria/lysosome/nucleus) of the MMP-CDs indicate three distinct MMP levels (normal/decrease/vanishing), which represent three different cell states (healthy/damaged/dead). The normal cells with high mitochondrial membrane potential causes the MMP-CDs to accumulate in the mitochondria due to the Nernstian effect. The MMP-CDs are released from the mitochondria and transported to the lysosome when cells are in apoptosis with the decreased mitochondrial membrane potential. In dead cells with a vanished mitochondrial membrane potential, the MMP-CDs are collocated with the nucleus due to the affinity with DNA. Thus, various cellular states can be visualized through the different localizations of the MMP-CDs. These MMP-CDs are successfully employed in the detection of autophagy and H 2 O 2 -induced mitochondria and mitochondrial membrane potential changes.

Transmembrane potential, an indicator in situ reporting cellular senescence and stress response in plant tissues

Liu Hai,Li Yufei,Peng Ting,Xue Shaowu

Background Plant cells usually sustain a stable membrane potential due to influx and/or efflux of charged ions across plasma membrane. With the growth and development of plants, different tissues and cells undergo systemic or local programmed decline. Whether the membrane potential of plasma membrane could report senescence signal of plant tissues and cells is unclear. Results We applied a maneuverable transmembrane potential (TMP) detection method with patch-clamp setup to examine the senescence signal of leaf tissue cells in situ over the whole life cycle in Arabidopsis thaliana. The data showed that the TMPs of plant tissues and cells were varied at different growth stages, and the change of TMP was higher at the vegetative growth stage than at the reproductive stage of plant growth. The distinct change of TMP was detectable between the normal and the senescent tissues and cells in several plant species. Moreover, diverse abiotic stimuli, such as heat stress, hyperpolarized the TMP in a short time, followed by depolarized membrane potential with the senescence occurring. We further examined the TMP of plant chloroplasts, which also indicates the senescence signal in organelles. Conclusion sThis convenient TMP detection method can report the senescence signal of plant tissues and cells, and can also indicate the potential of plant tolerance to environmental stress.

Spectroscopic, Polarographic, and Microcalorimetric Studies on Mitochondrial Dysfunction Induced by Ethanol

Ma Long,Dong Jia-Xin,Wu Can,Li Xue-Yi,Chen Jing,Zhang Hong,Liu Yi

Liver mitochondria are involved in several important life processes; mitochondrial dysfunction and disorders are implicated in several human diseases. Alcohol permeates all tissues of the body and exerts some intrinsic hepatotoxicity. In this work, our results demonstrated that ethanol caused a series of mitochondria permeability transition pore (MPTP) opening factors such as mitochondrial swelling, increased permeability of H + and K + , collapsed membrane potential, and increased membrane fluidity. Furthermore, mitochondrial ultrastructure alternation observed clearly by transmission electron microscopy and the release of Cytochrome c could explain the MPTP opening from another aspect. Moreover, ethanol damaged the mitochondrial respiration system and induced disturbance of mitochondrial energy metabolism which was monitored by polarographic and microcalorimetric methods, respectively. Considered together, these damages may promote both apoptotic and necrotic cell death and contribute to the onset or progression alcohol-induced liver diseases.

The adsorption behaviour of carbon nanodots modulated by cellular membrane potential

Shuyuan Zhang,Changqing Xiao,Hang He,Ziqiang Xu,Beibei Wang,Xueqin Chen,Cao Li,Bingbing Jiang,Yi Liu

Carbon nanodots (CDs) are fluorescent nanomaterials that have attracted widespread attention for biomedical applications. Since different cells possess different membrane potentials, the influence of the trans-membrane potential on the adsorption behaviour needs to be explored extensively. It was found that fewer CDs can be adsorbed by depolarized cells with decreased membrane potential, as compared to hyperpolarized cells with enhanced membrane potential that promotes the adsorption of CDs. A similar adsorption trend was observed for two cell lines, namely NIH3T3 and MCF-7. Since phosphocholine is the primary component of the cellular membrane, the adsorption behaviour of CDs is further complemented by the interactions between CDs and three kinds of liposomes with different surface charges via the thermodynamic and kinetic perspectives. The adsorption constants between negatively charged CDs and liposomes are positively related to their membrane potential, which is similar to the adsorption rate constants. It was further found that the adsorption of CDs onto liposomes was initiated by electrostatic attraction forces. The membrane integrity and fluidity of liposomes exposed to CDs have also been characterized. The membrane potential of cells is, therefore, particularly important when predicting the relative cytotoxicity of CDs and developing CDs-based nanomaterials for biomedical applications.

Hydrogels designed for preventing bacterial adhesion based on the response mechanism of Staphylococcus aureus to material stiffness

Leyi Xiao,Chen Chen,Mengge Feng,Yulan Wang,Tiange Zhang,Yueqi Ni,Fanyu Zhang,Jiaojiao Li,Qi Xiao,Yu Cui,Yufeng Zhang

Medical implants have opened up more possibilities in clinical medicine, while they may lead to severe complications due to biofilm formed by bacteria. Understanding the bacterial response mechanism to the material's stiffness is critical to preventing biofilm formation. This study used Staphylococcus aureus , one of the most common clinical pathogens, to explore the response mechanism. Based on the results, bacteria adhere more efficiently with lower membrane potential on stiff hydrogels than soft hydrogels. Combining experiments and molecular dynamics simulations, we found that the Arsenical pump membrane protein (ArsB) of S. aureus was upregulated on stiff hydrogels and can modulate the bacterial membrane potential and adhesion. The interface membrane curvature was the main morphology difference between bacteria on soft and stiff hydrogels. With the more significant membrane deformation of the bacteria on stiff hydrogels, we demonstrated that the changes in membrane curvature could open the ArsB ion channel. Finally, we screened the ArsB inhibitory drug Rapamycin and verified that Rapamycin-loaded hydrogels could efficiently prevent bacterial adhesion. It can be a way of inhibiting bacterial adhesion during biofilm formation and provide directions for the design of future medical materials.

Dual-Modality Imaging with a Zwitterionic Fluorescent Probe for Reversible Monitoring of Mitochondrial Membrane Potential Dynamics

Fei Peng,Xiangnan Ai,Xiaoyu Bu,Jing Sun,Zixuan Zhao,Zikang Yang,Xinmeng Qin,Baoxiang Gao

Traditional fluorescence intensity-based probes face challenges in accurately measuring mitochondrial membrane potential (MMP) due to intramolecular fluorescence quenching. In this work, we introduce a novel approach by incorporating quenching moieties within the zwitterionic probe to eliminate self-quenching interference, thus, enabling real-time and precise visualization of reversible MMP changes. We synthesized a zwitterionic fluorescent probe consisting of silicon-rhodamine (SiR) that was hydroxyl-substituted on the bay position of perylene diimides (PDIs) connected via a polyethylene glycol (PEG) linker. The lipophilic cationic SiR facilitates the entry of the PDI into the mitochondria, where the alkaline pH environment (pH = 8.0) ionizes the hydroxyl to a negatively charged species, affecting the quenching efficiency of SiR depending on the distance between the PDI and SiR moieties regulated by the MMP. The rigid aromatic ring of the PDI and strong hydrophobic interactions with the lipid bilayer, along with the inhibitory effect of the negatively charged hydroxyl on internalization, ensure the retention of PDI within the mitochondria. As the MMP decreases, SiR shifts outward, reducing quenching by phenolic anions and restoring fluorescence. Conversely, as the MMP increases, SiR moves inward, intensifying quenching by phenolic ions and reducing fluorescence, enabling reversible visualization monitoring of the MMP. This strategy overcomes the limitations of traditional intensity-based probes, providing a new avenue for reversible monitoring of the MMP.

Construction of the FRET Pairs for the Visualization of Mitochondria Membrane Potential in Dual Emission Colors

Ruiqing Feng,Lifang Guo,Jinglong Fang,Yue Jia,Xueying Wang,Qin Wei,Xiaoqiang Yu

Mitochondria membrane potential (MMP) play significant roles during metabolism, signaling, and other important bioevents. Visualization of MMP levels is essential for many biological researches. However, fluorescent probes for monitoring MMP levels in dual emission colors are still deficient, which greatly limited the development of relative research areas. In this work, a pair of fluorescent probes have been designed and synthesized to monitor the MMP levels in dual emission colors based on Forster resonance energy transfer (FRET) mechanism. The FRET donor (FixD) is constructed by linking a benzyl chloride group to a fluorophore with bright-green emission. The FixD could target mitochondria and be immobilized in mitochondria by linking to the thiol group of mitochondrial proteins. The FRET acceptor (LA) is designed with green absorption and deep-red emission. In live cells with high MMP levels, FixD and LA both target mitochondria, and deep-red (DR) emission could be detected with the excitation of 405 nm. Particularly, the spectral shift of fluorescence upon the decrease of MMP is up to 110 nm, which is greatly favorable for the clear observation of MMP levels. With the decrease of MMP, LA would be released from mitochondria while FixD would still be immobilized in mitochondria, and decreased DR emission and increased green fluorescence could be detected due to the absence of FRET. In this manner, the MMP levels could be monitored in dual emission colors.

Fluorescence Lifetime Super-Resolution Imaging Unveil the Dynamic Relationship between Mitochondrial Membrane Potential and Cristae Structure Using the Förster Resonance Energy Transfer Strategy

Fei Peng,Xiangnan Ai,Jing Sun,Xichuan Ge,Meiqi Li,Peng Xi,Baoxiang Gao

Mitochondrial cristae, invaginations of the inner mitochondrial membrane (IMM) into the matrix, are the main site for the generation of ATP via oxidative phosphorylation, and mitochondrial membrane potential (MMP). Synchronous study of the dynamic relationship between cristae and MMP is very important for further understanding of mitochondrial function. Due to the lack of suitable IMM probes and imaging techniques, the dynamic relationship between MMP and cristae structure alterations remains poorly understood. We designed a pair of FRET-based molecular probes, with the donor (OR-LA) being rhodamine modified with mitochondrial coenzyme lipoic acid and the acceptor (SiR-BA) being silicon-rhodamine modified with a butyl chain, for simultaneous dynamic monitoring of mitochondrial cristae structure and MMP. The FRET process of the molecular pair in mitochondria is regulated by MMP, enabling more precise visualization of MMP through fluorescence intensity ratio and fluorescence lifetime. By combining FRET with FLIM super-resolution imaging technology, we achieved simultaneous dynamic monitoring of mitochondrial cristae structure and MMP, revealing that during the decline of MMP, there is a progression involving cristae dilation, fragmentation, mitochondrial vacuolization, and eventual rupture. Significantly, we successfully observed that the rapid decrease in MMP at the site of mitochondrial membrane rupture may be a critical factor in mitochondrial fragmentation. These data collectively reveal the dynamic relationship between cristae structural alterations and MMP decline, laying a foundation for further investigation into cellular energy regulation mechanisms and therapeutic strategies for mitochondria-related diseases.

Simultaneous monitoring of mitochondrial viscosity and membrane potential based on fluorescence changing and location switching of carbon dots in living cells

Shuo Guo,Yuanqiang Sun,Zhaofan Wu,Ran Yang,Lingbo Qu,Zhaohui Li

Mitochondrial viscosity and mitochondrial membrane potential (MMP) are vital parameters for reflecting mitochondrial conditions and they are closely correlative in various biological events. Accordingly, simultaneous detection of mitochondrial viscosity and MMP is imperative for elucidating their interactions, further exploring the mechanism of diseases, and investigating numerous pathological procedures. However, such profound research remains a big challenge due to the lack of high-performing single fluorescent probes capable of simultaneously visualizing of these two parameters. In this study, we developed a new kind of fluorescent carbon dots (CDs) that can successfully realize simultaneous monitoring of mitochondrial viscosity and MMP in living cells. The fluorescence of CDs can be specifically turned on as the viscosity of solvents increases from 0.89 to 945 cP. Meanwhile, the CDs accumulate in mitochondria under normal MMP conditions, switch to exist in both mitochondria and nucleoli under reduced MMP, and present only in nucleoli with the vanishment of MMP. Thus, various MMP situations can be easily differentiated through the changed spatial distributions of CDs. More importantly, based on the developed CDs, an increased mitochondrial viscosity accompanying a decreased MMP was simultaneously tracked in drug-stimulated cells. We envision that the developed CDs has a great potential to be utilized as a promising fluorescent probe for mitochondrion-related biology studies.

Mitochondria-engine with self-regulation to restore degenerated intervertebral disc cells via bioenergetic robust hydrogel design

Juehan Wang,Yulin Jiang,Ce Zhu,Zheng Liu,Lin Qi,Hong Ding,Jing Wang,Yong Huang,Yubao Li,Yueming Song,Ganjun Feng,Li Zhang,Limin Liu

Previous studies have confirmed that intervertebral disc degeneration (IDD) is closely associated with inflammation-induced reactive oxygen species (ROS) and resultant cell mitochondrial membrane potential (MMP) decline. Clearance of ROS in an inflammatory environment is essential for breaking the vicious cycle of MMP decline. Additionally, re-energizing the mitochondria damaged in the inflammatory milieu to restore their function, is equally important. Herein, we proposed an interesting concept of mitochondrion-engine equipped with coolant, which enables first to “cool-down” the inflammatory environment, next to restore the MMP, finally to allow cells to regain normal energy metabolism through materials design. As such, we developed a multi-functional composite composed of a reactive oxygen species (ROS)-responsive sodium alginate/gelatin hydrogel infused into a rigid 3D-printed thermoplastic polyurethane (TPU) scaffold. The TPU scaffold was coated with conductive polypyrrole (PPy) to electrophoretically deposit l -arginine, which could upregulate the Mammalian target of rapamycin ( mTOR ) pathway, thus increasing MMP and energy metabolism to stimulate extracellular matrix synthesis for IVD repair. While the ROS-responsive hydrogel acting as the “mito-engine coolant” could scavenge the excessive ROS to create a favorable environment for IVD cells recovery. Demonstrated by in vitro and in vivo evaluations, the mito-engine system markedly promoted the proliferation and collagen synthesis of nucleus pulposus cells while enhancing the mitochondrial respiration and MMP under oxidative stress. Radiological and histological assessments in vivo revealed the efficacy of this system in IVD repair. This unique bioinspired design integrated biomaterial science with mitochondrial biology, presents a promising paradigm for IDD treatment.

Shiitake mushroom-derived extracellular nanovesicles: Preparation, characterization, and inhibition of Caco-2 cells

Xueli Yang,Zihao Zong,Ben Niu,Hangjun Chen,Weijie Wu,Xiangjun Fang,Ruiling Liu,Haiyan Gao,Honglei Mu

In this study, Shiitake mushroom-derived extracellular nanovesicles (SMDENVs) were isolated from fresh Shiitake mushrooms by ultracentrifugation and sucrose gradient ultracentrifugation. The morphological characteristics of SMDENVs were investigated via Transmission Electron Microscopy and Laser Scanning Confocal Microscopy. SMDENVs were spherical, hollow, and uniform in size, with an average diameter of 177.6 ± 51.4 nm. Based on the analysis of lipidomics and proteomics, 383 lipids species and 1290 proteins were identified in SMDENVs. Compared with the conventional liposomes, SMDENVs demonstrated higher stability in different environmental conditions. Furthermore, we observed that SMDENVs were cytocompatible and inhibited the proliferation of Caco-2 cells. SMDENVs could be phagocytized by Caco-2 cells in a time-dependent manner. Further, SMDENVs also inhibited the proliferation of Caco-2 cells in a dose-dependent manner, and the half-maximal inhibitory concentration (IC 50 ) was 236.2 ± 3.2 μg/mL. Additionally, SMDENVs induced cellular apoptosis by increasing the levels of reactive oxygen species and decreasing the mitochondrial membrane potential.

The interactions between CdSe quantum dots and yeast Saccharomyces cerevisiae: Adhesion of quantum dots to the cell surface and the protection effect of ZnS shell

Jie Mei,Li-Yun Yang,Lu Lai,Zi-Qiang Xu,Can Wang,Jie Zhao,Jian-Cheng Jin,Feng-Lei Jiang,Yi Liu

The interactions between quantum dots (QDs) and biological systems have attracted increasing attention due to concerns on possible toxicity of the nanoscale materials. The biological effects of CdSe QDs and CdSe/ZnS QDs with nearly identical hydrodynamic size on Saccharomyces cerevisiae were investigated via microcalorimetric, spectroscopic and microscopic methods, demonstrating a toxic order CdSe   >   CdSe/ZnS QDs. CdSe QDs damaged yeast cell wall and reduced the mitochondrial membrane potential. Noteworthy, adhesion of QDs to the yeast cell surface renders this work a good example of interaction site at cell surface, and the epitaxial coating of ZnS could greatly reduce the toxicity of Cd-containing QDs. These results will contribute to the safety evaluation of quantum dots, and provide valuable information for design of nanomaterials.
