NINJ1 regulates plasma membrane fragility under mechanical strain

Zhu Yunfeng,Xiao Fang,Wang Yiling,Wang Yufang,Li Jialin,Zhong Dongmei,Huang Zhilei,Yu Miao,Wang Zhirong,Barbara Joshua,Plunkett Christopher,Zeng Mengxue,Song Yiyan,Tan Tian,Zhang Ruibin,Xu Kezhen,Wang Zhongxing,Cai Changjie,Guan Xiangdong,Hammack Scott,Zhang Liang,Shi Zheng,Xiang Fu-li,Shao Feng,Xu Jie

Plasma membrane integrity is vital for nearly all aspects of cell functioning 1 . Mechanical forces can cause plasma membrane damage 2 , but it is not known whether there are large molecules that regulate plasma membrane integrity under mechanical strain. Here we constructed a 384-well cellular stretch system that delivers precise, reproducible strain to cultured cells. Using the system, we screened 10,843 siRNAs targeting 2,726 multi-pass transmembrane proteins for strain-induced membrane permeability changes. The screen identified NINJ1, a protein recently proposed to regulate pyroptosis and other lytic cell death 3,4 , as the top hit. We demonstrate that NINJ1 is a critical regulator for mechanical strain-induced plasma membrane rupture (PMR), without the need of stimulating any cell death programs. NINJ1 level on the plasma membrane is inversely correlated to the amount of force required to rupture the membrane. In the pyroptosis context, NINJ1 on its own is not sufficient to fully rupture the membrane, and additional mechanical force is required for full PMR. Our work establishes that NINJ1 functions as a bona fide determinant of membrane biomechanical properties. Our study also suggests that PMR across tissues of distinct mechanical microenvironments is subjected to fine tuning by differences in NINJ1 expression and external forces.

Nanopipette-Based Nanosensor for Label-Free Electrochemical Monitoring of Cell Membrane Rupture under H2O2 Treatment

Xiao-Yuan Wang,Jian Lv,Qin Hong,Ze-Rui Zhou,Da-Wei Li,Ruo-Can Qian

H2O2 is an essential signaling molecule in living cells that can cause direct damage to lipids, proteins, and DNA, resulting in cell membrane rupture. However, current studies mostly focus on probe-based sensing of intracellular H2O2, and these methods usually require sophisticated probe synthesis and instruments. In particular, local H2O2 treatment induces cell membrane rupture, but the level of cell membrane destruction is unknown because the mechanical properties of the cell membrane are difficult to accurately determine. Therefore, highly sensitive and label-free methods are required to measure and reflect mechanical changes in the cell membrane. Here, using an ultrasmall quartz nanopipette with a tip diameter less than 90 nm as a nanosensor, label-free and noninvasive electrochemical single-cell measurement is achieved for real-time monitoring of cell membrane rupture under H2O2 treatment. By spatially controlling the nanopipette tip to precisely approach a specific location on the membrane of a single living cell, stable cyclic membrane oscillations are observed under a constant direct current voltage. Specifically, upon nanopipette advancement, the mechanical status of the cell membrane can be sensibly displayed by continuous current versus time traces. The electrical signals are collected and processed, ultimately revealing the mechanical properties of the cell membrane and the degree of cell apoptosis. This nanopipette-based nanosensor paves the way for developing a facile, label-free, and noninvasive strategy to assay the mechanical properties of the cell membrane during external stimulation at the single-cell level.

Contactless Interfacial Thin-Film Breaking Caused Splash-Like Demulsification of Floating Micron Emulsions via Ionic Wind

Haoyu Dai,Linfeng Yang,Yaping Feng,Junhan Sun,Fenglin Chen,Xianfeng Luo,Zengyi He,Xuetao Xu,Bing Wang,Xubo Liu,Zhichao Dong,Lei Jiang

Emulsified oil leakage onto the bulk water surface causes severe issues on global ecology and health. Developing efficient, rapid, and universal separation methods of emulsions has been a significant topic in scientific studies. However, a contactless and additive-free strategy to achieve continuous floating emulsion separation and oil collection remains to be discovered. Herein, a universal contactless demulsification, transportation, and collection method to dispose floating emulsions by ionic wind, which contains active charged particles generated by corona discharge is reported. The splash-like demulsification process of floating emulsions is attributed to the rupture of water film enveloped on oil droplet surface, simultaneously and respectively. The evidence of this process recorded by a high-speed camera with 20 000 fps has a referential significance for other works. This study may clean up universal floating emulsions discharged on water in real situations such as oil stations, chemical plants, and restaurants, and furthermore increase the potential of remote control in liquid dynamics by corona discharge.

Understanding ultrafast free-rising bubble capturing on nano/micro-structured super-aerophilic surfaces

Hu Yue,Xu Zhenbo,Shi Haotian,Wang Benlong,Wang Liqiu,Zhang Lu-Wen

Rapid bubble capture is essential for collecting targeted gaseous media and eliminating floating impurities across aquatic environments. While the role of nanostructures during the collision of free-rising bubbles with super-aerophilic surfaces is well established, the fundamental contribution of microtextures in promoting initial capture, even before contact, has yet to be fully understood. We report the rising bubble-induced large deformation of the entrapped gas layer, rapidly thinning the liquid film to its rupture threshold and thus achieving an ultrafast bubble capture down to about 1 ms with an array of microcones, decorated with nanoparticles as a convenient example to obtain super-aerophilicity. This rapid capture is also very stable due to the hysteresis movement of three-phase contact lines that inspired a critical pressure criterion for ensuring gas-layer stability and capture efficacy. The present nano/microstructured surface supports prolonged, loss-free gas transport in challenging shear flow as well, providing robust bubble control strategies for diverse systems.

Calcium/P53/Ninjurin 1 Signaling Mediates Plasma Membrane Rupture of Acinar Cells in Severe Acute Pancreatitis

Chehao Lee,Guang Xin,Fan Li,Chengyu Wan,Xiuxian Yu,Lijuan Feng,Ao Wen,Yu Cao,Wen Huang

Ninjurin 1 (NINJ1) is a double-transmembrane cell-surface protein that might mediate plasma membrane rupture (PMR) and the diffusion of inflammatory factors. PMR is a characteristic of acinar cell injury in severe acute pancreatitis (SAP). However, the involvement of NINJ1 in mediating the PMR of acinar cells in SAP is currently unclear. Our study has shown that NINJ1 is expressed in acinar cells, and the expression is significantly upregulated in sodium-taurocholate-induced SAP. The knockout of NINJ1 delays PMR in acinar cells and alleviates SAP. Moreover, we observed that NINJ1 expression is mediated by Ca2+concentration in acinar cells. Importantly, we found that Ca2+overload drives mitochondrial stress to upregulate the P53/NINJ1 pathway, inducing PMR in acinar cells, and amlodipine, a Ca2+channel inhibitor, can reduce the occurrence of PMR by decreasing the concentration of Ca2+. Our results demonstrate the mechanism by which NINJ1 induces PMR in SAP acinar cells and provide a potential new target for treatment of SAP.Keywords:nerve-injury-induced protein 1;plasma membrane rupture;severe acute pancreatitis

The electrostatic interaction between nanoparticles and surfactants with complementary groups promotes the liquid film rupture

Haozhe Yi,Taotao Fu,Daofan Ma,Chunying Zhu,Youguang Ma

The stability of the liquid film is determined by the van der Waals force and the electrical double layer force, which dictates the termination of the liquid film drainage process. The liquid film drainage is the rate-determining step of the thermodynamic interactions between droplets and interfaces, including droplet coalescence, adsorption, and cross-phase transfer. The interaction between nanoparticles and surfactants has great potential in influencing the stability of liquid film. When nanoparticles and surfactants are adsorbed on the two interfaces of liquid film respectively, an electrical double layer force is generated to attract or repel the two interfaces based on the charges’ polarity. Consequently, we verified the electrostatic interaction between nanoparticles and surfactants experimentally. Subsequently, the effect of electrical double layer force on liquid film rupture was validated by analyzing the dynamics of droplet adsorption. Finally, the intensification of droplet adsorption in a microchannel was realized by utilizing this mechanism. This study establishes a fundamental framework for precise regulation of interfacial forces at the scale of liquid film thickness, thereby providing a novel approach to investigate soft matter.

Highly efficient sterilization of biogenic FeS2 nanoparticles: Mechanism and inhibition of antibiotic resistance

Siyu He,Jing Chen,Junqiao Zhao,Zhe Wang,Ruoyan Wu,Yongkui Zhang

As antibiotic-resistant bacteria rapidly render antibiotics ineffective, the development of antibacterial nanomaterials that can replace antibiotics is extremely urgent. Here, novel biogenic FeS 2 (bio-FeS 2 ) nanomaterials with broad-spectrum antibacterial activity were synthesized using biogenic FeS as a precursor, which can delay the onset of bacterial resistance. Both S. aureus and E. coli were completely inactivated by bio-FeS 2 under nutritional conditions. Mechanism studies revealed that bio-FeS 2 exhibited superior antibacterial performance than other iron sulfides attributed to the stronger release of iron and sulfur ions and crystal structure. The antibacterial potency of bio-FeS 2 active components follows the order as below: Fe 2+ >S 2 2- >S 2 2- disproportionation to generate S 2- >washed bio-FeS 2 precipitate. These components synergistically killed bacteria by membrane rupture, glutathione consumption, and lipid peroxidation accumulation. Six strains of antibiotic-resistant E. coli were found in the real municipal wastewater. Bio-FeS 2 kept the initial antibacterial activity against the resistant E. coli after ten passages, demonstrating the ability to impede bacterial resistance, whereas tetracycline induced bacterial resistance within four passages. Furthermore, bio-FeS 2 showed biocompatibility with mammalian cells and aquatic plants. This study proposes an efficient antibacterial agent and offers a promising strategy for the treatment of resistant bacteria.

Antibacterial effect and mechanism against Escherichia coli of polysaccharides from Armillariella tabescens mycelia

Kunfeng Zhang,Yuzhe Huang,Qianzhen Wu,Wenhua Guo,Hao Chen,Wenna Zhang,Yong Li,Yongming Lu,Qingxi Wu,Wenjuan Pan,Lei Chen,Yan Chen

The objectives of this study were (1) to prepare Armillariella tabescens mycelia polysaccharides (PAT) with remarkably growth inhibitory effect on typical food-borne pathogenic bacteria using a green and efficient polyamide method and (2) to explore the antibacterial mechanism of PAT for use as a natural antibacterial agent. The sugar and uronic acid contents of PAT were 93.41% and 12.24%, respectively. PAT could inhibit the growth of Escherichia coli , Proteus vulgaris , Bacillus subtilis , and Staphylococcus aureus cells, with minimum inhibitory concentrations of 0.5, 1.0, 4.0, and 4.0 mg/mL, respectively. Ultra-high-resolution field emission scanning electron microscopy and high-resolution transmission electron microscopy analysis revealed cell wall and membrane rupture of E. coli treated with PAT. Further, 0.5–4.0 mg/mL PAT was found to significantly ( P  < 0.01) and concentration-dependently increase the conductivity of the broth, exudation of the intracellular protein, and alkaline phosphatase and β-galactosidase activities. Confocal laser scanning microscopy revealed morphological changes in E. coli DNA after PAT treatment and intracellular reactive oxygen species accumulation; flow cytometry revealed E. coli cell apoptosis. Our findings provide a theoretical basis and technical support for the development of PAT as a natural antibacterial product.

NINJ1-mediated plasma membrane rupture of pyroptotic endothelial cells exacerbates blood-brain barrier destruction caused by neutrophil extracellular traps in traumatic brain injury

Zheng Xiao-Bo,Wang Xue,Gao Sheng-Qing,Gao Chao-Chao,Li Tao,Han Yan-Ling,Zhao Ran,Sun Yan,Miao Shu-Hao,Qiu Jia-Yin,Jin Wang-Xuan,Zhou Meng-Liang

Brain endothelial cell (bEC) dysfunction is the main factor of blood-brain barrier (BBB) breakdown, which triggers a vicious cycle of aggravating traumatic brain injury (TBI) pathogenesis. Previous studies have revealed that neutrophil extracellular traps (NETs) released by neutrophils can lead to BBB disruption, but there is a lack of research on the underlying mechanisms after TBI. Here, excessive NETs were found in both contused brain tissue and circulation following TBI. We found that NETs could activate the TLR4/NF-κB pathway to induce bEC pyroptosis, which led to BBB disruption after TBI. During this process, ninjurin-1 (NINJ1) was activated in pyroptotic bECs, and it mediated the release of high mobility group box 1 protein (HMGB1) via plasma membrane rupture (PMR) to promote NET formation. NINJ1-mediated release of HMGB1 aggravated NET accumulation by forming a vicious circle following TBI. Knockdown of NINJ1 rescued NET formation, attenuated BBB leakage, and improved neurological outcomes after TBI. NINJ1 may represent a promising target for alleviating NET-induced BBB destruction and other related injuries after TBI.

Size- and shape-dependent cytotoxicity of nano-sized Zr-based porphyrinic metal-organic frameworks to macrophages

Fang Hao,Zhu-Ying Yan,Xiu-Ping Yan

The wide utilization of nano-sized metal-organic frameworks (NMOFs) leads to inevitable health risks to humans. Previous studies on health risks of NMOFs mainly focus on the cytotoxic tests of typical NMOFs,but lack sufficient studies on the effects of physiochemical characteristics of NMOFs on the cytotoxicity and the related mechanisms. Here, four kinds of Zr-based porphyrinic NMOFs (PCNs), including spherical 30, 90, and 180 nm PCN-224 and rod-like 90 nm PCN-222, were taken as a proof of the concept to investigate the effects of the size and shape of NMOFs on the cytotoxicity and related mechanisms to macrophages. The 30 nm spherical PCN-224 induced significant rupture of cell membrane and dissolved in lysosome, leading to the most significant cell necrosis among the studied other nano-sized PCNs. However, other studied PCNs showed insignificant membrane rupture and their dissolution in lysosome. Furthermore, the 90 nm-sized PCN-224 led to much more significant cell necrosis by inducing lysosome damage and inhibiting of autophagy flux than the rod-like 90 nm PCN-222. These findings reveal the size- and shape-dependent cytotoxicity of PCNs and the related mechanisms and are helpful to the assessment of the potential health risks of NMOFs and the safe application of NMOFs.

Cationic engineered nanodiamonds for efficient antibacterial surface with strong wear resistance

Li Fu-Kui,Zhao Wen-Bo,Wang Yong,Huang Wen-Tao,Ku Ya-Lun,Liu Hang,Guo Rui,Yu Hui-Hui,Liu Kai-Kai,Shan Chong-Xin

The spread of diseases caused by bacterial adhesion and immobilization in public places constitutes a serious threat to public health. Prevention of bacteria spread by the construction of an antibacterial surface takes precedence over post-infection treatment. Herein, we demonstrate an effective antibacterial surface with strong wear resistance by constructing cationic engineered nanodiamonds (C-NDs). The C-NDs with positive surface potentials interact effectively with bacteria through electrostatic interactions, where the C-NDs act on the phospholipid bilayer and lead to bacterial membrane collapse and rupture through hydrogen bonding and residual surface oxygen-containing reactive groups. In this case, bactericidal rate of 99.99% and bacterial biofilm inhibition rate of more than 80% can be achieved with the C-NDs concentration of 1 mg/mL. In addition, the C-NDs show outstanding antibacterial stability, retaining over 87% of the antibacterial effect after stimulation by adverse environments of heat, acid, and external abrasion. Therefore, an antibacterial surface with high wear resistance obtained by integrating C-NDs with commercial plastics has been demonstrated. The antibacterial surface with a mass fraction of 1 wt.% C-NDs improved abrasion resistance by 3981 times, with 99% killing of adherent bacteria. This work provides an effective strategy for highly efficient antibacterial wear-resistant surface, showing great practical applications in public health environments.
