Pore formation can facilitate the release of various intracellular substances upon cell death, which is of critical benefit for tumor immunotherapy by immunogenic cell death (ICD). Given the effective endogenous antigen release is of primary importance for ICD-eliciting immunogenicity, other than by enhancing the cytotoxicity of ICD, we designed an immunogenic induction strategy via pore formation based on saponin, a composite medium of the membrane-disrupting agent. In this study, saponin/polyphenol (ZS-TA) at proper concentrations can directly cause membrane perforation by removal of cholesterol from the membrane, thereby lead to the release of intracellular substances. Simultaneously, in situ nano-antigens (nano-Ag) would be formed through the mechanism of protein-polyphenol interaction and the nano-Ag would be served as reservoirs of antigen to trigger long-term immune effects. Meanwhile, this membrane perforation could also enhance the uptake of chemotherapeutic drugs, which could be served as a general approach for drug delivery. Therefore, this work insights for the design of enhanced drug delivery system and in situ vaccines to sensitize tumor chemo-immunotherapy.

Tympanic membrane perforation (TMP) often leads to hearing loss and requires effective repair strategies. However, existing surgical options are invasive and lack ideal biomaterials for scaffold-based healing. Herein, we present a custom-engineered mechano-acoustic responsive hydrogel incorporating bFGF-loaded sodium alginate microspheres, designed for controlled drug release and tissue regeneration under dual stimulation: vibrational simulation and low-frequency acoustic waves (generated by radio devices) — representing its first transition from in vitro characterization to in vivo regenerative application. This polymer-based scaffold exhibits robust adhesion, biocompatibility, and mechanoresponsive release behavior. A rat acute TMP model was employed to evaluate the hydrogel's therapeutic efficacy. Compared with control and blank hydrogel groups, the bFGF-loaded mechanoresponsive hydrogel (SGM) noticeably accelerated tympanic membrane closure, reduced local inflammation, and enhanced early auditory recovery, as confirmed by otoscopic inspection, ABR tests, histological staining, and TEM imaging. Our findings demonstrate that the SGM hydrogel effectively promotes functional tissue regeneration and early hearing restoration in vivo. This work highlights the potential of polymer-based, stimuli-responsive biomaterials in advancing minimally invasive strategies for TMP treatment and offers valuable insights for future tissue engineering applications in otolaryngology.

The tympanic membrane plays an important role in the human hearing system, which is easily perforated under unfavorable conditions, leading to loss of hearing and otitis media. Many autologous materials and artificial materials have been used to repair a perforated tympanic membrane, but these materials sometimes can cause severe hearing loss because of their adhesion to the ossicle during the healing process and the postoperative process. Herein, we report Janus membranes with asymmetric cellular adhesion behaviors for regenerating the eardrum. These Janus membranes are constructed by co-depositing a tannic acid (TA)/3-aminopropyltriethoxysilane (APTES) coating on one surface of the polypropylene microfiltration membrane. Cellular experiments indicate that the Janus membranes have good biocompatibility and asymmetric cellular adhesion properties. The repair of the tympanic membrane perforation experiment and laser Doppler vibrometer (LDV) measurements prove that the hydrophilic surface of Janus membranes repairs perforated eardrums, and meanwhile the hydrophobic surface can avoid adhering to the inner ear tissue for reducing hearing loss. The Janus membranes have good prospects in the treatment of tympanic membrane perforation.
