Battery-free antenna sticker for wireless sensing joint rotation

Pengxiang Si,Mingrui Liang,Jihua Zou,Yu Kuang,Juntao Zhang,Qingsha S. Cheng,Yiqian Kong,Yuxin Cai,Guozhi Huang,Yun Wu,Zhu Long,Boxin Zhao,Dan Zhang

Bio-integrated electronic devices are skin-mounted accurate tools for acquiring biological signals. The knee joint is one of the most important joints for human movement, playing a crucial role in functions such as walking, standing, sitting and containing important physiological health information about orthopedic disease diagnosis, prevention, and rehabilitation. However, it is still challenging to precisely and directly monitor joint rotation via skin-mounted bio-integrated electronic devices because joints are hidden inside the skin. In this study, we design a reusable, sustainable, battery-free, wireless, and flexible antenna sticker to adhere to the skin that can directly measure knee joint rotation degree. The antenna sticker is adhered to the lower leg and powered by electromagnetic radiation from a radio frequency identification (RFID) reader. When rotating the knee, the distance between the antenna sticker and the reader changes, leading to an alteration in the received signal strength indicator (RSSI). Meanwhile, the 3D optical motion capture system collects joint rotation data as a “gold standard”. After matching two series of real-time data using the Gaussian method, one RSSI can represent one specific joint rotation degree. The antenna sticker is stable at a variety of humidity and temperatures. The antenna sticker shows great potential to sense joint rotation and human motions. The scientific insights and engineering principles revealed in this antenna sticker will be applicable to a broad range of applications including skin-mounted epidermal electronics and wearable devices.

Study on the Slow-Release Mometasone Furoate Injection of PLGA for the Treatment of Knee Arthritis

Liang Yutong,Zhang Jiaojiao,Zhao Xinghua,Wang Ming,Ding Shi,Wang Yang,Chen Ye,Liu Ju

Purpose: The purpose of this study is to develop a new PLGA based formulation for microspheres, which aims to release mometasone furoate for one month, so as to improve compliance. Methods: The microspheres containing mometasone furoate were prepared by oil in water emulsion and solvent evaporation. The microspheres were characterized by surface morphology, shape, size and encapsulation efficiency. The release in vitro was studied in 37°C phosphate buffer, and in vivo, pharmacodynamics and preliminary safety evaluation were conducted in male Sprague Dawley rats. Results: The morphology results showed that the microspheres have a smooth surface, spherical shape and an average diameter of 2.320-5.679μm. The encapsulation efficiency of the microspheres loaded with mometasone furoate was in the range of 53.1% to 95.2%, and the encapsulation efficiency of the microspheres could be greatly affected by the proportion of oil phase to the water phase and other formulation parameters. In vitro release kinetics revealed that drug release from microspheres was through non-Fick's diffusion and PLGA polymer erosion. Pharmacokinetic data showed that the initial release of microspheres was small and then sustained. The results of the pharmacodynamics study fully proved the long-term effectiveness of mometasone furoate microspheres. The results of in vivo safety evaluation showed that the preparation system possessed good in vivo safety. Conclusion: This study shows that the microspheres prepared in this study have sufficient ability to stable drug release at least for 35 days, with good efficacy and high safety. In addition, mometasone furoate can be used as a potential candidate drug for 35 days long-term injection.

Light-Driven Artificial Cell Micromotors for Degenerative Knee Osteoarthritis

Chao Gao,Ye Feng,Suyi Liu,Bin Chen,Miaomiao Ding,Dailing Du,Wenjing Zhang,Daniela A. Wilson,Yingfeng Tu,Fei Peng

Combining artificial cellular compartmentalization and intelligent motion benefits of micro/nanomotors, light is used as energy input to construct an artificial cell-based micromotor capable of photosynthetic anabolism and intelligent directional movement. This system is assembled from phospholipids functionalized with F-ATP synthase and molybdenum disulfide (MoS 2 ) nanoparticles (Vesical@MoS 2 -ATPase). The underlying mechanism involves the generation of protons (H + ) through photo-hydrolysis of MoS 2 nanoparticles within vesicles, which generates a local electroosmotic flow inside the vesicles and drives the negatively charged MoS 2 toward light. The established proton gradient across the phospholipid membrane, in turn, drives the ATP synthase to catalyze ATP production. Both in vitro and in vivo models demonstrate that the micromotor can elevate local intracellular ATP levels upon light and improve the metabolism of denatured chondrocytes. This cell mimicry, with capabilities of migration and biosynthesis, emerges as a promising platform for the next generation of functional bio-interface.

A novel composite meniscus scaffold with shape memory and adhesive properties alleviates knee cartilage degeneration through minimally invasive and sutureless approach

Yan Wenqiang,Tian Feng,Zhao Lingyu,Wang Pengyi,Zhang Manwen,Gao Zeyuan,Huang Wei,Ao Yingfang,Yu Ran

Three-dimensionally (3D) printed engineered meniscus scaffolds offer a promising therapeutic strategy for post-meniscectomy defects. Nevertheless, existing scaffolds face critical limitations impeding clinical implementation. Primarily, the transplantation procedure necessitates larger incisions in skin and joint capsules due to insufficient scaffold deformability. This invasive approach risks iatrogenic tissue damage, prolongs recovery timelines, and compromises aesthetic outcomes. Secondly, conventional fixation methods involving sutures to peripheral meniscal remnants or capsular tissues introduce dual complications: mechanical perforation of scaffolds reduces structural integrity and accelerates degradation, while the demanding technical requirements for suture placement increase surgical complexity. To address these challenges, we engineered a composite meniscus scaffold featuring an elastomeric skeleton fabricated via digital light processing (DLP) 3D printing integrated with a hydrogel matrix. This biomimetic design recapitulates the native meniscus architecture comprising an aqueous extracellular matrix reinforced by collagen fibrils. The composite scaffold demonstrates three critical functional attributes: 1) biomechanical properties approximating native meniscus tissue, 2) exceptional shape recovery capabilities facilitating trans-arthroscopic delivery through minimally invasive portals, and 3) strong interfacial adhesion enabling suture-free fixation via covalent bonding to host tissues. In vivo evaluation using a rabbit meniscectomy model confirmed the scaffold’s chondroprotective efficacy. Notably, preclinical validation in an ex vivo ovine knee under arthroscopic guidance demonstrated successful scaffold deployment through subcentimeter incisions with stable adhesion to residual meniscal tissue. This innovative design eliminates suture-related complications while reducing surgical invasiveness, thereby presenting a clinically translatable solution for irreparable meniscal injuries.

Injectable Nanocomposite Hydrogel System for miRNA-Based Cartilage Repair in Knee Osteoarthritis

Dianbo Long,Guping Mao,Hongyi Li,Huaiwen Chen,Rui Huang,Tianyu Zhao,Zhiqi Zhang,Xiaoyi Zhao

Knee osteoarthritis (KOA) is a common joint disorder characterized by progressive cartilage degeneration. MiR-455-3p has been identified as a promising therapeutic candidate to maintain cartilage homeostasis. However, clinical application of free miR-455-3p is limited by rapid degradation, poor targeting efficiency, and off-target effects. To overcome these limitations, an injectable nanocomposite hydrogel-based miRNA delivery system consisting of a cartilage affinity nanocarrier (CANC) encapsulated within a PCL-b-PEG-b-PCL hydrogel is developed. The CANC is formulated using 50% PEGylated G5 PAMAM dendrimers to load miR-455-3p and is further modified with chondrocyte-affinity peptides (CAP) and minimal “self” peptides (MSP) to enhance cartilage targeting and evade macrophage uptake. In vitro studies demonstrated that it exhibited excellent stability, low cytotoxicity, superior cartilage penetration and chondrocyte targeting. Moreover, in vivo experiments in miR-455-3p knockout mice and a destabilization of the medial meniscus (DMM) induced KOA model confirmed that the sustained release and targeted delivery of miR-455-3p effectively rescued cartilage degeneration and prevent KOA progression. Notably, the nanocomposite hydrogel demonstrated high biocompatibility without observed cytotoxicity in major organs. This study highlights the therapeutic potential of the nanocomposite hydrogel miRNA delivery system in promoting cartilage regeneration and presents a promising approach for the development of disease-modifying osteoarthritis drugs.

Observation of the Effects of Infrapatellar Fat Pad Excision on the Inflammatory Progression of Knee Osteoarthritis in Mice

Ya Li,Peizhi Lu,Haoyu Yao,Shuo Yang,Bizhi Tu,Lingchao Kong,Rende Ning

Background Knee osteoarthritis (KOA) is a degenerative joint disease characterized by cartilage degradation, synovial inflammation, and joint pain. The infrapatellar fat pad (IFP) has been suggested to play a role in modulating the inflammatory processes in KOA. Excision of the IFP is considered a potential therapeutic approach to reduce inflammation and slow disease progression. Methods A mouse model of KOA was used to evaluate the impact of IFP excision on inflammation. Mice were divided into five groups: sham (control), unexcised IFP, quarter excision, partial excision, and complete excision of the IFP. Knee joints were collected at early, middle, and late stages of KOA. Gait analysis, micro-computed tomography (micro-CT), HE staining, Safranin O-Fast Green staining, and immunohistochemistry (IHC) were performed to assess joint stability, bone changes, and inflammatory markers (MMP-3, IL-6, TNF-α, COL-2). qRT-PCR was conducted for cartilage tissue analysis. Results Partial IFP excision significantly improved joint stability, particularly in the middle and late stages of KOA. Micro-CT analysis showed increased bone volume fraction (BV/TV) and trabecular thickness (Tb.Th) in excised groups, with the most significant effects in the partial and complete excision groups. IHC and qRT-PCR indicated reduced MMP-3, IL-6, and TNF-α levels in excised groups, particularly in the partial and complete excision groups, suggesting reduced inflammation. COL-2 expression was higher in excised groups, particularly in late-stage KOA, indicating cartilage protection. The partial excision group exhibited the most balanced reduction in inflammation and improved cartilage integrity across all disease stages. Conclusion IFP excision, especially partial excision, significantly modulates the inflammatory response in KOA. Partial excision showed the most effective and balanced impact on joint stability, bone integrity, and cartilage protection, offering potential as a therapeutic approach for KOA.

Biomimetic cartilage scaffold with orientated porous structure of two factors for cartilage repair of knee osteoarthritis

Jianhua Wang,Yingying Wang,Xiaomin Sun,Deshuai Liu,Chenguang Huang,Jiulin Wu,Chunrong Yang,Qiqing Zhang

A dual-layer biomimetic cartilage scaffold was prepared by mimicking the structural design, chemical cues and mechanical characteristics of mature articular cartilage. The surface layer was made from collagen (COL), chitosan (CS) and hyaluronic acid sodium (HAS). The transitional layer with microtubule array structure was prepared with COL, CS and silk fibroin (SF). The PLAG microspheres containing kartogenin (KGN) and the polylysine-heparin sodium nanoparticles containing TGF-β1 (TPHNs) were constructed for the surface, transitional layer, respectively. The SEM result showed that the dual-layer composite scaffold had a double structure similar to natural cartilage. The vitro biocompatibility experiment showed that the biomimetic cartilage scaffold with orientated porous structure was more conducive to the proliferation and adhesion of BMSCs. A rabbit KOA cartilage defect model was established and biomimetic cartilage scaffolds were implanted in the defect area. Compared with the surface layer and transitional layer scaffolds group, the results of dual-layer biomimetic cartilage scaffold group showed that the defects had been completely filled, the boundary between new cartilage and surrounding tissue was difficult to identify, and the morphology of cells in repair tissue was almost in accordance with the normal cartilage after 16 weeks. All those results indicated that the biomimetic cartilage scaffold could effectively repair the defect of KOA, which is related to the fact that the scaffold could guide the morphology, orientation, and proliferation and differentiation of BMSCs. This work could potentially lead to the development of multilayer scaffolds mimicking the zonal organization of articular cartilage.

Synthesis of novel photothermal nanomaterial ZnSe-PAM@1 and its treatment of knee osteoarthritis cells

Li Zhigang,Liu Xuzhuo,Wang Chunlong,Song Hanbing,Zhang Jie

In this study, we synthesized zinc selenide (ZnSe)-polyacrylamide (PAM) nanomaterials for their potential application in treating knee osteoarthritis (KOA). Initially, we characterized the newly prepared material through various techniques including Fourier-transform infrared spectroscopy, scanning electron microscopy, transmission electron microscopy, and nitrogen adsorption–desorption to confirm its identity as the desired product. The results demonstrated that ZnSe-PAM nanomaterial exhibited excellent water solubility, biocompatibility, and stability, along with a large surface area, making it suitable for use as a drug carrier. Subsequently, the material was loaded with the drug 1 to obtain ZnSe-PAM@1. Cellular and thermosensitive experiments were then conducted, revealing that this nanomaterial could facilitate drug release by generating heat, thereby reducing the level of microRNA-146a-5P and blocking the phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin/extracellular (PI3K/Akt/mTOR/ERK) signal-regulated kinase signaling pathway, thus alleviating the symptoms of KOA. These findings present a new direction for drug intervention in KOA and highlight the potential of developing ZnSe-PAM nanomaterials as novel drug carriers.

Artesunate protects against surgery-induced knee arthrofibrosis by activating Beclin-1-mediated autophagy via inhibition of mTOR signaling

Qi Wan,Hui Chen,Guirun Xiong,Rui Jiao,Yun Liu,Xiaolei Li,Yu Sun,Jingcheng Wang,Lianqi Yan

Intraarticular fibrosis following knee surgery is a troublesome complication and remains a challenging problem for clinicians. Artesunate (ART), a classical anti-malarial drug extracted from the Chinese medicinal herb Artemisia annua L, has been associated with some fibrosis-related diseases. However, its effect and underlying mechanism on knee arthrofibrosis are still obscure. In the present study, we found that ART induced cellular autophagy flux and inhibited cell proliferation in fibroblasts. Intriguingly, genetic depletion of Beclin-1 abolished ART-triggered cellular autophagy and further attenuated the inhibitory effect of ART on fibroblasts proliferation. Moreover, at molecular level, our results demonstrated that ART-induced autophagy activation was associated with the inhibition of mTOR signaling through PI3K/AKT/mTOR pathway and AMPK/mTOR pathway. In vivo, ART treatment triggered autophagy activation and alleviated the severity of surgery-induced knee arthrofibrosis. Taken together, we concluded that ART exhibited anti-proliferation efficacy in fibroblasts and alleviated the severity of knee arthrofibrosis in rabbits by inducing Beclin-1-mediated autophagy via inhibition of mTOR signaling. These findings indicated that ART might be a potential therapeutic agent for preventing the progression of surgery-induced intraarticular fibrosis of knee.

Tribological and mechanical behavior of tantalum and its compounds reinforced PEEK coatings for TKR

Haoling Huo,Lin Cao,Jie Li,Huaijun Lin,Qiwei Wang,Xusheng Yang,Chuanjun Zang,Jingtao Zhang,Peng Zhang,Wei Li

In biomedical applications, poly-ether-ether-ketone (PEEK) is capable of modifying traditional metal components, but still lacks strength and wear resistance. In this study, a series of soft-hard phase structure coatings with modified PEEK are produced by combining PEEK as the soft-antifriction phase, as well as tantalum (Ta), tantalum carbide (TaC), and tantalum nitride (TaN) nanoparticles as the hard-wear resistance phase. The coatings of a total knee replacement system were evaluated by finite element analysis (FEA). As a result, the reinforcement of nanoparticles has a significant impact on stress distribution and transfer, improving mechanical behavior and wear resistance, whereas TaC nanoparticles have the greatest ability to enhance these properties. The implant with the modified coating (PEEK/TaC) can be expected to reduce the stress of the Ti implant substrate by about 38.3 % compared to unmodified PEEK coating. Combined the experimental and simulated results showed that the modified coatings have the potential to enhance the lifespan of TKR Ti-based implants.

“Integrated diagnosis and treatment” of upconversion nanocomposite hydrogel for osteoarthritis treatment

Zhao Luan,Xulu Ma,Qingxin Zhao,Anli Yang,Jinming Li

Osteoarthritis (OA) is a common degenerative joint disease for which there is no definitive treatment. Mesenchymal stem cells (MSCs) can be differentiated into many types of cells such as chondrocytes, which have been used for OA treatment. How to keep the cell viability and promote the chondrogenic differentiation after MSCs transplantation into the knee joint cavity   is   crucial. In this study, upconversion nanocomposite hydrogel was developed for integrated diagnosis and treatment of OA. The Er doped core–shell upconversion nanoparticles (UCNPs) were synthesized first and coated on mesoporous silica (UCNP@mSiO 2 ). Then, an H 2 O 2 -sensitive thioketal (TK) linker (MAL-PEG-TK-NHS) was attached to the UCNP@mSiO 2 surface to link capsular β-cyclodextrin (NH 2 -β-CD) and black hole quencher (NH 2 -BHQ 3 ) to form functionalized UCNPs. After loading the drug kartogenin (KGN) that can induce chondrogenic differentiation of MSCs, the UCNP/KGN nanocomposites were mixed with VitroGel to formed the upconversion nanocomposite hydrogel (UCNP/VitroGel). The UCNP/VitroGel could allow MSCs to be implanted well and maintain good survival in vitro. In the presence of hydrogen peroxide (H 2 O 2 ), the TK bond breakage and triggered release BHQ to recover the fluorescence of UCNPs for ROS detection, and the β-CD also left from the surface of UCNPs to release KGN for inducing the chondrogenic differentiation of MSCs. Thus, the UCNP/VitroGel could detect the ROS after inject into the knee joint cavity for OA diagnosis first via the fluorescence recovery of UCNPs under the 980 nm near-infrared light. And then the KGN release from UCNP/KGN nanocomposites in the environment of OA to induce the chondrogenic differentiation of MSCs for the treatment of OA. This UCNP/VitroGel will be a powerful tool for “integrated diagnosis and treatment” of OA.

In vitro antibacterial and In vivo osteogenesis of 3D-printed magnesium peroxide–doped calcium phosphate silicate scaffolds for revision total knee arthroplasty

Lisha Meng,Hao Li,Xujia Hao,Tao Wu,Jingqiu Zhou,Yadong Chen,Qiang Zheng,Xiuhong Cao,Juan Wang,Xinwei Liu,Tongmeng Jiang,Tianxing Gong,Wei Yuan

Revision total knee arthroplasty (RTKA) often encounters tibial bone defects and high infection risk, especially from methicillin-resistant Staphylococcus aureus (MRSA). Current strategies rely on bone grafts with antibiotics, but prolonged use promotes resistance. Here, we developed a 3D-printed magnesium peroxide (MgO 2 )–doped calcium phosphate silicate (CSP) scaffold to address both structural and antibacterial demands. The MgO 2 –CSP scaffold exhibited cancellous bone-like strength (∼7.95 MPa) and an interconnected macroporous structure conducive to cell migration and healing. In vitro , the 14 wt% MgO 2 scaffold (B14M) inhibited 80.4 % of Gram-negative bacteria and 74.6 % of MRSA via Mg 2+ and H 2 O 2 release, while both B0M (no MgO 2 ) and B14M promoted BMSC proliferation and osteogenic differentiation. In vivo , the B14M scaffold markedly enhanced bone regeneration in rat tibial defects, achieving a BV/TV of ∼73.09 % versus ∼29.84 % for B0M at 8 weeks. These findings highlight MgO 2 –CSP scaffolds as a promising strategy to promote osteogenesis while combating MRSA-associated infections in RTKA.
