Traditional oxygen-dependent photodynamic therapy (PDT) faces formidable challenges owing to abnormal angiogenesis and insufficient oxygen supply in solid tumors. Herein, a multifunctional nanoplatform (HRP-DPBF-Zn) consisting of photosensitizer zinc (II) phthalocyanine (ZnPc), singlet oxygen ( 1 O 2 ) probe 1,3-diphenylisobenzofuran (DPBF), and hydrogen peroxide (H 2 O 2 ) catalyst horseradish peroxidase (HRP) is developed. Such HRP-DPBF-Zn nanosystems are expected to possess catalase-like capacity for catalyzing intracellular H 2 O 2 to produce oxygen (O 2 ). The enhanced O 2 content further facilitates the production of cytotoxic 1 O 2 by ZnPc under laser irradiation. DPBF loading endows the nanoplatform with the high 1 O 2 sensitivity, which can promote precise PDT with fluorescent 1 O 2 feedback. HRP-DPBF-Zn exhibits excellent biocompatibility, efficient 1 O 2 generation, high 1 O 2 sensitivity, favorable H 2 O 2 -to-O 2 decomposition, and superior fluorescence imaging properties. Both in vitro and in vivo experimental results confirm that such HRP-DPBF-Zn can accomplish highly specific and efficient PDT against hypoxic tumors, which shows great potential in tumor theranostics.

In this study, a sealable fixed-bed reactor was developed to investigate the influence of gaseous products on lignin pyrolysis with a strategy of hydrogen self-supply, and lignin pyrolysis performances at various temperatures were monitored by the comprehensive characterization of the generated products, including gas, bio-oil and char. The results showed that the release of volatiles (both gas and bio-oil) from lignin was strengthened in this sealable system when the temperature rose, and therefore less char was left. Meanwhile no H 2 was detected under any of the operating conditions, which is a common component in pyrolysis gas from conventional reactors, indicating its participation in lignin pyrolysis. Furthermore, the formation of CO and CO 2 in the employed pyrolyzer benefited from not only the thermal evolution of feedstock, but also the interactions between char residues and generated gas components. Meanwhile, because of the hydrogen self-supply, compounds in bio-oil were enriched and only nine species were detected, which were all phenolic compounds. Vanillic acid was dominant, and its relative content was up to 38.86% for Alcell organosolv lignin and 34.41% for soda alkali lignin. This illustrates that highly selective production of phenolic compounds from lignin pyrolysis can be achieved by self-supplying hydrogen from pyrolysis gas.

The limited light penetration depth and tumor hypoxia are two natural shortcomings of photodynamic therapy (PDT). Overcoming these two issues within a single system is still a great challenge. Herein, photosensitizer (PS)-loaded and PEG-modified MnFe2O4-decorated large-pore mesoporous silica-coated β-NaYF4:20%Yb,2%Er@β-NaYF4 upconversion nanoparticles (UCMnFe-PS-PEG) as excellent PDT agents are successfully prepared for NIR light-mediated and O2 self-sufficient PDT. The large mesoporous structure observably increases PS loading efficiency (11.33 wt%) and the green luminescence from upconversion nanoparticles activated by NIR is able to activate PSs to generate ROS effectively. In addition, sub-10 nm MnFe2O4 nanoparticles work as a Fenton catalyst to generate O2in situ. In vivo experiments further prove that UCMnFe-PS-PEG not only provides magnetic guidance to the tumor, but also overcomes tumor hypoxia and dramatically enhances PDT efficiency. Furthermore, in vivo MR and UCL imaging are performed for accurate cancer therapy. We believe that the successful construction of the multifunctional UCMnFe-PS-PEG provides more revelations for developing advanced nano-drug systems for cancer therapy.

Catalytic therapy based on nanozymes is promising for the treatment of bacterial infections. However, its therapeutic efficacy is usually restricted by the limited amount of hydrogen peroxide and the weak acidic environment in infected tissues. To solve these issues, we prepared polyvinyl alcohol (PVA)–polyacrylic acid (PAA)–iron oxide (Fe3O4)/polyvinyl alcohol (PVA)–zinc peroxide (ZnO2) double-layer electrospun nanofibers (PPF/PZ NFs). In this design, PVA serves as the carrier for ZnO2 nanoparticles (NPs), Fe3O4 NPs, and PAA. The double-layer structure of nanofibers can spatially separate the PAA and ZnO2 to avoid their reaction with each other during preparation and storage, while in the wet wound bed, PVA can dissolve and PAA can provide H+ ions to promote the generation of hydrogen peroxide and subsequent conversion to hydroxyl radicals for bacteria killing. In vitro experimental results demonstrated that PPF/PZ NFs can reduce the methicillin-resistant Staphylococcus aureus by 3.1 log (99.92%). Moreover, PPF/PZ NFs can efficiently treat the bacterial infection in a mouse wound model and promote wound healing with negligible toxicity to animals, indicating their potential use as “plug-and-play” antibacterial wound dressings. This work provides a novel strategy for the construction of double-layer electrospun nanofibers as catalytic wound dressings with hydrogen peroxide/acid self-supplying properties for the efficient treatment of bacterial infections.

Photodynamic therapy (PDT) holds promise in biochemical study and tumor treatment. A novel multifunctional nanosystem CaO 2 /MnO 2 @polydopamine (PDA)-methylene blue (MB) nanosheet (CMP-MB) was designed. CaO 2 nanoparticles were encapsulated by MnO 2 nanosheet, and then PDA was coated on the surface of CaO 2 /MnO 2 nanosheets, which could adsorb photosensitizer MB through hydrophobic interaction or π-π stacking. In this nanosystem, CaO 2 /MnO 2 had the ability of self-production of oxygen, which solved the problem of tumor hypoxia largely. Moreover, it is worth mentioning that the fluorescence of MB was suppressed by MnO 2 , while its emission was triggered in the simulated tumor microenvironment. Therefore, CMP-MB nanosheet could be used to switch-control cell imaging potentially. 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyltetrazolium bromide testing and Live/Dead assay confirmed CMP-MB nanosheet had fewer side effects without illumination while it destroyed Hela cell with the illumination of light. Vitro cell experiment demonstrated CMP-MB nanosheet could achieve tumor microenvironment responsive imaging and inhibit tumor cell growth under illumination effectively. Therefore, the system has great potential for PDT application and switch-control tumor cell imaging. © 2018 Wiley Periodicals, Inc. J Biomed Mater Res Part B: Appl Biomater, 106B: 2544–2552, 2018.

Chemodynamic therapy (CDT), an innovative approach for treating bacterial infections, has garnered significant attention due to its ability to generate hydroxyl radicals (•OH) via Fenton/Fenton-like reactions. However, the effectiveness of CDT is considerably hindered by the limited availability of endogenous hydrogen peroxide (H2O2) and the overexpression of glutathione (GSH) within the infection microenvironment. To address these limitations, a multifunctional nanoplatform with self-supplying H2O2, GSH-depletion properties, and photothermal properties was developed through a straightforward and mild strategy. This platform employs calcium peroxide (CaO2) as the core, coated with silica (SiO2) to enhance stability and further modified with a Cu(II)-doped polydopamine (PDA) layer, forming a core–shell structured CaO2@SiO2@PDA-Cu (CSPC). The Cu(II) released by CSPC, combined with the H2O2 produced from CaO2 degradation, participates in a Fenton-like reaction to generate toxic •OH radicals. Additionally, Cu(II)-mediated redox reactions deplete overexpressed GSH, thereby enhancing CDT efficacy. Upon coordination with Cu(II), the photothermal properties of PDA are significantly enhanced, achieving a photothermal conversion efficiency of up to 43%. The hyperthermia induced by photothermal therapy (PTT) further increases •OH production, augmenting CDT. The CSPC nanomaterials demonstrated outstanding synergistic photothermal bactericidal activity against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) at 60 μg/mL, achieving complete eradication. Moreover, CSPC eliminated 65.90 ± 3.46% of the S. aureus biofilm under near-infrared (NIR) irradiation. In vivo experiments demonstrated that CSPC treatment effectively eradicated bacteria, with a bacterial survival rate of 6.56 ± 3.28%, and accelerated wound healing, reducing the relative wound size to 7.0 ± 2.6%. Therefore, this study successfully developed versatile nanomaterials that significantly enhance the PTT/CDT dual-mode antibacterial performance.

The hypoxic character of tumors and the poor targeting ability of photosensitizers often limit the efficacy of photodynamic therapy (PDT). In recent years, the discovery of metal nanoenzymes and nanocarriers has improved PDT. Thereby, to improve the effective utilization of photosensitizers and oxygen (O 2 ) in tumors, herein, a nanosystem (LS-HB@HvCeO 2 -NRP1 mAb, LHCN1) is reported, in which a hollow virus-like cerium oxide (HvCeO 2 ) is surface-decorated with tumor-targeting neuropilin-1 monoclonal antibody (NRP1 mAb), and loaded with a photosensitizer (chlorin e6-C-15-ethyl ester, LS-HB). In vitro and in vivo experiments demonstrate that LHCN1 can efficiently accumulate within the tumor sites via the targeting guidance of NRP1 mAb and is then rapidly endocytosed into cells. Furthermore, HvCeO 2 with catalase-mimetic activity can decompose the endogenous hydrogen peroxide (H 2 O 2 ) to promote O 2 via the valence transformation between Ce 4+ and Ce 3+ , relieving tumor hypoxia and improving the PDT efficacy. Upon near-infrared laser irradiation, LS-HB produces large amounts of cytotoxic reactive oxygen species. Moreover, LHCN1 is used in fluorescence/photoacoustic multimodal imaging for in vivo drug localization, and its use in PDT evidently helps inhibit tumor growth with no apparent toxicity to normal tissues. Thus, LHCN1 may provide a promising strategy for precise tumor-specific diagnosis and treatment.

The real-time tracking and efficacy evaluation of therapeutic nanoplatforms especially in deep-tissues is of great importance but faces challenges. Meanwhile, chemodynamic therapy (CDT), relying on Fenton reaction by converting H2O2 into toxic hydroxyl radicals (•OH), has drawn wide interests in the fabrication of nanozymes for tumor therapy, while endogenous H2O2 is usually insufficient for effective CDT. Here, we report the pH-responsive multifunctional nanoplatforms consisting of copper peroxide (CP) nanoparticles, paclitaxel (PTX) and perfluoro-15-crown-5-ether (PFCE), for 19F magnetic resonance imaging guided and enhanced chemo–chemodynamic synergetic therapy with self-supplied H2O2 stemmed from the decomposition of CP nanoparticles under acid conditions in tumor. The decomposition of CP nanoparticles further promotes the release of PTX for enhanced chemotherapy. Both in vitro and in vivo results indicate that the efficient generation of •OH and drug release effectively inhibits tumor growth. Furthermore, 19F MRI signal can clearly track the fate of nanoplatforms in tumor and guide tumor treatment. This work provides a promising strategy for the rational design and construction of multifunctional nanoplatforms for imaging-guided synergistic therapy of deep seated tumor.

Photodynamic therapy (PDT) and ferroptosis show significant potential in tumor treatment. However, their therapeutic efficacy is often hindered by the oxygen-deficient tumor microenvironment and the challenges associated with efficient intracellular drug delivery into tumor cells. Toward this end, this work synthesized perfluorocarbon (PFC)-modified Pluronic F127 (PFC-F127), and then exploits it as a carrier for codelivery of photosensitizer Chlorin e6 (Ce6) and the ferroptosis promoter sorafenib (Sor), yielding an oxygen self-supplying nanoplatform denoted as Ce6-Sor@PFC-F127. The PFCs on the surface of the micelle play a crucial role in efficiently solubilizing and delivering oxygen as well as increasing the hydrophobicity of the micelle surface, giving rise to enhanced endocytosis by cancer cells. The incorporation of an oxygen-carrying moiety into the micelles enhances the therapeutic impact of PDT and ferroptosis, leading to amplified endocytosis and cytotoxicity of tumor cells. Hypotonic saline technology was developed to enhance the cargo encapsulation efficiency. Notably, in a murine tumor model, Ce6-Sor@PFC-F127 effectively inhibited tumor growth through the combined use of oxygen-enhanced PDT and ferroptosis. Taken together, this work underscores the promising potential of Ce6-Sor@PFC-F127 as a multifunctional therapeutic nanoplatform for the codelivery of multiple cargos such as oxygen, photosensitizers, and ferroptosis inducers.

Chemodynamic therapy (CDT), which employs Fenton/Fenton-like agents to decompose hydrogen peroxide (H 2 O 2 ) into toxic hydroxyl radical (•OH) to induce cancer cell apoptosis and necrosis, holds great promise in tumor therapy due to its high selectivity . Nevertheless, its efficiency is impaired by the insufficient intracellular H 2 O 2 concentration and/or the insensitive response of Fenton/Fenton-like agents to the slightly acid tumor microenvironment (pH∼7.0–6.5). Herein, we develop a novel CDT reagent based on CeO 2 quantum dot (QD) decorated MgO 2 nanosheets engineered with cascade reactions to boost the intracellular H 2 O 2 level and high pH-activated (pH = 6.5) characteristic for an enhanced CDT. Under the tumor microenvironment (pH = 6.5), MgO 2 nanosheets that are highly reactive can react with H 2 O to produce nontoxic Mg 2+ and abundant H 2 O 2 , boosting the intracellular H 2 O 2 level. The self-generated H 2 O 2 is subsequently converted into •OH by CeO 2 QD, which is served as a relatively high pH-activated (pH = 6.5) Fenton-like agent. The sufficient intracellular H 2 O 2 supply and sensitive response to the slightly acid tumor sites significantly improve the Fenton reaction, leading to the excellent in vivo CDT results with tumor growth inhibition effect. Our work presents a distinctive paradigm for self-boosting CDT efficacy.

The emergence of bacterial resistance has a negative impact on the conventional antimicrobial treatments, and research into the development of the new antimicrobial materials is of great significance. Multimodal synergistic antimicrobial materials exert better antimicrobial effects compared with a single modality. In recent years, Polyoxometalate (POM) has shown great potential in the biomedical field due to its high catalytic activity and high photothermal conversion ability. However, owing to its small surface area, its applications have been greatly limited. Herein, we designed a H 2 O 2 self-supplied CDT and PTT bimodal nanogeneration material (i.e., CaO 2 /POM@MOF) possessing a larger surface area for the treatment of the bacterial infections, in which CaO 2 could release O 2 and H 2 O 2 in the weakly acidic microenvironment of bacterial infection and the dual catalytic site of POM@MOF could enhance the CDT reaction to generate ROS, resulting in the bacterial oxidative stress and the leakage of the bacterial contents, and the exposure to the NIR light generates localized high temperature that cause the rupture of bacterial cell membranes and the denaturation of their proteins. Meanwhile, the superior stability of POM@MOF can release fewer metal ions to improve its biocompatibility. The in vitro antimicrobial experiments demonstrated that CaO 2 /POM@MOF possessed the combined CDT and PTT effect and exhibited excellent antimicrobial efficacy. This work presents a promising strategy to combat the bacterial infections.

Premature drug release in chemotherapy and hypoxic conditions in photodynamic therapy (PDT) are perplexing problems in tumor treatment. Thus, it is of great significance to develop the novel therapeutic system with controllable drug release and effective oxygen generation. Herein, a pH-responsive oxygen self-sufficient smart nanoplatform (named DHCCC), integrating hollow mesoporous silica nanoparticles (HMSNs), chitosan (CS), doxorubicin hydrochloride (DOX), chlorin e6 (Ce6) and catalase (CAT), is fabricated to enhance the tumor therapeutic efficacy efficiently through avoiding premature drug release and mitigating hypoxia of tumor microenvironment (TME). The drug DOX can be efficiently loaded into the HMSNs with large cavity and be controllable released because of the pH responsiveness of CS to the weak acidic TME, thereby elevating the chemotherapy efficacy. Meanwhile, CAT can catalyze the decomposition of endogenous hydrogen peroxide in situ generating oxygen to alleviate the hypoxia and enhance the PDT efficiency considerably. In vitro and in vivo results demonstrate that the combined chemo-photodynamic therapy based on the DHCCC nanoplatform exerts more effective antitumor efficacy than chemotherapy or PDT alone. The current study provides a promising inspiration to construct the pH-responsive oxygen self-sufficient smart nanomedicine with potentials to prevent premature drug leakage and overcome hypoxia for efficient tumor therapy.
