AA/MA/AMA copolymer and chitosan as self-gelling powders for rapid hemostasis and robust tissue adhesion

Wan Peng,Yefeng Jiang,Yang Zhang,Youjin Lai,Yiming Zhu,Huixia Wang,Zilin Kan,Pingsheng Liu

Bleeding control by traditional hemostatic materials poses a risk of continuing oozing bleeding, while existing self-gelling hemostatic powders still face challenges in simultaneously achieving robust wound sealing and effectively accelerating blood coagulation. Herein, we reported acrylic acid/methacrylic acid/2-aminoethyl methacrylate copolymer (PAMA) and chitosan (CS) as a new type of self-gelling composite powder (PAMA-CS) for acute bleeding wound sealing with reduced blood leakage. Upon rapid blood absorption, PAMA-CS powders could quickly form an adhesive hydrogel that strongly binds to bleeding tissues, exhibiting high burst pressure tolerance (up to 452.6 mmHg) for wound sealing to prevent bleeding. The incorporation of chitosan endowed significantly enhanced liquid uptake ability and accelerated blood coagulation (clot index of 77.92 % within 30 s) to the PAMA-CS powder. Based on the synergistic effect of tight adhesive sealing and accelerating intrinsic blood coagulation, PAMA-CS powders could achieve effective hemostasis within 20 s for acute bleeding liver and stomach on rats, outperforming commercial hemostatic chitosan powders. Combined with rapid hemostasis, tight & stable wound sealing, and favorable biodegradation, PAMA-CS powders may serve as a promising hemostatic candidate for emergency bleeding control and wound closure.

Ultrafast self-gelling powder mediates robust wet adhesion to promote healing of gastrointestinal perforations

Xin Peng,Xianfeng Xia,Xiayi Xu,Xuefeng Yang,Boguang Yang,Pengchao Zhao,Weihao Yuan,Philip Wai Yan Chiu,Liming Bian

Achieving strong adhesion of bioadhesives on wet tissues remains a challenge and an acute clinical demand because of the interfering interfacial water and limited adhesive-tissue interactions. Here we report a self-gelling and adhesive polyethyleneimine and polyacrylic acid (PEI/PAA) powder, which can absorb interfacial water to form a physically cross-linked hydrogel in situ within 2 seconds due to strong physical interactions between the polymers. Furthermore, the physically cross-linked polymers can diffuse into the substrate polymeric network to enhance wet adhesion. Superficial deposition of PEI/PAA powder can effectively seal damaged porcine stomach and intestine despite excessive mechanical challenges and tissue surface irregularities. We further demonstrate PEI/PAA powder as an effective sealant to enhance the treatment outcomes of gastric perforation in a rat model. The strong wet adhesion, excellent cytocompatibility, adaptability to fit complex sites, and easy synthesis of PEI/PAA powder make it a promising bioadhesive for numerous biomedical applications.

A Self-Gelling Powder Based on Polyacrylic Acid/Polyethyleneimine/Polyethylene Glycol for High-Performance Hemostasis and Antibacterial Activity

Jia Li,Shu Li,Aozhen Zhong,Jun Xing,Ling Li,Cai Wang,Min Zheng

Powder-based hemostatic materials have offered unprecedented opportunities for the effective sealing and repair of irregularly shaped wounds and high-pressure, noncompressible arterial bleeding wounds caused by surgeries, traffic accidents, and wartime injuries. However, inadequate adhesion to bleeding wounds and poor hemostasis in biological tissues remains challenging. Herein, we report a self-gelling hemostatic powder based on polyacrylic acid/polyethyleneimine/polyethylene glycol (named PPG) for rapid hemostasis and effective antibacterial ability. When deposited on bleeding wounds, PPG powder can absorb interfacial liquid and rapidly swell into a physically cross-linked hydrogel in situ within 2 s to form a pressure-resistant physical barrier. Furthermore, the in vivo and in vitro results indicate that, as an effective sealant, the PPG powder possesses ease of use, excellent hemocompatibility, strong antibacterial abilities, and superior blood clotting abilities. The effective hemostatic sealing capability of the PPG powder is demonstrated in a variety of injury models in rats and rabbits. All of these factors show that, with its superior wound treatment abilities, PPG powder is a profound biomaterial for surgical applications.

Immobilization of polyoxometalates via in-situ protonation and self-gelation of PEG-b-PDMAEMA-b-PTEPM triblock copolymer and its application in selective oxidation

Jing Yan,Jianghao Yao,Yinghui Hu,Diping Huang,Dongdong Yao,Fangzhou Wu,Qiuyu Zhang,Yi Yan

To enrich the building block and function as well as the preparation methodology of metallopolymer system, functioned inorganic polyoxometalate clusters were introduced by using their intrinsic acidity induced multiple interactions with triblock copolymers . In the designed triblock copolymer PEG- b -PDMAEMA- b -PTEPM, the POMs clusters can be immobilized through in-situ protonation of the PDMAEMA block induced electrostatic interaction and the self-gelation of the PTEPM block, while the PEG block can improve the water solubility and dispersion of the final polyoxometalate-based hybrid catalyst (PHC). The selective oxidation activity of PHC is comparable with the homogeneous POMs catalyst as demonstrated by the model reaction of the selective oxidation of methyl phenyl sulfide (MPS) to methyl phenyl sulfoxide (MPSO). By optimizing the ratio between PHC and oxidant H 2 O 2 , the selectivity for MPSO can achieve as high as 99% with conversion above 95%. Furthermore, the multiple interactions between POMs and the polymer substrate also enhance the stability of PHC and enable the recycling of PHC. Therefore, the methodology of in-situ protonation and self-gelation by the acidity of POMs provides a facile and general strategy to prepare robust POMs-based metallopolymers, paving a new avenue towards cluster-based materials and devices.

Simple component self-gelling powders as multifunctional tissue adhesives for sutureless wound healing

Wan Peng,Youjin Lai,Jin Fan,Chuanchao Dai,Jian Shen,Pingsheng Liu

For decades, bio-adhesives have gained great attention due to their potency to replace sutures. However, facile fabrication of asymmetric bioadhesives simultaneously with robust adhesion to non-planar wet tissues, effective resistance to postoperative tissue adhesion and bacterial associated infection remains a great challenge. Here, we report a simple component & ready-to-use self-gelling adhesive powder (copolymer of acrylic acid and 2-aminoethyl methacrylate, AMA) combined with an anti-adhesive powder (a mixture of zwitterionic sulfobetaine copolymer and an acrylic homopolymer, AASB) as the multifunctional tissue adhesive for sutureless wound closure & healing. The adhesive powder is capable of forming a physically cross-linked hydrogel within 5 s by absorbing water and strongly sticking to diverse non-planar wet tissues (adhesion strength up to 63 kPa, bursting pressure up to 350 mmHg within 24 h) with a broad-spectrum bactericidal efficiency (99.4 %, 96.2 % and 99.9 % against S. aureus , E. coli and MRSA). The integrated AASB hydrogel layer can effectively prevent cell/tissue adhesion and suppress hemolysis ratio & inflammation of the adhesive. Moreover, the AMA/AASB Janus hydrogel adhesive exhibited durable adhesive/anti-adhesive capability (at least for 30 days) and good biodegradability (within 60 days) in vivo , comparable sealing & repairing of gastric defects but superior anti-postoperative tissue adhesion to the clinic used fibrin glue. Such a simple-component, ready-to-use and multifunctional powder based adhesive offer new opportunities for minimal invasive sutureless tissue sealing in many clinical applications.

Superwettable calcium ion exchanged carboxymethyl cellulose powder with self-gelation, tissue adhesion and bioabsorption for effective hemorrhage control

Kaitao Yao,Shuyang Li,Xuzhou Zheng,Qian Zhang,Jinhui Liu,Cheng Liang,Ke Duan,Junwu Ye,Yiran Yin,Xingtao Chen

Water-soluble polymers can be used to develop self-gelling hemostatic powders, and powder wettability potentially plays the key role in determining their hemostatic efficacy. Herein, a superwettable calcium ion exchanged carboxymethyl cellulose (Ca-ex-CMC) powder is reported for effective hemorrhage control. Despite being water-soluble, Ca-ex-CMC powder can form a physically crosslinked hydrogel upon contact with water (self-gelation), and the rate of water permeating the hydrogel to achieve dissolution is extremely slow. With increased calcium content, Ca-ex-CMC powder shows not only improved procoagulant activity, but enhanced wettability which can facilitate the fusion of the powders with water, resulting in promoted water uptake and in-situ formation of strong hydrogel barriers with enhanced tissue adhesion. Applying Ca-ex-CMC powder to non-compressible (liver and tail) and massive (artery) hemorrhage wounds in rat and rabbit models can stanch bleeding instantly, because of complete sealing of the bleeding sites by enhanced in-situ self-gelation. Moreover, owing to water-solubility, Ca-ex-CMC powder shows outstanding bioabsorbability and histocompatibility after implantation into rabbit liver and gluteus. These findings not only support Ca-ex-CMC powder as a promising hemostatic agent for extensive application but identify the critical role of wettability in promoting hemostasis for self-gelling powders.

Shape-adapted self-gelation hydrogel powder for high-performance hemostasis and wound healing

Zhaoyang Wang,Tong Lyu,Qianlin Xie,Yusongjia Zhang,Haotian Sun,Yucen Wan,Ye Tian

In the battlefield, accidents, first aid, and hospital environments, uncontrollable bleeding caused by severe wound infections pose a serious risk of death and high costs. However, performing ultrafast hemostasis without any external conditions remains challenging. In this paper, polyacrylic acid, polyacrylamide, tea polyphenols, and Yunnan Baiyao (PAA/PAAm/T/YNBY, PPTY) are used to prepare self-gelling powder with shape adaptability. When the mass fraction of PAA and PAAm is 10 wt% and 8 wt%, the hydrogel can form the most intermolecular hydrogen bonds, exhibiting the best adhesion ability and mechanical strength. When the amount of YNBY was 0.1 wt%, the hydrogel had the most suitable cellular compatibility and hemostatic ability. At this time, the hydrogel powder can achieve the fastest self-gelling (< 3 s), the strongest adhesion (interface adhesion > 499.4 N m  −   1 ), and the fastest hemostatic speed (< 30 s). With the addition of a small amount of YNBY (0.1 wt%), the hemostatic efficiency of the hydrogel can be greatly improved. The combination of hydrogel and YNBY not only prolongs the duration of YNBY ingredient efficacy but also enhances its biocompatibility. This easy-to-use but effective hydrogel can stop bleeding, promote wound healing in emergencies, and has opportunities in functional materials, tissue engineering, biomedical engineering, and clinical medicine.

Self-gelling, tunable adhesion, antibacterial and biocompatible quaternized cellulose/tannic acid/polyethylene glycol/montmorillonite composite powder for quick hemostasis

Xuanyu Liu,Yinchun Hu,Junjie Hu,Miaomiao Bai,Min Kang,Yan Wei,Di Huang

Hemostatic powders are widely used in incompressible or irregularly shaped bleeding wounds, but traditional hemostatic powders exhibit low adhesion, unsatisfactory hemostatic effect, limited infection control, and are not suitable for clinical or emergency situations. This study developed a novel self-gelling hemostatic powder (QTPM) consisting of quaternized cellulose (QC)/ tannic acid (TA)/ polyethylene glycol (PEG)/ montmorillonite (MMT). QTPM could absorb interfacial liquid hydrating to a stable hydrogel which form a switchable adhesion to tissues. Moreover, QTPM exhibits excellent antibacterial property by the synergistic effect of QC and TA. Furthermore, QTPM directly activate intrinsic and extrinsic coagulation hemostatic pathways to enhance hemostasis, and it concentrate coagulation factors. In vivo hemostasis study results show that QTPM significantly accelerated hemostasis and reduced blood loss compared with the blank group (>75 % reduction in hemostatic time, >85 % reduction in blood loss) in liver bleeding model (hemostasis time of 71.67 ± 7.09 s, blood loss of 19.23 ± 2.60 mg) and tail amputation model (hemostasis time of 91.03 ± 12.05 s, blood loss of 15.24 ± 1.77 mg). Therefore, the advantages of QTPM including rapid and effective hemostasis, easy usage, easy storability and adaptability make it a potential biomaterial for rapid hemostasis direction in the clinical setting.

A self-gelling powder based on polyacrylic acid/polyacrylamide/quaternate chitosan for rapid hemostasis

Zhouying Tan,Xi Li,Chaojie Yu,Mengmeng Yao,Zhongming Zhao,Bingyan Guo,Lei Liang,Yuping Wei,Fanglian Yao,Hong Zhang,Junjie Li

In order to improve the hemostatic effect of the hemostatic dressing for non-compressible wounds, unknown bleeding points and irregularly shaped wounds, a self-gelling hemostasis powder based on polyacrylic acid/polyacrylamide/quaternate chitosan (PAA/PAM/QCS) is prepared in this study. When in contact with water, the PAA/PAM/QCS can fuse and rapidly form a stable hydrogel in a short time (< 0.25 min). The PAA/PAM ratio is the main parameter that modulates the formation of the self-gel. The PAA/PAM self-gel can be formed only when the PAA/PAM ratio is 5:5, and the introduction of QCS does not influence the self-gelling behaviors and hydrogel stability. Moreover, the PAA/PAM/QCS self-gel shows good adhesive properties on wet tissue surfaces. In addition, the introduction of QCS improves the antibacterial activity of the self-gelling hemostasis powder. Furthermore, the prepared PAA/PAM/QCS powder can rapidly adsorb lots of blood, aggregate blood cells and platelets. The hemostatic results in vivo show that PAA/PAM/QCS powder is superior to the control group and commercial product groups (chitosan powder) with performance similar to hemostatic zeolite in terms of the amount of bleeding and hemostatic time. Therefore, the PAA/PAM/QCS self-gelling powder shows great application prospects for rapid hemostasis.

A Self-Gelling Powder Directly Co-Assembled by Natural Small Molecules for Traumatic Brain Injury

Shutong Yang,Weikang Luo,Xianwen Song,Quan Chen,Jingjing Liu,Pingping Gan,Chuntai Liu,Teng Li,Gang Xu,Yi Zhang,Jun Zheng,Yang Wang

The prognosis for traumatic brain injury (TBI) depends largely on prompt hemostasis and effective pharmacologic interventions. Natural all-small-molecule self-gelling powder, integrating the advantages of self-assembled small-molecule hydrogels and powders, is expected to provide timely and effective prehospital management of TBI. However, the synthesis and application of natural all-small-molecule self-gelling powder is still uncharted territory. In this study, an all-small-molecule co-assembled MGF-H 3 BO 3 -RUT (MBR) self-gelling powder is fabricated through the co-assembly of mangiferin (MGF) and rutin (RUT) in H 3 BO 3 /NaOH aqueous solution. Both compounds can bind with boric acid, leading to co-assembling into hydrogels through hydrogen-bonding interactions and π – π stacking. MBR self-gelling powder is then obtained by drying the as-prepared hydrogels, thus integrating hemostasis and pharmacodynamics into one. Remarkably, it displays robust regeneration capabilities, while retaining excellent self-healing properties and injectability after drying-hydration cycles. Moreover, MBR self-gelling powder not only achieves rapid effective hemostasis but also attenuates conspicuously cerebral edema and inflammatory response after TBI by in situ spraying, exhibiting notable neuroprotective effects without discernible toxic side effects. This study provides a novel assembly strategy and application form for self-assembled gel materials originating from natural small molecules, offering promising avenues for the treatment of TBI in the acute phase.

Strong tissue adhesive polyelectrolyte complex powders based on low molecular weight chitosan for acute hemorrhage control

Mi Wu,Shun Mao,Xu Liu,Yunen Liu,Peifang Cong,Jianhua Lv,Huaqin Tian,Yan Zhao

Self-gelling and bioadhesive powders offered promising effective hemostats to suit irregularly shaped, complex and non-compressible wounds for clinical applications. In the current study, chitosan based polyelectrolyte complex coacervate were simply prepared by mixing high concentrations (10 %) of low molecular weight chitosan (CS) and polyacrylic acid (PAA) solutions. Obtained by lyophilization, the physical cross-linked polyelectrolyte complex powders would form a gel within 5 s upon hydration, which demonstrated excellent mechanical properties, significant antibacterial activities, strong and lasting adhesion on wet tissues in physiological environment. In vitro blood clotting assays showed that the CS/PAA powders could remarkably aggregate blood cells and accelerate blood clotting process. As studied by diverse hemorrhage models, including rat tail, liver and heart injuries and dog incision, CS/PAA powders significantly facilitated the decrease of blood loss as well as hemostatic time by creating robust physical barriers and promoting blood clot formation on the bleeding sites. These outstanding properties in terms of easy preparation, rapid self-gelling, strong wet adhesion, effective hemostasis and shape-adaptability endowed CS/PAA polyelectrolyte complex powders with great potential in managing acute hemorrhage of non-compressible trauma.

Ultrafast Self-Gelling, Adhesive, Anti-Bacterial Coacervate-Based Powders for Enhanced Hemostasis and Wound Healing

Hui Zhou,Bin Kong,Yi Cheng,Si Meng,Haifeng Dong,Cheng Qi,Tiantian Kong,Yuanjin Zhao,Zhou Liu

Uncontrolled hemorrhage, especially in non-compressible and deep wounds, remains a critical issue in emergency and surgical care. Existing hemostatic powders often lack rapid gelation, mechanical robustness, and adequate adherence, increasing the risk of rebleeding under high-pressure blood flow. To address these limitations, PQPP, a novel self-gelling hemostatic material composed of polyacrylamide/quaternized chitosan coacervates and polydopamine nanoparticles is developed. PQPP can rapidly absorb blood within 2 s, undergoes in situ gelation, and forms a robust adhesive hydrogel to effectively seal wounds. Its efficacy stems from the electrostatic adsorption and catechol functional groups of polydopamine nanoparticles. Importantly, PQPP exhibits high burst pressure resistance, excellent blood cell aggregation capability, outstanding biocompatibility, and antibacterial properties. Comprehensive in vitro and in vivo studies, including cytotoxicity and blood compatibility tests, as well as trials in mouse liver, heart, and vascular injury models, demonstrate PQPP's superior hemostatic performance under high-pressure conditions without causing inflammation. With its rapid gelation, robust adhesion, and mechanical integrity, PQPP represents a promising hemostatic material for immediate wound management in surgical and emergency applications.
