4D Printing of Self-Folding Hydrogel Tubes for Potential Tissue Engineering Applications

Yu-Dong Zhao,Jia-Hui Lai,Min Wang

In recent years, 4D printing has gained increasing attention in the tissue engineering field since this advanced manufacturing platform can create stimulus-responsive structures, which can change their shapes, functions, and/or properties when appropriate external stimulus/stimuli is/are applied. A number of hydrogels with swellable/shrinkable abilities have been explored for 4D printing to fabricate different shape-morphing structures for tissue engineering. Among them, gelatin methacryloyl (GelMA) has been 4D printed, which can self-fold into microtubular structures. Currently, the self-folding ability of 4D printed GelMA hydrogels is mainly based on the different cross-linking degrees (which control and govern the swelling degrees) across the thickness of hydrogels. However, this strategy alone can only form self-folding GelMA tubes with diameters at the micrometer level and cannot create self-folding GelMA tubes with diameters at the millimeter level, which is mainly due to the insufficient internal force generated in 4D printed GelMA hydrogels when they are exposed to water. To overcome this limitation, this study has investigated a new strategy to fabricate self-folding GelMA tubes with large diameters at the millimeter level for tissue engineering applications. The new strategy introduced a cross-linking degree gradient across the GelMA plane in addition to its thickness by printing a second layer of strips on the first 4D printed GelMA film. In the aqueous environment, under the current fabrication condition, such bilayer GelMA hydrogels could self-fold into tubes of larger diameters up to 6 mm. The in vitro release behavior of heparin incorporated into the 4D printed GelMA was also studied. It was shown that heparin release could be controlled by the GelMA concentration and heparin content in 4D printed GelMA. The 4D printed GelMA hydrogels with the improved self-folding ability and controlled release of a drug are promising for targeted tissue engineering applications.

Integrated Generation and Manipulation of Microcapsules via Optically Induced Dielectrophoresis

Wenguang Yang,Wenhao Wang,Xiangyu Teng,Zezheng Qiao,Zhixing Ge,Zheng Yuan

Microcapsules have a wide range of applications in the medical field, but their fabrication process is often complicated and inefficient. This study presents a method that uses polystyrene (PS) microspheres and polyethylene glycol diacrylate (PEGDA) to achieve both particle positioning and polymerization in a single step through optically induced dielectrophoresis (ODEP), enabling the fabrication of microcapsules with different shapes. The maximum speed and dielectrophoretic force of 1 and 10 μm PS microspheres are investigated in solutions with different PEGDA concentrations and frequencies. After the PS microspheres are collected, the polymerization reaction of PEGDA is completed in the original solution. In addition, by extending the irradiation time of the light source, the self-folding of the two-dimensional capsule is induced, thereby producing a capsule with particles for a tubular three-dimensional shape. This innovation holds promise for various applications in the targeted treatment of diseases and medical diagnosis.

Thermally resistant thermadapt shape memory crosslinked polymers based on silyl ether dynamic covalent linkages for self-folding and self-deployable smart 3D structures

Zhenjie Ding,Li Yuan,Guozheng Liang,Aijuan Gu

Thermadapt shape memory polymers with high glass transition temperature and high tensile strength are greatly important for developing high performance self-folding and self-deployable smart 3D structures. Herein, starting from an epoxy resin, ethanolamine, 4,4′-diaminodiphenylmethane and 3-isocyanatopropyltrimethoxysilane, new thermadapt shape memory polymers (EPSis) with highly tunable crosslinked networks and silyl ether dynamic covalent linkages were synthesized. The crosslinked structures and comprehensive performances of EPSis were investigated systematically. The results show that EPSis not only exhibit high glass transition temperatures, which can be adjusted from 118.1 to 156.4 °C, about 38.1–76.4 °C higher than those reported in the literature, but also have high tensile strengths. In addition, EPSis show good thermadapt shape memory behaviours (the shape retention ratio is 80.5–86.3%, the shape fixity ratio is 97.1–98.9%, and the shape recovery ratio is 95.6–99.8%), and they can convert from planar films into various shapes such as a “six-claw stool”, “wave-like rocking chair”, “windmill” and “paper airplane” under heating stimulation, proving that EPSis have great potential for developing high performance self-folding and self-deployable 3D structures. The mechanism study proves that these excellent properties originate from the crosslinked networks based on dynamic silyl ether bonds with both robust toughness and thermal stability, and the topological rearrangement of the network induced by dynamic exchange of silyl ether bonds is responsible for the outstanding thermadapt behaviour. This study provides a unique strategy for developing high performance smart 3D structures.

Highly sensitive humidity-driven actuators based on metal–organic frameworks incorporating thermoplastic polyurethane with gradient polymer distribution

Yi He,Jiayu Guo,Xiazhen Yang,Bing Guo,Hangyan Shen

Ambient humidity plays an important role in the fields of industrial and agricultural production, food and drug storage, climate monitoring, and maintenance of precision instruments. To sense and control humidity, humidity-responsive actuators that mimick humidity responsive behavior existing in nature, have attracted intense attention. The most common and important class of humidity actuators is active bilayer structures. However, such bilayer structures generally show weak interfacial adhesion, tending to delaminate during frequent bending and restoration cycles. In this work, to address this problem, a novel monolayer humidity-driven actuator with no adhesive issue is developed by integrating the swellable metal–organic frameworks (MIL-88A) into thermoplastic polyurethane films. The proposed actuators display excellent humidity response that under the conditions of relative humidity simulated with saturated salt solution, the MIL-88A/polyurethane composite films show good self-folding response and stability for recycling use. In addition, a deep insight into the self-folding of the composite films is also provided and a new response mechanism is proposed. In this case, the results show that both the preparation method and response properties of the humidity actuators are improved. Therefore, it suggests a new promising way to develop and design flexible humidity actuators.

Protein fibers with self-recoverable mechanical properties via dynamic imine chemistry

Sun Jing,He Haonan,Zhao Kelu,Cheng Wenhao,Li Yuanxin,Zhang Peng,Wan Sikang,Liu Yawei,Wang Mengyao,Li Ming,Wei Zheng,Li Bo,Zhang Yi,Li Cong,Sun Yao,Shen Jianlei,Li Jingjing,Wang Fan,Ma Chao,Tian Yang,Su Juanjuan,Chen Dong,Fan Chunhai,Zhang Hongjie,Liu Kai

The manipulation of internal interactions at the molecular level within biological fibers is of particular importance but challenging, severely limiting their tunability in macroscopic performances and applications. It thus becomes imperative to explore new approaches to enhance biological fibers’ stability and environmental tolerance and to impart them with diverse functionalities, such as mechanical recoverability and stimulus-triggered responses. Herein, we develop a dynamic imine fiber chemistry (DIFC) approach to engineer molecular interactions to fabricate strong and tough protein fibers with recoverability and actuating behaviors. The resulting DIF fibers exhibit extraordinary mechanical performances, outperforming many recombinant silks and synthetic polymer fibers. Remarkably, impaired DIF fibers caused by fatigue or strong acid treatment are quickly recovered in water directed by the DIFC strategy. Reproducible mechanical performance is thus observed. The DIF fibers also exhibit exotic mechanical stability at extreme temperatures (e.g., −196 °C and 150 °C). When triggered by humidity, the DIFC endows the protein fibers with diverse actuation behaviors, such as self-folding, self-stretching, and self-contracting. Therefore, the established DIFC represents an alternative strategy to strengthen biological fibers and may pave the way for their high-tech applications.
