Diabetic wounds, characterized by excessive exudate accumulation and dysregulated inflammation, pose a global healthcare challenge due to their persistent non-healing nature. Traditional therapies often fail to simultaneously address three critical healing requirements: precise exudate management, coordinated inflammation-immune modulation, and pro-regenerative microenvironment establishment. Here, we present an innovative self-pumping Janus fibrous dressing integrating curcumin-loaded polycaprolactone nanofibers with cotton gauze. Its gradient architecture enables spontaneous exudate drainage and dynamic microenvironment modulation, establishing an optimal milieu for accelerated wound healing. Comprehensive in vitro investigations validate the dressing’s multifunctionality, including sensitive colorimetric pH response, antibacterial properties (>99 % inhibition against S. aureus ), and antioxidant capabilities (>90 % DPPH radical scavenging). In vivo studies in a diabetic mice model demonstrate the dressing’s superior wound healing efficacy, with ∼47 % reduction in wound edge maceration, mitigated inflammatory responses, and enhanced angiogenesis compared to commercial cotton dressings. This novel approach provides a promising foundation for the development of advanced wound dressings tailored for the management and healing of chronic wounds.

The directional management of exudate drainage and drug delivery assumes paramount importance in the realm of precision therapy for diabetic wounds. Drawing inspiration from the water-collecting and transporting abilities of cactus spines and the water-drinking behavior of spiny lizards, this work designed a robust self-pumping medical microneedle array composed of cellulose nanofibril-reinforced microneedles by non-covalent cooperative bond networks and a Janus backing layer with bidirectional fluid transport capability. Microneedles reinforced with nanofiber exhibit exceptional photothermal conversion efficiency while maintaining sufficient mechanical strength and toughness for epidermal penetration without deformation. The bidirectional Janus backing encompasses a drug delivery zone and a UV-reversibly induced exudate pumping/wettability maintenance zone, enabling staggered transport of the drug and exudates to guarantee long-term efficacy in sustained drug release, controlled exudate removal and microenvironment wettability maintenance. Crucially, it effectively combines broad-spectrum antibacterial, biofilm elimination, antioxidant, biocompatibility and anti-inflammatory functions with edema reduction in diabetic wounds, significantly accelerating the healing process. These findings present an innovative strategy for designing precision therapeutic materials with multimodal wound management capabilities.

The application of solar interfacial evaporation technology in clean water production has garnered increasing attention in recent years, with thin-film evaporation being considered one of the most efficacious strategies to address the growing demand for high evaporation rates at moderate temperatures. This investigation presents the design of a Janus nanofiber film based on a self-pumping effect, comprising a hydrophobic layer (TPU) and a hydrophilic layer (PVA-(ε-PL)@MXene). This structure creates a self-pumping effect that promotes unidirectional transfer of liquids, effectively increasing water transfer efficiency and evaporation, achieving a maximum evaporation rate of 1.56 kg m–2 h–1 while maintaining 1.52 kg m–2 h–1 after multiple cycles. This Janus nanofiber film exhibits high performance in the 200–2500 nm range, demonstrating strong light absorption with an absorption efficiency of 87.69%, and rapidly increasing from room temperature to 57.8 °C in 3 min under 1 kW m–2. Furthermore, TPU/PVA-(ε-PL)@MXene exhibited substantial antimicrobial activity against E. coli (89.77%) and S. aureus (53.96%), attributed to the inhibitory effect of ε-PL on the biosynthesis of proteins and enzymatic biomolecules.

Digital bioassays are powerful methods to detect rare analytes from complex mixtures and study the temporal processes of individual entities within biological systems. In digital bioassays, a crucial first step is the discretization of samples into a large number of identical independent partitions. Here, we developed a rapid and facile sample partitioning method for versatile digital bioassays. This method is based on a detachable self-digitization (DSD) chip which couples a reversible assembly configuration and a predegassing-based self-pumping mechanism to achieve an easy, fast, and large-scale sample partitioning. The DSD chip consists of a channel layer used for loading the sample and a microwell layer used for holding the sample partitions. Benefitting from its detachability, the chip avoids a lengthy oil flushing process used to remove the excess sample in loading channels and can compartmentalize a sample into more than 100,000 wells of picoliter volume with densities up to 14,000 wells/cm 2 in less than 30 s. We also demonstrated the utility of the proposed method by applying it to digital PCR and digital microbial assays.

Optimized management of wound exudate is pivotal in modulating the wound healing microenvironment and facilitating tissue regeneration. Conventional dressings, however, are constrained by limited absorption capacity and a slow drainage rate, frequently resulting in excessive exudate accumulation that hinders the healing process. In this study, we developed hydrophilic kapok fiber gauze (H-KG) and Janus kapok fiber gauze (J-KG) through a sustainable and scalable hydroentanglement forming process combined with chemical modification, leveraging the unique hollow tubular structure of kapok fibers. H-KG and J-KG achieved rapid absorption, high capacity storage, and efficient evaporation through synergistic wettability modulation and capillary action-enhanced liquid transport within the hollow structure. The kapok fiber-based gauze demonstrated remarkable sustainable liquid self-pumping capabilities, exhibiting a saturation absorption rate of approximately 1346% with complete liquid evaporation achieved within 1 h. In vivo experiments revealed that both H-KG and J-KG significantly mitigated wound exudate accumulation compared with cotton gauze (CG), thereby accelerating wound healing. Quantitative analysis showed that on day 12 post-treatment, the H-KG and J-KG exhibited residual wound areas measuring 48.23% and 34.41%, respectively, of CG. This work elucidated the mechanistic role of hollow fiber in exudate management while proposing an innovative wound dressing with dynamic fluid regulation capabilities.

Excessive exudate secreted from diabetic wounds often results in skin overhydration, severe infections, and secondary damage upon dressing changes. However, conventional wound dressings are difficult to synchronously realize the non-maceration of wound sites and rapid exudate transport due to their random porous structure. Herein, a self-pumping Janus hydrogel with aligned channels (JHA) composed of hydrophilic poly (ethylene glycol) diacrylate (PEGDA) hydrogel layer and hydrophobic polyurethane (PU)/graphene oxide (GO)/polytetrafluoroethylene (PTFE) layer is designed to rapidly export exudate and accelerate diabetic wound healing. In the design, the ice-templating process endows the hydrophilic hydrogel layer with superior liquid transport ability and mechanical strength due to the formation of aligned channel structure. The hydrophobic layer with controlled thickness functions as an effective barrier to prevent exudate from wetting the skin surface. Experiments in diabetic rat model show that JHA can significantly promote re-epithelialization and collagen deposition, shorten the inflammation phase, and accelerate wound healing. This unique JHA dressing may have great potential for real-life usage in clinical patients.
