Bioinspired micro/nanomotor with visible light energy–dependent forward, reverse, reciprocating, and spinning schooling motion

Jintao Tong,Dalei Wang,Ye Liu,Xin Lou,Jiwei Jiang,Bin Dong,Renfeng Dong,Mingcheng Yang

In nature, microorganisms could sense the intensity of the incident visible light and exhibit bidirectional (positive or negative) phototaxis. However, it is still challenging to achieve the similar biomimetic phototaxis for the artificial micro/nanomotor (MNM) counterparts with the size from a few nanometers to a few micrometers. In this work, we report a fuel-free carbon nitride (C3N4)/polypyrrole nanoparticle (PPyNP)-based smart MNM operating in water, whose behavior resembles that of the phototactic microorganism. The MNM moves toward the visible light source under low illumination and away from it under high irradiation, which relies on the competitive interplay between the light-induced self-diffusiophoresis and self-thermophoresis mechanisms concurrently integrated into the MNM. Interestingly, the competition between these two mechanisms leads to a collective bidirectional phototaxis of an ensemble of MNMs under uniform illuminations and a spinning schooling behavior under a nonuniform light, both of which can be finely controllable by visible light energy. Our results provide important insights into the design of the artificial counterpart of the phototactic microorganism with sophisticated motion behaviors for diverse applications.

GSH-responsive Pt-based nanomotor with improved doxorubicin delivery for synergistic osteosarcoma chemotherapy

Sheng Xu,Ziwei Hu,Weihao Zheng,Chaozhen Qin,Xiaoyu Bai,Qinghua Yang,Tao Zeng,Dandan Mo,Bo Zhou,Chun Lu,Xiaomin Chen,Biying Tan,Jinmin Zhao,Li Zheng

Osteosarcoma (OS), a highly malignant primary tumor, poses significant threats. Chemotherapy remains the main treatment approach but is limited by low drug bioavailability, poor permeability, and notable side effects. Herein, a near-infrared light (NIR)-driven and GSH-responsive poly(ethylene glycol)-SS-polystyrene-doxorubicin and platinum nanoparticles (PSPDP) nanomotor, wherein disulfide bonds served as GSH sponsors and platinum nanoparticles as producers of reactive oxygen species (ROS) to induce cell apoptosis, combined with NIR-driven propulsion to enhance the inhibitory effect of encapsulated doxorubicin (DOX). The results demonstrated that the PSPDP nanomotor can be effectively driven due to its good photothermal properties, with its movement speed increased 2.10 times under NIR laser exposure. Additionally, the efficiency of DOX release increased with the increase in GSH concentration, demonstrating favorable GSH responsiveness. Pt-NPs also exhibited good photothermal properties, enabling self-thermophoresis to drive. Minimal cytotoxic effects of PSPDP were observed on a series of cell lines compared with DOX solution and Pt-NPs. Notably, the Pt-NPs generated a significant amount of ROS, synergistically enhancing the therapeutic effect of DOX, as evidenced by a 5.53-fold increase in OS cell growth inhibition and evident osteosarcoma growth inhibition in the nude mice model. Thus, the NIR-driven, localized, and low-toxic nanomotor may offer a promising therapeutic strategy for OS intervention. Statement of significance Enhancing drug penetration efficiency and developing delivery systems that respond to the tumor microenvironment to release drugs are effective strategies for treating osteosarcoma (OS). Here, a near-infrared (NIR) light-driven and glutathione (GSH)-responsive nanomotor, integrating poly(ethylene glycol)-SS-polystyrene-doxorubicin and platinum nanoparticles (PSPDP), was produced and used for OS treatment. This PSPDP nanomotor exhibits significant advancements in photothermal activation and self-thermophoresis, enabling a 2.10-fold increase in movement speed under NIR exposure. Such enhanced motility improves the localized delivery and controlled release of doxorubicin, thus increasing drug bioavailability and minimizing systemic toxicity. Additionally, the nanomotor's ability to generate reactive oxygen species significantly amplifies its therapeutic impact, evidenced by a remarkable 5.53-fold increase in tumor growth inhibition. These features make the PSPDP nanomotor a promising candidate for effective and targeted OS treatment strategies.

Bioinspired Jellyfish-like Carbon/Manganese nanomotors with H2O2 and NIR light Dual-propulsion for enhanced tumor penetration and chemodynamic therapy

Yi Xing,Mengyun Zhou,Xiangmei Liu,Minghang Qiao,Liping Zhou,Tailin Xu,Xueji Zhang,Xin Du

Chemodynamic therapy (CDT), which catalyzes the production of highly toxic reactive oxygen species (ROS), has been widely used in tumor therapy in the past five years. However, most of the current nanomaterials for CDT are difficult to penetrate into the tumor effectively due to the lack of motility, resulting in an unsatisfactory CDT effect. Herein, bioinspired jellyfish-like carbon/manganese nanomotors (JCMNs) are designed to achieve hydrogen peroxide (H 2 O 2 ) and NIR light dual-propulsion for the enhanced CDT therapy. Based on the jellyfish-like asymmetric structure, the manganese component can catalyze H 2 O 2 to generate chemical concentration gradient, propelling JCMNs to dynamically generate ROS in cells by self-diffusiophoresis. In addition, the carbon component of JCMNs can absorb near-infrared (NIR) light to produce a temperature gradient, driving JCMNs to enhance tumor penetration by self-thermophoresis. The combination of chemical and NIR light propulsions significantly improves the efficiency of tumor CDT, revealing that it should be a reasonable strategy to promote the development of nanobiomedicine by endowing nanosystems with propulsion capacity.

Light-Driven Biomimetic Nanomotors for Enhanced Photothermal Therapy

Hong Wang,Junbin Gao,Cong Xu,Yuejun Jiang,Meihuan Liu,Hanfeng Qin,Yicheng Ye,Li Zhang,Wanxian Luo,Bin Chen,Lingli Du,Fei Peng,Yingjia Li,Yingfeng Tu

Nanotechnology-based strategy has recently drawn extensive attention for the therapy of malignant tumors due to its distinct strengths in cancer diagnosis and treatment. However, the limited intratumoral permeability of nanoparticles is a major hurdle to achieving the desired effect of cancer treatment. Due to their superior cargo towing and reliable penetrating property, micro-/nanomotors (MNMs) are considered as one of the most potential candidates for the coming generation of drug delivery platforms. Here, near-infrared (NIR)-actuated biomimetic nanomotors (4T1-JPGSs-IND) are fabricated successfully and we demonstrate that 4T1-JPGSs-IND selectively accumulate in homologous tumor regions due to the effective homing ability. Upon laser irradiation, hyperthermia generated by 4T1-JPGSs-IND leads to self-thermophoretic motion and photothermal therapy (PTT) to ablate tumors with a deep depth, thereby improving the photothermal therapeutic effect for cancer management. The developed nanomotor system with multifunctionalities exhibits promising potential in biomedical applications to fight against various diseases.

Light/gas cascade-propelled Janus micromotors that actively overcome sequential and multi-staged biological barriers for precise drug delivery

Xiang Zhou,Xiuhong Huang,Bincheng Wang,Lihui Tan,Yi Zhang,Yanpeng Jiao

Passive diffusion-based drug delivery systems have indeed shown impressive performance to overcome a cascade of biological barriers, but the delivery efficiency is still modest. Although artificial micro/nanomotors are capable of autonomous movement, oriented navigation and seeking target actively represent an alternative and attractive strategy to revolutionize drug delivery patterns potentially, the separated and single propulsion mode of motors still hampers their further biomedical application that needs to overcome successive biological barriers. Herein, we demonstrated a cascaded NIR-light/gas-driven Janus calcium carbonate particle micromotors (JCPMs) to overcome multi-staged biological barriers and achieved active drug delivery. These JCPMs exhibited NIR light-driven thermophoretic motion with precisely steered start/stop switch, speed, and motion trajectory in biological media and displayed CO 2 -bubbles-powered autonomous propulsion in an acidic environment. With the collaborative use of cascaded light/gas double-mode driving, JCPMs displayed a multistage self-adapted strategy to overcome sequential biological barriers during tumor-targeted drug delivery in a cascaded manner, involving of light-driven active tumor cells seeking, facilitated tumor penetration and cellular membranes traversal by virtue of photo-thermomechanical force and self-thermophoretic propulsion, gas-driven lysosomes escape and intracellular drug release successively. Such a strategy guaranteed trajectory-controllable active drug delivery to exert the selective tumor therapeutic effect as well as avoided side effects. We envisioned that the proposed cascaded driving mode of micromotors for traversing biological barriers would boost the delivery efficiency for tumor treatment, which may pave a convenient and versatile way to develop next generation active vehicles in the biomedical field.

Bimetallic Nanozymes-Integrated Parachute-Like Au2Pt@PMO@ICG Janus Nanomotor with Dual Propulsion for Enhanced Tumor Penetration and Synergistic PTT/PDT/CDT Cancer Therapy

Xiaolei Zhang,Yangsai Lyu,Jia Li,Xiaohan Yang,Ziwei Lan,Zhixu Chen

Nanocatalytic therapeutic agents with triple synergistic treatment modes of chemodynamic therapy (CDT), photothermal therapy (PTT) and photodynamic therapy (PDT) are emerging nanomaterials in malignancy treatment. Nevertheless, the passive diffusion and transport of nanomaterials result in poor permeability in tumor lesions, severely affecting the effectiveness of synergistic therapy. Herein, a dual-source-driven parachute-like Au 2 Pt@PMO@ICG Janus nanomotor (APIJNS) is prepared by an interfacial energy-mediated anisotropic growth strategy for PTT-mediated CDT/PDT triple synergistic cancer therapy. Such nanomotor can realize self-thermophoresis drive under the trigger of near-infrared light, which can effectively enhance the active permeability and uptake of the APIJNS in the tumor tissue, thus achieving efficient PTT/PDT/CDT synergistic therapeutic effect. In addition, parachute-like APIJNS exhibits high-efficiency catalase (CAT)-like activity and can catalyze the overexpressed H 2 O 2 in the tumor microenvironment (TME) to generate oxygen (O 2 ), not only alleviating the hypoxia in the tumor lesion, but also converting it into singlet oxygen ( 1 O 2 ) to activate the photosensitizer ICG, achieving PDT. This work provides new ideas for enhancing triple synergistic cancer therapy by improving tumor permeability with dual-drive Janus nanomotors based on dual noble metal nanozymes.

Site-Selective Superassembly of a Multilevel Asymmetric Nanomotor with Wavelength-Modulated Propulsion Mechanisms

Tianyi Liu,Miao Yan,Shan Zhou,Qirui Liang,Yanjun He,Xin Zhang,Hui Zeng,Jian Liu,Biao Kong

Micro-/nanomotors with advanced motion manipulation have recently received mounting interest; however, research focusing on the motion regulation strategies is still limited, as the simple construction and composition of micro-/nanomotors restrict the functionality. Herein, a multifunctional TiO2-SiO2-mesoporous carbon nanomotor is synthesized via an interfacial superassembly strategy. This nanomotor shows an asymmetric matchstick-like structure, with a head composed of TiO2 and a tail composed of SiO2. Mesoporous carbon is selectively grown on the surface of TiO2 through surface-charge-mediated assembly. The spatially anisotropic distribution of the photocatalytic TiO2 domain and photothermal carbon domain enables multichannel control of the motion, where the speed can be regulated by energy input and the directionality can be regulated by wavelength. Upon UV irradiation, the nanomotor exhibits a head-leading self-diffusiophoretic motion, while upon NIR irradiation, the nanomotor exhibits a tail-leading self-thermophoretic motion. As a proof-of-concept, this mechanism-switchable nanomotor is employed in wavelength-regulated targeted cargo delivery on a microfluidic chip. From an applied point of view, this nanomotor holds potential in biomedical applications such as active drug delivery and phototherapy. From a fundamental point of view, this research can provide insight into the relationship between the nanostructures, propulsion mechanisms, and motion performance.
