Self-Cascade of the Cu/Ce-MOF-808 Nanozyme for One-Step Colorimetric Sensing

Yuan Qin,Huimei Liang,Jia Wu,Zheyang Chi,Shulin Zhao,Fanggui Ye

Natural enzyme cascades, including the same and different kinds of enzymes, have been widely used in biosensing. However, it is still a challenge for nanozymes, the mimics of natural enzymes, to achieve biosensing with different types of enzyme-like activities based on cascade reactions, such as hydrolase and oxidoreductase. Considering the characteristics of the multienzyme-like activity of some nanozymes, as a proof of concept, this study demonstrates a Cu/Ce-MOF-808 nanozyme as a self-cascade catalyst with hydrolase-like and catechol oxidase-like activities. Taking organophosphorus pesticides as an example, we constructed a self-cascade catalytic reaction for the one-step colorimetric detection of isocarbophos. Specifically, the hydrolase-like activity of Cu/Ce-MOF-808 can hydrolyze isocarbophos to produce colorless isopropyl salicylate, which is further oxidized by the catechol oxidase-like activity of Cu/Ce-MOF-808 and complexed with 4-aminoantipyrine (4-AAP) to produce red quinoneimine under neutral conditions with illumination. Thus, a colorimetric strategy for sensitive isocarbophos analysis is developed, with a linear range of 0.90–400.00 μg·mL–1 and a detection limit of 0.42 μg·mL–1. This research presents a new approach to the design and synthesis of nanozymes and has great application potential in the field of nanozyme cascade biosensing.

Self-cascade reaction triggered colorimetric detection of choline in triple-negative breast cancer

Xiao-Jie Shi,Xin-Yi Dai,Yu Wu,Ting-Ting Zhang,Tian-Shun Song,Xiang-Ling Li,Jing-Jing Xie

Achieving a simple process, detailed information, and visual response for cancer diagnosis and treatment is desirable but challenging. Herein, we develop a self-cascade system-triggered colorimetric array for the sensitive detection of choline, a promising biomarker in triple-negative breast cancer (TNBC). The system utilizes a ChOx-Ce 3+ probe, where ChOx catalyzes the oxidation of target choline to betaine, generating H 2 O 2 , which then in situ reacts with Ce 3+ to form a CeO 2 nanozyme. This nanozyme exhibits robust oxidase (OXD)-like activity, oxidizing tetramethylbenzidine (TMB) to oxTMB, resulting in a visible color change that correlates with choline concentration. The self-triggered cascade mechanism significantly simplifies the detection process by eliminating the need for expensive materials and complex equipment. Additionally, it enhances the selectivity, resistance to protein interference, and long-term stability of the probe. The system demonstrated high accuracy in measuring choline concentrations in MDA-MB-231 cells and real samples, with recovery rates ranging from 97.4 % to 101 %. These results illustrate the potential of the self-cascade system-triggered colorimetric array for early TNBC detection and therapeutic monitoring, offering a cost-effective, user-friendly diagnostic tool for clinical applications.

Inbuilt self-cascade catalysis of the bimetal-confined structural nanozyme CoNi@CNTs-N/GO for increased bienzymatic activity and H2O2-free smartphone-based visual assay of total antioxidant capacity in foods

Hailong Gong,Xiaofeng Hou,Binrong Li,Siyu Shen,Chen Wang,Qinqin Ma,Tingting Liu,Xuedong Wang

The level of total antioxidant capacity (TAC) reflects the overall ability of food to resist oxidative damage, maintain quality and nutritional stability. In this work, we pioneered a Co Ni alloy-confined N-doped carbon nanozyme (CoNi@CNT-N/GO) with a self-cascade catalysis. Compared to other nanozymes, the synergistic effect of OXD and POD realized self-sustained generation of H 2 O 2 , eliminating the need of exogenous addition, and further decomposition of H 2 O 2 into ·OH and oxidizes colorless 3,3′,5,5′- tetramethylbenzidine (TMB) to blue oxTMB as an efficient catalyst. The integration of multiple components and the built-in unique mechanism enhance bienzymatic activity. A “Thing Identify” APP was utilized to construct a smartphone-based visualization platform, demonstrating satisfactory linearity (0.01–1.2 mM) and low detection limit (3.3 μM) in TAC detection of real-world foods. This platform yielded data comparable to those from commercially colorimetric kits. Overall, it proposes a novel idea for engineering multi-functional and non-additional H 2 O 2 nanozymes in on-site food-quality monitoring.

Self-cascade reaction catalyzed by CuO nanoparticle-based dual-functional enzyme mimics

Ai-Ling Hu,Hao-Hua Deng,Xiao-Qing Zheng,Yan-Yu Wu,Xiu-Ling Lin,Ai-Lin Liu,Xing-Hua Xia,Hua-Ping Peng,Wei Chen,Guo-Lin Hong

It is desirable but challenging to assemble various biomimetic properties into a functional catalytic cascade system . In this work, cupric oxide nanoparticles were investigated as oxidase mimics for the aerobic oxidation of cysteine to cystine with the generation of hydrogen peroxide. Coupling this property with the peroxidase-like activity of CuO nanoparticles, we constructed a self-organized cascade reaction system based on a single-component nanozyme , which includes the oxidation of cysteine to yield cystine and hydrogen peroxide and the hydrogen peroxide-mediated oxidation of terephthalic acid to produce a fluorescence change. Based on this artificial enzymatic cascade reaction system, a fluorometric assay method with a low detection limit of 6.6   nM was established for cysteine determination. This platform was then applied for the detection of cysteine in pharmaceutical products and human plasma, which yielded satisfactory results. Our investigations open up a new route and holds promise for the development and applications of multifunctional nanomaterials as enzyme mimics .

Engineering Single-Atom Iron Nanozymes with Radiation-Enhanced Self-Cascade Catalysis and Self-Supplied H2O2 for Radio-enzymatic Therapy

Xianyu Zhu,Jiabin Wu,Ruixue Liu,Huandong Xiang,Wenqi Zhang,Qingchao Chang,Shanshan Wang,Rui Jiang,Feng Zhao,Qiqiang Li,Liang Huang,Liang Yan,Yuliang Zhao

Single-atom nanozymes (SAzymes), with individually isolated metal atom as active sites, have shown tremendous potential as enzyme-based drugs for enzymatic therapy. However, using SAzymes in tumor theranostics remains challenging because of deficient enzymatic activity and insufficient endogenous H2O2. We develop an external-field-enhanced catalysis by an atom-level engineered FeN4-centered nanozyme (FeN4-SAzyme) for radio-enzymatic therapy. This FeN4-SAzyme exhibits peroxidase-like activity capable of catalyzing H2O2 into hydroxyl radicals and converting single-site FeII species to FeIII for subsequent glutathione oxidase-like activity. Density functional theory calculations are used to rationalize the origin of the single-site self-cascade enzymatic activity. Importantly, using X-rays can improve the overall single-site cascade enzymatic reaction process via promoting the conversion frequency of FeII/FeIII. As a H2O2 producer, natural glucose oxidase is further decorated onto the surface of FeN4-SAzyme to yield the final construct GOD@FeN4-SAzyme. The resulting GOD@FeN4-SAzyme not only supplies in situ H2O2 to continuously produce highly toxic hydroxyl radicals but also induces the localized deposition of radiation dose, subsequently inducing intensive apoptosis and ferroptosis in vitro. Such a synergistic effect of radiotherapy and self-cascade enzymatic therapy allows for improved tumor growth inhibition with minimal side effects in vivo. Collectively, this work demonstrates the introduction of external fields to enhance enzyme-like performance of nanozymes without changing their properties and highlights a robust therapeutic capable of self-supplying H2O2 and amplifying self-cascade reactions to address the limitations of enzymatic treatment.

A self-cascade system based on Ag nanoparticle/single-walled carbon nanotube nanocomposites as an enzyme mimic for ultrasensitive detection of L-cysteine

Feifei Li,Congcong Hu,Wenen Su,Hao Liang,Fubing Xiao,Jinquan Liu,Yan Tan,Shengyuan Yang

L-Cysteine, widely used in medicine and the food industry, is of great essentiality to organisms and the food quality. Given that current detection approaches require exacting lab conditions and tedious sample treatment, there is a pressing demand for developing a method that possesses advantages of user friendliness, prominent performance, and cost-effectiveness. Herein, a self-cascade system was developed for the fluorescence detection of L-cysteine based on the ingenious performance of Ag nanoparticle/single-walled carbon nanotube nanocomposites (AgNP/SWCNTs) and DNA-templated Ag nanoclusters (DNA-AgNCs). The fluorescence of DNA-AgNCs could be quenched on account of the adsorption of DNA-AgNCs on AgNP/SWCNTs by π–π stacking. With the cooperation of Fe2+, AgNP/SWCNTs with oxidase and peroxidase-like activities could catalyze the oxidation of L-cysteine to produce cystine and hydrogen peroxide (H2O2) and then break the O–O bond of H2O2 to generate a hydroxyl radical (·OH), which could cleave the DNA strand into different sequence fragments which subsequently peeled off from the AgNP/SWCNTs, resulting in a “turn-on” fluorescence response. In this paper, AgNP/SWCNTs with multi-enzyme activities was synthesized enabling the reaction to proceed in just one step. The successful preliminary applications for the L-cysteine detection in pharmaceutical, juice beverage, and serum samples indicated that the developed method exhibited great potential in medical diagnosis, food monitoring, and the biochemical field, which also broadened the horizon for follow-up research.

Efficiently selective inactivation of Gram-positive bacteria by MCM-41 molecular sieve-supported copper-based self-cascading nanozymes

Xue-Yao Pang,Fan-Xiang Meng,Cheng Ma,Si-Yang Ni,Zi-Han Jia,Bo Li,Yu-Xiang Liu,Wei-Wei Gao

Gram-positive bacteria can cause a variety of infectious diseases. Selectively inhibiting Gram-positive bacteria will avoid the misuse of broad-spectrum antibiotics, achieve precise treatment, and delay the development of antibiotic resistance. In this work, we successfully developed an MCM-41 molecular sieve-supported copper-based nanozyme composites (MCM-41-Cu 2 O) for selectively combating Gram-positive bacteria. MCM-41, with its porous structure and excellent biocompatibility, not only serves as a carrier but also neutralizes the positive charges of the metal nanozyme, endowing MCM-41-Cu 2 O with negative zeta potentials. This allows it to interact electrostatically with Gram-positive bacteria, facilitating the penetration of free radicals into the bacterial cell membrane, thereby killing the bacteria. MCM-41-Cu 2 O continuously generates endogenous H 2 O 2 through its glutathione oxidase (GSHOx)-like activity·H 2 O 2 serves as the substrate for peroxidase (POD)-like catalytic reactions, producing •OH radicals, thereby enabling a self-cascade reaction. This approach avoids the toxic side effects associated with excessive exogenous H 2 O 2 and offers higher biosafety with potential for antibacterial applications. DFT calculations reveal that compared to Cu 2 O, MCM-41-Cu 2 O exhibits greater adsorption and decomposition capacity for H 2 O 2 , accelerating reaction rates and enhancing POD-like activity. MCM-41-Cu 2 O selectively targets Gram-positive bacteria, achieving a bactericidal rate of up to 99.6% against S. aureus and B. subtilis while leaving E. coli and P. aeruginosa colonies largely unaffected. Reactive oxygen species (ROS) generated by MCM-41-Cu 2 O-5% disrupt bacterial membranes, leading to depolarization, protein leakage, and bacterial death. Additionally, the MCM-41-Cu 2 O nanocomposites exhibit excellent biocompatibility and demonstrate superior in vivo antibacterial efficacy compared to antibiotics, effectively treating wound infections caused by S. aureus . These findings position MCM-41-Cu 2 O as a promising alternative for treating infections caused by Gram-positive bacteria and provide new insights into the antimicrobial applications of molecular sieves.

Self-cascade catalytic system constructed using carbon dots and Au nanoparticles co-assembled on MIL-53(Fe)-NH2 as a three-in-one fluorescent nanozyme for multimodal detection of glucose and maltose in food

Junxue Liu,Yufei Wang,Mingqin He,Xiaohong Chen,Chenyu Zhou,Jiyang Li

Determination of glucose and maltose is crucial for food production and human health. Herein, a novel Au/CD@MIL-53(Fe)-NH 2 self-cascade nanozyme was constructed via host–guest assembly with “three-in-one” features, including blue fluorescence, H 2 O 2 production as an oxidase mimic, and •OH generation as a peroxidase mimic. Theoretical and experimental results proved that the incorporation of carbon dots renders the composite with high peroxidase-like activity and extremely high affinity for H 2 O 2 (K m : 0.011 mM), 336 times higher than that of the natural enzyme. The self-cascade catalytic process could oxidize colorless 3,3′,5,5′-tetramethylbenzidine (TMB) to blue ox-TMB, which further quenched the fluorescence and produced a photothermal effect. Consequently, colorimetric/fluorescence/photothermal-based multimodal detection of glucose and maltose was first achieved, obtaining low detection limits of 12/7.5/1.1 and 18/11.3/1.4 μM, respectively. This study offers a promising strategy for designing efficient nanozyme-based cascade systems that help enhance food quality determination.

Enhanced biomimetic catalysis via self-cascade photocatalytic hydrogen peroxide production over modified carbon nitride nanozymes for total antioxidant capacity evaluation

Shengzhen Li,Shushu Chu,Mingyuan Xia,Hengya Wei,Yizhong Lu

The peroxidase mimics usually requires the addition of exogenous hydrogen peroxide (H 2 O 2 ), which greatly hinder their practical applications. Herein, through rational co-modification of multiple elements (potassium (K), chlorine (Cl) and iodine (I)), the modified carbon nitride nanomaterials (KCl/KI-CN) could serve as efficient bifunctional catalysts. The multiple elements doping and the incorporation of cyano groups ( C N) are deemed to enhance their photocatalytic and peroxidase-like activity, respectively. Based on the photocatalytic function, H 2 O 2 can be produced continuously and steadily via two-electron oxygen reduction over modified carbon nitride under visible light irradiation. Subsequently, the KCl/KI-CN could catalyze the chromogenic substrate by the in-situ produced H 2 O 2 . Taking advantage of the bifunctional properties of modified carbon nitride, we for the first time demonstrate a self-cascade catalytic process and apply successfully for the ascorbic acid (AA) detection and versatile total antioxidant capacity (TAC) evaluation. This paper not only prepares an efficiently bifunctional catalyst but also provides a new self-cascade photocatalytic H 2 O 2 production strategy for the peroxidase-like application.

Self-Cascade MOF@MOF nanozyme for ultrasensitive and Low-Background detection of Multidrug-Resistant bacteria

Ruiwen Li,Guoqing Qian,Hao Shen,Shaoning Yu

The accurate, timely, and convenient detection of multidrug-resistant bacteria (MDRB) is of enormous value in clinical treatment and food safety. Traditional methods include plate culture and polymerase chain reaction, but these strategies are time-consuming, costly, or cumbersome to operate, making it difficult to meet the increasingly severe demands. Here, we describe the synthesis of a metal–organic framework (MOF) heterostructure hybrid (MOF@MOF) self-cascade nanozyme that promotes signal amplification for colorimetric MDRB aptasensing. The MOF@MOF was composed of copper-functionalized MOF-808 with catechol oxidase-mimicking activity and an iron porphyrin MOF with peroxidase-mimicking activity; MOF-808 with open copper sites hydrolyzed the substrate 3,5-ditert-butylcatechol, producing H 2 O 2 in situ , and the iron porphyrin MOF catalyzed H 2 O 2 to further oxidize 3,3′,5,5′-tetramethylbenzidine into a colored product. Benefitting from the nanoproximity effect and confinement effect, the biocatalytic ability of the biomimetic cascade was significantly facilitated, generating a strengthened colorimetric signal. Taking methicillin-resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa (PA) as examples, the MOF@MOF nanozyme coordinated with aptamers to construct a colorimetric aptasensing platform for ultrasensitive detection of MDRB. Compared to frequently used probes containing H 2 O 2 , the inactive substrate did not oxidize TMB to induce a color change, resulting in low background signal. The limit of detection was 5 CFU/mL for MRSA and 2 CFU/mL for PA, with a wide linear range for both (10–10 8 CFU/mL). The proposed aptasensing platform was used to analyze MDRB in biological samples, indicating excellent practicability. The self-cascade MOF@MOF may offer valuable insights into the rational construction of biomimetic sensors based on nanozymes.

A Radiation-Activated and Self-Cascade Nano-Immunostimulator Triggers a Potent Abscopal Effect Against Distant Tumors

Bianfei Shao,Xiuqi Liang,Zhongzheng Xiang,Fang Liu,Yuanyuan Zeng,Tao He,Shunyao Zhu,Xinchao Li,Huile Gao,Peng Mi,Lei Liu,Changyang Gong

Radiotherapy (RT) inhibits local tumors by stimulating reactive oxygen species (ROS) generation, and sometimes suppresses distant tumors known as the abscopal effect. However, the abscopal effect is rarely observed in clinic, due to insufficient tumor-associate antigens (TAAs) and weak immune stimulation. Here, a radiation-activated and self-cascade nano-immunostimulator (RAIDER) is fabricated to trigger a potent abscopal effect against distant tumors by unleashing cascade-amplified ROS generation and boosting antigen presentation. After accumulation in tumors, RAIDER undergoes HAase-triggered deshielding and charge-reversal, and GE11 enriched liposomal core and CpG are both released. Under following RT, protoporphyrin IX-induced Cherenkov radiation-photodynamic therapy (CR-PDT) elicits ROS generation, leading to disintegration of RAIDER and catalase (CAT) burst release. Meanwhile, by the combination of RT, CR-PDT, and CAT-induced O 2 supply, self-cascade ROS generation is achieved to enhance lysis of tumor cells and release of massive TAAs. Subsequently, the co-stimulation of TAAs and the released CpG activates dendritic cell maturation to boost antigen presentation, resulting in a strong systemic immune response. Combined with RT, RAIDER exhibits a potent abscopal effect, with an average distant tumor growth inhibition of ≈70.3%. Overall, the strategy amplifies antitumor effect of RT from local to systemic, holding a potential against local and distant tumors.

Bicatalytic Cupric Metal–Organic Framework Nanosheets for Self-Cascade Conversion of Methyl Parathion to 4-Aminophenol

Caiwen Pan,Xiaoyan Dong,Qinghong Shi,Linling Yu,Yan Sun

Methyl parathion (MP) is a typical organophosphate pesticide widely used in agriculture, causing serious pollution. MP hydrolysis generates a toxic 4-nitrophenol (4-NP) with environmental risks, but a subsequent reduction of 4-NP produces a nontoxic 4-aminophenol (4-AP), which serves as a resource for further use in chemical synthesis. Thus, the detoxification and resourcilization of MP are of great significance for environmental remediation and sustainable development. However, most reports on MP cascade conversion require two different catalysts, each of which is single-catalytic. Herein, we report the first self-cascade nanocatalyst with bicatalytic activities of hydrolysis and reduction based on cupric metal–organic framework (Cu-AZB) nanosheets for the conversion of MP to 4-AP. It was found that Cu-AZB could completely convert MP to 4-AP in the presence of NaBH4 within 10 min. Moreover, hydrolysis was 19-fold accelerated by the NaBH4-mediated reduction in the self-cascade catalysis, so a catalyst with a high cascade conversion efficiency against MP with an activity factor of 513.2 min–1 g–1, a reaction rate constant of 0.25 min–1, and a half-life of 2.82 min was achieved, which was more efficient than literature-reported cascade catalysts. The results demonstrated the application potential of the bicatalytic Cu-AZB in the complete detoxification and conversion of the toxic pollutant into a nontoxic resource through one-pot cascade catalysis.
