Topology-Directed Self-Locking of Colloidal Suprastructures

Yanqiu Du,Yunhan Zhang,Jing Jin,Shiyan Xiao,Haojun Liang,Wei Jiang

The robustness and stability of self-assembled colloidal suprastructures are pivotal and challenging in the field of soft condensed matter and nanotechnology. Herein, we develop a novel approach to “self-locking” the colloidal suprastructures to achieve excellent stability by taking advantage of the topology of polymer brushes on nanoparticles. Compared to linear polymer brushes, looped polymer brushes can significantly improve the structural stability of the suprastructures. A combination of experiments and simulations reveals that the suprastructures achieve their significant stabilities through a “self-locking” mechanism by forming folded entanglement between looped polymer molecules. The stabilized suprastructures can stay intact in various organic solvent environments or under heating and mechanical oscillations. Furthermore, the interparticle spacing of suprastructures can be modulated by controlling polymer conformations in an aqueous solution through varying salt concentrations. This new approach of “self-locking” provides an easy-to-operate strategy to stabilize suprastructures and modulate interparticle distances without adding chemicals, which allows the potentially wide applications of colloidal suprastructures under harsh environments.

Dual-Action Psoriasis Therapy: Antiproliferative and Immunomodulatory Effects via Self-Locking Microneedles

Zi Yi Wang,Ze Qiang Zhao,Yu Jun Sheng,Ke Jun Chen,Bo Zhi Chen,Xin Dong Guo,Yong Cui

Psoriasis is a chronic, immune-mediated disorder characterized by immune regulation disorders and abnormal keratinocyte proliferation. Deucravacitinib (Deu), a selective oral Tyrosine Kinase 2 (TYK2) inhibitor, shows promise in treating psoriasis but may cause systemic side effects and fail to address persistent localized thickened lesions. Herein, a self-locking microneedle (MN) patch with a polyvinyl alcohol (PVA) inner ring loaded with Deu is developed, designed to penetrate the transdermal barriers and dissolve rapidly, downregulating the IL-23/IL-17 pathway and serve as the first line of defense against the spread of skin-originated inflammation. Additionally, Calcipotriol (Cal), a vitamin D derivative, is incorporated into a methacrylated hyaluronic acid (HAMA) backing layer and outer ring that mimics occlusive administration, maintaining localized skin surface retention for prolonged anti-proliferative therapy. The Deu@Cal MN demonstrates satisfactory adhesiveness due to swelling-mediated mechanical interlocking via the outer ring, ensuring targeted drug release at lesion site. Besides its effectiveness in alleviating both skin inflammation and proliferation, it inhibits the differentiation of Th17 cells in the spleen, suggesting potential to reduce systemic inflammation. These findings offer a new therapeutic approach for treating psoriasis and other autoimmune and inflammatory conditions.

A honeybee stinger-inspired self-interlocking microneedle patch and its application in myocardial infarction treatment

Yuwen Lu,Tanchen Ren,Hua Zhang,Qiao Jin,Liyin Shen,Mengqi Shan,Xinzhe Zhao,Qichao Chen,Haoli Dai,Lin Yao,Jieqi Xie,Di Ye,Tengxiang Lin,Xiaoqian Hong,Kaicheng Deng,Ting Shen,Jiazhen Pan,Mengyan Jia,Jun Ling,Peng Li,Yue Zhang,Huanan Wang,Lenan Zhuang,Changyou Gao,Jifu Mao,Yang Zhu

Weak tissue adhesion remains a major challenge in clinical translation of microneedle patches. Mimicking the structural features of honeybee stingers, stiff polymeric microneedles with unidirectionally backward-facing barbs were fabricated and embedded into various elastomer films to produce self-interlocking microneedle patches. The spirality of the barbing pattern was adjusted to increase interlocking efficiency. In addition, the micro-bleeding caused by microneedle puncturing adhered the porous surface of the patch substrate to the target tissue via coagulation. In the demonstrative application of myocardial infarction treatment, the bioinspired microneedle patches firmly fixed on challenging beating hearts, significantly reduced cardiac wall stress and strain in the infarct, and maintained left ventricular function and morphology. In addition, the microneedle patch was minimally invasively implanted onto beating porcine heart in 10 minutes, free of sutures and adhesives. Therefore, the honeybee stinger-inspired microneedles could provide an adaptive and convenient means to implant patches for various medical applications. Statement of significance Adhesion between tissue and microneedle patches with smooth microneedles is usually weak. We introduce a novel barbing method of fabricating unidirectionally backward facing barbs with controllable spirality on the microneedles on microneedle patches. The microneedle patches self-interlock on mechanically dynamic beating hearts, similar to honeybee stingers. The micro-bleeding and coagulation on the porous surface provide additional adhesion force. The microneedle patches attenuate left ventricular remodeling via mechanical support and are compatible with minimally invasive implantation.

A 3D-printed bioactive polycaprolactone scaffold assembled with core/shell microspheres as a sustained BMP2-releasing system for bone repair

Weida Zhuang,Genlan Ye,Jiachang Wu,Leyu Wang,Guofang Fang,Zhuofeng Ye,Guohua Lai,Xiaozhong Qiu,Hongxun Sang

Integration of biological factors and hierarchical rigid scaffolds is of great interest in bone tissue engineering for fabrication of biomimetic constructs with high physical and biological performance for enhanced bone repair. Core/shell microspheres (CSMs) delivering bone morphogenetic protein-2 (BMP-2) and a strategy to integrate CSMs with 3D-printed scaffolds were developed herein to form a hybrid 3D system for bone repair. The scaffold was printed with polycaprolactone (PCL) and then coated with polydopamine. Shells of CSMs were electrosprayed with alginate. Cores were heparin-coated polylactic acid (PLA) microparticles fabricated via simple emulsion and heparin coating strategy. Assembly of microspheres and scaffolds was realized via a self-locking method with the assistance of controlled expansion of CSMs. The hybrid system was evaluated in the rat critical-sized bone defect model. CSMs released BMP-2 in a tunable manner and boosted osteogenic performance in vitro . CSMs were then successfully integrated inside the scaffolds. The assembled system effectively promoted osteogenesis in vitro and in vivo . These observations show the importance of how BMP-2 is delivered, and the core/shell microspheres represent effective BMP-2 carriers that could be integrated into scaffolds, together forming a hybrid system as a promising candidate for enhanced bone regeneration.
