Carbon dioxide (CO 2 ) plays a crucial role in the host search/localization process of bed bugs, Cimex lectularius L., and Cimex hemipterus (F.). Current research on the CO 2 perception by bed bugs mainly focuses on their behavioral responses, and the molecular mechanisms are still unclear. In addition, existing research has mainly been conducted on C. lectularius with very little research on C. hemipterus . In this study, we investigated the behavioral response of C. hemipterus to CO 2 and analyzed the role of antennae in the CO 2 sensing process. Then, potential CO 2 receptor genes were screened through antennal RNA sequencing and tissue-specific expression profiling analysis. Finally, the function of CO 2 receptors was further validated using RNA interference. Results indicated that increasing the CO 2 concentration in the air not only stimulated the activity of C. hemipterus , but also presented a directional attraction effect on them. CO 2 , with a concentration of 0.06%–1.8%, had a significant attraction effect on C. hemipterus . Removal of the antennae led to the loss of bed bugs’ preference for CO 2 . Four candidate CO 2 receptor genes ( ChGR1 , ChGR2 , ChGR3 , and ChGR4 ) were identified through transcriptomic analysis of the bed bug antennae, and 3 of them ( ChGR2 , ChGR3 , and ChGR4 ) were highly expressed in the antennae. Silencing ChGR2 or ChGR4 individually led to a significant reduction or even loss of CO 2 sensing ability in C. hemipterus . In conclusion, CO 2 can induce the host searching behavior of C. hemipterus ; moreover, ChGR2 and ChGR4 are crucial for detecting CO 2 in C. hemipterus .

BACKGROUND Over the past two decades, bed bugs ( Cimex spp.) have resurged as common urban pests around the world. The search for efficient and safe control measures has become a key interest among researchers, manufacturers, and pest control professionals. In this study, we evaluated and compared the efficacy of steam, diatomaceous earth (DE) dust, and a combination of both against tropical bed bugs ( Cimex hemipterus (F.)) under laboratory and field conditions. RESULTS In the laboratory study, the mortality of bed bugs after 2 days of exposure to DE dust was 100%. When bed bugs stayed on the surface of an object or in cracks, a brief steam treatment (1 s) caused 100% mortality. However, when bed bugs were hidden under a fabric cover, steam application for 10 s only caused 89 ± 6% mortality. Bed bugs that survived steam treatment exhibited reduced feeding activity. In a 14-week long study, there was no significant difference in the reduction rate of bed bugs between steam treatment and DE dust treatment. A 37-week long control study showed that steam and steam plus DE dust treatments eliminated 97–100% of the infestations. CONCLUSION Applying steam and DE dust are effective strategies for eliminating natural tropical bed bug infestations. Continuous follow-up monitoring and treatment until no bed bugs are found are crucial in completely eliminating the infestation of tropical bed bugs. © 2024 Society of Chemical Industry.

The widespread adoption of Bt crops expressing insecticidal proteins derived from Bacillus thuringiensis has created a need to assess the potential effects of these toxins on non-target organisms, especially species such as Arma custos , a generalist predator that provides important biological control services in many field crops in Asia. Direct dietary exposure of A. custos to Cry1Ah and Vip3Aa proteins produced no adverse effects on life history traits, despite continuous exposure throughout development and early adult life to concentrations significantly higher than the Bt protein concentration likely encountered by A.custos in the field, even when feeding directly on Bt plants. Enzyme-linked immunosorbent assay confirmed the presence of Bt proteins in A. custos midguts, but quantitative real-time PCR analysis of 12 genes associated with detoxification, antioxidative responses, immune responses, and metabolism revealed no significant changes in expression in adult bugs. Indirect exposure to these toxins via consumption of intoxicated prey, larvae of Helicoverpa armigera (Hübner), likewise produced no negative impacts on survival, development, adult weight, or female fecundity in either the F0 (exposed) or F1 (unexposed) generation, but female fresh weight was reduced in the F0 generation by the Cry1Ah (50 μg/g) treatment. Finally, a competitive binding assay with labelled protein and a ligand blotting assay both demonstrated that the Cry1Ah protein could not bind to receptors on the midgut brush border membrane vesicles (BBMVs) of A. custos adults. Therefore, we conclude that Cry1Ah and Vip3Aa proteins are unlikely to have significant negative effects on A. custos populations if employed as plant-incorporated protectants in field crops.

BACKGROUND The bites and blood sucking of bed bugs ( Cimex spp.) (Hemiptera: Cimicidae) pose a serious threat to human physical and mental health. Application of an effective repellent can prevent or reduce bed bug bites. Previous studies on repellent screening mainly focused on Cimex lectularius L. In this study, we investigated the repellent effect of two safe food additives, ethyl anthranilate (EA) and butyl anthranilate (BA), against Cimex hemipterus (F.), and also explored the role of antennae and mouthparts on C. hemipterus perception of repellents. RESULTS Both EA and BA had a strong repellent effect against tropical bed bugs and their repellency was similar or lower than that of N , N -diethyl-3-methyl benzoyl amide, depending on whether or not a CO 2 source was present. EA had higher repellency than BA and exhibited repellency to C. hemipterus for 3 days when a CO 2 source was present. C. hemipterus avoided resting on 20% EA- and BA-treated harborages. Applying 20% EA and BA on rabbit skin significantly reduced the blood intake of C. hemipterus within 2 h. C. hemipterus could perceive EA and BA after their antennae or mouthparts or both antennae and mouthparts were removed. CONCLUSION Both EA and BA had strong repellency against C. hemipterus , with EA being more repellent. Ablation of antennae and mouthparts did not affect the perception of EA and BA by C. hemipterus . © 2023 Society of Chemical Industry.

Desiccant dusts have been shown to be effective materials in the management of bed bugs ( Cimex spp.). Past research primarily focused on exploring the direct lethal effect of dusts against bed bugs, whereas information on their sublethal effects on bed bugs are limited. In this study, we evaluated the lethal effect of 3 dust products (Johnson's ® Baby powder, Vazor DE powder ™ , and CimeXa™; abbreviated as Baby powder, DE, and CimeXa) against the tropical bed bug, Cimex hemipterus (F.), under laboratory conditions. Results showed that the 3 dust products caused 97%–100% mortality to tropical bed bugs within 14 d, both in no-choice and choice experiments. However, in a brief exposure experiment (crossing a 2.5-cm-wide band), Baby powder exposure caused a significantly lower mortality (23%) than DE (88%) and CimeXa (100%). The more effective dusts (DE and CimeXa) were selected for further evaluation of their sublethal effects on C. hemipterus . DE and CimeXa caused significantly higher mortality (48%–58%) through horizontal transfer than that of control (6%). Brief exposure to DE and CimeXa dusts did not affect tropical bed bug activity and their response to CO 2 . However, CimeXa-exposed tropical bed bugs exhibited reduced feeding and lowered survival rate after feeding. Moreover, both DE and CimeXa dust bands showed strong barrier effects on the crawling of tropical bed bugs. Our results indicate that both DE and CimeXa have great potential for preventing C. hemipterus from reaching protected areas and for controlling C. hemipterus .
