Composition and migration of microbial community in the ice–water–sediment interface of saline lakes in cold and arid regions

Xuezheng Yu,Weiying Feng,Jing Liu,Jiayue Gao,Tengke Wang,Yunping Han,Yingnan Cao,Jianguo Liu

Ecosystems in extreme environments, such as saline lakes in cold and arid regions, are greatly affected by high salinity, significant temperature fluctuations, and low nutrient availability, resulting in unique microbial community structures. However, the spatiotemporal dynamics and migration mechanisms of microbial communities in such environments remain unclear. Here, we investigated the composition and migration of the microbial communities at the ice–water–sediment interfaces of Daihai Lake (high salinity) and Ulansuhai Lake (low salinity) in Inner Mongolia, China. The changes in microbial communities under varying salinity conditions were analysed and potential pathogenic genera with implications for fish and other aquatic animals were identified. Ice and water were dominated by Proteobacteria and Bacteroidetes, whereas sediments mainly contained Chloroflexi and Nitrospirota. Additionally, non-parametric Kruskal–Wallis rank-sum tests and linear discriminant analysis effect size (LEfSe) analysis were conducted to detect significant differences in microbial abundance. Significant seasonal and site-specific variations in indicator genera were observed in the sediments. Sulfur-oxidising bacteria, such as Thioalkalispira-Sulfurivermis and Thiobacillus, were predominant in the summer. The indicator genus at point D1 was Thiobacillus , while the indicator genus at point D2 was Bacillus . Furthermore, the microbial migration efficiency was substantially lower in high-salinity lakes (sediment-to-water body: 9.8 %; water body-to-ice: 15.9 %) than in low-salinity lakes (sediment-to-water body: 27.1 %; water body-to-ice: 22.7 %). Seven pathogenic genera were identified in the saline lake, which could affect fish health and cause acute gastrointestinal inflammation in humans upon consumption. This study will allow us to fully understand the compositional characteristics of microbial species under different salinities and provide valuable information for subsequent saline lake management. In particular, it has important practical applications in water quality monitoring and ecological restoration.

Effects of quaternary ammonium disinfectants on human pathogenic bacteria in anaerobic sludge digestion: Dose-response and resistance variation

Wan Yang,Yunfei Liang,Shengsen Wang,Chen Cai,Xiaozhi Wang,Xiaohu Dai,Xiang Chen

Sewage sludge is a critical reservoir for biological pollutants, and its harmless disposal remains a major issue. Quaternary ammonium compounds (QACs) as typical household disinfectants are inevitably concentrated in sewage sludge, and have the potential to affect human pathogenic bacteria (HPBs) that remain poorly understood. This study found that the relative abundance of HPBs in digesters was decreased by 10 − 20 % at low QACs dose, but increased by 238 − 591 % at high QACs dose. Mechanistic analysis revealed that low QACs doses promoted functional hydrolytic/fermentative bacteria and their metabolism by stimulating extracellular polymeric substances secretion and enhancing resistance to QACs. Conversely, high QAC doses decreased microbial biomass and developed QACs and antibiotic resistance of HPBs by increasing cell membrane permeability and triggering oxidative stress, resulting in deteriorating sanitation performance. These findings provide advanced insights into the potential function and hazards of exogenous QACs on the biosafety of digestate.

A dual-mode aptasensor for foodborne pathogens detection using Pt, phenylboric acid and ferrocene modified Ti3C2 MXenes nanoprobe

Wenhai Wang,Shu Xiao,Zhijian Jia,Hongzhen Xie,Tianhua Li,Qiqin Wang,Ning Gan

Foodborne pathogens could result in serious health issues to public and thus some rapid analysis methods are urgent developed to monitor them. Herein, a dual-mode aptasensor based on electrochemical and colorimetric modes was constructed for the on-site detection of Vibrio parahaemolyticus ( V.P) in shrimps as a model. A novel nanoprobe ( [email protected] @MXenes) was developed to generate both electrochemical and colorimetric signals by modifying phenylboronic acid and ferrocene on Pt-MXenes nanocomposite . The nanoprobe exhibited both features of peroxidase mimic and specific recognition. Simultaneously, a V.P aptamer-functionalized electrode interface was fabricated to firstly specifically capture V.P, then conjugated with [email protected] @MXenes to form a sandwich complex. The complex could catalyze tetramethylbenzidine (TMB)-H 2 O 2 to produce a visible signal, and generate electrochemical signal simultaneously. Under the optimal condition, the aptasensor could determine as low as 5 CFU mL −1 V.P by electrochemical mode and 30 CFU mL −1 V.P by colorimetric mode. These superior performances may be attributed to that the synergistic catalysis and conductive effects deriving from Pt and MXenes on the nanoprobe can significantly amplify the signals to improve detection sensitivity. In particular, the dual detection system could mutually verify to achieve high accuracy. The aptasensor could also be expanded to rapid analysis of other food-borne pathogens through changing the aptamer on the electrode.

Antimicrobial activity and mechanisms of carbon quantum dot decorated modified zinc oxide nanoparticles against oral pathogenic bacteria

Yayu Chen,Pengcheng Huang,Yuqiong Wu,Chaojin Liu

Caries represents one of the most prevalent bacterial infectious diseases in the oral cavity. Over decades, researchers have been actively seeking nanomaterials capable of inhibiting oral pathogenic bacteria. We utilized a one-pot hydrothermal method to synthesize CQDs with multicolor luminescence and CQD-loaded ZnO composite nanomaterials. FE-SEM revealed that the rod-like structure of CQDs/ZnO becomes finer with increased CQDs content. XRD and XPS confirmed the presence of all components within the nanocomposites. We compared the antimicrobial activities of ZnO, 7.7% and 14.3% CQDs/ZnO against Streptococcus mutans , Enterococcus faecalis , Escherichia coli , and Staphylococcus aureus . The 7.7% CQDs/ZnO composite exhibited the most effective antibacterial activity. The 7.7% CQDs/ZnO composite exhibited the most effective antibacterial activity. FE-SEM and ONPG assays demonstrated that 7.7% CQDs/ZnO disrupted the membrane structure and alters membrane permeability. ESR, DPBF and ROS assayed further confirmed that 1 O 2 produced by 7.7% CQDs/ZnO accumulated at the cell membrane and disrupts its structure.

Portable Saliva Sensor Based on Dual Recognition Elements for Detection of Caries Pathogenic Bacteria

Yanan Li,Hao Zhao,Guanghong Han,Ze Li,Samuel M. Mugo,Hongda Wang,Qiang Zhang

Dental caries is one of the most common diseases affecting more than 2 billion people’s health worldwide. In a clinical setting, it is challenging to predict and proactively guard against dental cavities prior to receiving a confirmed diagnosis. Streptococcus mutans (S. mutans) in saliva has been recognized as the main causative bacterial agent that causes dental caries. High sensitivity, good selectivity, and a wide detection range are incredibly important factors to affect S. mutans detection in practical applications. In this study, we present a portable saliva biosensor designed for the early detection of S. mutans with the potential to predict the occurrence of dental cavities. The biosensor was fabricated using a S. mutans-specific DNA aptamer and S. mutans-imprinted polymers. Methylene blue was utilized as a redox probe in the sensor to generate current signals for analysis. When S. mutans enters complementarily S. mutans cavities, it blocks electron transfer between methylene blue and the electrode, resulting in decreases in the reduction current signal. The signal variations are associated with S. mutans concentrations that are useful for quantitative analysis. The linear detection range of S. mutans is 102–109 cfu mL–1, which covers the critical concentration of high caries risk. The biosensor exhibited excellent selectivity toward S. mutans in the presence of other common oral bacteria. The biosensor’s wide detection range, excellent selectivity, and low limit of detection (2.6 cfu mL–1) are attributed to the synergistic effect of aptamer and S. mutans-imprinted polymers. The sensor demonstrates the potential to prevent dental caries.

Ultrasensitive self-powered biosensors with visual self-checking function for pathogenic bacteria detection

Wen Yu,Xinke Kong,Chengcheng Gu,Panpan Gai,Feng Li

We proposed an enzymatic biofuel cells (EBFCs)-based biosensing platform with visual self-checking function for the ultrasensitive detection of Vibrio parahaemolyticus (Vp) with low detection limit of 2 CFU mL −1 (S/N = 3). In detail, the as-proposed biosensor was composed of the aptamer/glucose dehydrogenase/poly-thionine/carbon nanotubes/gold nanoparticles bioanode , the mediated prussian blue (PB)/ prussian white (PW) electrode with the visual self-checking function and the bilirubin oxidase/carbon nanotubes (BOD/CNTs) biocathode. Initially, the successful electron transfer from the bioanode to the PB/PW electrode could produce a high open circuit voltage ( E OCV ), accompanying with the reduction of PB to PW. Once in the presence of the target Vp , it was recognized and captured by the aptamer at the bioanode, and the E OCV obviously decreased while the color of the mediated electrode would return back to blue. Thus, the signal E OCV difference could be checked by color change, which could to a great extent avoid possible false-positive readout. In addition, the self-checking function could be recovered when connected with BOD/CNTs electrode. Overall, this work presented a prototype of an on-site tool with visual self-checking function.

Effects of several flavonoids on human gut microbiota and its metabolism by in vitro simulated fermentation

Pan Lixia,Ye Hangyu,Pi Xionge,Liu Wei,Wang Zhao,Zhang Yinjun,Zheng Jianyong

Introduction: Flavonoids have antiviral, antitumor, anti-inflammatory, and other biological activities. They have high market value and are widely used in food and medicine fields. They also can regulate gut microbiota and promote human health. However, only a few flavonoids have been reported for their regulatory effects on human gut microbiota. Methods: The effects of hesperidin, hesperetin-7-O-glucoside, hesperetin, naringin, prunin, naringenin, rutin, isoquercitrin, and quercetin on gut microbiota structural and metabolic differences in healthy subjects were studied by means of in vitro simulated fermentation technology. Results: Results showed that the nine kinds of flavonoids mentioned above, especially hesperetin-7-O-glucoside, prunin, and isoquercitrin, were found to have more effect on the structure of human gut microbiota, and they could significantly enhance Bifidobacterium (p < 0.05). After 24 h of in vitro simulated fermentation, the relative abundance of intestinal probiotics (e.g., Lactobacillus) was increased by the three flavonoids and rutin. Furthermore, the relative abundance of potential pathogenic bacteria was decreased by the addition of hesperetin-7-O-glucoside, naringin, prunin, rutin, and isoquercitrin (e.g., Lachnoclostridium and Bilophila). Notably, prunin could also markedly decrease the content of H2S, NH3, and short-chain fatty acids. This performance fully demonstrated its broad-spectrum antibacterial activity.Discussion: This study demonstrates that flavonoids can regulate the imbalance of gut microbiota, and some differences in the regulatory effect are observed due to different structures. This work provides a theoretical basis for the wide application of flavonoids for food and medicine.

Artificial simulated saliva, gastric and intestinal digestion and fermentation in vitro by human gut microbiota of intrapolysaccharide from Paecilomyces cicadae TJJ1213

Juanjuan Tian,Xiaomeng Wang,Xueliang Zhang,Xiaohong Chen,Mingsheng Dong,Xin Rui,Qiuqin Zhang,Mei Jiang,Wei Li

In this study, the in vitro digestion and fermentation of two intra-polysaccharide fractions (IPS1 and IPS2) from Paecilomyces cicadae TJJ1213 were investigated. The constituent monosaccharides of IPS1 and IPS2 were not changed after simulated saliva, gastric and small intestinal digestion. However, they can be hydrolyzed and utilized by gut microbiota, and short-chain fatty acids (SCFAs) level were increased after IPS1and IPS2 treatments. Furthermore, 16S rRNA sequencing analysis of fermentation samples were performed. Alpha-diversity, beta-diversity and taxonomic composition differences analysis revealed that IPS1 and IPS2 promoted the proliferation of beneficial bacteria and modulated the overall structure of gut microbiota. Taxonomic comparison analysis found that IPS1 increased the relative abundances of beneficial bacteria including Megamonas , Bifidobacterium and Lactobacillus , while IPS2 could increase the abundance of Bacteroides , Parabacteroides and Phascolarctobacterium. In addition, they can also decrease the levels of pathogenic bacteria containing Escherichia-Shigella , Klebsiella and Fusobacterium . These results indicated that IPS from P. cicadae TJJ1213 could be used as potential candidates for new functional foods.

Dual-FRET platform based on dsDNA-functionalized Ti3C2Tx MXene for highly sensitive detection of bacteria

Langjin Zhang,Yingwen Wang,Yubing Jiao,Min Mao,Weiliang Xiong,Yingjie Zhang,Youning Wang,Songbai Zhang,Wei Zhang,Jinguang Yang

Pathogenic bacteria represent a considerable threat to public health and food safety, necessitating the development of advanced detection methodologies. In this study, a novel fluorescence sensing platform based on fluorescence resonance energy transfer (FRET) was established to detect Pathogenic bacteria with rapid response, exceptional sensitivity, and stable selectivity. Using carboxyl-modified Ti 3 C 2 T x MXene (Ti 3 C 2 T x -COOH) as nanocarriers, double-stranded DNA (dsDNA) were assembled to functionalize the MXene for rapidly efficient quenching of fluorescence dye-labeled aptamers and specific recognition of target bacteria. Upon the incorporation of target bacteria, the aptamer preferentially bound to bacteria, dissociating from the MXene and resulting in a significant and rapid increase in fluorescence intensity. Using E. coli O157:H7 and S. aureus as exemplar biotargets, the sensor demonstrated ultra-sensitive detection of E. coli O157:H7 and S. aureus with limits of detection (LOD, S/N = 3) as low as 0.61 CFU/mL and 1.62 CFU/mL, respectively, within a rapid response time of 15 min, thereby demonstrating superior performance in comparison to the majority of existing FRET-based biosensors. Moreover, this method demonstrates excellent reproducibility and stability, having been successfully applied to spiked milk samples. The recovery rates for S. aureus ranged from 90.2 % to 114.3 %, with acceptable RSDs of 1.4 % to 7.4 %, while those for E. coli O157:H7 ranged from 91.7 % to 93.6 %, with acceptable RSDs of 5.2 % to 8.7 %, confirming its strong potential for practical sample analysis. The present study establishes a foundational framework for subsequent research into biosensors for detecting targets and provides a new pathway for the analysis of other biological factors.

Diagnosis of Bacterial Plant Diseases via a Nitroreductase-Activated Fluorescent Sensor

Yi-Xuan Fu,Wu-Yingzheng Guo,Nan Wang,Yi-Jie Dai,Zi-Ye Zhang,Xin-Lin Sun,Wen-Chao Yang,Guang-Fu Yang

Plant diseases caused by bacteria have become one of the serious problems that threaten human food security, which led to the remarkable reduction of agricultural yields and economic loss. Nitroreductase (NTR), as an important biomarker, is highly expressed in bacteria, and the level of NTR is closely related to the progression of pathogen infection. Therefore, the design of small-molecule fluorescent sensors targeting NTR is of great significance for the detection and diagnosis of plant pathogenic bacteria. In this study, a new fluorescent sensor targeting NTR was discovered and then successfully applied to the imaging of zebrafish and pathogenic bacteria. Most importantly, the developed sensor achieved the real-time diagnosis of Brassica napus L. infected with bacteria, which provides a promising tool for examining the temporal and spatial infection of plant pathogens in precision agriculture.

Magnetic biochar/quaternary phosphonium salt reduced antibiotic resistome and pathobiome on pakchoi leaves

Zhi Mei,Yuhao Fu,Fang Wang,Leilei Xiang,Fang Hu,Jean Damascene Harindintwali,Mingyi Wang,Marko Virta,Syed A. Hashsham,Xin Jiang,James M. Tiedje

Antibiotic resistance genes (ARGs) and human pathogenic bacteria (HPB) in leafy vegetable is a matter of concern as they can be transferred from soil, atmosphere, and foliar sprays, and poses a potential risk to public health. While traditional disinfection technologies are effective in reducing the presence of ARGs and HPB in soil. A new technology, foliar spraying with magnetic biochar/quaternary ammonium salt (MBQ), was demonstrated and applied to the leaf surface. High-throughput quantitative PCR targeting 96 valid ARGs and 16 S rRNA sequencing were used to assess its efficacy in reducing ARGs and HPB. The results showed that spraying MBQ reduced 97.0 ± 0.81% of “high-risk ARGs”, associated with seven classes of antibiotic resistance in pakchoi leaves within two weeks. Water washing could further reduce “high-risk ARGs” from pakchoi leaves by 19.8%− 24.6%. The relative abundance of HPB closely related to numerous ARGs was reduced by 15.2 ± 0.23% with MBQ application. Overall, this study identified the potential risk of ARGs from leafy vegetables and clarified the significant implications of MBQ application for human health as it offers a promising strategy for reducing ARGs and HPB in leafy vegetables.

Ga3+-doped coenzyme-based adhesives for periodontitis treatment

Ying Qi,Jing Zheng,Qian Zhang,Yage Sun,Jiaxing Shao,Hongying Wang,Rong Yang,Ziyang Xu,Chunyan Cui,Wenguang Liu

Chronic periodontitis is a long-standing thorny issue for oral health, characterized primarily by the excessive production and accumulation of reactive oxygen species, pathogenic bacterial infection, and the continuous loss of alveolar bone. In the present study, a poly(lipoic acid) (PolyLA) coenzyme-based tissue adhesive (PolyLA/Arg/Ga) was developed for the treatment of periodontitis. It functions by combining the nonantibiotic antimicrobial ion gallium ion (Ga3+ ) to resist pathogenic bacteria and promote osteogenesis; while the basic arginine not only stabilizes PolyLA at room temperature by forming salt bridge hydrogen bonds with its carboxyl groups but also serves as a cofactor, effectively regulating the release behavior of LA-based active molecules and Ga3+ by partially deprotonating the carboxyl groups on PolyLA. This, in turn, imparts significant antioxidant and antimicrobial properties to the adhesive. Thanks to the rich adhesive carboxyl groups on the surface of the adhesive and the structural stability of the hydrophobic PolyLA skeleton in a wet environment, The PolyLA/Arg/Ga exhibits long-lasting wet-resistant adhesion properties to biological tissues. Moreover, in vitro cell experiments and in vivo subcutaneous implantation studies demonstrate that the PolyLA/Arg/Ga adhesive has better cytocompatibility and histocompatibility. The PolyLA/Arg/Ga is explored as a tissue adhesive to treat a periodontitis model in rats. The results demonstrate that the PolyLA/Arg/Ga adhesive more effectively inhibited alveolar bone resorption and promoted periodontal tissue regeneration within 4 weeks compared to commercial patch. Thus, the simply designed yet multifunctional PolyLA/Arg/Ga bioadhesive offers an innovative approach for periodontitis management.
