Protective mechanism of lutein combined with EGCG against N-methyl-N nitrosourea-induced retinitis pigmentosa

Huili Jing,Meimei Nie,Xiaoqin Wang,Zhongyuan Zhang,Yayuan Xu,Guodong Zhang,Dajing Li,Zhuqing Dai

In this study, N-methyl-N nitrosourea (MNU)-induced retinitis pigmentosa (RP) model mice were used to investigate the protective effect of lutein (LUT) combined with epigallocatechin-3-gallate (EGCG) treatment. Compared to the LUT group, after the LUT combined with EGCG (LUT-EGCG) treatment, the accumulation content of LUT was significantly increased by 50.24% in the liver. The histopathological results showed that LUT-EGCG treatment significantly improved the structural damage of the retina and recovered the thickness of the outer nuclear layer of the retina to 185.28 ± 0.29 μm, which was no significant difference compared to the control group. The LUT-EGCG treatment also increased the production of short-chain fatty acids such as acetic and propionic acids. Compared with LUT individual treatment, the LUT-EGCG treatment significantly increased the relative abundance of Lachnospiraceae and Helicobacteraceae by Metastats analysis. RT-qPCR results showed that LUT-EGCG treatment significantly increased the antiapoptotic gene Bcl-2 expression. In addition, the expression level of IL-6 in the LUT-EGCG group was down-regulated significantly, but there was no significance in NF-κβ, TNF-α, IL-1β, and IL-18 compared with LUT treatment. Coupled with correlation analysis, results showed that LUT combined EGCG might protect against RP by regulating antiapoptotic gene expression and improving intestinal flora. These results provided helpful information for RP intervention.

Photoreceptor-Mimetic Microflowers for Restoring Light Responses in Degenerative Retina through a 2D Nanopetal/Cell Biointerface

Gerile Oudeng,Seema Banerjee,Qin Wang,Ding Jiang,Yadi Fan,Honglian Wu,Feng Pan,Mo Yang

Retinitis pigmentosa is the main cause of inherited human blindness and is associated with dysfunctional photoreceptors (PRs). Compared with traditional methods, optoelectronic stimulation can better preserve the structural integrity and genetic content of the retina. However, enhancing the spatiotemporal accuracy of stimulation is challenging. Quantum dot-doped ZnIn 2 S 4 microflowers (MF) are utilized to construct a biomimetic photoelectric interface with a 0D/3D heterostructure, aiming to restore the light response in PR-degenerative mice. The MF bio interface has dimensions similar to those of natural PRs and can be distributed within the curved spatial region of the retina, mimicking cellular dispersion. The soft 2D nano petals of the MF provide a large specific surface area for photoelectric activation and simulate the flexibility interfacing between cells. This bio interface can selectively restore the light responses of seven types of retina ganglion cells that encode brightness. The distribution of responsive cells forms a pattern similar to that of normal mice, which may reflect the generation of the initial “neural code” in the degenerative retina. Patch-clamp recordings indicate that the bio interface can induce spiking and postsynaptic currents at the single-neuron level. The results will shed light on the development of a potential bionic subretinal prosthetic toolkit for visual function restoration.

The impacts of resveratrol on the retinal degeneration in a rat model of retinitis pigmentosa induced by alkylation: an in-vivo study

Weiming Yan,Yan Sun,Yutong Wang,Wangjiao Liang,Yuxin Xia,Weihua Yan,Meizhu Chen,Tao Chen,Dongliang Li

Upregulation of Sirtuin Type 1 (SIRT1), a nicotinamide adeninedinucleotide (NAD+)-dependent deacetylase, has been proved to protect against ample ocular diseases, while its effect on retinitis pigmentosa (RP) has not been illustrated. The study was aimed to explore the impacts of resveratrol (RSV), a SIRT1 activator, on the photoreceptor degeneration in a rat model of RP induced by N-methyl-N-nitrosourea (MNU), an alkylation. The rats were induced RP phenotypes via the intraperitoneal injection of MNU. The electroretinogram was conducted and revealed that RSV could not prevent the decline of retinal function in the RP rats. The optical coherence tomography (OCT) and the retinal histological examination were performed and showed that the reduced thickness of the outer nuclear layer (ONL) was not preserved by RSV intervention. The immunostaining technique was applied. Afther the MNU administration, the number of the apoptotic photoreceptors in the ONL throughout the retinasand the number of microglia cells present among the outer part throughout the retinas were not significantly reduced by RSV. Western blotting was also performed. The data showed that the level of SIRT1 protein was decreased after MNU administration, while RSV was not able to obviously alleviate the downregulation. Our data together indicated that RSV was not able to rescue the photoreceptor degeneration in the MNU-induced RP rats, which might be due to the MNU-induced consumption of the NAD+.

Synergistic effect of chitosan and β-carotene in inhibiting MNU-induced retinitis pigmentosa

Enjuan Shi,Xiaoqin Wang,Huili Jing,Yayuan Xu,Lei Feng,Fatao He,Dajing Li,Zhuqing Dai

In this study, N -Methyl-N-nitrosourea (MNU) was intraperitoneally injected to construct a mouse retinitis pigmentosa (RP) model to evaluate the protective effect of chitosan and β-carotene on RP. The results demonstrated that chitosan synergized with β-carotene significantly reduced retinal histopathological structural damage in RP mice. The co-treatment group of β-carotene and chitosan restored the retinal thickness and outer nuclear layer thickness better than the group treated with the two alone, and the thickness reached the normal level. The content of β-carotene and retinoids in the liver of chitosan and β-carotene co-treated group increased by 46.75 % and 20.69 %, respectively, compared to the β-carotene group. Chitosan and β-carotene supplement suppressed the expressions of Bax, Calpain2, Caspase3, NF-κB, TNF-α, IL-6, and IL-1β, and promoted the up-regulation of Bcl2. Chitosan and β-carotene interventions remarkably contributed to the content of SCFAs and enhanced the abundance of Ruminococcaceae , Rikenellaceae , Odoribacteraceae and Helicobacteraceae . Correlation analysis demonstrated a strong association between gut microbiota and improvement in retinitis pigmentosa. This study will provide a reference for the study of the gut-eye axis.
