Herein, by the pyrolysis and alkali leaching of zeolitic imidazolate framework-8 (ZIF-8)@SiO2, followed by Fe and Co doping, a novel Fe1Co1/NC nanozyme, with adjacent Fe and Co dual-atom pairs decorated on an N-doped carbon support, is exactly built. Due to the synergistic effect of adjacent Fe and Co dual-atom pairs, the peroxidase-like activity of Fe1Co1/NC is significantly enhanced, far exceeding those of Fe1/NC and Co1/NC. Based on this, we propose a simple colorimetric electronic tongue consisting of Fe1Co1/NC, chromogenic substrates (3,3′,5,5′-tetramethylbenzidine) (TMB), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and o-phenylenediamine (OPD) and H2O2 to identify seven active substances in licorice. The mechanism of action of this tongue is that different active substances have differential degrees of inhibition on the three nanozyme-catalyzed reactions (Fe1Co1/NC-TMB-H2O2, Fe1Co1/NC-ABTS-H2O2, and Fe1Co1/NC-OPD-H2O2). The collected absorbance variation data, i.e., the colorimetric fingerprint patterns, are analyzed by linear discriminant analysis, realizing the identification of the seven active substances with 100% accuracy. This work provides insights into the rational design of dual-atom catalysts with nanozyme activities for the discrimination of multiple targets.

Quality control and screening of active substances in traditional Chinese medicines have been performed using fingerprint analysis. The spectrum-effect relationship between chromatography fingerprints and efficacy of herbal drugs is considered as a potentially useful method for determining active ingredients in complex mixtures. The study was designed to develop a method for determining the bioactive components of a compound Chinese medicine called Tiefeng based on spectrum-effect relationships between high-performance liquid chromatography (HPLC) fingerprints and anticoccidial activities. Four peaks of the established HPLC fingerprint indicate the main bioactive components of this medicine. In addition, pharmacodynamic atlas was defined and used to assess the anticoccidial activity of Tiefeng from different sources for the first time. We found that the level of anticoccidial activity of Tiefeng was consistent with the degree of similarity between the pharmacodynamic atlas and chromatogram of any sample. Furthermore, effect of this medicine was related with the main active constituents, along with the origin and the harvesting time.

Introduction Huangma Tincture (HMT) is a Chinese patent medicine with a history of clinical use for more than 60 years, widely used for treating dermal chronic ulcer such as diabetic foot ulcer. However, the overall quality evaluation and control method of HMT has not yet been researched. Objective The aim of this study is to establish a comprehensive quality evaluation and control method for HMT based on high-performance liquid chromatography (HPLC) fingerprint, chemometrics, and multicomponent content determination. Methods Establishing chemical fingerprint of HMT and carrying out similarity analysis comprehensively reflect the consistency of the formulation in terms of chemical composition. Chemometrics analyses, including hierarchical cluster analysis (HCA), principal component analysis (PCA), and orthogonal partial least squares-discriminant analysis (OPLS-DA), were performed to identify components crucial to quality differences. The content of five potential bioactive ingredients (brucine, strychnine, jatrorrhizine, coptisine, and berberine hydrochloride) was determined as a supplementary quality control measure. Results HPLC fingerprint of HMT with similarity index > 0.990 was established, in which five common compounds (brucine, strychnine, jatrorrhizine, coptisine, and berberine hydrochloride) were identified. HCA, PCA, and OPLS-DA results, validated through 200 permutation tests, were basically consistent. The contents of brucine, strychnine, jatrorrhizine, coptisine, and berberine hydrochloride in 20 batches of HMT samples were 0.2088–0.5556, 0.2599–0.9868, 0.1358–0.2092, 0.2634–0.6843, and 1.8301–2.7826 mg/mL, respectively. Conclusion HPLC fingerprint combined with chemometrics and multicomponent content determination considering both effect and toxicity provides a robust method for the comprehensive quality evaluation and control of HMT.

Angelicae Sinensis Radix is a world-renowned herbal medicine originating in China. Owing to many environmental and geographical factors, Angelicae Sinensis Radix from various origins may have a difference in the content of ingredients, which made the confusion in the clinical practice and market. Herein, a binary chromatographic fingerprinting analysis method is developed via hydrophilic interaction chromatography and reversed-phase liquid chromatography to obtain more chemical information. Following that, an ultra-performance liquid chromatography with a triple quadrupole mass spectrometry method is furnished to simultaneously detect 17 ingredients of Angelicae Sinensis Radix gathered from six geographic zones in China. Eventually, the principal component analysis is successfully carried out to classify and differentiate the Angelicae Sinensis Radix from different origins, meanwhile the quantitative volcano plots was used to observe the changes of ingredient trends vividly. Accordingly, the proposed binary chromatography and triple quadrupole tandem mass spectrometry coupled with multivariate statistical analysis can be utilized as a facile and reliable method for origin tracing and quality control of Angelicae Sinensis Radix .

Background Many studies have assessed the fingerprint and quantitative analysis of Ginkgo biloba preparations, but the fingerprint mainly focuses on flavonoid glycosides. However, according to our previous study, the differences among diverse manufacturers mainly involve organic acids. Methods A novel reverse-phase liquid chromatography assay using diode array detection was developed for evaluating Ginkgo biloba preparations for quality based on a chromatographic fingerprint allowing the simultaneous assessment of eleven compounds, including four organic acids, six flavonol glycosides and one flavonoid aglycone. And the method was applied to 51 batches of Ginkgo biloba preparations from manufacturers in China. Chemometric approaches were performed for evaluating 51 batches of Ginkgo biloba preparations from various manufacturers. Results The similarity values among the chromatograms of 51 samples ranged from 0.45 to 1.00, showing that the quality of Ginkgo biloba preparations produced by different manufacturers varied greatly. Data analysis of the 51 batches of GBP samples suggested significant variations of the total contents of all 11 targets, also demonstrating the quality difference of GBP samples. There were significant differences in organic acids in particular. Conclusion Combining the chemical fingerprint and quantitative assessment revealed significant variations in the examined commercial products with regard to organic acids. Thus, this study provided a more comprehensive tool for monitoring the quality consistency of Ginkgo biloba preparations.

Polygonatum sibiricum(P. sibiricum) is a significantly health-promoting plant unique in medicine and food. Currently, research on the bioactive components ofP. sibiricumprimarily focuses on polysaccharides. According to theChinese Pharmacopoeia, the polysaccharide content inP. sibiricummust be at least 7.0%, which is the only criterion used for determining its content. In contrast, this study aims to thoroughly investigate and clarify the various components that contribute to the therapeutic and functional properties ofP. sibiricum. We seek to broaden the focus beyond polysaccharides to identify other potentially significant bioactive substances. We established High-Performance Liquid Chromatograph (HPLC) fingerprints for wine-processedP. sibiricumfrom various regions and identified 17 common peaks. The antioxidant activities of these components were assessed using ABTS and DPPH methods. The spectrum–effect relationship was elucidated through partial least squares regression and grey relational analysis. The results revealed that the antioxidant active components in wine-processedP. sibiricuminclude 5-hydroxymethylfurfural,p-hydroxycinnamic acid, myricetin, caffeic acid, vanillic acid, and adenosine. This research not only clarifies the antioxidant material basis of wine-processedP. sibiricumbut also establishes a scientific foundation for enhanced quality control in future applications.

This study integrates HPLC fingerprinting and NIR spectroscopy to evaluate the antioxidant capacity of Rheum tanguticum . HPLC fingerprinting comprehensively characterized the complex chemical composition, identifying 13 compounds. Meanwhile, NIR spectroscopy provided a rapid, non-destructive approach to predict antioxidant capacity. The combination of these techniques offers a holistic and efficient strategy for evaluating antioxidant activity. The spectrum-effect relationships were analyzed by correlating the results of three antioxidant assays (ABTS, DPPH, and FRAP) with the peak areas of the common peaks identified by HPLC. Glycosylated anthraquinones, particularly conjugated forms like rhein-8-O-glucoside, were found to synergistically enhance antioxidant activity with free aglycones. NIR models optimized using the PLS method with Python-based algorithms demonstrated strong predictive abilities, achieving RPD values of 2.43 for ABTS, 2.63 for DPPH, and 2.43 for FRAP. This research provides a rapid method to evaluate the antioxidant capacity of Rh. tanguticum , offering a reference for future applications in other fields.

Guang Dilong is a Traditional Chinese Medicine prepared from the dried body of Pheretima aspergillum (E. Perrier), a species of earthworm. However, preparations of Guang Dilong may be adulterated by other species and a method of quality control is needed. A method was developed to analyze and authenticate commercial Guang Dilong, utilizing ultra-high performance liquid chromatography (UHPLC) coupled with diode array detection (DAD). Equipment included an Acquity UPLC HSS T3 column (100 mm × 2.1 mm, 1.8 μm). The mobile phase consisted of acetonitrile and 0.01% formic acid, pumped at 0.3 mL/min. Wavelength detection was at 260 nm. Twenty-two batches of confirmed P. aspergillum samples (reference) from different sources and 20 batches of adulterated samples were analyzed to establish a reference fingerprint for commercial Guang Dilong. Five peaks in the fingerprints of the reference batches were identified as characteristic; six characteristic peaks in the fingerprints of the adulterants were identified by comparing their retention time with those of the references. The total 42 batches of samples were compared with the reference fingerprint, and the fingerprints of the P. aspergillum samples were similar. The UHPLC-DAD method can simultaneously determine the contents of six compounds (hypoxanthine, xanthine, uridine, inosine, guanosine, and adenosine) in the reference and adulterated batches. The six compounds showed good regression (r > 0.9999) within test ranges. The recovery (accuracy) was 98.25 to 101.68%, with relative standard deviation <2.67%. In summary, this UHPLC-DAD method combines chromatographic fingerprint with quantification analysis and could be readily used as an efficient quality control method for Guang Dilong.

Traditional Chinese medicine consists of complex phytochemical constituents. Selecting appropriate analytical markers of traditional Chinese medicine is a critical step in quality control. Currently, the combination of fingerprinting and efficacy evaluation is considered as a useful method for screening active ingredients in complex mixtures. This study was designed to develop an orthogonal partial least squares model for screening bioactive quality control markers of QishenYiqi dripping pills based on the fingerprint–efficacy relationship. First, the chemical fingerprints of 49 batches of QishenYiqi dripping pill samples were established by ultra-high performance liquid chromatography coupled with a photodiode array detector. Second, ultra-high performance liquid chromatography coupled with quadrupole-time-of-flight mass spectrometry was exploited to systematically investigate the 36 copossessing fingerprint components in QishenYiqi dripping pills. The vascular protective activity of QishenYiqi dripping pills was determined by using a cell counting kit-8 assay. Finally, fingerprint–efficacy relationship was established by orthogonal partial least squares model. The results indicated that ten components exhibited strong correlation with vascular protective activity, and these were preliminarily screened as quality control markers. The present study provided a novel idea for the study of the pharmacodynamic material basis and quality evaluation of QishenYiqi dripping pills.

Nigella glandulifera Freyn et Sint seeds (NG) as well as Nigella sativa L. seeds (NS) have been employed as folk remedies for centuries. However, little work has been done on their quality control. In order to assess the quality and guarantee therapeutic effects, the chemical constituents of the two herbs were extensively characterized by ultra-performance liquid chromatography coupled with electrospray time-of-flight mass spectrometry (UPLC-Q-TOF/MS), and a simple, practical, and reliable HPLC fingerprinting method was established and applied to determination of 20 batches of samples. With their chromatographic fingerprints, the two species were successfully differentiated by pattern recognition methods, namely, principal component analysis (PCA) and partial least square discriminant analysis (PLS-DA). Results from this study show that the proposed analytical methods can be applied to the quality control of the seeds and have high potential to ensure their safety and efficacy.

A classification method of nutritional food Rana chensinensis ovum (RCO) was established on the basis of unsaturated fatty-acid (UFA) profiling and high-performance liquid chromatography (HPLC) fingerprints in combination with quantitative analysis of multi-components by single-marker (QAMS). The 19 batches of RCO samples were collected from the main breeding forests in northeast China. HPLC analytical method of UFAs in RCO was established by optimizing chromatographic conditions. The RCO samples from different forest regions were used to establish two kinds of HPLC fingerprints, which matched chromatographic peaks with the peak area ratio >0.5% and 7 identified UFAs. A total of 11 common peaks were obtained, of which 7 peaks were identified as eicosapentaenoic acid (EPA), α-linolenic acid (ALA), docosahexaenoic acid (DHA), arachidonic acid (ARA), docosapentaenoic acid (DPA), linoleic acid (LA) and oleic acid (OA). RCO samples were classified into three groups by hierarchical cluster analysis (HCA) and principal component analysis (PCA). Partial least squares discriminant analysis (PLS-DA) predicted differential components that may affect grade classification. The QAMS method established here showed good robustness and feasibility. ALA was designated as an internal reference, and the relative correction factors (RCFs) of the other six UFAs were calculated. Compared with the external standard method (ESM), there was no significant difference (P > 0.05, RE% between ±5.00% and cos θ > 0.9999) in the quantitative analysis of UFAs in RCO by QAMS, but the newly established method is more economical and timesaving. This work provides a comprehensive evaluation method for controlling the quality of RCO.
