TheArtemisia argyileaf (AL) has been used as a traditional medicine and food supplement in China and other Asian countries for hundreds of years. Phytochemical studies disclosed that AL contains various bioactive constituents. Among bioactive constituents, phenolic acids have been recognized as the main active compounds in AL. To the best of our knowledge, no research has been focused on extraction method for the bioactive phenolic acids from AL. Nowadays, deep eutectic solvents (DESs) are emerging as a new type of green and sustainable solvent for efficient extraction of bioactive compounds from natural products. In the present study, an environmentally friendly extraction method based on DESs was established to extract bioactive phenolic acids from ALs. Diverse tailor-made solvents, including binary and ternary DESs, were explored for simultaneous extraction of four phenolic acids (3-caffeoylquinic acid, 3,4-di-O-caffeoylquinic acid, 3,5-di-O-caffeoylquinic acid, and 4,5-di-O-caffeoylquinic acid) from AL. The results indicated that the ternary DES composed of a 2:1:2 molar ratio of choline chloride, malic acid, and urea showed enhanced extraction yields for phenolic acids compared with conventional organic solvents and other DESs. Subsequently, the extraction parameters for the four phenolic acids by selected tailor-made DESs, including liquid–solid ratios, water content (%) in the DESs, and extraction time, were optimized using response surface methodology and the optimal extraction conditions were: extraction time, 23.5 min; liquid–solid ratio, 57.5 mL/g (mL of DES/g dry weight of plant material); water content, 54%. The research indicated that DESs were efficient and sustainable green extraction solvents for extraction of bioactive phenolic acids from natural products. Compared to the conventional organic solvents, the DESs have a great potential as possible alternatives to those organic solvents in health-related areas such as food and pharmaceuticals. Keywords:deep eutectic solvents;Artemisia argyileaves;phenolic acids;extraction;response surface methodology

Artemisia argyi leaf is a well-known species in traditional Chinese medicine. However, the anti-inflammatory and activating blood stasis activities of its essential oil (AAEO) have not been explored in vivo. The present study measured the contents of three chemical components by gas chromatography (GC). The anti-acute inflammatory effects of AAEO were investigated in dimethyl benzene, glacial acetic acid and carrageenan-induced animals through skin administration or by oral gavage, respectively. The effects of AAEO on haemorheology were studied in a rat acute blood stasis model. The contents of eucalyptol, camphor and borneol in AAEO were 254.4, 51.6 and 58.7 mg/g, respectively. All dosages of AAEO by skin administration significantly decreased the swelling in dimethyl benzene-induced ear oedema and carrageenan-induced paw oedema, and reduced the permeability in glacial acetic acid-induced abdominal blood capillary ( p < 0.01). Meanwhile, haemorheology indexes such as whole blood viscosity and the erythrocyte aggregation index significantly decreased only in the high dosage group. In addition, the effects of AAEO by oral gavage were weaker than skin administration at the medium dose in the experiments. It suggests that AAEO has better absorption bioavailability and pharmacological effects through skin administration due to the better skin permeability of essential oil than gastrointestinal absorption.

To reveal the utilization value of leaf, stem, and root of Artemisia argyi , a rapid online liquid microextraction combined with a high-performance liquid chromatography coupled with 2,2-nitrogen-di (3-ethyl-benzothiazole-6-sulfonic acid) diammonium salt antioxidant assay system was established for analysis of antioxidants in the leaf, stem, and root of A. argyi , and a calibration quantitative method of antioxidant activity with equivalent chlorogenic acid was proposed. Thirty-three positive peaks were identified; among them, 12 compounds were found that possess good antioxidant activity including eleven organic acids (components 2–4, 8, 11–14, 17, 19, and 21) and one flavonoids (component 22). The proposed calibration quantitative method avoided the influence of content of compound and compared the extent of radical scavenging capacity of five antioxidant compounds, which were ranked as follow: 3,5-dicaffeoylquinic acid > 3,4-dicaffeoylquinic acid ≈ 4,5-dicaffeoylquinic acid > 1,4-dicaffeoylquinic acid > chlorogenic acid. In conclusion, this study provided composition and biological potential for the future development of the leaf, stem, and root of A. argyi . It is believed that the online liquid microextraction combined with high-performance liquid chromatography based antioxidant assay system can be widely used for the rapid screening of natural antioxidant components in the different parts of natural products.

Nanotechnology contributes to the development of biopesticides with reduced toxicity, increased stability, improved biosafety, and higher activity against targets. It has demonstrated great potential as a solution for practical applications of essential oils. Herein, the HLB method developed two Artemisia argyi essential oil-loaded nanoemulsions (AEOnano). Further systematically optimizing the ratio led to small droplet size (<200 nm) and low PDI (<0.3). As anticipated, the AEOnano demonstrated robust physical stability when subjected to various stressors. Notably, both indoor and outdoor pot experiments demonstrated that AEOnano outperformed ordinary A. argyi essential oil aqueous emulsions (AEOAE) in weed control. These nanoemulsions allowed a 2–4 times reduction in AEO dosage while maintaining the same weed-killing efficacy. The superior improvement attributed to the nanoemulsions enhanced leaf-wetting capability, improved leaf retention, and resistance to wash-off. In addition, biosafety assessment indicated that AEOnano did not affect the plants ( Artemisia argyi , Chrysanthemum morifolium , cotton), which provide orientation for its practical application. Therefore, this study introduces a promising approach to enhance the effectiveness of AEO-based herbicides, which will have a broad application prospect in sustainable agriculture.

Background The hepatotoxicity induced by acetaminophen (APAP), a commonly used antipyretic, analgesic and anti-inflammatory drug in clinical practice, has received accumulated attention. Artemisia argyi essential oil (AAEO), a volatile oil component extracted from traditional Chinese medicine Artemisia argyi H.Lév. & Vaniot, has great hepatoprotective effects. However, the potential role of AAEO in APAP-induced hepatotoxicity has not been characterized. The present study aimed to investigate the effects of AAEO on hepatic metabolic changes in mice exposed to APAP. Methods In this study, 300.00 mg/kg acetaminophen was used to establish liver injury model in C57BL/6 J mice. Hepatoprotective effect of AAEO on APAP-induced hepatotoxicity in mice was investigated by detecting liver function enzymes and histopathological examination. Secondly, UPLC-MS/MS was used to analyze the to analyze the small molecule metabolites and metabolic pathways induced by AAEO treatment; In addition, the effect of AAEO on APAP-induced oxidative stress and inflammation were evaluated by detecting the levels of glutathione peroxidase 4, malondialdehyde, reactive oxygen species and inflammatory factors. Finally, the active components of AAEO were preliminarily screened by cellular assays. The hepatoprotective effect of AAEO against APAP-induced hepatotoxicity was examined through the Western blotting, after the CYP2E1 gene was knocked down in AML12 cells by siRNA transfection. Results Compared with the APAP group, AAEO could reduce the abnormal increase in the levels of liver function enzymes caused by APAP. AAEO could enhance the antioxidant capacity by down-regulating the biosynthesis pathway of unsaturated fatty acids and promoting the activity of antioxidant enzymes SOD and CAT in liver tissue induced by APAP. Our study revealed that AAEO promoted GSH synthesis and covalently combined to form APAP-GSH conjugates to reduce the accumulation of APAP in liver tissue. In addition, the chemical constituents in AAEO were analyzed by GC–MS/MS, and it was determined to identify that dihydro-beta-ionone and (-)-verbenone in AAEO might have a significant protective effect on hepatocyte survival after APAP exposure. Further studies on the hepatoprotective mechanism of AAEO indicated that it might reduce the production of toxic metabolites by regulating CYP2E1 levels. Conclusion AAEO exerted hepatoprotective effects on acetaminophen-induced hepatotoxicity in mice via regulating the activity of CYP2E1 and regulating the γ-glutamyl cycle pathway.

In this study, Artemisia argyi essential oil (AAEO), extracted from the leaves of Artemisia argyi , was employed as a green extraction solvent for in-syringe kapok fiber-supported liquid-phase microextraction (KF-SLME) of hydrophobic compounds in aqueous samples. The wheat of natural kapok fiber as the support medium was 2 mg, and the optimum volume of the extraction solvent AAEO was 12.5 μL. The volume of green desorption solvent 75 % ethanol was 100 μL, which realized a highly environmentally friendly sample preparation method. By simply pulling and pushing the syringe plunger five times, the extraction process enabled rapid and convenient enrichment of analytes while removing most interferences. Coupled with liquid chromatography-tandem mass spectrometry (LC-MS/MS), the AAEO-based in-syringe SLME method was applied as a proof-of-concept study to determine triazole fungicides (TFs) residues in fruit juices, herbal oral liquids and herbal extract granules. Employing in-syringe KF-SLME prior to LC-MS/MS detection significantly reduced the limits of quantification (LOQs) by 5–50-fold. The integrated AAEO-based in-syringe KF-SLME-LC-MS/MS method demonstrated satisfactory linearity (R 2 ≥ 0.995), accuracies (86.5 %–115.1 %), and precisions (1.2 %–12.7 %) using an isotope calibration method. Furthermore, three greenness assessment metrics uniformly highlighted its outstanding environmental friendliness. The proposed method offers competitive advantages in terms of its environmentally friendly approach, detection sensitivity, analysis cost, and operational convenience.

Artemisia argyi H.Lév. & Vaniot, a traditional Chinese medicine with a history spanning over two millennia, has been extensively used in folk medicine to treat dysmenorrhea, uterine bleeding and inflammation. Recent studies have demonstrated that the essential oil extracted from Artemisia argyi H.Lév. & Vaniot, known as AAEO, exhibits significant anti-tumor properties against liver and lung cancers. There is a scarcity of research on the potential impact of AAEO on pancreatic cancer (PC) cells. In this study, UPLC-MS/MS-based metabolomics method was established to evaluate the effect of AAEO on the proliferation of PC cells. The differential compounds included 5-oxoproline, glutamate, γ-glutamylcysteine, glutathione, arachidonic acid, adrenal acid and linoleic acid were detected by metabolomics, enriching in the γ-glutamyl cycle and polyunsaturated fatty acid metabolism, which were closely related to ferroptosis. Meanwhile, AAEO dramatically increased the levels of intracellular iron ion via up-regulation of TFR1, augmented reactive oxygen species and malondialdehyde in a dose-dependent manner by down-regulation of γ-glutamyl cycle through decreasing expressions of SLC7A11. Additionally, β-caryophyllene oxide, one of the main components of AAEO, could covalently bind to Cys in SW1990 cells to form a conjugate Cpo-Cys, resulting in the inhibition of glutathione synthesis. Importantly, the ferroptosis inhibitor deferoxamine significantly blocked the inhibitory effect of AAEO on SW1990 cells. Meanwhile, β-caryophyllene oxide, dihydro-β-ionone and α-bisabolol had strong binding force with GPX4, SLC7A11 and TFR1, respectively. These findings showed that AAEO induced ferroptosis via regulation of γ-glutamyl cycle by SLC7A11 and iron disorders by TFR1. Our study discovered AAEO as a potential therapeutic approach to induce ferroptosis to prevent or treat PC.

Artemisia argyi , commonly known as wormwood, is a traditional Chinese herbal food and medicine celebrated for its notable antibacterial and anti-inflammatory properties. This study explores a novel delivery method for wormwood, aiming for more convenient and versatile applications. Specifically, we present the first investigation into combining wormwood with microstructures to create a microneedle (MN) patch for wound healing. The wormwood microneedle (WMN) patch is formulated with milled wormwood sap, calcium carbonate, and sodium hyaluronate. The addition of 0.3% (w/v) sodium hyaluronate enhances the mechanical strength of the WMN patch. Pectin, derived from wormwood, is combined with calcium carbonate to create a gelatinous and solidified substance. The WMN patch exhibits a well-defined shape and sufficient mechanical strength to penetrate the epidermis, as confirmed by our results. In vitro experiments demonstrate the biocompatibility of the WMN patch with fibroblasts and highlight its antibacterial and anti-inflammatory properties. Furthermore, the patch facilitates collagen deposition at the wound site. In an excisional rat model, the WMN patch significantly accelerates the wound closure rate compared to the control group. Our findings suggest that the WMN patch has the potential to serve as a natural treatment for wound healing. Additionally, this approach can be extended to other biologically active substances with similar physiochemical characteristics in future applications. Graphical Abstract

A thrombin-activity-based electrochemiluminescence (ECL) biosensor was successfully constructed using tungsten oxide quantum dots (WO 3-x QD S ) as the co-reactant and thrombin-cleavable peptides as the recognizer. Specifically, Ru(bpy) 3 2+ were doped on silica nanoparticles (Ru@SiO 2 ), which greatly enhanced the ECL potential. AuNPs@WO 3-x QDs composite was then prepared to accelerate electron transfer and improve the ECL signal by 219 times. Under ideal conditions, the limit of detection for thrombin in serum was determined to be 0.28 μU/mL with a linear range from 1 μU/mL to 1 U/mL. In addition, the developed ECL biosensor was used to screen for thrombin inhibitors from 12 compounds in Artemisiae Argyi Folium. Among the compounds tested, it was observed that 100 μmol/L luteolin exhibited a significantly higher inhibition rate (exceeding 80%) compared to apigenin , isorhamnetin , naringin , or eriodictyol . In an in-vitro anticoagulation experiment, luteolin (100 μmol/L) prolonged APTT by 49%, and the molecular docking assay indicated that luteolin had binding sites of Gly219 and Asp189 in the active pockets of thrombin. This may have been the main reason underpinning luteolin's anticoagulation effects. Overall, the Ru@WO 3-x QD S ECL biosensor provided a reliable strategy for thrombin activity assay and screening of anticoagulant agents.

Medical wound dressing with natural antibacterial efficacy is an important issue. However, few studies on Artemisia argyi oil based wound dressing have been studied because of the volatility of the Artemisia argyi oil. In this study, Artemisia argyi oil -microcapsules (AAO-MC)/PVC fibrous membrane were fabricated through emulsification-internal gelation and electrospinning method. The membranes were characterized by SEM, Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), and particle diameter, hydrophilicity, breathability, water vapor transmission rate (WVTR), and antibacterial performance were evaluated. The results show that AAO-MC microcapsules excellent stability, and the slow-release property. Moreover, the prepared AAO-MC/PVC fibrous membrane indicated good WVT (72.47 g/(m²·h)), high hydrophilicity (30.5°), and outstanding antibacterial rate (94.3%). This study provides a new strategy to prepare microcapsule nanofibrous membrane with excellent performance and is suggested to be a promising candidate for medical wound dressings.

The environmental pollutant bisphenol A (BPA), used in the manufacture of plastic packaging materials for various diets, is widely distributed in the environment and causes severe hepatotoxicity by inducing oxidative stress. Artemisia argyi essential oil (AAEO), a volatile oil component isolated from Artemisia argyi H.Lév . & Vaniot , has pharmacological effects, especially for hepatoprotective actions. However, the potential effect of AAEO in BPA induced hepatotoxicity has not been characterized. First, we analyzed the chemical composition in AAEO by gas chromatography–mass spectrometry. Herein, we investigated the effect of AAEO on hepatic metabolic changes in mice exposed to BPA. Results showed that compared with the BPA group, AAEO could reduce the level of liver function enzymes in BPA mice serum, and ameliorate hepatic lesions and fibrosis. Additionally, 20 differential metabolites screened by metabolomics were mainly involved in the reprogramming of glutathione metabolism, purine metabolism, and polyunsaturated fatty acid synthesis. Moreover, AAEO could reduce hepatic ferroptosis induced by BPA, as demonstrated by reducing xanthine oxidase activity, up-regulating the activities of glutathione peroxidase 4 (GPX4), superoxide dismutase, and catalase and the expression of SLC7A11 to promote the glutathione synthetic, while inhibiting transferrin receptor 1 (TFR1) expression to reduce the accumulation of Fe 2+ in cells. Therefore, our study identified AAEO as a hepatic protectant against BPA-induced hepatotoxicity by reversing the occurrence of ferroptosis.
