Hypertrophic scars (HS) still remain an urgent challenge in the medical community. Traditional Chinese medicine (TCM) has unique advantages in the treatment of HS. However, due to the natural barrier of the skin, it is difficult for the natural active components of TCM to more effectively penetrate the skin and exert therapeutic effects. Therefore, the development of an efficient drug delivery system to facilitate enhanced transdermal absorption of TCM becomes imperative for its clinical application. In this study, we designed a compound Salvia miltiorrhiza–Blumea balsamifera nanoemulsion gel (CSB-NEG) and investigated its therapeutic effects on rabbit HS models. The prescription of CSB-NEG was optimized by single-factor, pseudoternary phase diagram, and central composite design experiments. The results showed that the average particle size and PDI of the optimized CSB-NE were 46.0 ± 0.2 nm and 0.222 ± 0.004, respectively, and the encapsulation efficiency of total phenolic acid was 93.37 ± 2.56%. CSB-NEG demonstrated excellent stability and skin permeation in vitro and displayed a significantly enhanced ability to inhibit scar formation compared to the CSB physical mixture in vivo. After 3 weeks of CSB-NEG treatment, the scar appeared to be flat, pink, and flexible. Furthermore, this treatment also resulted in a decrease in the levels of the collagen I/III ratio and TGF-β1 and Smad2 proteins while simultaneously promoting the growth and remodeling of microvessels. These findings suggest that CSB-NEG has the potential to effectively address the barrier properties of the skin and provide therapeutic benefits for HS, offering a new perspective for the prevention and treatment of HS.

Adinandra nitida tea (Shiyacha) is a traditional eminent and flourishing tea with a long history in Southeast Asia. However, transformation of phytochemicals of A. nitida tea infusion during in vitro simulated digestions is vague. Herein, we investigated cellular antioxidant and antiproliferative activity of phytochemicals obtained from A. nitida tea infusion subjected to in vitro gastrointestinal conditions. Phenolic, flavonoid, flavonol, and anthocyanin contents found in A. nitida tea infusion were evaluated by normal colorimetric methods. Subsequently, the main compounds were determined by high-performance liquid chromatography. The antioxidant activity was evaluated based on peroxyl radical scavenging capacity (PSC), oxygen radical absorbance capacity (ORAC), and cellular antioxidant activity (CAA). The antiproliferative activity towards Caco-2 cells was determined by the cell methylene blue colorimetric assay. It was found that in simulated gastrointestinal fluids, the contents of phenolics and flavonols increased, whereas the contents of flavonoids and anthocyanins decreased. Moreover, the content of (−)-epigallocatechin and camellianin B increased, whereas that of L-epicatechin, quercitrin, and camellianin A decreased. Furthermore, the oxygen radical scavenging capacity and cellular antioxidant activity values increased, whereas the PSC values decreased. Most importantly, the gastrointestinal fluids of A. nitida tea infusion exhibited a strong antiproliferative activity towards Caco-2 cells and affected caspase 3/9 signaling pathway, and apoptosis induction might serve as the underlying mechanism for this antiproliferative activity. It can be concluded that A. nitida tea infusion may act as a potential intracellular antioxidant and colon cancer cell proliferation inhibitor.

Essential oil is one of the most important bioactive substances of Artemisia argyi Levl. et Van. var. argyi cv. Qiai, a traditional Chinese herb used in folks for centuries. An essential oil was prepared by steam distillation method. Chemical components of the essential oil were explored by gas chromatography-mass spectrometry. Antimicrobial activities of the essential oil were determined by agar disc diffusion assay and double-broth dilution method. Also, antimicrobial mechanisms were investigated based on cytomembrane permeability, protein and K + leakage and electron microscopy. The results showed that the essential oil yield was (0.7 ± 0.02)% (w/w). A total of 32 components were identified, and thujone was found to be the major compound (14.94%), followed by globulol (11.6%), 1,8-cineole (11.31%), 1-caryophyllene (8.11%), camphor (6.87%), α-terpineol (4.69%), caryophyllene oxide (4.05%), borneol (3.95%), eugenol (3.01%), and germacrene D (2.81%). The oil showed antimicrobial activities against both Gram-positive and Gram-negative bacteria. The minimum inhibition concentrations of the essential oil against Staphylococcus aureus , Bacillus cereus , Bacillus subtilis , Listeria monocytogenes , Escherichia coli , Pseudomonas aeruginosa , and Salmonella Typhimurium were 16.0, 16.0, 32.0, 32.0, 32.0, 64.0, and 32.0 μg/mL, respectively. The minimum bactericidal concentrations of the essential oil against bacteria mentioned above were 32.0, 16.0, 64.0, 64.0, 64.0, 128.0, and 64.0 μg/mL, respectively. Supposedly, the possible antimicrobial mechanism of the essential oil involved simultaneous cytomembrane disruption, leading to the leakage of intracellular substances, such as protein and K + . The results indicated that the cytomembrane might be the target of essential oil.
