UPLC-MS Analysis, Quantification of Compounds, and Comparison of Bioactivity of Methanol Extract and Its Fractions from Qiai (Artemisia argyi Lévl. et Van.)

Ting Zhang,Dingrong Wan,Yuanyuan Li,Sisi Wang,Xiuteng Zhou,Fatemeh Sefidkon,Xinzhou Yang

TheArtemisia argyiLévl. et Van. growing in the surrounding areas of Qichun County in China are called Qiai (QA). Qiai is a crop that can be used both as food and in traditional folk medicine. However, detailed qualitative and quantitative analyses of its compounds remain scarce. The process of identifying chemical structures in complex natural products can be streamlined by combining UPLC-Q-TOF/MS data with the UNIFI information management platform and its embedded Traditional Medicine Library. For the first time, 68 compounds in QA were reported by the method in this study. The method of simultaneous quantification of 14 active components in QA using UPLC-TQ-MS/MS was reported for the first time. Following a screening of the activity of QA 70% methanol total extract and its three fractions (petroleum ether, ethyl acetate, and water), it was discovered that the ethyl acetate fraction enriched with flavonoids such as eupatilin and jaceosidin had the strongest anti-inflammatory activity, while the water fraction enriched with chlorogenic acid derivatives such as 3,5-di-O-caffeoylquinic acid had the strongest antioxidant and antibacterial activity. The results provided the theoretical basis for the use of QA in the food and pharmaceutical industries. Keywords:Artemisia argyiLévl. et Van.;UPLC-MS;quantification;anti-inflammatory;antioxidant

Preparation and Characterization of Long-Term Antibacterial and Temperature-Responsive Chitosan-g-PNIPAm @ Mugwort Oil Microcapsules

Li Jing,Liu Wan,Jiang Ping

First, biocompatible temperature-responsive polymer chitosan- graft - N -isopropylacrylamide (CS- g -PNIPAm) synthesized via ATRP. Second, mugwort oil (MO) was encapsultated with CS- g -PNIPAm as shell materials to form long-term anti-bacterial agents and temperature responsive microcapsules. The CS- g -PNIPAm@MO (CSPN@MO) microcapsules were prepared via single coacervation method and solvent evaporation method. The chemical structures of CS 0.1 - g -PNIPAm 22 was confirmed by 1 H NMR and FTIR. And the copolymer micelles were characterized by UV, laser particle-size analysis, and TEM. The results indicated that the copolymer had temperature-responsive behaviors. The microcapsules were characterized by laser particle-size analyzers (LPSA), scanning electron microscopy (SEM), TG and anti-bacterial agents performance testing. The results showed that the average particle size of microcapsules was 0.28 μm, and the encapsulation efficiency and drug loading efficiency of MO reached 60.2 and 44.5%. After 48 h of release in media at different temperature (20, 40 and 60 °C) still sequestered 79.2, 92.2 and 96.1% of MO which approved the shell of microcapsules responded to different temperature. The morphology of CSPN@MO microcapsules revealed that the microcapsules presented smooth and firm structure. Antibacterial test for Artemisia argyi of CSPN@MO implied that the bacteriostatic rate reached 99.2 and 96.2% after 1 day and 10 days respectively. Bio-based macromolecular modification strategies can provide inspiration for the development of green microcapsules. Graphical Abstract

Solvent extraction of caffeoylquinic acids from Artemisia selengensis Turcz leaves and their in vitro inhibitory activities on xanthine oxidase

Yizhen Li,Yin Wan,Ruyi Li,Laojiu Xu,Mingyong Xie,Guiming Fu

In this study, the caffeoylquinic acid (CQA) extract solution of the Artemisia selengensis Turcz (AST) leaves prepared by 70% ethanol reflux method was separated with petroleum ether, ethyl acetate, and water-saturated n -butanol to obtain the proper solvent to purify CQAs. Results showed that the ethyl acetate fraction (EAF) exhibited the highest total chlorogenic acid content (TCC) (241.46 ± 0.00 mg/g dry fraction), followed by the water-saturated n -butanol fraction ( n BuF) (TCC = 62.30 ± 0.82 mg/g dry fraction). Ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry and high-performance liquid chromatography photodiode array were used to qualitatively and quantitatively determine the CQAs in these two fractions. In EAF, four CQAs, namely, 3-CQA, 3,5-diCQA, 3,4-diCQA, and 4,5-diCQA, were identified with the contents of 5.69 ± 0.03, 106.65 ± 0.83, 15.65 ± 0.24, and 28.08 ± 0.82 mg/g, respectively. Six CQAs, namely, 3-CQA, 4-CQA, 1,3-diCQA, 3,5-diCQA, 3,4-diCQA, and 4,5-diCQA, with contents of 16.57 ± 0.23, 3.22 ± 0.55, 4.49 ± 0.01, 16.86 ± 0.24, 7.35 ± 0.11, and 6.41 ± 0.13 mg/g, respectively, were identified in n BuF. Quantitative analysis of the six CQAs in AST leaves have not been reported, and 1,3-diCQA has been found in AST leaves for the first time. Moreover, either EAF or n BuF and the six CQA reference compounds showed certain inhibitory activity on xanthine oxidase (XOD) in vitro . EAF demonstrated a stronger XOD inhibitory effect (IC 50  = 1.67 mg/mL) than n BuF. Among the six CQA compounds, 3,4-diCQA had the lowest IC 50 of 12.80 μM, whereas 1,3-diCQA and 3-CQA had IC 50 values of 23.69 and 28.29 μM, respectively. The inhibitory activity of 4-CQA was weak. These findings indicated that CQAs are the major components inhibiting the XOD activity in vitro in AST leaves.

Fabrication and Biological Activities of All-in-One Composite Nanoemulsion Based on Blumea balsamifera Oil-Tea Tree Oil

Yue Zhu,Teng Chen,Tingting Feng,Jiaojiao Zhang,Zejing Meng,Ning Zhang,Gang Luo,Zuhua Wang,Yuxin Pang,Ying Zhou

Nanoemulsion is a new multi-component drug delivery system; the selection of different oil phases can give it special physiological activity, and play the role of “medicine and pharmaceutical excipients all-in-one”. In this paper, we used glycyrrhizic acid as the natural surfactant, andBlumea balsamiferaoil (BB) and tea tree oil (TTO) as the mixed oil phase, to obtain a new green functional composite nanoemulsion. Using the average particle size and polydispersion index (PDI) as the evaluation criteria, the effects of the oil ratio, oil content, glycyrrhizic acid concentration, and ultrasonic time on the nanoemulsion were systematically investigated. The stability and physicochemical properties and biological activities of BB-TTO NEs prepared via the optimum formulation were characterized. The optimal prescription was BB: TTO = 1:1, 5% oil phase, 0.7% glycyrrhizic acid, and 5 min ultrasonication time. The mean particle size, PDI, and zeta potential were 160.01 nm, 0.125, and −50.94 mV, respectively. The nanoemulsion showed non-significant changes in stability after centrifugation, dilution, and 120 days storage. These nanoemulsions were found to exhibit potential antibacterial and anti-inflammatory activities. The minimal inhibitory concentration (MIC) of BB-TTO NEs againstEscherichia coli,Staphylococcus aureus, andPseudomonas aeruginosais 2975 μg/mL, 2975 μg/mL, and 5950 μg/mL, respectively. A lower level of inflammatory cell infiltration and proportion of fibrosis were found in the synovial tissue of AIA rats treated with BB-TTO NEs. These findings demonstrate that the BB-TTO NEs produced in this study have significant potential for usage in antibacterial and anti-inflammatory areas. Keywords:nanoemulsion;Blumea balsamiferaoil;tea tree oil;glycyrrhizic acid;anti-inflammatory;antibacterial

Preparation and Characterization of a One-Step Electrospun Poly(Lactic Acid)/Wormwood Oil Antibacterial Nanofiber Membrane

Xiaoyan Tang,Xun Guo,Yongchao Duo,Xiaoming Qian

With the continuous improvement of the standard of living, people are increasingly inclined towards natural, green, and environmentally friendly products. Plant-based products that are safe, natural, non-toxic, and beneficial to human health are often more favored. Poly(lactic acid) (PLA) is a polymer obtained through lactate polymerization using renewable plant resources such as corn and has excellent biocompatibility and biodegradability. It is widely used in the field of food packaging. Wormwood oil (WO) is an oil extracted from the stems and leaves of Artemisia plants, and it has broad-spectrum antibacterial properties. In this article, through electrospinning technology, wormwood oil was directly incorporated into PLA, giving the PLA nanofiber membrane antioxidant and antibacterial functions. Various parameters such as voltage (11 KV, 13 KV, 15 KV), spinning solution concentration (8%, 10%, 12%), distance (15 cm, 17 cm, 19 cm), and feeding rate (0.4 mL/h, 0.5 mL/h, 0.6 mL/h) were explored, and the resulting spun fibers were characterized. Through SEM characterization, it was found that when the spinning voltage was 13 KV, the spinning solution concentration was 10%, the distance was 17 cm, and the feeding rate was 0.5 mL/h, the nanofiber membrane had a smooth morphology without bead formation, with an average diameter of 260 nm. The nanofiber membrane was characterized using FTIR, TG, and SEM, confirming the successful incorporation of artemisia essential oil into PLA. The prepared antimicrobial nanofilm was subjected to antimicrobial testing, and the results showed that as the concentration of the essential oil increased, the inhibition zones also increased. When wormwood oil concentration was 4%, the diameter of the inhibition zone forStaphylococcus aureusincreased from 1.0 mm to 3.5 mm, while the diameter of the inhibition zone forEscherichia coliincreased from 2.0 mm to 4.5 mm.Keywords:electrospinning;nanofiber;poly(lactic acid);wormwood oil;natural antimicrobial

Preparation and characterization of wormwood-oil-contained microcapsules

Lei Wang,Tingxiao Li,Binjie Xin,Yan Liu,Fuli Zhang

In recent years, wormwood has been successfully applied in various fields, such as flavouring agent, plant dye, insect repellent and medical product. Wormwood oil encapsulated microcapsules with continuous release behaviours would be further applied in the health care textiles. Here, wormwood oil encapsulated microcapsules with different core-wall materials ratios (gelatine: gum arabic: wormwood oil = 1:1:1, 1:1:2, 1:1:3, 1:1:4) were fabricated. The morphology and physicochemical properties of microcapsules were explored by scanning electron microscopy, ultrasonic wave particle size analysis, thermo gravimetric analysis and so on. The size analysis results indicated that the microcapsule diameter could be controlled less than 1 um. In vitro testing showed that the microcapsules were capable of extending the releasing period to over 6 days. The thermo gravimetric analysis proved that microcapsules had superior thermal stability compared with pure wormwood oil, showing broad application prospects in functional health care textile.

Bio-Evaluation of the Wound Healing Activity of Artemisia judaica L. as Part of the Plant’s Use in Traditional Medicine; Phytochemical, Antioxidant, Anti-Inflammatory, and Antibiofilm Properties of the Plant’s Essential Oils

Hamdoon A. Mohammed,Kamal A. Qureshi,Hussein M. Ali,Mohsen S. Al-Omar,Omar Khan,Salman A. A. Mohammed

Artemisia judaica(ArJ) is a Mediterranean aromatic plant used traditionally to treat gastrointestinal ailments, skin diseases, atherosclerosis, and as an immuno-stimulant. This study describes ArJ essential oil constituents and investigates their wound healing activity. The in vitro antioxidant and antibiofilm activities of ArJ essential oil were investigated. The in vivo pro/anti-inflammatory and oxidative/antioxidant markers were compared with standard silver sulfadiazine (SS) in a second-degree skin burn experimental rat model. The gas chromatography-equipped flame ionization detector (GC-FID) analysis of ArJ essential oil revealed the major classes of compounds as oxygenated monoterpenes (>57%) and cinnamic acid derivatives (18.03%). The antimicrobial tests of ArJ essential oil revealed thatBacillus cereus,Candida albicans, andAspergillus nigerwere the most susceptible test organisms. Two second-degree burns (each 1 inch square in diameter) were created on the dorsum of rats using an aluminum cylinder heated to 120 °C for 10 s. The wounds were treated either with ArJ or SS ointments for 21 days, while the negative control remained untreated, and biopsies were obtained for histological and biochemical analysis. The ArJ group demonstrated a significant increase in antioxidant superoxide dismutase (SOD) and catalase (CAT) enzymatic activities, while lipid peroxide (LP) levels remained insignificant compared to the negative control group. Additionally, ArJ and SS groups demonstrated a significant decrease in inflammatory levels of tumor necrosis factor α (TNF-α) compared to the negative group, while interleukin 1 beta (IL-1b) and IL-6 were comparable to the negative group. At the same time, anti-inflammatory IL-10 and transforming growth factor beta 1 (TGF-b1) markers increased significantly in the ArJ group compared to the negative control. The ArJ results demonstrated potent wound healing effects, comparable to SS, attributable to antioxidant and anti-inflammatory effects as well as a high proportion of oxygenated monoterpenes and cinnamate derivatives. Keywords:Artemisia judaica;essential oil;wound healing;inflammatory markers;antibiofilm activity;antioxidant activity

Preparation and Characterization of a Novel Artemisia Oil Packaging Film and Its Application in Mango Preservation

Xiaohan Meng,Ze Lv,Tianzhen Jiang,Yifei Tan,Shaoyang Sun,Jianguo Feng

In this work, a new food packaging film was synthesized via blending Artemisia oil (AO) into soybean protein isolate (SPI) and gelatin (Gel) for the postharvest storage of mango. The morphological architecture and mechanical properties of the films were characterized using scanning electron microscopy (SEM), atomic force microscopy (AFM), Fourier transform infrared spectroscopy (FTIR), X-ray diffractometer (XRD), and other technologies. The results show that the prepared films had relatively flat surfaces with good mechanical properties. AO enhanced the light-blocking ability of the film, increased the hydrophobicity, and affected the moisture content and water solubility of the film to a certain extent. Furthermore, the antioxidant performance and antifungal (Colletotrichum gloeosporioides) capacity of the films increased with higher AO concentration due to the presence of the active components contained in AO. During mango storage applications, the films showed good freshness retention properties. The above results indicate that SPI–Gel films containing AO have excellent physicochemical and application properties and have great potential in the field of food packaging. Keywords:soybean protein isolate;gelatin;Artemisia oil;mango preservation;active packaging film

Wormwood (Artemisia absinthium L.) as a promising nematicidal and antifungal agent: Chemical composition, comparison of extraction techniques and bioassay-guided isolation

Ting-ting Liu,Hai-bin Wu,Hai-bo Wu,Jing Zhang

The ethyl acetate, ethanol and water extracts of different organs (flowers, leaves, stems and roots) of wormwood ( Artemisia absinthium L.) were investigated to examine nematicidal and antifungal activities in an effort to promote future agricultural applications of the wormwood and its potential as an industrial crop. The results showed that highest concentration of total thiophene content (TTC), as well as nematicidal and antifungal activities were found in ethyl acetate extract of roots. TTC and biological activities were highly correlated, indicating that thiophenes played an important role in nematicidal and antifungal activities. Furthermore, four extraction techniques (Maceration, Soxhlet extraction, Supercritical fluid extraction and Ultrasound-assisted extraction) were compared by evaluating the yield of thiophenes. Supercritical fluid extraction (SFE) appeared to be the best method for extracting thiophenes with the highest TTC and superior activities. In addition, eight thiophenes ( 1 – 8 ) including two new compounds ( 1 and 2 ) were isolated from the SFE extract by the bioassay-guided method. All thiophenes showed potent nematicidal activities. In particular, 1 – 4 (50% lethal concentration (LC 50 ) values of 2.69, 4.17, 6.13 and 7.65 mg/L, respectively) were more effective than commercial nematicide abamectin (LC 50 , 9.47 mg/L). Also, these thiophenes showed antifungal activities against four soil-borne fungi at different degrees, especially for compounds 1 and 2 which exhibited noticeable antifungal effects (minimum inhibitory concentration (MIC) values ranging from 8–16 mg/L). Findings of this work suggest that wormwood root and its thiophenes could be used in nematicide and fungicide agent industry.

Bioactivities of phytochemicals in Araiostegia yunnanensis (Christ)

Longsheng Chen,Wei Xu,Rong Shao,Xianfeng Du

The profile and bioactivity of phytochemicals in Araiostegia yunnanensis ( Christ ) Cop were investigated. The total flavonoids content in A. yunnanensis is about 84.90   mg/g. By means of HPLC–DAD–ESI-MS, the main flavonoids in A. yunnanensis were tentatively identified as myricetin 3- O -rhamnosylglucoside, eriodictyol 7- O -rutinoside, quercetin 3- O -rutinoside, luteolin-7- O -apiosylglucoside, quercetin 3- O -rhamnosylgalactoside, and luteolin 7- O -glucoside. The extract (0.268   mg/ml total flavonoids) from A. yunnanensis showed very strong superoxide anion radical scavenging potential and reducing power, which are higher than those of rutin (0.25   mg/ml). The extract (0.268   mg/ml total flavonoids) from A. yunnanensis exhibited similar DPPH scavenging activity with rutin (0.25   mg/ml). However, rutin (0.25   mg/ml) showed a significantly higher ABTS radical scavenging effect than that of the extract (0.268   mg/ml total flavonoids) from A. yunnanensis. The methanol extract from A. yunnanensis showed obviously cytotoxic effects on A549 cells and it had no effect against acetylcholinesterase.

Extraction of flavonoids from Artemisia argyi using deep eutectic solvents and its antioxidant activities

Huibing Dai,Maoqian Hu,Ning Zhu,Lijun Song,Hailong Hong

Deep eutectic solvents (DES) are regarded as eco-friendly alternatives for extracting bioactive substances from natural products. In this study, the DES with varying compositions were employed to extract flavonoids from Artemisia argyi H.Lév. & Vaniot ( A. argyi ) with the assistance of ultrasound, and optimized the extraction conditions through a Box-Behnken design (BBD). The results indicated that the choline chloride (ChCl) / lactic acid (LAC) (molar ratio 1:2) was the best DES for extracting flavonoids from A. argyi with an extraction power of 150 W. The optimal process condition was identified as DES with a water content of 40.0 %, a material-liquid ratio of 1:22.4 g·mL −1 , and the extraction temperature of 44.8 °C. This method demonstrated superior performance compared to the traditional ethanol method. The biological activities in vitro revealed that the extracts exhibited desirable antioxidant activity, and the extraction mechanism of flavonoids was investigated by density functional theory. Furthermore, five distinct types of macroporous resins (HPD826, AB8, YKDH-3, D101, and NKA-9) were selected for the recovery of DES. YKDH-3 exhibited the strongest adsorption capacity, and HPD826 the weakest. The extraction rate of recycled DES reached 84.5 %, the number of recycling was up to more than 6 times. In short, this method, combined with macroporous resin enrichment, can be used as an alternative method for green and efficient extraction of bioactive flavonoids from plants.
