Rapid and nondestructive determination of sesamin and sesamolin in Chinese sesames by near-infrared spectroscopy coupling with chemometric method

Zhenzhen Xia,Tian Yi,Yan Liu

Sesame was one of the most important crops in Africa and east Asia. The sesamin and sesamolin in sesames have shown various pharmacological, biological and physiologic activities. In this study, a rapid and nondestructive method for determination of sesamin and sesamolin in Chinese sesames by near-infrared spectroscopy coupled with chemometric method was proposed. The near infrared spectra of sesame samples from three different Chinese areas were collected and the partial least squares (PLS) was used to construct the quantitative models. The spectral preprocessing and variable selection methods were adopted to improve the predictability and stability of the model. Reasonable quantitative results can be obtained when the samples used for model construction and prediction were harvested in same years. For sesamin and sesamolin, the correlation coefficient (R) and root mean square error prediction (RMSEP) were 0.9754, 0.9636 and 151.2951, 39.7720, respectively. The optimized models seem less effective when they were used to predict the samples harvested in other years or countries. However, acceptable results can still be obtained.

Sesamin Attenuates Lipopolysaccharide-Induced Acute Lung Injury by Inhibition of TLR4 Signaling Pathways

Qiang Li,Yuan Jiang,Shouyin Jiang,Yulin Li,Libing Jiang,Jian-an Wang

Recent studies suggested that TLR4 signaling pathways played an important role in the development of LPS-induced acute lung injury (ALI). Sesamin, a sesame lignan exacted from sesame seeds, has been shown to exhibit significant anti-inflammatory activity. The purpose of this study was to investigate the anti-inflammatory effects of sesamin on LPS-induced ALI in mice. Mice ALI model was induced by intratracheal instillation of LPS. Sesamin was given 1 h after LPS challenge. Our results showed that sesamin inhibited LPS-induced lung pathological change, edema, and myeloperoxidase (MPO) activity. Sesamin suppressed LPS-induced inflammatory cytokines TNF-α, IL-6, and IL-1β production. Furthermore, sesamin inhibited LPS-induced TLR4 expression and NF-κB activation. In conclusion, the results of this study indicated that sesamin protected against LPS-induced ALI by inhibition of TLR4 signaling pathways.

The inhibitory effects of sesamol and sesamolin on the glycidyl esters formation during deodorization of vegetables oils

Lipeng Han,Jiahui Li,Shujie Wang,Weiwei Cheng,Lukai Ma,Guoqin Liu,Dongxue Han,Li Niu

BACKGROUND Glycidyl esters (GEs) have attracted worldwide attention for their potential harm to human health. The GEs in edible oils mainly form during the deodorization of the oil refining processes. We used sesamol and sesamolin to inhibit the formation of GEs in model corn oil (MCO), model palm oil (MPO) and model rice bran oil (MRO) during a deodorization process. RESULTS The results showed that, in the three model oils, the total GE content was in the following order from highest to lowest: MRO (1437.98 μg kg −1 ) > MPO (388.64 μg kg −1 ) > MCO (314.81 μg kg −1 ). The inhibitory effect of the three antioxidants on the formation of GEs in the MCO was in the following order from strongest to weakest: tert-butylhydroquinone (TBHQ) > sesamol > sesamolin. CONCLUSION When the mass percentage of sesamol was 0.05%, its inhibition percentage on GEs was close to the inhibition percentage of 0.02% added TBHQ. The present study provides a foundation for understanding how to inhibit the formation of GEs in oils by adding sesamol during the deodorization process.

Sesamin attenuates liver inflammation caused by PFOS via regulating SAP130-mediated hepatocyte-macrophage crosstalk

Jingyi Ren,Longfei Li,Ziyi Wang,Siqi Zhu,Huanting Pei,Yadong Zhang,Bowen Yin,Zhenao Zhang,Xiaoya Sun,Huicai Guo,Yuxia Ma

Background Perfluorooctane sulfonate (PFOS) is a prototypical persistent organic pollutant that has been linked to liver inflammation. Sesamin (Ses), a major lignan in sesame, has been shown to exhibit health-promoting properties. However, the role and specific mechanism of Ses in PFOS-induced liver inflammation remain largely unclear. Purpose This study aimed to investigate the hepatoprotection and potential mechanism of Ses against PFOS-induced inflammation. Methods Wild-type (WT), Mincle knockout (Ko-mincle), and Hepatocyte-specific knockout Foxo1 (Hko-Foxo1) mice were intragastrically administered Ses and/or PFOS (10 mg/kg) for 4 weeks. In vitro, HepG2 cells were pretreated with Ses for 1 h, then exposed to PFOS for 24 h to establish a cell injury model. Results We revealed that Ses markedly alleviated PFOS-induced liver injury and inflammation, as evidenced by noticeable histopathological improvements, increased cell activity, and alterations in serum liver enzyme and inflammatory factor levels. Moreover, Ses pretreatment reduced the expression of Mincle and the phosphorylation of its downstream targets, SYK and p65, in macrophages. The reducing effect of Ses on liver inflammation in wild-type mice is significantly better than that in Mincle knockout mice. We also found that Ses decreased the expression of spliceosome-associated protein 130 (SAP130) and Foxo1. Further experiments revealed that SAP130 may serve as a mediator between hepatocytes and macrophages. Hepatocyte-specific knockout Foxo1 reduced the release of SAP130 from hepatocytes and inhibited Mincle/Syk signaling activation in macrophages. Conclusion This study demonstrates that Ses protects against PFOS-induced liver inflammation in mice by modulating crosstalk between hepatocytes and macrophages. Those findings support the potential of Ses as a promising natural compound for counteracting liver inflammation.

Sesamin protects against renal ischemia reperfusion injury by promoting CD39-adenosine-A2AR signal pathway in mice

Ke Li,Xia Gong,Ge Kuang,Rong Jiang,Jingyuan Wan,Bin Wang

Ischemia reperfusion injury (IRI) is a leading cause of acute kidney injury with high morbidity and mortality due to limited therapy. Here, we examine whether sesamin attenuates renal IRI in an animal model and explore the underlying mechanisms. Male mice were subjected to right renal ischemia for 30 min followed by reperfusion for 24 h with sesamin (100 mg/kg) during which the left kidney was removed. Renal damage and function were assessed subsequently. The results showed that sesamin reduced kidney ischemia reperfusion injury, as assessed by decreased serum creatinine (Scr) and Blood urea nitrogen (BUN), alleviated tubular damage and apoptosis. In addition, sesamin inhibited neutrophils infiltration and pro-inflammatory cytokines tumor necrosis factor (TNF)-α and interleukin (IL)-1β production in IR-preformed kidney. Notably, sesamin promoted the expression of CD39, A2A adenosine receptor (A2AAR), and A2BAR mRNA and protein as well as adenosine production. Furthermore, CD39 inhibitor or A2AR antagonist abolished partly the protection of sesamin in kidney IRI. In conclusion, sesamin could effectively protect kidney from IRI by inhibiting inflammatory responses, which might be associated with promoting the adenosine-CD39-A2AR signaling pathway.

Sesamin protects against Acetaminophen-induced nephrotoxicity by suppressing HMOX1-mediated apoptosis and ferroptosis

Siqi Zhu,Jingyi Ren,Yadong Zhang,Xiaoya Sun,Huanting Pei,Bowen Yin,Ziyi Wang,Zhenao Zhang,Shenghe Li,Ruonan Zhang,Ziqian Zeng,Yuxia Ma

Background Acetaminophen (APAP) is a widely used antipyretic and analgesic agent, and acute exposure can lead to renal injury. Sesamin (Ses) is known for its various health benefits. However, it remains unclear whether Ses exerts a protective effect against APAP-induced kidney injury. Methods In vivo, C57BL/6 mice were pretreated with Ses and injected intraperitoneally with APAP. In vitro, human kidney proximal tubule cells 2 were pretreated with Ses, and then models of kidney injury induced by APAP were established. Kidney damage was evaluated by morphological, inflammation, oxidative stress and protein analyzes. Results Ses significantly improved APAP-induced nephrotoxicity in vitro and in vivo models. Transcriptomic analysis revealed that the differentially expressed genes were enriched in ferroptosis and apoptosis signaling pathways, identifying heme oxygenase 1 (HMOX1) as a core protein. In the Ses-treated group, ferroptosis and apoptosis were significantly inhibited, while HMOX1 was effectively restored. In cell experiments, both the HMOX1 agonist hemin and Ses attenuated ferroptosis and apoptosis. HMOX1 inhibitor Zinc Protoporphyrin significantly eliminated the protective effect of Ses.Conclusion Ses alleviates APAP-induced renal injury by mediating the inhibition of ferroptosis and apoptosis via HMOX1. This study provides a new strategy for the prevention and treatment of drug-induced renal injury.

Yushu Sui,Liben Pan,Nan Yang,Xiaorui Guo,Zhonghua Tang

In the present study, developed a dependable HPLC technique to quantify asarinin and sesamin in Asarum heterotropoides Fr. Schmidt var. mandshuricum (Maxim.) Kitag. The optimal conditions for the ultrasonic-assisted extraction were determined by conducting single-factor experiment, Plackett-Burman design experiment, and Box-Behnken response surface method . Based on actual operation factors and analysis, the optimal extraction parameters were methanol volume fraction of 80% (v/v), liquid-to-material ratio of 10:1 (mL/g), ultrasonic time of 40 min, ultrasonic temperature of 51°C, and ultrasonic power of 300 W. When subjected to these conditions, the average asarinin and sesamin content were found to be 13.40 mg/g and 2.39 mg/g, respectively, with a relative error of 1.04% and 3.46%, which was consistent with the theoretical value. This study can offer a theoretical reference for the extraction of asarinin and sesamin in Asarum .

Simultaneous and rapid determination of sesamin and sesamolin in sesame oils using excitation-emission matrix fluorescence coupled with self-weighted alternating trilinear decomposition

Yan Liu,Qian Wu,Zhenzhen Xia,Yang Wu,Yan Li,Zhiyong Gong

BACKGROUND Sesamin and sesamolin are two typical and important lignans isolated from sesame oil. Various studies have shown the bioactivity, physiological activity, and potential health benefits of the two components. In this study, a rapid method for the simultaneous determination of sesamin and sesamolin in sesame oils was proposed. The excitation-emission fluorescence spectra of the oils were obtained after a simple pretreatment, then self-weighted alternating trilinear decomposition was used to extract the quantitative information from the very overlapping spectra. RESULTS It was found that reasonable quantification results could be obtained with the limits of detection for the two lignans. These limits were 0.05 mg/g and 0.24 mg/g, and the limits of quantitation were 0.14 mg/g and 0.74 mg/g, respectively. The average recoveries for sesamin and sesamolin were 99.05% and 94.97%. CONCLUSION The results indicate that, with simple sample pretreatment, the application for combining excitation-emission fluorescence spectra and self-weighted alternating trilinear decomposition can be a useful and sensitive tool for the determination of lignans in sesame oil. © 2020 Society of Chemical Industry

Sesamin Protects the Femoral Head From Osteonecrosis by Inhibiting ROS-Induced Osteoblast Apoptosis in Rat Model

Deng Shuang,Zhou Jian-Lin,Fang Hong-Song,Nie Zhi-Gang,Chen Sen,Peng Hao

Glucocorticoids intake has become the most common pathogenic factor for osteonecrosis of the femoral head (ONFH). Annually, tens of millions of patients suffer from pain related to ONFH. Researchers have proposed several underlying mechanisms of ONFH, including osteocyte apoptosis, cell differentiation disorder, and angiogenesis hindrance. Sesamin, isolated from Sesamum indicum seeds, was reported could affect osteocyte inflammation and differentiation in osteoarthritis and osteoporosis. We investigated the underlying influence of sesamin on ONFH rat model. Fifteen male Sprague-Dawley rats were randomly divided into three groups. The ONFH model group only received the methylprednisolone (MPS) and lipopolysaccharide (LPS) injection to promote the development of ONFH. The sesamin treatment group was injected with sesamin, MPS, and LPS. The control group was untreated. Rats from above groups were sacrificed 4 weeks later. The effect of sesamin on ONFH rats was validated by H&E staining. TUNEL staining showed that femoral head necrosis was attenuated by sesamin. Furthermore, the phosphorylation of Akt was increased and the downstream cellular apoptosis signal pathway was inhibited. Intracellular ROS level was decreased after sesamin treatment. In conclusion, our findings suggest that sesamin protects the femoral head from osteonecrosis by inhibiting ROS-induced osteoblast apoptosis.

Exploring the mechanism of sesamin for the treatment of PM2.5-induced cardiomyocyte damage based on transcriptomics, network pharmacology and experimental verification

Yadong Zhang,Rui Wen,Jingyi Ren,Fan Zhang,Huanting Pei,Jinshi Zuo,Yuxia Ma

Introduction: Exposure to fine particulate matter (PM2.5) is known to be associated with cardiovascular diseases. Sesamin (Ses) is a natural phenolic compound found in sesame seeds and sesame oil. Ferroptosis is a novel mode of cell death characterised by iron-dependent lipid peroxidation. This study aims to explore whether PM2.5 can induce ferroptosis in H9C2 cells and to investigate the precise protective mechanism of Ses.Methods: Based on transcriptomic data, PM2.5 may induce ferroptosis in cardiomyocytes. The ferroptosis inducer erastin and ferroptosis inhibitor ferrostatin-1 (Fer-1) were used to illustrate the mechanisms involved in PM2.5-induced H9C2 cell injury. Using network pharmacology, the pharmacological mechanism and potential therapy targets of Ses were explored for the treatment of PM2.5-induced cardiomyocyte injury. H9C2 cells were cultured and pretreated with Fer-1 or different concentrations of Ses, and then cardiomyocyte injury model was established using erastin or PM2.5. Indicators of oxidative responses, including total superoxide dismutase, reduced glutathione, glutathione peroxidase and malondialdehyde, were measured. The expression levels of ferroptosis-related proteins were determined through Western blot analysis. Results: Results demonstrate that PM2.5 induces ferroptosis in H9C2 cells and Ses exerts a protective effect by suppressing ACSL4-mediated ferroptosis.Discussion: Overall, these findings elucidate a novel mechanism by which Ses ameliorates the detrimental effects of PM2.5 on cardiomyocytes.

Sesamin attenuates PM2.5-induced cardiovascular injury by inhibiting ferroptosis in rats

Jing-yi Ren,Bo-wen Yin,Xiang Li,Si-qi Zhu,Jin-liang Deng,Yi-ting Sun,Zhen-ao Zhang,Zi-hao Guo,Huan-ting Pei,Fan Zhang,Rui-qiang Li,Feng-ge Chen,Yu-xia Ma

Objective: This study aimed to elucidate the pharmacological effects of sesamin (Ses) and its mechanism of action towards PM2.5-induced cardiovascular injuries. Method: Forty Sprague Dawley (SD) rats were randomly divided into five groups: a saline control group; a PM2.5 exposure group; and low-, middle-, and high-dose Ses pretreatment groups. The SD rats were pretreated with different concentrations of Ses for 21 days. Afterward, the rats were exposed to ambient PM2.5 by intratracheal instillation every other day for a total of three times. The levels of inflammatory markers, including tumor necrosis factor-alpha (TNF-α), interleukin-1beta (IL-1β), and interleukin-6 (IL-6), and indicators related to oxidative responses, such as total superoxide dismutase (SOD), reduced glutathione (GSH), glutathione peroxidase (GSH-Px), and malondialdehyde (MDA), were measured in the blood and heart. The expression of ferroptosis-related proteins in heart tissues was determined via western blot and immunohistochemistry. Results: Ses pretreatment substantially ameliorated cardiovascular injuries in rats as evidenced by the decrease in the pathological score and collagen area. The decreased levels of SOD, GSH, and GSH-Px in the heart and serum were inhibited by Ses. In addition, Ses not only notably increased the activity of antioxidant enzymes but also reduced the levels of MDA, CK, LDH, CK-MB, IL-6, TNF-α, IL-1β, and IL-6. Furthermore, Ses pretreatment upregulated the expression levels of GPX4, SLC7A11, TFRC, and FPN1 and inhibited the expression levels of FTH1 and FTL. Conclusion: Ses pretreatment could ameliorate PM2.5-induced cardiovascular injuries perhaps by inhibiting ferroptosis. Therefore, Ses pretreatment may be a novel strategy for the prevention and treatment of PM2.5-induced cardiovascular injury.

Sesamin ameliorates lipopolysaccharide/d-galactosamine-induced fulminant hepatic failure by suppression of Toll-like receptor 4 signaling in mice

Li Ma,Xia Gong,Ge Kuang,Rong Jiang,Rongchun Chen,Jingyuan Wan

Sesamin has been described to exert anti-oxidant and anti-inflammatory properties. In present study, we investigated the potential effects and mechanisms of sesamin on lipopolysaccharide (LPS)-induced fulminant hepatic failure (FHF) in d -galactosamine (D-GalN)-sensitized mice. Our results showed that pretreatment with sesamin dose-dependently improved LPS/D-GalN-induced mortality and liver injury as indicated by reduced serum levels of aminotransferases and alleviated pathological damage as well as hepatocyte apoptosis in mice. Additionally, sesamin markedly attenuated LPS/D-GalN-induced adhesion molecules expression, and decreased neutrophils recruitment. Furthermore, sesamin inhibited LPS-induced tumor necrosis factor-alpha (TNF-α) production, p38 mitogen-activated protein kinases (MAPK) and NF-κB activation, and Toll like receptor (TLR) 4 expression in mice and in RAW264.7 macrophage cells. In summary, these results demonstrate that sesamin protects mice from LPS-induced FHF and the molecular mechanisms may down-regulate the expression of TLR4, block MAPK and NF-κB activation, decrease the production of TNF-α.
