Sesame was one of the most important crops in Africa and east Asia. The sesamin and sesamolin in sesames have shown various pharmacological, biological and physiologic activities. In this study, a rapid and nondestructive method for determination of sesamin and sesamolin in Chinese sesames by near-infrared spectroscopy coupled with chemometric method was proposed. The near infrared spectra of sesame samples from three different Chinese areas were collected and the partial least squares (PLS) was used to construct the quantitative models. The spectral preprocessing and variable selection methods were adopted to improve the predictability and stability of the model. Reasonable quantitative results can be obtained when the samples used for model construction and prediction were harvested in same years. For sesamin and sesamolin, the correlation coefficient (R) and root mean square error prediction (RMSEP) were 0.9754, 0.9636 and 151.2951, 39.7720, respectively. The optimized models seem less effective when they were used to predict the samples harvested in other years or countries. However, acceptable results can still be obtained.

A simple and fast normal-phase high performance liquid chromatography method for simultaneous detection of α-tocopherol, β-tocopherol, γ-tocopherol, δ-tocopherol, sesamin, sesamolin, sesamol, and asarinin in sesame oil (SO) was investigated. Conditions were a silica gel column as the separation unit, n -hexane and tetrahydrofuran (93:7, v/v) as the mobile phase, isocratic elution, a flow rate of 0.8 mL/min, column temperature of 30 °C, and florescence detector with excitation and emission wavelengths of 295 and 330 nm. SO samples were directly dissolved in n -hexane for HPLC analysis. Results showed that the method performed wide linear ranges and good linearity (0.27–100.0 μg/mL, R 2 > 0.9996), low limits of detection (0.09–0.16 μg/mL) and quantification (0.27–0.41 μg/mL), good precision and accuracy (biases of <2.83 % and relative standard deviations of <3.77 %), good separation performance [resolution factor (1.66–6.20), separation factor (1.11–3.94), retention factor (1.08–1.42), and height of theoretical plates (0.01–0.04 mm)], and satisfactory recoveries (87.3–109.8 % for SO and 96.5–106.9 % for lard). The method was successfully applied for the simultaneous determination of the eight analytes in eleven SO samples.

BACKGROUND Glycidyl esters (GEs) have attracted worldwide attention for their potential harm to human health. The GEs in edible oils mainly form during the deodorization of the oil refining processes. We used sesamol and sesamolin to inhibit the formation of GEs in model corn oil (MCO), model palm oil (MPO) and model rice bran oil (MRO) during a deodorization process. RESULTS The results showed that, in the three model oils, the total GE content was in the following order from highest to lowest: MRO (1437.98 μg kg −1 ) > MPO (388.64 μg kg −1 ) > MCO (314.81 μg kg −1 ). The inhibitory effect of the three antioxidants on the formation of GEs in the MCO was in the following order from strongest to weakest: tert-butylhydroquinone (TBHQ) > sesamol > sesamolin. CONCLUSION When the mass percentage of sesamol was 0.05%, its inhibition percentage on GEs was close to the inhibition percentage of 0.02% added TBHQ. The present study provides a foundation for understanding how to inhibit the formation of GEs in oils by adding sesamol during the deodorization process.

Sesaminol triglucoside (STG), the most abundant lignan glycoside existing in sesame cake/meal, has exhibited various biological activities. However, little information about its in vitro fermentation with intestinal microbiota is available. Therefore, the effect of STG from sesame cake on the fermentation of human fecal microbiota was evaluated. First, high-purity STG was successfully prepared from defatted sesame cake by extraction with 80% ethanol and simple purification procedures of polyamide column chromatography and Toyopearl HW-40S column chromatography. Then the influence of STG on intestinal microbiota was conducted by monitoring bacterial populations and analyzing the concentrations of short-chain fatty acids (SCFA). We found that STG could significantly induce an increase in numbers of Lactobacillus–Enterococcus group and Bifidobacterium in fermentation in vitro with human fecal microbiota, while it did not stimulate the bacterial growth of Eubacterium rectale–Clostridium coccoides group, Clostridium histolyticum group, and Bacteroides–Prevotella group. Furthermore, it was found that concentrations of formic, acetic, propionic, and butyric acids in STG culture increased significantly during the fermentation, and its total SCFA concentration was relatively higher than those of the control and glucose cultures at 6 and 12 h fermentation. Our findings provided further evidence for the importance of human intestinal bacteria in the bioactivity of STG and its metabolites in the maintenance of human health.

BACKGROUND Sesamin and sesamolin are two typical and important lignans isolated from sesame oil. Various studies have shown the bioactivity, physiological activity, and potential health benefits of the two components. In this study, a rapid method for the simultaneous determination of sesamin and sesamolin in sesame oils was proposed. The excitation-emission fluorescence spectra of the oils were obtained after a simple pretreatment, then self-weighted alternating trilinear decomposition was used to extract the quantitative information from the very overlapping spectra. RESULTS It was found that reasonable quantification results could be obtained with the limits of detection for the two lignans. These limits were 0.05 mg/g and 0.24 mg/g, and the limits of quantitation were 0.14 mg/g and 0.74 mg/g, respectively. The average recoveries for sesamin and sesamolin were 99.05% and 94.97%. CONCLUSION The results indicate that, with simple sample pretreatment, the application for combining excitation-emission fluorescence spectra and self-weighted alternating trilinear decomposition can be a useful and sensitive tool for the determination of lignans in sesame oil. © 2020 Society of Chemical Industry

CYP46A1 is a brain-specific enzyme responsible for cholesterol homeostasis. Inhibition of CYP46A1 activity serves as a therapeutic target for excitatory neurotoxicity. Sesame is a common medicine and food resource; its component lignans possess various pharmacological activities. In this study, the inhibitory effects of sesame lignans on CYP46A1 activity were investigated. Inhibition kinetics analyses revealed that sesamin and sesamolin produce mixed partial competitive inhibition of CYP46A1, while sesamol produces non-competitive inhibition. Notably, molecular simulations revealed that the sesame lignans have excellent orientations within the active cavity of CYP46A1. Importantly, the sesame lignans had high permeability coefficients and low efflux ratios. Furthermore, sesamin significantly reduced the levels of 24S-hydroxycholesterol in rat plasma and brain tissues, and down-regulated the protein expressions of CYP46A1, NMDAR2A, NMDAR2B, and HMGCR. Collectively, sesame lignans exhibit significant inhibitory effects on CYP46A1 activity, highlighting their potential therapeutic role in treating excitatory neurotoxicity.

Mikania, a well-accepted invasive plant in nature, has been difficult to be completely removed even based on chemical herbicides. Moreover, the employment of chemical herbicides will lead to hazardous results to health or environment. In this study, the whole-new carbon polymer quantum dots (CPDs) have been afforded in the light of Mikania as the carbon source. Moreover, sesame has been selected as a plant model and the effect of such kind of nanoscale fertilizer on its growth has been explored. Results support that CPDs at a concentration of 0–0.4 mg/mL possess a physiological response to the germination and no toxicity has been observed. Confocal images demonstrate that CPDs can spread through the roots of sesame seeds to the stems and leaves. The utilization of micrantha-derived carbon quantum dots with fluorescence characteristics in the growth of sesame will give rise to new ideas for waste-plant management and biological control, and strengthen the importance of nanomaterials in related fields.

β-carbolines (harman and norharman) are potentially mutagenic and have been reported in some vegetable oils. Sesame seed oil is obtained from roasted sesame seeds. During sesame oil processing, roasting is the key procedure to aroma enhancement, in which β-carbolines are produced. Pressed sesame seed oils cover most market share, while leaching solvents are used to extract oils from the pressed sesame cake to improve the utilization of the raw materials. β-carbolines are nonpolar heterocyclic aromatic amines with good solubility in leaching solvents (n-hexane); therefore, the β-carbolines in sesame cake migrated to the leaching sesame seed oil. The refining procedures are indispensable for leaching sesame seed oil, in which some small molecules can be reduced. Thus, the critical aim is to evaluate the changes in β-carboline content during the refining of leaching sesame seed oil and the key process steps for the removal of β-carbolines. In this work, the levels of β-carbolines (harman and norharman) in sesame seed oil during chemical refining processes (degumming, deacidification, bleaching and deodorization) have been determined using solid phase extraction and high performance liquid chromatography-mass spectrometry (LC-MS). The results indicated that in the entire refining process, the levels of total β-carbolines greatly decreased, and the adsorption decolorization was the most effective process in reducing β-carbolines, which might be related to the adsorbent used in the decolorization process. In addition, the effects of adsorbent type, adsorbent dosage and blended adsorbent on β-carbolines in sesame seed oil during the decolorization process were investigated. It was concluded that oil refining can not only improve the quality of sesame seed oil, but also reduce most of the harmful β-carbolines.Keywords:adsorbent;β-carbolines;harman;norharman;sesame seed oil

The Sesamum indicum bast contains more hemicellulose, lignin, and flaky parenchymal cells than the commonly used ramie bast, which are rigid for the degumming process. We explored a facile and greener method by using glycolic acid microwave-assisted treatment combined with alkaline treatment to effectively and low-cost prepare the high-quality Sesamum indicum single fibers. The chemical and physical properties of fibers were analyzed by chemical composition analysis, SEM, FTIR, XRD, TG, and GPC analysis. The residual gum content was decreased from 34.4% to 4.2%, resulting in a cleaner micro-surface of the Sesamum indicum fibers with increased cellulose crystallinity (75.2%–82.5%) and thermal stability (357°C–365°C). The fiber fineness (2.5 dtex) and breaking tenacity (4.9 cN/dtex) of the fibers produced by the novel treatment method exhibited the same level compared with the traditional two-step alkaline fibers. Besides, both the consumption of water and the chemicals was decreased compared with traditional degumming. This work provided a novel green and facile degumming method for processing the natural bast fibers.

BACKGROUND Sesame oil has an excellent flavor and is widely appreciated. It has a higher price than other vegetable oils because of the high price of its raw materials, and different processing techniques also result in products of different quality levels, which can command different prices. In the market, there is a persistent problem of adulteration of sesame oil, driven by economic interests. The screening of volatile markers used to distinguish the authenticity of sesame oil raw materials and production processes is therefore very important. RESULTS In this work, six markers related to the production processes and raw materials of sesame oil were screened by gas chromatography–tandem mass spectrometry (GC-MS/MS) combined with chemometric analysis. They were 3-methyl-2-butanone, 2-ethyl-5-methyl-pyrazine, guaiacol, 2,6-dimethyl-pyrazine, 5-methyl furfural, and ethyl-pyrazine. The concentration of these markers in sesame oil is between 10 and1000 times that found in other vegetable oils. However, only 3-methyl-2-butanone and 2-ethyl-5-methyl-pyrazine differed significantly as the result of the use of different production processes. Except for guaiacol, which was mainly derived from raw materials, the other five compounds mentioned above all result from the Maillard reaction during thermal processing. The six compounds mentioned above are sufficient to distinguish fraud involving sesame oil raw materials and production processes, and can identify accurately adulteration levels of 30% concentration. CONCLUSION In this study, the classification markers can identify the adulteration of sesame oil accurately. These six compounds are therefore important for the authenticity of sesame oil and provide a theoretical basis for the rapid and accurate identification of the authenticity of sesame oil. © 2021 Society of Chemical Industry.

Sesamol is a momentous marker for evaluating the quality of sesame or sesame products. In this study, a new strategy for fabricating portable smartphone sensor based on violet phosphorene (VP)/two-dimensional transition metal borides (MBene) nanozyme modified with Au − Ag bimetallic nanoparticles (Au − AgNPs) on the surface of the screen-printed electrode, which is synthesized under the guidance of machine learning (ML) with eXtreme Gradient Boosting (XGBoost) algorithms, is described for intelligent sensing of sesamol in farmland and agricultural fields. A fluorine-free hydrothermal-assisted alkane etching strategy is used to prepare the fence-like MoAlB-MBene. Highly environmentally stable VP/MoAlB-MBene nanozyme decorated by Au − Ag bimetallic nanoparticles is obtained by ultrasonic-assisted liquid-phase exfoliation with XGBoost assistance. The ML models including artificial neural network, support vector machine and XGBoost for the synthesis of nanozyme are contrasted and discussed, and the XGBoost algorithm is found as the most accurate model for synthesizing nanozyme due to the highest value of correlation coefficient. The nanozyme-modified screen-printed carbon electrode coupled with the palm-sized smartphone-controlled wireless electrochemical device as a portable wireless intelligent sensing platform for the determination of sesamol in a wider linear range of 0.1 ∼ 1000 μmol/L with a lower limit of detection of 21 nmol/L, which enable the on-site determination of 10 varieties of sesame seeds. This work will provide a new reference for the one-step preparation of bimetallic nanoparticles decorated graphene-like nanozyme guided by ML for cost-effective, simple, fast detection of phytochemicals in agricultural products using a portable smartphone sensor.

Recently, we found that vanillin commonly exists in sesame oils, while vanillin compounds are not allowed to be used as food additives in edible vegetable oils according to Chinese standards for Food Additives (GB2760-2014). To trace the source of vanillin in sesame oil, an accurate and reliable method was developed and validated for the determination of vanillin, methyl vanillin and ethyl vanillin in sesame samples based on high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The limit of determination (LOD) and limit of quantification (LOQ) of the established method were in the range of 17.4–33.1 and 52.2–99.2 μg kg −1 , respectively. The calibration curves of the three vanillin compounds exhibited good linearity between 0 and 400 ng mL −1 , with a high correlation coefficient greater than 0.999. The recoveries ranged from 91.7% to 112%, with a low relative standard deviation (RSD) less than 5.55%. Then this method was applied for the detection of sesame seed and intermediate samples. The results showed that all samples contained vanillin in a concentration range of 382–3804 μg kg −1 , while methyl vanillin and ethyl vanillin were not detected in those samples. Hence, the vanillin in sesame oil may originate from its raw materials, indicating that vanillin was not a good detection marker for illegal addition of flavor enhancers in sesame oils.
