Unraveling the molecular interactions and antioxidant mechanism of β-lactoglobulin with aloe’s hydroxyanthracene derivatives: A focus on aloin and aloe-emodin

Mohan Li,Na Gan,Dian Zhou,Yi He,Fang Geng,Wei Wang,Di Wu

Aloe dairy products have occupied a certain market share, and the interaction between the small bioactive molecules contained in aloe and milk proteins during product processing profoundly affects the performance of dairy products. In this study, two typical and potentially dangerous bioactive small molecules of aloe, aloin (AA) and aloe-emodin (AE), were selected to investigate their interaction mechanism with β -lactoglobulin ( β -La). Computer simulations showed that AA/AE had the potential to form complexes with β -La, and the β -La-AA system exhibited a stronger binding affinity than the β -La-AE system. Fluorescence analysis showed that the binding affinity values of AA to β -La at 298 K were deduced to be (4.184 ± 1.06) × 10 4  M −1 , meaning medium-strength interactions, comparatively, AE to β -La was weaker, were deduced to be (3.15 ± 1.25) × 10 3  M −1 . In addition, the thermodynamic analysis found that ΔH 0 and ΔS 0 were positive in both the β -La-AA system and the β -La-AE system, which implies that the main driving force for the combination of AA/AE and β -La to form the ground state complex is the hydrophobic force. The conformation changes in β -La brought about by binding to AA/AE may alter the properties and functions of the protein, such as surface hydrophobicity and oxidation resistance. Analysis shows that β -La is potentially present in whey as an AA/AE carrier during the production of aloe milk.

Main anthraquinone components in Aloe vera and their inhibitory effects on the formation of advanced glycation end-products

Shimo Kang,Xin Zhao,Lu Yue,Ling Liu

Food heating process causes increased protein glycation and the formation of advanced glycation endproducts (AGEs) which related with diabetes and ageing. Aloe vera ( A. vera ) extract is a potent antioxidant with established effects attributed largely to a key ingredient anthraquinone. This study investigated the inhibitory ability of the three anthraquinones from A. vera on the formation of AGEs in a heated model system at 70°C. Anthraquinones, including aloe-emodin (AED), emodin (ED), and chrysophanol (CSN), were extracted from A. vera and analyzed by ultra-high performance liquid chromatography-tandem mass spectrometry. Under optimal extraction conditions, the extraction yield of the anthraquinones was 4.377 mg/g. Anthraquinones showed good inhibition toward the formation of glycation products; the inhibition rates of AED, ED, and CSN on crosslinked AGEs were 46.48%, 66.38%, and 57.32% at 10 μM concentrations. Sodium dodecyl sulfate–polyacrylamide gel electrophoresis images also proved that the anthraquinones could significantly inhibit crosslinked AGE formation in a time-dependent manner. ED showed higher inhibitory ability than AED and CSN on the formation of N ɛ -(carboxymethyl)lysine and crosslinked AGEs. The results indicate that the antiglycation properties of these anthraquinones are likely related to the phenolic hydroxyl groups in their structures, but this ability is not related to their antioxidant activity, which probably depends on the whole structure of the quinone. Generally, the anthraquinones in A. vera , especially ED, can effectively inhibit AGE formation and reduce protein crosslinking during food processing, even at low concentration. Practical applications The inhibitory effects of three anthraquinones extracted from Aloe vera on the formation of AGEs were investigated in a heated model system. The results indicate Aloe can be used as an effective inhibitor of glycation products in food processing, moreover, this research can be useful for finding suitable nature inhibitors of AGEs for food processing.

Yilei Wang,Yingbing Zhang,Zhuangsheng Lin,Ting Huang,Wen Li,Wanxin Gong,Yuhui Guo,Jianmin Su,Jinyi Wang,Qin Tu

Past commercial and clinical research on wound dressings generally only focused on their therapeutic activities, however, how to make products environmentally friendly with reduced production of hazardous waste while ensuring product functionality is becoming a crucial and burning issue in the manufacturing industry. To address this problem, we developed a natural, degradable and environmentally friendly hydrogel wound dressing using aloe vera (AV) as an active ingredient. The hydrogel wound dressing was prepared using all natural ingredients, comprised of sodium hyaluronate (SH), dopamine (DA), chitosan (CS), and AV, and using a green natural deep eutectic solvent (DES) as a green solvent. The resultant DES-SH-CS/DA/AV hydrogel was easy to degrade and the preparation process was environmentally friendly. The chemical and physical properties of the hydrogel samples were characterized. The DES-SH-CS/DA/AV hydrogel showed good cytocompatibility tested using NIH-3T3 fibroblast cells, and antibacterial properties against both gram-positive ( S. aureus ) and gram-negative ( E. coli ). Furthermore, the hydrogel promoted skin tissue regeneration healed the wound of mouse skin after surgery within 12 days. The H&E staining was used to assess the integrity of the regenerated epidermis and dermis and the new generation of skin attachments in this work. Overall, our study showed the DES-SH-CS/DA/AV hydrogel we developed has the potential to be used as a natural and green wound dressing.

Studies on polysaccharides from leaf skin of Aloe barbadensis Miller: Part II. Structural characteristics and molecular properties of two lower molecular weight fractions

Xiao-Dan Shi,Jun-Yi Yin,Liu-Jing Zhang,Ou-Ye Li,Xiao-Jun Huang,Shao-Ping Nie

Aloe barbadensis Miller ( Aloe vera ) has long been used in food , cosmetic and pharmaceutical industries. The solid wastes such as the spikes and the outer green skins are usually separated from the inner gel and discarded during the process of Aloe vera leaves. Three kinds of polysaccharides (designated as ASP-4N, ASP-6N and ASP-8N) were successfully purified from the leaf skin of Aloe vera in our previous work. The present study aimed to analyze the structural and molecular features of ASP-6N and ASP-8N using methylation and GC-MS, NMR, high performance size-exclusion chromatography coupled with multi-angle laser light scattering (HPSEC-MALLS) and scanning electron microscopy (SEM) techniques. Results showed that ASP-6N and ASP-8N were β-(1→4)-glucomannans with acetyl groups, which may attach to O -2, O -3 or O -6 of mannopyranosyl residues in the backbone as mono-, di-, or tri-acetylated forms. HPSEC-MALLS analysis indicated that the polysaccharide fractions from Aloe leaf skin adopted various conformations in the aqueous solution depending on their molecular mass. The polysaccharides had fibrous, ribbon-like and spherical morphology by SEM observation.

A Novel Aloe Vera-Loaded Ethylcellulose/Hydroxypropyl Methylcellulose Nanofibrous Mat Designed for Wound Healing Application

Mohebian Zahra,Tajmohammadi Issa,Yavari Maroufi Leila,Ramezani Soghra,Ghorbani Marjan

Newly, the usage of nanofibers (NFs) as wound dressings with the aim of their rapid healing and prevention of bacterial infection has been considered by researchers. In this regard, we produced the ethylcellulose/hydroxypropyl methylcellulose NFs incorporated with Aloe-vera (EC/HPMC/Alv) by the electrospinning technique. The produced NFs were investigated for their chemical structure, morphological, mechanical, degradation, swelling, cell viability, and antibacterial properties. Amongst the produced NFs, the NF samples containing 10% Alv illustrated the appropriate mechanical properties. The produced NFs did not show any cell cytotoxicity which indicates their good compatibility. Also, NFs containing Alv significantly (P < 0.05) increased cell proliferation and adhesion. In addition, the NFs/Alv sample was indicated antibacterial ability against Staphylococcus aureus (10.21 ± 1.21 mm) and Escherichia coli (5.06 ± 1.3 mm) pathogenic bacteria. As a result, these findings suggest that the produced NFs could be applied as an active mat for wound dressing application.

Determination of four salicylic acids in aloe by in vivo solid phase microextraction coupling with liquid chromatography-photodiode array detection

Xu’an Fang,Guosheng Chen,Junlang Qiu,Jianqiao Xu,Junhui Wang,Fang Zhu,Gangfeng Ouyang

In recent years, great concerns have been raised about salicylic acid (SA) and its derivatives as plant regulators. Therefore, precise determination of the distribution of SAs in the living plants is necessary for not only fundamental researches but also the regulating mechanisms. In this study, a custom-made solid phase microextraction (SPME) fiber based on diallyl dimethyl ammonium chloride-assembled graphene oxide-coated C18 composite (C18@GO@PDDA) was proposed for in vivo detection of salicylic acid, acetylsalicylic acid (ASA), 4-methyl salicylic acid(4-SA)and 3-methyl salicylic acid (3-SA) in aloe plants. Under the optimized conditions, the analytical performance evaluated in homogenized aloe plant tissues exhibited low detection limits (1.8–2.8 μg g −1 ), wide linear ranges (10–5000 μg g −1 ), and satisfactory reproducibility (relative standard deviations less than 8.4% and 9.3% for inter-fiber and intra-fiber assays, respectively). Under cadmium stress, the developed method was applied for the in vivo tracing of four salicylic acids in aloe plants. A 48-h in vivo tracing revealed that salicylic acids were involved in the pathway of cadmium stress tolerance. To our best knowledge, it is the first effort to realize the in vivo analysis of SA and its derivatives in plants, and it has a made a great step forward in the area of plant hormone analysis.

Chemical Investigation of Major Constituents in Aloe vera Leaves and Several Commercial Aloe Juice Powders

Zhang Yuehong,Bao Zhichao,Ye Xiaoyan,Xie Zhaoyang,He Kan,Mergens Bill,Li Wenjie,Yatcilla Mike,Zheng Qunyi

Background: There are a substantial number of papers in the scientific literature reporting on the chemical composition of the Aloe vera plant. None of these investigations are truly comprehensive nor address the differences in composition that occur through processing variations in fresh leaves and commercially available product forms. Objectives: This work was to analytically examine a range of these forms and compile the findings. Methods: Fresh A. vera leaves and a number of commercial aloe juice powders were investigated for their major chemical constituents. Samples included fresh leaves from China and Mexico, plus commercial powders from different manufacturers made from different plant parts and/or manufacturing processes. The test results include moisture, ash, fiber, protein, lipids, minerals, organic acids, free sugars, and polysaccharides. The analytical methods employed comprise inductively coupled plasma-optical emission spectroscopy for minerals, high-performance anion-exchange chromatography equipped with pulsed amperometric detection for free sugars, HPLC for organic acids, and size exclusion chromatography (SEC)–multi-angle laser light scattering (MALS)–differential refractive index (dRI) for polysaccharide analyses. The absolute MW and MW distribution were determined using MALS measurement. Results: The major constituents of A. vera fresh leaf are fibers, proteins, organic acids, minerals, monosaccharides, and polysaccharides, which accounted for 85–95% of the total composition determined. In the commercial powdered aloe juice samples, four major components—organic acids, minerals, monosaccharides, and polysaccharides—accounted for 78–84% of the total composition. Apart from the four major components, products manufactured by ethanol precipitation contained high amounts of fiber and protein, while the free sugars were removed. In ethanol-precipitated products, the polysaccharide MW was less affected by manufacturing conditions and the concentration of aloe polysaccharides was higher than in products made in the nonethanol manufacturing processes. The overall chemical profiles were found to be consistent, except for the MW and content of polysaccharides in the commercial aloe samples analyzed, which were largely dependent on the types of manufacturing processes employed. Conclusions: This present study provides a comprehensive investigation of the major chemical composition of A. vera leaf and commercially derived products. The use of the SEC combined with MALS and differential RI detectors has proved to be an improved tool for the accurate determination of polysaccharide MW and contents of the various commercially available A. vera products in this study.

Study on fecal fermentation characteristics of aloe polysaccharides in vitro and their predictive modeling

Chang Liu,Peng Du,Yuliang Cheng,Yahui Guo,Bin Hu,Weirong Yao,Xuan Zhu,He Qian

Aloe polysaccharides (APs) are well-known plant polysaccharides, but little is known about their digestion and fermentation characteristics in vitro . In this study, the molecular weight of APs had no significant changes after gastric and intestinal digestion. During the fecal fermentation, the content of volatiles and pH value decreased continuously, while the short-chain fatty acids (SCFAs) concentration increased significantly. Additionally, the abundance of the microbiota associated with the metabolism of SCFAs was increased, including Prevotella , Catenibacterium , Lachnospiraceae , and Coprococcus , while the harmful microbiota was decreased, like Escherichia - Shigella , and Veillonella . Moreover, bioinformatics analysis indicated that APs boosted fructose and mannose metabolism, and the gene expressions of enzymes, containing mannose-6-phosphate isomerase [EC:5.3.1.8]. Structural equation modeling also highlighted that SCFAs-producing microbiota were primary degraders of APs, suggesting APs may facilitate the manufacture of functional foods with the purpose of maintaining intestinal health.

Cadmium removal from rice protein via synergistic treatment of rhamnolipids and F127/PAA hydrogels

Chong Shen,Shi Tang,Qin Meng

Heavy metal ions can accumulate in the area’s crop harvest, such as rice, via a tight binding with protein. Such binding paves great difficulties to remove heavy metal ions. This study aims to remove cadmium from contaminated rice protein using rhamnolipid biosurfactant together with Pluronic F127/poly (acrylic acid) (i.e., F127/PAA) hydrogels. The two-step consequential washing removed 92% of cadmium in rice protein, making the residual cadmium content below the safety level (<0.2 mg/kg) without impairing the main components and structure of rice protein. Interestingly, the washing by either rhamnolipids or hydrogels failed to effectively remove cadmium, indicating the synergistic effect of the two materials. Mechanistically, rhamnolipids with higher Cd 2+ binding constant compete with protein on binding with cadmium and then deliver it to F127/PAA hydrogels that present highest Cd 2+ adsorptive capability for immobilization. In considering that the F127/PAA hydrogels are reusable, this treatment is low-cost, safe and effective for removal of cadmium from polluted rice products.

Aloe vera polysaccharides mitigate high-fat high-cholesterol diet-induced atherosclerosis in ApoE-/- mice via regulation of lipid metabolism and gut microbiota

Shenji Yang,Xiaojun Huang,Xiajialong Li,Jing Feng,Peng Chen,Zhikang Cao,Luyao He,Chaojun Li,Zhipeng Li,Ming-Yong Xie

Cardiovascular diseases are leading causes of death globally, often manifesting after years of atherosclerosis (AS) progression. In this study, we investigated the atheroprotective effects of three different sources of glucomannan, Dendrobium officinale polysaccharide, Konjac glucomannan, and Aloe vera polysaccharide (AVP) using an in vitro ox-LDL induced foam cell model and an in vivo high-fat high-cholesterol diet-fed ApoE-/- mouse model. Both settings indicate that AVP exerts the most significant atheroprotective effects. It inhibits lipid absorption and enhances the regulation of lipid homeostasis by Liver X Receptor, thereby suppressing the formation of foam cells. It can also alleviate ox-LDL-induced oxidative stress and apoptosis in RAW 264.7 cells. Animal experiments show that AVP can prevent the formation of atherosclerotic plaques and coronary artery fibrosis, while also reducing circulating IL-1β levels. Furthermore, liver transcriptomic analysis shows that AVP inhibits inflammation, and promotes bile acid excretion and transport by upregulating Farnesoid X Receptor. Additionally, metagenomic analysis indicates that AVP can significantly reverse the microbial alterations associated with AS. Specific gut microbes, such as Prevotella, may partially mediate the effects of AVP through the gut-liver axis. This is the first study to report the atheroprotective effects of AVP, demonstrating that it alleviates atherosclerosis by restoring lipid metabolism homeostasis and modulating the gut microbiome.

Evaluation on the antiviral activity of aloe emodin against Micropterus salmoides rhabdovirus in vitro and in vivo

Yao Wang,Mengmeng Wang,Zhiyang Lin,Guangshuo Wang,Yunlong Qin,Mingzhu Liu,Fei Ling,Haifeng Jiang,Tianqiang Liu,Gaoxue Wang

Micropterus salmoides rhabdovirus (MSRV) is a highly pathogenic rhabdovirus that has caused significant economic losses in the aquaculture industry of largemouth bass. To date, there is no approved treatment for MSRV infection. In order to address this urgent need, virtual screening of the natural product monomer compound library targeting MSRV glycoprotein was conducted and the antiviral activity of 13 natural compounds identified in the screening was studied. Among these compounds, Aloe emodin (AE) exhibited high binding affinity and antiviral activity, inhibiting MSRV replication in grass carp ovary (GCO) cells with a half-maximal inhibitory concentration (IC 50 ) of 2.55 μM. Additionally, viral titers and cytopathic effects (CPE) were significantly reduced in the presence of AE. AE was demonstrated to directly destroy MSRV virions and inhibit their adsorption to GCO cells according to ultracentrifugation assay and viral binding assay. In vivo studies demonstrated that AE injection increased the survival rate of MSRV-infected largemouth bass by 63.3 %, which was 46.7 % higher than that of the virus group. RT-qPCR and pathological tissue section results demonstrated that AE treatment could significantly reduce viral loads in the liver, spleen, and kidney on days 1, 4, and 7 post-infection, and AE effectively mitigated tissue damage caused by MSRV infection. Circular dichroism (CD) spectroscopy and Isothermal titration calorimetry (ITC) showed that AE induced changes in the secondary structure of the MSRV glycoprotein and exhibited high affinity for the glycoprotein (Kd = 2.803 × 10 −5  M, ΔG = −25.99 kJ/mol). These results suggested that AE was a natural product with potent anti-MSRV activity through direct targeting of the glycoprotein.

Interaction mechanism of aloe-emodin with trypsin: molecular structure–affinity relationship and effect on biological activities

Guoyan Ren,He Sun,Gen Li,Jinling Fan,Lin Du,Guoting Cui

The molecular mechanism of interaction between aloe-emodin (AE) and trypsin was investigated, exhibiting remarkable outcomes. To detect the interaction mechanism, the binding of AE with trypsin was examined by a multi-spectroscopy and molecular docking method. Results showed that the binding of AE and trypsin would lead to static quenching and their binding forces were van der Waals forces and hydrogen bonding. The results of simultaneous and three-dimensional fluorescence spectroscopy showed that the combination of AE and trypsin caused changes in the microenvironment around the trypsin fluorophore, which might change the spatial structure of trypsin. FT-IR spectroscopy showed that the contents of α-helix and β-turn in trypsin were decreased and the contents of β-sheet, random coil and antiparallel β-sheet were increased. Moreover, all these experimental results were verified and reasonably explained by molecular docking results. We also investigated the enzyme activity of trypsin and the antioxidant activity of AE. The results showed that both the enzyme activity of trypsin and the antioxidant activity of AE were decreased after interaction between AE and trypsin. The findings outlined in this study should elucidate the molecular mechanisms of interaction between AE and trypsin and contribute to making full use of AE in the food industry.
