The problem of opioid abuse has become a global problem. Thus, creating an urgent need for highly sensitive detection of opioid substances. In this work, we developed a method for the controllable preparation of Ag@Au nanocrystals (Ag@Au NCs) for highly sensitive SERS detection of fentanyl and its analogs. By regulating the concentration of ligands on the surface of silver seed, we successfully prepared Ag@Au NCs with three different morphologies, including core-satellite, yolk shell and hollow structure. Firstly, we explored the SERS-enhancing effect of Ag@Au NCs with different morphology using rhodamine 6G as the molecule to be tested. The results show that the core-satellite Ag@Au NCs has the best SERS effect, and the lowest detection concentration for R6G reached to 10 -10 M. Furthermore, we used the prepared core-satellite Ag@Au NCs to detect fentanyl and its five analogs, including carfentanyl, furanylfentanyl, thiofentanyl, 4-fluorobutyrfentanyl and N-4-piperidylacetanilide. Trace detection was achieved for the above six substances. For the environmental water samples spiked with fentanyl, the calculated recovery was 89.2% with an RSD value of 7.3%. Moreover, in order to realize the qualitative analysis of the characteristic peaks of different fentanyl analogs, we performed DFT calculations on the Raman spectra of the above-mentioned 6 substances. By analyzing the DFT calculation results, conventional Raman spectroscopy and SERS spectroscopy, we realized the distinction of six fentanyl analogs with similar structures.

Electrochemical determination of synthetic opioids such as fentanyl is meaningful but still challenging no matter from a social or academic perspective. Herein, we report a series of novel electrocatalysts based on Fe-containing metal–organic frameworks and multi-walled carbon nanotubes (Fe-MOF/MWCNTs). The obtained Fe-MOF/MWCNT electrode materials all show ultrasensitivity on fentanyl determination. In particular, MOF-235/MWCNTs even exhibit an ultra-low limit of detection (LOD) of 0.03 μM with a wide linear range from 0.1 to 50 μM. Besides, this series of Fe-MOF/MWCNTs also displays excellent repeatability, selectivity, and stability. Moreover, they show good performance in real sample detection and achieve good recovery of 95.47%–102.41% and 96.62%–103.15% in blood and urine samples, respectively. This high performance in fentanyl determination is mainly contributed by the Fenton-like process and the adsorption function of the Fe-MOF. Therefore, these novel Fe-MOF/MWCNTs are promising electrocatalysts for point-of-care device fabrication and also have potential applications in fentanyl rapid test technology.

Recently, the phenomenon of fentanyls overdose leading to death is emerging in an endless stream. There is an urgent requirement to quickly identify fentanyl content in body fluids for medical and judicial purposes. With this in mind, we present a paper-based SERS substrate decorated with uniform gold nanospheres treated by chloride ion for the detection of fentanyl citrate in urine and serum. In particular, the paper-based SERS sensor was prepared by liquid/liquid self-assembly technique and chloride ion was introduced to clean and modify the substrate surface, which improved the sensitivity of the solid substrate with an enhancement factor (EF) as high as 1.64 × 10 5 . Moreover, the uniformity of each paper-based substrate and the repeatability on different batches of substrate were excellent, and there was no obvious change in the intensity response of Raman spectra within a month. As a result, the quantitative analysis of fentanyl citrate in artificial urine and rat serum were performed based on the modified paper-based substrate with the limit of detection as low as 0.59 μg/mL and 2.78 μg/mL, respectively. Both the concentrations of the two biological samples with the Raman signal intensity were linearly plotted and the recovery of the spiked samples with different concentrations was collected to verify the accuracy of the quantitative curves. All the results suggest that this work makes SERS method available for the rapid identification and quantitative analysis of illicit drug in the real biological samples.

Bimetallic nanoparticles have received extensive attention due to their unique physical and chemical properties, including enhanced optical properties, chemical stability, and better catalytic activity. In this article, we have successfully achieved the controllable preparation of Ag@Au nanoparticles via a seed-mediated growth method. By regulating the amount of seeds—silver nanospheres—we realized that Ag@Au nanoparticles gradually changed from spherical to a sea-urchin-like structure. The structure and composition of the prepared nanoparticles were characterized via scanning electron microscopy (SEM), transmission electron microscopy (TEM), energy-dispersive X-ray spectroscopy (EDS), and high-angle circular dark field scanning transmission electron microscopy (HAADF-STEM). In addition, we use the prepared Ag@Au nanoparticles as a substrate material for highly sensitive surface-enhanced Raman spectroscopy (SERS). Using 4-aminothiophenol (4-ATP) as the test molecule, we explored the SERS enhancement effects of Ag@Au nanoparticles with different structures. Furthermore, we used Ag@Au nanoparticles for SERS to detect the drug fentanyl, and realized the label-free detection of fentanyl, with the lowest detection concentration reaching 10−7M. This research not only provides a method for preparing bimetallic Ag@Au nanoparticles with different structures, but also provides a reference for the application of Ag@Au nanoparticles in the field of detection technology.Keywords:Ag@Au nanoparticle;core–shell structure;sea-urchin-like structure;SERS;fentanyl

Colorimetric assays are inexpensive and attractive tools for the detection of fentanyl (FTN), yet further enhancing their sensitivity remains a major challenge. Herein, we develop a halogen-bond mediated visual competitive colorimetric assay for FTN using Erythrosine B (EB) as a probe. The addition of poly(ethylene oxide)-block-poly(propylene oxide)-block-poly(ethylene oxide (F-127) induces the EB aggregation, strongly suppressing the background signal. Upon the introduction of FTN, the subsequent competitive reaction displaces F-127 to generate EB-FTN charge-transfer complexes via N … I halogen bonds between electron-rich amine groups and electron-deficient iodine sites, accompanying a significant absorbance wavelength shift and pink-to-purple color change. The limits of detection of this approach for FTN are 2 mg·L −1 by the naked eye and 0.19 mg·L −1 by UV–vis spectroscopy, which are approximately 3.7-fold to 4 orders of magnitude more sensitive than the reported colorimetric assays. Meanwhile, the present method is well applied for FTN-spiked domestic sewage samples, and an easy-to-use smartphone-based digital image colorimetry is also fabricated. It is expected that such an assay can play a key role in alleviating the worldwide opioid overdose crisis.

In this study, a biochar-based magnetic solid-phase microextraction method, coupled with liquid chromatography–mass spectrometry, was developed for analyzing fentanyl analogs from urine sample. Magnetic biochar was fabricated through a one-step pyrolysis carbonization and magnetization process, followed by an alkali treatment. In order to achieve desired extraction efficiency, feed stocks (wood and bamboo) and different pyrolysis temperatures (300–700°C) were optimized. The magnetic bamboo biochar pyrolyzed at 400°C was found to have the greatest potential for extraction of fentanyls, with enrichment factors ranging from 58.9 to 93.7, presumably due to H-bonding and π–π interactions between biochar and fentanyls. Various extraction parameters, such as type and volume of desorption solvent, pH, and extraction time, were optimized, respectively, to achieve the highest extraction efficiency for the target fentanyls. Under optimized conditions, the developed method was found to have detection limits of 3.0–9.4 ng/L, a linear range of 0.05–10 μg/L, good precisions (1.9–9.4% for intrabatch, 2.9–9.9% for interbatch), and satisfactory recoveries (82.0–111.3%). The developed method by using magnetic bamboo biochar as adsorbent exhibited to be an efficient and promising pretreatment procedure and could potentially be applied for drug analysis in biological samples.

A cost-effective and highly reproducible capillary-based surface-enhanced Raman scattering (SERS) platform for sensitive, portable detection and identification of fentanyl is presented. Through encapsulating gold trisoctahedra (Au TOH) in the capillary tube for the first time, the SERS platform was constructed by combining the superior SERS properties of Au TOH and the advantages of capillaries in SERS signal amplification, facile sample extraction, and portable trace analysis. The effects of the size and density of Au TOH on the SERS performance were investigated by experiments and simulations, which showed that the maximum SERS enhancement was obtained for Au TOH with the size of 75 nm when particle density reached 74.54 counts/μm2. The proposed SERS platform possesses good reproducibility with a relative standard deviation (RSD) of less than 5%. As a demonstration, the platform was applied to detect fentanyl spiked in aqueous solution and serum samples with a limit of detection (LOD) as low as 1.86 and 40.63 ng/mL, respectively. We also validated the feasibility of the designed platform for accurate identification of trace fentanyl adulterated in heroin at mass concentration down to 0.1% (10 ng in 10 μg total). Overall, this work advances more explorations on capillary-based SERS platform to benefit portable trace analysis.

Surface-enhanced Raman spectroscopy (SERS) is a rapidly emerging technology that offers a fast, extremely sensitive, and quantitative approach to trace chemical detection. Such a technique incorporated into a portable device is attractive to law enforcement and emergency room personnel for rapid accurate on-site screening of illicit and abused drugs. Toward this, we are developing an analyzer in a hand-held unit, based on SERS that is integrated with a capillary sensor embedded with activated gold nanoparticles in a porous glass matrix. In this preliminary study, we have used this gold sol–gel capillary to measure aqueous solutions of 14 high-priority drugs to define sensitivity (lowest measured concentration [LMC] and estimated limit of detection [LOD]), determine concentration dependence and quantification capabilities by constructing calibration curves, examine effects of pH at 3, 7, and 11 on SERS measurements, and perform multicomponent analysis of fentanyl laced drug mixtures and spectral identification on our portable and handheld units. Of particular significance, fentanyl was detected with an LMC of 1 ng/ml and estimated LOD at 0.11 ng/ml, while other representative drugs such as cocaine and phenylcyclidine produced LMCs and LODs at 5.0 and 0.77 ng/ml, and 10.0 and 0.62 ng/ml, respectively. These sub-nanogram per milliliter detection limits are comparable or lower than previously reported and confirm that this gold sol–gel has the potential to meet the sensitivity requirements for saliva analysis. Most importantly, these sensors can be manufactured easily and cheaply and when integrated with our portable Raman units produce high-quality spectra and accurate identification within 1.2 s.

In this work, semi-automated filter micro-solid phase extraction (SA-FμSPE) packed by urea–formaldehyde (UF) microspheres coupled with corona discharge ionization-ion mobility spectrometry (CD-IMS) was developed for the extraction and determination of fentanyl and nor-fentanyl in urine samples. UF microspheres, synthesized by aqueous polymerization, can be used to achieve effective extraction of analytes through multiple adsorption interactions such as hydrophobic, dipole–dipole and hydrogen bonding interactions. UF microspheres dispersed in methanol were packed in the filter by syringe, and the extraction process was performed by a portable automatic filter extraction device. The parameters affecting extraction efficiency (i.e., extraction time, the flow rate of extraction, pH value, amount of sorbent, ionic strength, cycle times of desorption, and type of eluent) were investigated. Then, the eluate containing fentanyl and nor-fentanyl was simultaneously analyzed by CD-IMS in positive mode. Under the optimal extraction conditions, the detection limits for the target compounds were 5.00 ng mL −1 and 8.00 ng mL −1 , respectively. The spiked recoveries were in the range of 81.97 % −112.37 % by testing the real samples and validated by UHPLC-MS. Thus, the satisfactory results reveal that SA-FμSPE coupled with CD-IMS has great potential for on-site extraction and detection of fentanyl and nor-fentanyl in urine samples.

The in vitro detection applications of europium complex-doped microspheres mainly rely on strong fluorescence intensity and a well-defined morphology. In this work, using methyl methacrylate-modified polystyrene microspheres has been proven an effective strategy to enhance the fluorescence and morphology of Eu-complexes. The experimental results showed that the modification resulted in the formation of a porous structure within the polystyrene microspheres, enhancing the doping uniformity and facilitating a more significant accumulation of fluorescent molecules. Furthermore, because of their encapsulation ability, microspheres efficiently confine the fluorescent molecules within them. In addition, the nano-scale porous structure endowed the microspheres with enhanced properties without compromising solvent swelling capability, thereby significantly boosting the fluorescence performance of porous PSMMA. In lateral flow immunoassays (LFIAs), PSMMA-Eu microspheres were effectively utilized to detect fentanyl with exceptional sensitivity by capitalizing on these benefits, capable of detecting concentrations as low as 0.10 ng mL−1. This technology has significant potential for rapid point-of-care screening and clinical applications.

Objective: This study aims to determine the drug concentration of etomidate, remifentanil, and rocuronium bromide for general anesthesia in fetus as well as the placental transport rate between term and preterm delivery, twins, and singleton. Study design: Sixty parturients with 72 fetuses undergoing cesarean section under general anesthesia were included. According to whether the fetus was a twin or premature, parturients were divided into Group I (term singleton), Group II (premature singleton), Group III (term twins), and Group IV (premature twins). The preoperative demographic characteristics and laboratory examination of parturients, hemodynamic indicators, the Apgar score of neonates at 1, 5, and 10 min after delivery and at specific assigned values, umbilical artery blood gas analysis results, neonatal weight, and resuscitative measures were recorded. Anesthetic drug concentrations in maternal arterial (MA), umbilical arterial (UA), and umbilical venous (UV) blood were detected by Ultra Performance Liquid Chromatography Tandem Mass Spectrometry (UPLC-MS/MS). Result: No significant differences were observed in the concentrations of etomidate, remifentanil, and rocuronium bromide in MA, UV, and UA blood, or in the UV/MA and UA/UV ratios between term and preterm infants, twins, and singletons. Moreover, there was no variation in the anesthetic drug concentration among each pair of twins. Additionally, no correlation was found between the neonatal weight and the plasma concentrations of anesthetic drugs in UV and UA blood, except for remifentanil in UA blood. Conclusion: Preterm or twin deliveries do not affect the neonatal concentration of etomidate, remifentanil, and rocuronium bromide used in general anesthesia for cesarean sections. In clinical diagnostics, human breath presents an alternative and more convenient sample than biofluids for detecting the ingestion of nonvolatile drugs. Surface-enhanced Raman spectroscopy (SERS) is a powerful vibrational spectroscopy technique with high sensitivity based on molecular fingerprinting. However, the low affinity of traditional SERS substrates for aerosols and the stochastic fluctuation of the SERS signal at low concentrations limit their application in breath aerosol analysis. In this study, we synthesized hydrogel microsphere SERS substrates with highly reversible swelling/shrinking properties that enhance target analyte accumulation in breath aerosols and promote plasmonic nanoparticle aggregation for intense Raman hotspot formation. Furthermore, these hydrogel microsphere SERS substrates function as a three-in-one system, enabling multilevel selectivity based on size, charge, and hydrophilicity for target molecules simultaneously without pretreatment. Notably, by “digitizing” the SERS signal of each individual hydrogel microsphere and calculating the proportion of positive microspheres, the hydrogel microspheres can serve as a digital SERS platform that circumvents the low stability issues resulting from fluctuations in SERS signal intensity. Consequently, the digital SERS platform achieved a detection limit of 0.5 ppm for fentanyl in simulated breath aerosols. This innovative sensing strategy not only demonstrates a promising approach for screening nonvolatile drugs but also simplifies the sampling process, holding great potential for clinical diagnosis of breath aerosols.
