Morphological, physiological and biochemical changes and effects of polyamines on regulation of Anoectochilus roxburghii during flowering period

Weiwei Jiang,Xiaoyun Yan,Chenfei Lu,Xiaowei Xu,Enting Xu,Dandan Chen,Yuqiu Huang,Qingsong Shao,Aimin Lv

Objective Anoectochilus roxburghii is a valuable medicinal and ornamental plant. The aim of this study is to investigate the morphological and biochemical responses during the flower development stages of A. roxburghii , and to assess the effects of exogenous polyamines (PAs) on bud differentiation and metabolism, thereby providing a theoretical basis for understanding the flowering form and physiology of A. roxburghii . Methods In this study, morphological and biochemical responses in flower development stages of A. roxburghii were investigated using paraffin sections and stereomicroscope. A. roxburghii was divided into five periods, including vegetative growth period, flower bud period, flowering period, late flowering period and fruiting period. During the flowering phase, specific biochemical parameters were measured, including soluble sugar content, superoxide dismutase (SOD) activity, soluble protein content, peroxidase (POD) activity, and catalase (CAT) activity. These measurements were conducted to understand the biochemical changes occurring within A. roxburghii during its flowering process. Furthermore, the effects of PAs on bud differentiation were examined. Additionally, the activities of S-adenosylmethionine decarboxylase (SAMDC) and polyamine oxidase (PAO), as well as the content of polyphenols, polysaccharides, and flavonoids in A. roxburghii , were measured after PA treatment to evaluate the metabolic changes induced by exogenous PAs. Results During the flowering phase of A. roxburghii , soluble sugar content and SOD activity were steadily declining. Soluble protein content was initially increasing and then reducing, and POD and CAT activities showed opposite pattern. In addition, the effects of exogenous PAs on bud differentiation were investigated. Results showed that 3 mmol/L putrescine or 0.3 mmol/L spermidine significantly promoted the bud differentiation of A. roxburghii and advanced the flowering. The activities of SAMDC, PAO, and the content of polyphenols, polysaccharides and flavonoids in A. roxburghii significantly increased after PA treatment, demonstrating that exogenous PA can accelerate metabolism and improved the active ingredients content. Conclusion The flower development of A. roxburghii was divided into five stages, with significant changes in soluble sugar, protein, POD, SOD, CAT, MDA, and PRO levels. Exogenous putrescine and spermidine enhanced bud differentiation and accelerated flowering, increasing SAMDC and PAO activities, suggesting accelerated PA metabolism. PAs also improved active component content. These findings provide a theoretical basis for studying flower morphology and PA-induced flowering regulation of A. roxburghii .

Accumulation patterns of major bioactive components in two chemotypes of Agastache rugosa during flower development evaluated by GC-QQQ-MS/MS and UPLC-QTOF-MS/MS analyses

Hui-Dan Hou,Cheng-Ying Wu,Jing Zhou,Fang Long,Hong Shen,Jin-Di Xu,Shan-Shan Zhou,Qian Mao,Ying-Jie Wei,Song-Lin Li

Agastache rugosa (AR) is a wildly cultivated medicinal and aromatic crop, which usually recommended to be harvested when the plant is exuberant with not flowering or in full flower stage. But there is no scientific basis for the support of the cognition. Therefore, the accumulation patterns of major active components during flower development (inflorescences with buds, full bloom and after full bloom stages) in two chemotypes (menthone type and estragole type) of AR were investigated by newly developed GC–QQQ–MS/MS and UPLC–QTOF–MS/MS methods. Five volatile (pulegone, menthone , estragole, methyleugenol and β -caryophyllene) and nine non-volatile components (rosmarinic acid, tilianin, acacetin 7- O -(6"- O -malonyl)- β - D -glucopyranoside, apigenin , agastachoside, isoagastachoside, acacetin 7- O -(2"- O -acetyl-6"- O -malonyl)- β - D -glucopyranoside, cirsimaritin and acacetin) in two chemotypes of AR samples at three stages were simultaneously quantified. It was found that the relatively higher contents of main volatile components were present in menthone type of AR collected at the full flower stage and in estragole type of AR at the after full bloom stage, whereas the relatively higher contents of main non-volatile components were present in menthone type of AR collected at the inflorescences with buds stage and in estragole type of AR at the after full bloom stage. Considering major active volatile and non-volatile components with higher levels, full bloom stage and after full bloom stage should be optimal harvesting time for menthone type and estragole type of AR, respectively.

Untargeted metabolomics analysis revealed metabolite dynamics during the development and processing of Rosa rugosa flowers

Xi Cheng,Xin Li,Dongliang Chen,Qiong Wang,Hongli Wang,Kang Gao,Yanchao Luo,Yanni Sun,Conglin Huang

Rosa rugosa , which originated in China, is an important industrial plant, which have been used for landscaping, while its components have been included in foods, pharmaceuticals, and beauty and skin care products. As a potential available oil rose variety, the limited comprehensive understanding of R. rugosa ‘Han Xiang’ has hindered their development and utilization. In this study, UPLC-MS/MS and GC-MS techniques were applied to analyze metabolite dynamics during the development and processing of R. rugosa flowers. In total, 1816 non-volatile metabolites and 1029 volatile metabolites were identified in the development and processing of R. rugosa flowers. Significant differences in the non-volatile and volatile metabolites were detected in the petals and hydrosol. The differentially regulated non-volatile metabolite contents were highest in the bud stage petals, the differentially regulated volatile metabolite contents were highest in the full-bloom stage petals. Some key metabolites affect the presentation of rose color, flavor, and taste, as well as the efficacy of the product. The aim of this article is to provide a comprehensive understanding of R. rugosa ‘Han Xiang’ and to serve as a basis for further research and development. This study provides useful data for assessing rose flower quality as well as new insights into R. rugosa development and utility.

Dynamic Changes in Flavonoids’ Accumulation Pattern in Tilia miqueliana Flowers at Different Developmental Stages Based on Widely Targeted Metabolomic Analysis

Wenqin Bao,Yongbao Shen,Julian C. Verdonk

Tilia miquelianais an endemic species belonging to the genusTiliaL. (Tiliaceae) in China, which is known for its fragrant flowers and nectar, but the dynamic changes in metabolites during its growth and development are still unclear. In this study, the metabolic profiles fromT. miquelianaflowers at three developmental stages were detected by performing an ultra-performance liquid chromatography–electrospray ionization–tandem mass spectrometry (UPLC-ESI-MS/MS)-based widely targeted metabolomic analysis. A total of 1138 metabolites were detected, with 288 Differentially Accumulated Metabolites (DAMs) determined, flavonoids accounting for the largest proportion. The trend analysis showed that DAMs present seven distinctive patterns, and subclass 5 obtained the largest amount with continuously increased relative content during flower development. The Kyoto Encyclopedia of Genes and Genomes (KEGG) annotation and enrichment analysis of DAMs showed different overlap and variability in metabolic pathways, indicating different directions of flavonoids’ metabolic flux in the three developmental stages. A correlation network analysis further revealed five core metabolites that played essential roles in flavonoid biosynthesis. This research provides comprehensive insights into the exploitation and utilization ofT. miquelianaas well as a scientific basis for phylogenetic studies of the genusTilia.

Global lysine succinylation analysis unveils post-translational regulation effect on phenylpropanoid metabolism remodeling during Lonicera japonica flower development

Yao Chen,Luhuizi Shan,Wenxi Zheng,Jie Chen,Linfang Deng,Xu Tian,Ruili Xie,Yunhong Yang,Lin Zhang,Bingxian Yang

Lonicera japonica plays a significant role in traditional Chinese medicine and as a food source, making it a focus of studies on protein succinylation and its potential role in regulating secondary metabolism during flower development. This study aimed to clarify the regulatory mechanism of protein succinylation on phenylpropanoid-related phenotypic changes by conducting a global lysine succinylation proteomic analysis across different flowering stages. A total of 586 lysine succinylated peptides in 303 proteins were identified during early and late floral stages. Functional enrichment analysis revealed that succinylated proteins primarily participated in the tricarboxylic acid (TCA) cycle, amino acid metabolism, and secondary metabolism. The abundance of succinylated aspartate transaminase (AT), 4-coumarate-CoA ligase (4CL), and phenylalanine N-hydroxylase (CYP79A2) in phenylpropanoid metabolism varied during flower development. In vitro experiments demonstrated that succinylation increased AT activity while inhibited 4CL activity. Decreased levels of total flavonoids and phenolic acids indicated significant alterations in phenylpropanoid metabolism during later floral stages. These results suggest that succinylation of TCA cycle proteins not only influences flower development but also, together with AT-4CL-CYP79A2 co-succinylation, redirects phenylpropanoid metabolism during flower development in L. japonica .
