Microwave-assisted hydrodistillation extraction based on microwave-assisted preparation of deep eutectic solvents coupled with GC-MS for analysis of essential oils from clove buds

Yao Chen,Fangxiang Xu,Mingmei Pang,Xiangzi Jin,Huihao Lv,Zuguang Li,Mawrong Lee

In recent years, deep eutectic solvents (DESs) as green and sustainable solvents have been widely used in the effective extraction of natural products. Usually, DESs are synthesized by heating and stirring method which takes a long time and energy. In this case, a microwave assisted preparation technique was used to form DESs for saving time and energy. And the DESs as pretreatment solvents were combined with microwave-assisted hydrodistillation (MAHD) for the extraction of essential oils (EOs) from the clove buds in this work. The maximum essential oils yield of 4.60% was obtained with the microwave power of 600 W, the pretreatment time of 5 min and the hydrodistillation time of 40 min using a deep eutectic solvent composed of choline chloride and lactic acid (molar ratio with 1:2). In addition, the essential oil was analyzed by gas chromatography-mass spectrometry (GC-MS), a total of 40 compounds were identified. Compared with MAHD, water-based MAHD and traditional hydrodistillation (HD), the content and composition of essential oil extracted by DES-based MAHD were higher. Thus, the combination of deep eutectic solvents with a microwave-assisted technique in this study provided an eco-friendly way of extracting essential oils.

Ethanol extract of Sophora japonica flower bud, an effective potential dietary supplement for the treatment of hyperuricemia

Danni Song,Honghui Zhao,Lingling Wang,Fuqi Wang,Lina Fang,Xu Zhao

Hyperuricemia (HUA) is a common metabolic condition associated with a variety of diseases, mainly manifested by the increase in uric acid (UA) levels. As xanthine oxidase (XOD) is the key enzyme in UA production, it has become a target for treating HUA. In this study, an extract named EESJ was extracted from Sophora japonica using 80% ethanol solution and administered to HUA mice to verify its UA-lowering effect. The results confirmed that administering EESJ could reduce the UA levels in vivo by inhibiting XOD activity, and ameliorating renal injury in a dose-dependent manner. To further study the active components in EESJ, 243 chemical components and 32 blood components were identified by the UHPLC-Q-TOF-MS/MS technology, of which flavonoids were the main constituents. The molecular docking results showed that the following six compounds, namely hydroxygenkwanin, genistein , tectorigenin, kaempferol , sophoricoside, and quercetin , had strong binding properties with XOD; among which hydroxygenkwanin had the best binding capacity. In conclusion, this study confirmed that EESJ could effectively reduce UA levels, and speculated the active compounds playing a role in lowering UA levels, which has certain guiding significance for Sophora japonica and natural compounds as potentially effective treatment options for HUA.

Bioactive sesquiterpenoids from the flower buds of Tussilago farfara

Xiu-Qing Song,Jin-Hai Yu,Jia Sun,Kai-Lu Liu,Jun-Sheng Zhang,Hua Zhang

Eleven new compounds including five bisabolane ( 1 – 5 ) and three oplopane ( 6 – 8 ) sesquiterpenoids, a pair of benzopyran enantiomers ( 9 & 10 ) and a benzofuran derivative ( 11 ), along with six known sesquiterpenoid co-metabolites ( 12 – 17 ), have been obtained from the flower buds of Tussilago farfara . Their structures were elucidated by comprehensive spectroscopic analyses and comparison with structurally related known analogues. The absolute configurations of all the compounds except 11 were unequivocally assigned by various techniques, including Mosher’s method and time-dependent density functional theory (TD-DFT) based calculations of 13 C NMR and electronic circular dichroism (ECD) data. The C-8 absolute configuration on the sidechain of this group of bisabolane sesquiterpenoids was assigned for the first time. Our bioassays have established that compounds 3 , 4 , 13 and 14 showed significant α -glucosidase inhibitory activities, while 6 , 8 and 14 displayed moderate antiproliferative effects against two human tumor cell lines A549 and MDA-MB-231. Further flow cytometric analysis revealed that 14 effectively induced cell apoptosis and arrested cell cycle at the S/G2 phases in A549 cells, in a dose-dependent manner.

Prenylated indole alkaloids and lignans from the flower buds of Tussilago farfara

Xiu-Qing Song,Jia Sun,Jin-Hai Yu,Jun-Sheng Zhang,Jie Bao,Hua Zhang

Six new compounds including four prenylated indole alkaloids ( 1 – 4 ) and two lignans ( 5 – 6 ), along with eight known cometabolites ( 7 – 14 ), were isolated from the flower buds of Tussilago farfara . Structures of the new compounds were elucidated by comparison with structurally related known analogues and also by comprehensive spectroscopic analyses. Their absolute configurations were determined by a variety of means including Mosher's method, Marfey's analysis, electronic circular dichroism (ECD) exciton chirality method and ECD calculations. Our bioassays have established that compounds 1 and 2 showed potent α -glucosidase inhibitory activity with IC 50 values of 105 ± 4.7 and 35.2 ± 3.2 μM, respectively, while the known 13 and 14 exerted moderate DPPH radical scavenging activity with IC 50 values of 45.2 ± 2.9 and 29.2 ± 2.0 μM, respectively.

Integrated UHPLC-Q-Orbitrap-MS/MS Method and Network Pharmacology for Exploring the Active Components and Potential Mechanisms of Neuroprotective Effect of the n-Butanol Part of Mume Flos

Xue Wang,Hongsu Zhao,Mengying Wu,Yijie Gan,Shurong Zhang,Deling Wu,Jinmei Ou,Chuanshan Jin,Wei Zhang

Background: Mume Flos is a common Qi-regulating medication used in traditional Chinese medicine, modern study has discovered that Mume Flos and its components have neuroprotective properties. However, the effective components and molecular mechanisms of Mume Flos are still unclear. Methods: In this study, the current study chose Caenorhabditis elegans as a model to evaluate the anti-Alzheimer disease (AD) effect of the active parts of Mume Flos using life span, paralysis rate, oxidative stress capacity, and heat stress capacity as indicators. Then, UHPLC-Q-Orbitrap-MS/MS and network pharmacology combined with GEO database chip were used for joint analysis to predict the active components and potential molecular mechanisms, which were validated using C. elegans models. Results: The results showed that the n-butanol part of Mume Flos slowed down nematode paralysis and possessed anti-AD activity, and we tentatively identified a total of 45 constituents, mainly including flavonoids and phenylpropanoids as well as a small number of amino acids, and determined that 39 active components and 130 targets are related to neuroprotection. Further analysis identified the main active ingredients such as rutin, neochlorogenic acid, and chlorogenic acid, all of which could reduce Aβ deposition in nematodes and slow down the rate of paralysis in transgenic nematodes. Conclusion: This study revealed the neuroprotective effects of MFB and identified its active components as rutin, neochlorogenic acid, and chlorogenic acid. They may play a role in reducing Aβ deposition by regulating the targets of GAPDH, TNF, IL6, INS, and TP53. This study provides a basis for further in-depth research on the mechanism of MFB to improve neurological diseases.

Microwave bag cooking affects the quality, glucosinolates content and hydrolysate production of broccoli florets

Wenxiu Zheng,Wenhao Wang,Desheng Fu,Tianyu Zhang,Zhengrui Liang,Ling Yan,Changhong Liu,Lei Zheng

Appropriate processing and cooking technologies can effectively improve the content of bioactive compounds in vegetables. The effects of microwave bag cooking on broccoli floret quality attributes, glucosinolates (GLSs) content and hydrolysate production were investigated in this study. Microwave bag cooking not only preserved the color of the florets, but also enhanced the total phenolic and flavonoid content, as well as total chlorophyll and ascorbic acid content. Furthermore, the majority of the microorganisms were inactivated, and the structure of the florets was greatly destroyed, thereby increasing antioxidant capacity and promoting the release of GLSs and myrosinase activity in the florets. Moreover, microwave bag cooking significantly enriched the sulforaphane (SFN) and indole-3-carbinol (I3C) production of broccoli florets in the meantime, demonstrating that it was a convenient and quick cooking option to satisfy the requirements of modern consumers.

Comparative Analysis of Volatile Compounds in the Flower Buds of Three Panax Species Using Fast Gas Chromatography Electronic Nose, Headspace-Gas Chromatography-Ion Mobility Spectrometry, and Headspace Solid Phase Microextraction-Gas Chromatography-Mass Spectrometry Coupled with Multivariate Statistical Analysis

Yang Yue,Jiaxin Yin,Jingyi Xie,Shufang Wu,Hui Ding,Lifeng Han,Songtao Bie,Wen Song,Ying Zhang,Xinbo Song,Heshui Yu,Zheng Li

The flower buds of threePanaxspecies (PGF:P. ginseng; PQF:P. quinquefolius; PNF:P. notoginseng) widely consumed as health tea are easily confused in market circulation. We aimed to develop a green, fast, and easy analysis strategy to distinguish PGF, PQF, and PNF. In this work, fast gas chromatography electronic nose (fast GC e-nose), headspace-gas chromatography-ion mobility spectrometry (HS-GC-IMS), and headspace solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) were utilized to comprehensively analyze the volatile organic components (VOCs) of three flowers. Meanwhile, a principal component analysis (PCA) and heatmap were applied to distinguish the VOCs identified in PGF, PQF, and PNF. A random forest (RF) analysis was used to screen key factors affecting the discrimination. As a result, 39, 68, and 78 VOCs were identified in three flowers using fast GC e-nose, HS-GC-IMS, and HS-SPME-GC-MS. Nine VOCs were selected as potential chemical markers based on a model of RF for distinguishing these three species. Conclusively, a complete VOC analysis strategy was created to provide a methodological reference for the rapid, simple, and environmentally friendly detection and identification of food products (tea, oil, honey, etc.) and herbs with flavor characteristics and to provide a basis for further specification of their quality and base sources. Keywords:Panax;flower bud;fast GC e-nose;HS-GC-IMS;HS-SPME-GC-MS;multivariate statistical analysis
