Effects of polysaccharides on colonic targeting and colonic fermentation of ovalbumin-ferulic acid based emulsion

Hesheng Liu,Zhenzhen Huang,Ting Xin,Lihong Dong,Mei Deng,Lipeng Han,Fei Huang,Dongxiao Su

Rutin is a polyphenol with beneficial pharmacological properties. However, its bioavailability is often compromised due to low solubility and poor stability. Encapsulation technologies, such as emulsion systems, have been proven to be promising delivery vehicles for enhancing the bioavailability of bioactive compounds. Thus, this study was proposed and designed to investigate the colonic targeting and colonic fermentation characteristics of rutin-loaded ovalbumin-ferulic acid-polysaccharide (OVA-FA-PS) complex emulsions. The results indicate that OVA-FA-PS emulsion effectively inhibits the degradation of rutin active substances and facilitates its transport of rutin to the colon. The analysis revealed that the OVA-FA-κ-carrageenan emulsion loaded with rutin exhibited superior elasticity and colon targeting properties compared to the OVA-FA-hyaluronic acid or OVA-FA-sodium alginate emulsions loaded with rutin in the composite emulsion. Additionally, it was observed that the rutin loaded within the OVA-FA-κ-carrageenan emulsion underwent degradation and was converted to 4-hydroxybenzoic acid during colonic fermentation.

High-performance liquid chromatography micro-fraction bioactive evaluation combined with countercurrent chromatographic separation of antioxidants from Citrus peel and their tyrosinase inhibition activities

Liqun Fang,Tingting Lin,Ben Chen,Haibo You,Chunyan Wu,Chu Chu,Shengqiang Tong

In the present study, high-performance liquid chromatography micro-fraction bioactive evaluation and high speed countercurrent chromatography were performed on screening, identification and isolation of antioxidants from Citrus peel. Three compounds were screened as antioxidants and tyrosinase inhibitors using 2,2′-azino-bis (3-ethyl-benzothiazoline-6-sulfonic acid) radical cation scavenging assay and tyrosinase activity test, then they were identified as eriocitrin, narirutin and hesperidin. Moreover, the solvent system ethyl acetate- n -butanol-water (6:4:10, v/v/v) was used for separation of ethyl acetate extract of Citrus peel by high speed countercurrent chromatography. In total, 0.45 mg of eriocitrin with 87.10% purity, 2.04 mg of narirutin with 95.19% purity and 1.35 mg of hesperidin with 95.19% purity were obtained from 20 mg of ethyl acetate extract of Citrus peel in a single run and then each component was subjected to 2,2′-azino-bis (3-ethyl-benzothiazoline-6-sulfonic acid) radical cation scavenging assay and tyrosinase inhibition assay. Eriocitrin showed great antioxidant activity (the half-maximum concentration: 3.65 µM) and tyrosinase inhibition activity (the half-maximum concentration: 115.67 µM), while narirutin and hesperidin exhibited moderate activity. Tyrosinase inhibition activity for eriocitrin in vitro was reported for the first time. Furthermore, molecular docking between eriocitrin and mushroom tyrosinase was also studied.

Ultra-sensitive electrochemical sensor based on in situ grown ultrafine HKUST-1 nanoparticles @ graphite nanosheets and core–shell structured MoO3-polypyrrole nanowires for the detection of rutin in orange juice

Wang Yuefan,Chen Yinzhi,Zhou Yuhe,Wang Yilin,Wu Yingjie,Xie Yixi,Zhao Pengcheng,Hu Xiayi,Fei Junjie

Rutin extracted from natural plants has important medical value, so developing accurate and sensitive quantitative detection methods is one of the most important tasks. In this work, HKUST-1@GN/MoO 3 -Ppy NWs were utilized to develop a high-performance rutin electrochemical sensor in virtue of its high conductivity and electrocatalytic activity. The morphology, crystal structure, and chemical element composition of the fabricated sensor composites were characterized by SEM, TEM, XPS, and XRD. Electrochemical techniques including EIS, CV, and DPV were used to investigate the electrocatalytic properties of the prepared materials. The electrochemical test conditions were optimized to achieve efficient detection of rutin. The 2-electron 2-proton mechanism, consisting of several rapid and sequential phases, is postulated to occur during rutin oxidation. The results show that HKUST-1@GN/MoO 3 -Ppy NWs have the characteristics of large specific surface area, excellent conductivity, and outstanding electrocatalytic ability. There is a significant linear relationship between rutin concentration and the oxidation peak current of DPV. The linear range is 0.50–2000 nM, and the limit of detection is 0.27 nM (S/N = 3). In addition, the prepared electrode has been confirmed to be useful for rutin analysis in orange juice. Graphical Abstract

Degradation behavior of polyphenols in model aqueous extraction system based on mechanical and sonochemical effects induced by ultrasound

Pengxu Wang,Chuanxiang Cheng,Yaqin Ma,Meng Jia

In this study, the temporal and spatial variation of mechanical effects (ME) and sonochemical effects (SE), the degradation behavior of polyphenols as well as the correlation coefficients were comprehensively analyzed. The ME and SE showed inverse dependence along with the axial direction of transducer. Higher irradiation distance (50 mm) enhanced the ME but inhibited the generation of hydroxyl radicals (8.29 μmol/L). Rutin and cyanidin-3-glucoside, compared with thermal degradation (70°C, 6.41%, and 11.45%), were more susceptible to free radical degradation due to the boost of SE at low temperature (10°C, 20.23%, and 34.41%). The concentration of hydroxyl radicals at 70°C (18.38 μmol/L) was about 5.77 times lower than that at 10°C (3.22 μmol/L). No significant differences in the ME were observed for a wide range of temperatures (10–70°C). The impacts of increasing duty cycle on further promoting the ME and SE were limited. The concentration of hydroxyl radicals (0–23.37 μmol/L) increased linearly with sonication time (0–45 min) and the degradation of caffeic acid only occurred on a time scale (25 min). The free radical scavenger can minimize the degradation of polyphenols by inhibiting the hydroxyl radicals (23.37 μmol/L without t-BuOH; 12.17 μmol/L with t-BuOH). The above results showed that the degradation of polyphenols attacked by hydroxyl radicals presented a dose-response relationship. The correlation between the hydroxyl radicals and polyphenols as well as the mechanical effects and extraction yield indicated that the stability of polyphenols exposed to ultrasonic field depended on the structure-radical scavenging activity relationship and the extraction may be accompanied by the degradation.

Preparation and properties of inclusion complex of resveratrol with epichlorohydrin-modified β-cyclodextrin

Shide Li,Hongsheng Luo,Ronghua Qiu,Kaibin Huang,Ziying Chen,Xiangjun Fu

In order to improve the aqueous solubility and enhance stability of resveratrol (RES), an inclusion complex (RES-ECH-βCD) of RES with epichlorohydrin-modified β-cyclodextrin (ECH-βCD) was prepared using freeze-drying. The effects of RES-ECH-βCD on the solubility and stability of RES were investigated. Phase solubility studies indicated that ECH-βCD significantly enhanced the solubility of RES compared to β-cyclodextrin (β-CD). Thermal stability tests showed that the inclusion complex exhibited superior thermal stability compared to free RES. Furthermore, the application of RES-ECH-βCD in sunscreen formulations was validated. The formulation exhibited sun protection properties, with RES-ECH-βCD working synergistically with α-glucosyl rutin (Rutin-G) to significantly enhance both SPF and PFA. These findings suggest that RES-ECH-βCD not only improves the solubility and stability of RES but also has promising potential for use in cosmetic and sunscreen products.

Taraxacum-like Mg-Al-Si@porous carbon nanoclusters for electrochemical rutin detection

Yalikun Nuerbiya,Mamat Xamxikamar,Li Yongtao,Hu Xun,Wang Ping,Hu Guangzhi

The authors describe a method for synthesis of a nanomaterial consisting of porous carbon encapsulated Mg-Al-Si alloy (denoted as Mg-Al-Si@PC) nanocluster. The nanocluster was synthesis by a solvothermal reaction, followed by high-temperature annealing. The nanoclusters were used as a novel immobilization platform for electrochemical sensing of rutin. The electrochemical behavior of rutin at a modified electrode was investigated by cyclic voltammetry and differential pulse voltammetry. The modified electrode demonstrates a high electrocatalytic activity toward rutin oxidation at a relatively low working potential (0.6 vs. Ag/AgCl). Under optimal conditions, the sensor has a linear response in the 1–10 μM rutin concentration range, and a 0.01 μM lower detection limit (at an S/N ratio of 3). It was successfully applied to the quantification of rutin in pharmaceutical tablets, and satisfactory results were obtained. Furthermore, the results correspond with those with the standard method and with the amounts indicated by the producer, respectively. Schematic diagram of the Mg-Al-Si@PC nanocluster preparation process and electroanalysis mechanism.

Selective enrichment of Rutin in sunscreen by boronate affinity molecularly imprinted polymer prior to determination by high performance liquid chromatography

Jinyue Chai,Xue Chen,Chengcheng Jin,Fang Chai,Miaomiao Tian

In this study, AgCl was used as the carrier matrix, TiO 2 was introduced on the substrate surface, carboxyphenylboronic acid (CPBA) was used as the boronate affinity functional monomer , and Rutin (RT) as template to prepare molecular imprinted adsorbent AgCl@TiO 2 @B-MIPs (B-MIPs). The effects of synthesis conditions, solid-phase extraction (SPE) conditions, selectivity, competitiveness, reproducibility, and reusability were evaluated in detail. Under the optimum adsorption conditions, the maximum adsorption capacity is 31.70 mg g −1 . The utility of B-MIPs as a SPE adsorbent for RT was supported by an abundance of pore structure, as well as boric acid affinity sites that facilitate rapid binding of adsorbents to templates. The linear range is 0.05–50 mg L −1 , and the limit of detection (LOD) is 0.023 mg L −1 . The method was used for the determination of RT in sunscreen and isolation cream by high-performance liquid chromatography (HPLC). The target analytic recoveries are in the range of 84.7–98.0% (relative standard deviation is less than 2.4%, n  = 3). Furthermore, this work provides a potential possibility to detect other cis -diol containing analytes in the complex matrix.

Effects of dietary cyanidin-3-diglucoside-5-glucoside complexes with rutin/Mg(II) against H2O2-induced cellular oxidative stress

Wei Xu,Nan Zhang,Zhentao Zhang,Pu Jing

Anthocyanins exist likely as complexes with flavones/metal ions in some plants. However, few studies indicate the complexation effects on bioactivities, i.e. antioxidant activities. The aim of this study was to evaluate effects of complexes of cyanidin-3-diglucoside-5-glucoside (CY3D5G) with rutin/Mg(II) on intrinsic antioxidant enzymes and mRNA expressions. All complexes exhibited cellular antioxidant activity in RAW264.7 cells, whereas half maximal effective concentration (EC 50 ) values of CY3D5G complexes were significantly lower than its alone. Combination indexes of CY3D5G-rutin, CY3D5G-Mg(II), and CY3D5G-rutin-Mg(II) complexes were < 1 ( p  < .05), indicating of synergistic effects. Additionally, CY3D5G-Mg(II) rather than others significantly enhanced total antioxidant capacity, superoxide dismutase activity, and glutathione peroxidase activity while reducing malonic dialdehyde contents in H 2 O 2 -induced RAW264.7 cell model ( p  < 0.05), which were further confirmed with the up-regulated mRNA expression of nuclear factor E2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1), and the down-regulated p21 ( p  < 0.05). Our results demonstrated that CY3D5G and its complexes might enhance cellular antioxidant activities via the Nrf2/HO-1 signaling pathway. Additionally, a synergistic antioxidant effect of CY3D5G in the present of rutin/ Mg(II) as regular food components was evident, suggesting anthocyanin might interact with dietary components such as Mg(II) or rutin in food matrix or biological systems and exhibit a stronger antioxidant activity than it alone. The Mg (II) that exhibited no antioxidant activities alone shows potential as a co-adjuvant for the anthocyanin antioxidant activity.

Electrochemical synthesis of a nanocomposite consisting of carboxy-modified multi-walled carbon nanotubes, polythionine and platinum nanoparticles for simultaneous voltammetric determination of myricetin and rutin

Liu Chaoqiao,Huang Jianzhi,Wang Lishi

The authors describe a method for electrochemical synthesis of a nanocomposite consisting of carboxylated multi-walled carbon nanotubes, polythionine and platinum nanoparticles (denoted as cMWCNT@pTh@Pt). The composite was placed on a glassy carbon electrode (GCE) to obtain a sensor for simultaneous determination of myricetin and rutin by differential pulse voltammetry, with typical peak voltages of 0.16 and 0.34 V (vs. SCE). Under optimal conditions, the modified GCE has a linear response in the range of 0.01–15 μM myricetin concentration range, and a 3 nM lower detection limit. For rutin, the data are 0.01–15 μM and 1.7 nM, respectively. The sensor is selective, stable and reproducible. It was successfully applied to the simultaneous determination of myricetin and rutin in spiked juice samples, and satisfactory results were obtained. A nanocomposite consists of carboxy multi-walled carbon nanotube (cMWCNT), polythionine (pTh) and platinum nanoparticles (Pt) was presented for the simultaneous determination of myricetin and rutin. The nanocomposite shows excellent electrochemical determination performances based on a synergistic effect among cMWCNT, pTh and Pt.

Metal-Phenolic Network-Coated Nanoparticles as Stabilizers for the Engineering of Pickering Emulsions with Bioactivity

Caiyun Shan,Xiaomiao Cui,Zhiliang Gao,Mengqi Li,Xunhui Zhang,Muthupandian Ashokkumar,Aixin Song,Jiwei Cui

Pickering emulsions stabilized by functional nanoparticles (NPs) have received considerable attention for improving the physical stability and biological function of NPs. Herein, hydrophobic polyphenols were chosen as phenolic ligands to form metal-phenolic network (MPN) coatings on NPs (e.g., silica, polystyrene) mediated by the sono-Fenton reaction. The MPN coatings modulated the surface wettability and charges of NPs and achieved emulsification behavior for preparing Pickering emulsions with pH responsiveness and oxidation resistance. A series of polyphenols, including resveratrol, rutin, naringin, and curcumin, were used to form MPN coatings on NPs, which served as stabilizers for the engineering of functionalized oil-in-water (O/W) Pickering emulsions. This work provides a new avenue for the use of hydrophobic polyphenols to modulate NP emulsifiers, which broadens the application of polyphenols for constructing Pickering emulsions with antioxidant properties.

Impact of the solubility of phenolic compounds from Highland Barley (Hordeum vulgare L.) on their antioxidant property and protein binding affinity

Wangyan Qin,Ying Wang,Fall Mouhamed,Bruce Hamaker,Genyi Zhang

The impact of the solubility of phenolic compounds (PCs) from highland barley (HB) on their antioxidant property and protein binding affinity was investigated. HB-PCs from the phenolic extract (HBE) were analyzed through the UPLC-QTOF-MS, and five fractions were further divided based on their water solubility. Representative compounds of proanthocyanidins, (+)-catechins, and (−)-epicatechin from water fraction (HB–W, 197.67 ± 4.8 μmol GAE/100 g) were significantly correlated with the antioxidant activity of the HBE. In contrast, the PCs enriched in the acetone fraction (HB-A, 60.2 ± 3.1 μmol GAE/100 g) with a structure of 2-phenyl chromogen ketone group (such as rutin, kaempferol, hesperidin, and quercetin) were positively correlated with the protein binding affinity. Further analysis showed that the water-insoluble HB-A fraction significantly lowered the starch digestibility (37.5%) and postprandial glycemia (17.8%) compared to HB-W fractions. Thus, the solubility of PCs is intimately associated with their biological functions as antioxidants or protein-binding ligands, indicating PCs with strong antioxidant properties or high protein binding affinity are structurally distinct from each other, and the physical properties of phenolic compounds might be an important factor to further the understanding of their health functions and mechanisms.

A comparison of chemical composition, bioactive components and hypoglycemic activity of Stigma maydis obtained from different growing times

Yi Zhang,Jinxin Pang,Lianmou Yao,Chunfang Wang,Bingjie Chen,Qi Zheng,Yingxiong Hu,Yongjin Qiao

Stigma maydis (SM) possesses remarkable nutritional value due to the presence of health-protective chemical constituents in it. Despite its commercial uses, there is a lack of information on changes in the nutritional profile of SM during Shenke 601 waxy corn development by following four different maturity stages (silking stage; blister stage; milky stage; dough stage). In this study, the ideal times for the development of seven active compounds (polysaccharides, saponin, rutin, luteolin, quercetin, kaempferol and chlorogenic acid) were at the silking stage. The total flavonoid and total phenol content increased as the SM matured. Except for the above-mentioned active compounds, the content of major nutritional components of dough stage was higher than that of three other stages. The extract of Stigma maydis exhibited good α-amylase, α-glucosidase and pancreatic lipase inhibition activities. The ethanolic SM extracts showed better inhibitory effects than aqueous SM extracts with a dose–effect relationship. The hypoglycemic activity of ethanolic extract was higher than that of aqueous extract at each stage. This provided an important basis for the application of SM.
