Broccoli sprouts are rich in secondary metabolites, especially glucosinolates (GLs). The synthesis of GLs can be modified by light. In this study, extractive electrospray ionization-mass spectrometry (EESI-MS) was used to analyze the broccoli sprouts extracts (BSE) under different light quality treatments. Multivariable statistical methods were used to analyze the difference of different light quality treatments. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to analyze the gene expression of key enzymes in the aliphatic-GLs biosynthesis pathway. Nineteen compounds, including 7 GLs, 3 phenolic acids, 6 amino acids and 3 organic acids were simultaneously detected in one minute by EESI-MS. Relative expression of MYB28 , BCAT4 , CYP79F1 , CYP83A1 and AOP3 were mostly upregulated in R5B5 (light ratios of red/blue 5:5) treatment. These findings suggest that blue light improving the accumulation of secondary metabolites in broccoli sprouts, and the R5B5 treatment was most beneficial. Furthermore, we established EESI-MS method to simultaneous and rapid detection of secondary metabolites in broccoli sprouts, especially GLs. This study could provide a reference for selecting proper LED lighting conditions to produce broccoli sprouts with high content of bioactive components, and provides light regulation technology and theoretical basis for the industrialized production of sprouts.

Broccoli microgreens represent a novel type of vegetable characterized by high levels of bioactive compounds. Here, we investigated the effects of light-emitting diode (LED) light quality on the development and nutritional uptake of broccoli microgreens. The plants were exposed to five distinct light treatments: white, single red, and red: blue at 5:1, 5:3, and 5:5 ratios. The soluble sugars, nutrients, and secondary metabolites of broccoli sprout extracts were examined using high-performance liquid chromatography and ultra-high-performance liquid chromatography quadrupole time-of-flight mass spectrometry under various light-quality conditions. In addition, the expression levels of genes encoding important enzymes in the glucosinolates (GLs) biosynthesis pathway were examined using quantitative real-time polymerase chain reaction. Overall, the findings demonstrated that mixed red-blue light, especially the 5:1 red: blue ratio, was beneficial for broccoli microgreens in terms of their hypocotyl length, fresh weight, edible rate, soluble proteins, vitamin C, total phenolics, total flavonoids, and GLs. Our findings provide a foundation and reference point for selecting the most appropriate LED light ratio in microgreens production.

A bioinspired Au@Ag nanodome-cones array (Au@Ag NDCA) surface-enhanced Raman scattering (SERS) chip was developed for efficient residue analyses of food samples. The cicada wing inspired Au@Ag NDCA chip was fabricated by a bottom-up method, Au nanocones array was firstly grown onto nickel foil by displacement reaction and cetyltrimethylammonium bromide guidance growth, and then silver shell with controllable thickness was coated onto the Au nanocones array by magnetron sputtering . The Au@Ag NDCA chip exhibited good SERS performances with high enhancement factor of 1.2 × 10 8 , good uniformity with relative standard deviation (RSD) less than 7.5% (n = 25), good inter-batch reproducibility with RSD less than 9.4% (n = 9), and long-term stability over 9 weeks. By adapting a minimized sample preparation, Au@Ag NDCA chip combined with a 96-well plate could realize high-throughput SERS analyses of 96 samples with average analysis time less than 10 min. The substrate was applied for quantitative analyses of two food projects. One was 6-benzylaminopurine auxin residue in sprout samples with detection limit of 38.8 μg/L, recoveries of 93.3–105.4% and RSDs of 1.5–6.5%, and the other was an edible spice of 4-amino-5,6-dimethylthieno (2,3-d) pyrimidin-2(1H)-one hydrochloride additive in beverage samples with detection limit of 18.0 μg/L, recoveries of 96.2–106.6% and RSDs of 3.5–7.9%. All the SERS results were well confirmed by conventional high-performance liquid chromatographic methods with relative errors less than 9.7%. The robust Au@Ag NDCA chip exhibited good analytical performances possessed great potential for convenient and reliable analyses of food quality and safety.

The ultrasonic cell disruption method was used to efficiently extract isothiocyanates and other volatile compounds from radish microgreens. A total of 51 volatiles were identified and quantified by headspace solid-phase micro-extraction and gas chromatography–mass spectrometry (HS-SPME/GC–MS) in four radish microgreen cultivars, mainly including alcohols, aldehydes, isothiocyanates, sulfides, ketones, esters, terpenes, and hydrocarbons. The correlation between cultivars and volatile compounds was determined by chemometrics analysis, including principal component analysis (PCA) and hierarchical clustering heat maps. The aroma profiles were distinguished based on the odor activity value (OAV), odor contribution rate (OCR), and radar fingerprint chart (RFC) of volatile compounds. This study not only revealed the different flavor characteristics in four cultivars but also established a theoretical basis for the genetic improvement of radish microgreen flavors. Keywords:flavor;volatile compounds;radish microgreens;ultrasonic cell disruption;GC–MS

Sweet corn is one of the most widely planted crops in China. Sprouting of grains is a new processes to increase the nutritional value of grain products. The present study explores the effects of light on the nutritional quality of sweet corn sprouts. Gene expression of phenolic biosynthesis, phytochemical profiles and antioxidant activity were studied. Two treatments (light and dark) were selected and the morphological structure of sweet corn sprouts, as well as their biochemical composition were investigated to determine the effects of light on the regulation of genes responsible for nutritional compounds. Transcription analyses for three key-encoding genes in the biosynthesis of the precursors of phenolic were studied. Results revealed a negative regulation in the expression ofZmPALwith total phenolic content (TPC) in the light group. TPC and total flavonoid content (TFC) increased during germination and this was correlated with an increase in antioxidant activity (r= 0.95 and 1.0). The findings illustrate that the nutritional value of sweet corn for the consumer can be improved through germination to the euphylla stage. Keywords:germination;sweet corn;phenolic biosynthesis;antioxidant activity
