This study aims to explore the corrosion behavior of bitter almond oil (BAO) during processing. Hydrocyanic acid (HCN) and benzoic acid in oil are speculated as the main components that corrode the screw oil expeller. Immersion and simulation corrosion tests are carried out to verify this hypothesis. Immersion test results show that crude BAO (CBAO) containing 5.07 ± 0.06 mg kg −1 benzoic acid and 4.25 ± 0.04 mg kg −1 HCN exhibits corrosive effects on steel. Simulation corrosion test results indicate that the addition of CN − or benzoic acid has significant effect on the corrosion of steel. Specifically, at ≥1 × 10 4 mg kg −1 benzoic acid concentration or ≥5 mg kg −1 CN − concentration, the mass of steel blocks in oil decreases and the contents of Fe, Zn, Al, and Mg ions in oil increase significantly. Therefore, HCN in CBAO is the main cause of screw oil expeller corrosion in BAO processing and benzoic acid is the secondary cause. Practical Applications : The corrosion behavior of CBAO not only damages the integrity of the screw oil expeller but also increases the dissolution of toxic metals from the components into oil. The study of corrosion has practical value in taking effective measures, such as detoxification before pressing and addition of natural inhibitors, to reduce corrosion and ensure the quality of CBAO. Corrosion study provides the basis for the selection of steel materials of screw oil expeller.

Bitter apricot kernel (BAK) is a good source of amygdalin and oil. However, the current production process faces challenges including the inability to simultaneously extract amygdalin and oil, and the risk of toxicity due to amygdalin contamination in the oil. A novel, green and efficient ultrasonication-assisted extraction (UAE) method based on a natural deep eutectic solvent (NADES) composed of choline chloride-lactic acid (CC-LA) was established for simultaneous extraction of amygdalin and oil from BAK for the first time. Under optimizing conditions of a liquid-to-solid ratio of 18 mL/g, ultrasonic power of 420 W, temperature of 42 °C, and extraction time of 32 min, the highest yields of amygdalin and oil were achieved at 10.22 ± 0.04 % and 45.64 ± 0.24 %, respectively. The yield of amygdalin in UAE based on CC-LA was twice and nineteen times that of methanol (90 %, v/v) and water, respectively. Meanwhile, the yield of oil obtained by this new method was comparable to that by organic solvent extraction methods. The fatty acid composition of oil obtained by this method was consistent with that by conventional extraction methods. Scanning electron microscopy (SEM) analysis indicated that CC-LA and UAE had a synergistic effect on the destruction of BAK’s microstructure, which facilitated the recovery of amygdalin and oil. Molecular dynamics simulation results showed that amygdalin in CC-LA reached a higher solvent-accessible surface area (SASA) and formed more hydrogen bonds with longer lifetimes and greater stability. Furthermore, this new method demonstrated potential for industrial scalability.

Background Deciphering the in vivo processes of traditional Chinese medicine (TCM) is crucial for identifying new pharmacodynamic substances and new drugs. Due to the complexity and diversity of components, investigating the exposure, metabolism, and disposition remains a major challenge in TCM research. In recent years, a number of non-targeted smart mass-spectrometry (MS) techniques, such as precise-and-thorough background-subtraction (PATBS) and metabolomics, have realized the intelligent identification of in vivo components of TCM. However, the metabolites characterization still largely relies on manual identification in combination with online databases. Results We developed a scoring approach based on the structural similarity and minimal mass defect variations between metabolites and prototypes. The current method integrates three dimensions of mass spectral data including m/z , mass defect of MS1 and MS2, and the similarity of MS2 fragments, which was sequentially analyzed by a R-based mass dataset relevance bridging (MDRB) data post-processing technique. The MDRB technology constructed a component relationship network for TCM, significantly improving metabolite identification efficiency and facilitating the mapping of translational metabolic pathways. By combining MDRB with PATBS through this non-targeted identification technology, we developed a comprehensive strategy for identification, characterization and bridging analysis of TCM metabolite in vivo. As a proof of concept, we adopted the proposed strategy to investigate the process of exposure, metabolism, and disposition of Semen Armeniacae Amarum (CKXR) in mice. Significance The currently proposed analytical approach is universally applicable and demonstrates its effectiveness in analyzing complex components of TCMs in vitro and in vivo. Furthermore, it enables the correlation of in vitro and in vivo data, providing insights into the metabolic transformations among components sharing the same parent nucleus structure. Finally, the developed MDRB platform is publicly available for ( https://github.com/933ZhangDD/MDRB ) for accelerating TCM research for the scientific community. Graphical

Amygdalin can be decomposed into hydrocyanic acid, which is the primary source of Persicae Semen toxicity, by gut flora. Here, the inhibitory activity of β-glucosidase for test herb extracts was first determined and compared. In turn, optimization of the ratio of substrate and inhibitor in vitro and LD 50 values of extracts, serum and liver contents of amygdalin in vivo was measured. Lycii Cortex was found to be the best inhibitory activity for β-glucosidase. The ratio of amygdalin-to-Lycii Cortex extract of 7.19:8.18 (mmol L −1 /mg mL −1 ) can be relatively suitable for inhibiting β-glucosidase activity in test in vitro reaction system. After mixed with Lycii Cortex extract, the toxicity of Persicae Semen ethanol extract in mice is significantly reduced and more amygdalin can be absorbed into the bloodstream. The study provides useful information for reducing toxicity of Persicae Semen and suggests how to better use these natural β-glucosidase inhibitors in the utilization of glycosides and aglycones. Background Evidence has demonstrated that Chinese medicine formula Xuefu Zhuyu decoction can markedly promote the formation of new hair in patients and mice with alopecia areata (AA). Amygdalin is one of the active components of Xuefu Zhuyu decoction, but its therapeutic effects and the underlying mechanisms on AA remains largely unrevealed. Purpose Therefore, this study aims to investigate the therapeutic effects and to probe its molecular mechanisms of inflammation and immune regulation on AA model of C3H/HeJ mice. Study design The C3H/HeJ female mice were divided into control, AA, rusolitinib (60 mg/kg), and amygdalin groups (60, 90, and 120 mg/kg, 0.2 ml/10 g, i.g.). Methods The optical microscope was used to observe the feature of the local skin, and the number of lanugo and terminal hair. H&E staining was performed to determine the degree of pathological damage to the skin. ELISA was performed to detect levels of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) and interferon-γ (IFN-γ) in mice serum. Flow cytometry was carried out to analyze the CD4 + CD25 + FOXP3 + , CD4 + and CD8 + of skin tissue. And the levels of CD4 + and CD8 + , p-JAK/JAK2, p-STAT3/STAT, and SOCS3 were detected by immunohistochemistry. Western blot and qRT-PCR were employed to examine the expression levels of IL-6, TNF-α, IFN-γ, JAK2, p-JAK, STAT, p-STAT3 and SOCS3 proteins and genes in skin tissues. Results Compared with AA group, amygdalin immensely increased the number of vellus hairs and decreased the number of terminal hairs determined by skin microscopy and H&E staining. ELISA, Western blot and qRT-PCR data showed that the levels of IL-6, TNF-α and IFN-γ in serum and skin tissues of AA mice were significantly increased, while amygdalin administration dramatically restrained the contents of the three pro-inflammatory factors. Flow cytometry and immunohistochemistry hinted that amygdalin observably enhanced the number of CD4 + CD25 + FOXP3 + and CD4 + cells, while inhibited the number of CD8 + positive cells in mice with AA. Moreover, amygdalin signally reduced JAK2/STAT3 pathway-related protein and gene levels in AA mice. Conclusion Amygdalin could inhibit inflammatory response and improve immune function in the treatment of AA. The underlying molecular mechanism may be related to inhibition of JAK2/STAT3 pathway.

Accurate, specific, and cost-effective detection of toxic cyanogenic glycosides is crucial for ensuring biological health and food safety. In this study, a novel biosensor based on co-immobilized multi-enzyme system was constructed by artificial antibody-antigen-directed immobilization for the colorimetric detection of amygdalin through a cascade reaction catalyzed by β-glucosidase, glucose oxidase, and horseradish peroxidase. Artificial antibodies and antigens were prepared using catechol and 3,4-dihydroxybenzaldehyde, respectively, to generate mutual affinity recognition ability for enzyme immobilization. On this basis, the biosensing system showed a complete response to amygdalin within 4 min, with a linear range from 2 to 10 μM, a detection limit of 0.18 μM, and a quantification limit of 0.6 μM. In addition, this sensor had good precision, reproducibility, stability, and reusability. This study proposed a method for detecting cyanogenic glycosides, providing a successful case for the application of cascade biosensors in food safety detection.

Amygdalin, a cyanogenic glycoside abundant in apricot kernels, poses a food safety risk, requiring rapid detection that conventional chromatographic methods cannot fulfill for on-site application. Thus, this study aims to construct a novel colorimetric sensing platform and improve detection performance through a multivalent aptamer strategy. This pioneering work introduces a novel bivalent aptamer strategy for ultra-sensitive on-site amygdalin detection. The aptamers specific to amygdalin were screened using a five-segment single-stranded DNA library and Capture-SELEX, with truncated variants optimized to obtain a high-affinity core sequence. A key innovation involves the first design and synthesis of a bivalent aptamer that exploits synergistic binding to enhance target affinity. The bivalent aptamer (Bapt 4-4) exhibited doubled affinity for amygdalin compared to its monovalent truncated counterpart (Amyg 4–26), achieving a dissociation constant ( K d ) of (2.19 ± 1.03) nM. By integrating the bivalent aptamer with the laccase-like activity of silver oxide nanoparticles (Ag 2 O NPs), an aptasensor with high sensitivity and specificity was developed, yielding a detection limit of 0.008 μg mL −1 , which is lower than that of the monovalent aptamer-based sensor (0.0137 μg mL −1 ). This multivalent aptamer strategy provides an effective approach for highly sensitive amygdalin detection. Furthermore, the constructed bivalent aptasensor provides a robust technical tool for the on-site monitoring of plant-derived cyanogenic glycoside toxins, with broad application prospects in the domains of food safety and traditional Chinese medicine safety.

ObjectivePenicillin-resistant Streptococcus pneumoniae (PRSP), for which novel treatment medicines are required, has expanded extensively due to the overuse of antibiotics. This study aimed to detect the optimal ratio of the combination of the main components based on Yinhuapinggan granules (YHPG) to generate novel treatment concepts for PRSP-induced lung injury. Methods Three representative main components: chlorogenic acid (C), amygdalin (A), and puerarin (P) were selected, and the optimal combination of these three components was determined by an orthogonal experiment. Investigations were conducted on the potential mechanisms underlying the protective effect of this optimized combination against PRSP-induced lung epithelial cell damage. Meanwhile, the bacteriostatic effect was further explored through the optimized combination of these natural products combined with penicillin G (PG).Results The optimized combination CAP (C: 16 µg/mL, A: 24 µg/mL, P: 24 µg/mL) screened by the orthogonal experimental design reduced cell damage in a model of human lung epithelial cells infected by PRSP, and the combination of CAP and PG had a synergistic effect. At the cellular level, CAP attenuated lung epithelial cell injury by modulating the TLRs/MyD88 inflammatory pathway. At the bacterial level, CAP modulated the virulence and drug resistance of PRSP, resulting in enhanced bacterial inhibition by the combination of CAP and PG.Conclusion Taken together, our results suggest that CAP can modulate or synergize with PG to modulate the TLRs/MyD88 pathway and attenuate PRSP-induced lung injury, and can be used as a potential drug for treating PRSP infection.

Larvae and adults of the Colorado potato beetle ( Leptinotarsa decemlineata ), a major pest of potato crops, display conspicuous coloration to advertise their toxicity to predators. However, the identity of the toxic compounds remains unclear. Here, we show that larvae and adults release toxic hydrogen cyanide (HCN) from the degradation of mandelonitrile and other cyano-compounds, which are produced by commensal bacteria. We isolate the bacterium Proteus vulgaris Ld01 from the insect’s gut, and show that it produces HCN and a mandelonitrile-producing cyanoglucoside, amygdalin. Knockout of a gene ( hcnB ) encoding putative hydrogen cyanide synthase impairs HCN production in P. vulgaris Ld01. Antibiotic treatment of larvae, to eliminate their commensal bacteria, leads to a substantial reduction of HCN emission in larvae and adults. HCN release by bacteria-deprived beetles can be restored by addition of mandelonitrile or by re-infection with P. vulgari s Ld01 (but not with its ∆ hcnB1 or ∆ hcnB2 mutants). Finally, we use dual-choice experiments to show that domestic chicks prefer to eat bacteria-deprived larvae over control larvae, larvae re-colonized with P. vulgaris Ld01, or mandelonitrile-injected larvae. Our work highlights the role of the beetle’s intestinal bacteria in the production of the cyanoglucoside amygdalin and its derived metabolites, including mandelonitrile and HCN, which protect the insect from predation.

Medicinal almonds have been used for over 2 000 years and its clinical efficacy includes relieving cough and asthma. The domestic market in China is flooded with different kinds of dried almonds, such as bitter almond (Armeniacae Semen Amarum, AAS), sweet almond (Armeniacae Semen Dulce, ADS), salted almond (Armeniacae Semen Salsa, ASS), and their sulfur-fumigating products (Armeniacae Semen Sulphur Fumabat, ASFS). Wide varieties of almonds may lead to uncertain efficacy, aberrant quality, and even increased safety risk. However, the authentication method for medicinal almonds has not been reported, although imposters may lead to ineffective medical response. In the present study, Fourier transform infrared spectroscopy (FTIR) and the 2-dimensional infrared (2D-IR) spectroscopy were used to identify different almonds, which were extracted with different solvents including water, methanol, ethanol, chloroform and ethyl acetate, respectively. A new simple FTIR method was developed in the present study. According to the gradient solvent polarity, a new 2D IR method was first developed, and the commodities of almonds in China were analyzed by using the FTIR spectroscopy supported by hierarchical clustering of characteristic peaks. Moreover, 5-hydroxymethyl-2-furfural could be used as a detection index and control target in the quality control of medicinal almonds.

BACKGROUND Grapholita molesta is an important pest of rosaceous fruit trees. Female oviposition affects larval infestation levels in orchards. This behavior, shaped by larval feeding experiences and female olfactory responses, points to an experience-modulated semiochemical strategy based on host plant secondary metabolites. To elucidate the combined effects of non-volatile and volatile components on female oviposition, we analyzed the volatile profiles of peach, pear, and apple shoots/fruits under intact, mechanically injured, and larval damage states throughout the growing season, along with female oviposition preferences, and compared olfactory responses (electroantennogram (EAG) and wind-tunnel experiments) and oviposition bioassays with benzaldehyde (amygdalin hydrolysate) between females with two larval amygdalin-feeding experiences. RESULTS Female oviposition preference and benzaldehyde emissions, both modulated by host factors, exhibited synchronous fluctuations. In EAG and wind-tunnel experiments, females not fed amygdalin during larval stages exhibited a strong attraction to benzaldehyde at concentrations of 10 and 0.1 μg/μL; however, females from larvae fed amygdalin showed a weaker long-distance orientation but greater responses to benzaldehyde during close-range approach and landing behaviors. Females fed amygdalin during larval stages were more likely to be responsive to lower concentrations of benzaldehyde (0.01, 0.1, and 1.0 μg/μL) for oviposition. In contrast, close-range attraction and oviposition behaviors were stronger for females that developed from amygdalin-fed larvae than those that were not fed amygdalin. CONCLUSION Dietary exposure to amygdalin during the larval stages increased the olfactory responses of females to benzaldehyde. Female oviposition behavior is co-regulated by larval experience derived from amygdalin ingestion and benzaldehyde as a host semiochemical signal. © 2025 Society of Chemical Industry.

Amygdalin is a phytotoxin that releases hydrocyanic acid, a highly toxic substance, under the action of enzymes or acids, posing a threat to human health. In this study, a highly sensitive and specific monoclonal antibody (mAb) 3D11, with a half-maximal inhibitory concentration of 0.66 ng/mL, was developed through similarity calculation and molecular structure, molecular spatial overlap, electrostatic potential distribution, and HOMO-LUMO orbitals analyses of hapten design principles. A gold nanoparticle-based lateral flow immunoassay (GNP-LFIA) was developed to detect amygdalin in almonds and almond-based products, with a visual detection limit of 2–10 μg/kg and a detection limit of 0.27–1.27 μg/kg. The recoveries of GNP-LFIA ranged from 94.02 % to 109.50 %, and the accuracy and reliability were further confirmed by testing positive samples, demonstrating that GNP-LFIA can be a practical tool for the rapid on-site detection of amygdalin in almonds and almond-based products.
