Early detection of Huanglongbing with EESI-MS indicates a role of phenylpropanoid pathway in citrus

Ahui Xue,Yongquan Liu,Haoxing Li,Meng Cui,Xueyong Huang,Wenjing Wang,Dong Wu,Xiali Guo,Yingbin Hao,Liping Luo

Huanglongbing (HLB), a devastating disease for citrus worldwide, is caused by Candidatus Liberibacter asiaticus ( C Las). In this study, we employed a novel extractive electrospray ionization-mass spectrometry (EESI-MS) method to analyze the metabolites in leaves of uninfected and HLB-infected Newhall navel orange. The results showed that uninfected and HLB-infected leaves could be readily distinguished based on EESI-MS combined by multivariable analysis. Nine phenolic compounds involved in phenylpropanoid pathway, such as p -coumaric acid, naringin , and apigenin , were principal components to distinguish the leaves of uninfected and HLB-infected Newhall navel orange. Gene expression was also conducted to further explore the molecular mechanism of phenylpropanoid branch pathway in HLB. The expression of genes ( 4CL , HCT , CHI , CHS , CYP , and C12R ) involved in phenylpropanoid branch pathway was increased in asymptomatic and early period of HLB-infected leaves, while decreased in later period of HLB-infected leaves. This study provides a novel method for early detection of citrus HLB and suggests the regulation mechanism of phenylpropanoid pathway in the interaction between citrus and C Las.

Exogenous methyl jasmonate regulates phenolic compounds biosynthesis during postharvest tomato ripening

Xiaoya Tao,Qiong Wu,Jiayin Li,Suqing Huang,Luyun Cai,Linchun Mao,Zisheng Luo,Li Li,Tiejin Ying

Methyl jasmonate (MeJA) acts as an elicitor to stimulate phenolic compounds in fruit and vegetables. To explore the regulation of MeJA on phenolic compounds biosynthesis during postharvest tomato ripening, mature green tomato fruit were treated by 0.5 mM MeJA or deionized water under vacuum, and then kept in darkness for 16 d. Total phenolics and total flavonoids contents, phenolic compounds contents, enzymatic activities, as well as expression levels of genes in response to phenylpropanoid pathway were determined in the present study. Results showed exogenous MeJA enhanced total phenolics and flavonoids contents, augmented the contents of most phenolic compounds, as well as promoted enzymatic activities associated with phenylpropanoid pathway (PAL, C4H, 4CL, CHS, and CHI) during tomato ripening. Furthermore, MeJA up-regulated the expressions of the corresponding genes (1.16- to 19.32-fold) involved in phenylpropanoid pathway during the first 7 d. Results suggested MeJA enhanced phenolic compounds biosynthesis during postharvest tomato ripening via influencing the enzymatic activities and gene expressions. These results may provide valuable information for exploring technical means for the regulation of phenylpropanoid pathway during tomato ripening.

Exogenous nanoselenium alleviates imidacloprid-induced oxidative stress toxicity by improving phenylpropanoid metabolism and antioxidant defense system in Perilla frutescens (L.) Britt

Qinyong Dong,Dong Li,Yangliu Wu,Chunran Zhou,Yongxi Lin,Peijuan Miao,Jiaqi Li,Canping Pan

Few studies have been conducted to investigate the impact of pesticides on the secondary metabolism of traditional Chinese medicine and strategies to mitigate the toxicity of pesticide-induced oxidative stress. The current study focuses on evaluating the potential impacts of nano selenium (NSe) and imidacloprid (IMI) on the quality, physiological biochemistry, and secondary metabolites in Perilla frutescens (L.) Britt . ( P. frutescens ). The study utilized metabolome analysis to explore the toxicity mechanism of IMI. The study noted that IMI-induced stress could emerge with detrimental effects by targeting the destruction of the phenylpropanoid biosynthesis pathway. IMI-induced phenylpropanoid metabolism disorder resulted in an 8%, 17%, 25%, 10%, 65%, and 29% reduction in phenylalanine, coniferyl aldehyde, ferulic acid, cafestol, p-coumaraldehyde, and p-coumaric acid levels, respectively. Under the treatment of exogenous NSe, the levels of these metabolites were increased by 16%, 32%, 22%, 22%, 92%, and 29%, respectively. The application of exogenous NSe increased the levels of these metabolites and improved the biochemical disorder and quality of P. frutescens leaves by optimizing the phenylpropanoid metabolic pathway and enhancing the antioxidant system. Overall, the results suggest that foliar application of NSe could alleviate the oxidative stress toxicity induced by IMI and improve the quality of P. frutescens .

Wei Ping,Tang Qing-fa,Huo Hui-ling,Chen Fei-long,Song Shuai,Xing Xue-feng,Luo Jia-bo

Objective To investigate the pharmacokinetic characteristics of three phenylpropanoids (cinnamic acid, cinnamic alcohol and coumarin) in Ramulus Cinnamomi (GZ) and Ramulus Cinnamomi-Ephedrae Herba (MH) herb-couple (GZMH). Methods Twelve male Sprague-Dawley rats were randomly and equally divided into the GZ and GZMH herb-couple groups. Blood samples were collected at 0, 0.08, 0.25, 0.5, 0.75, 1.5, 3, 4, 6, 8, 12, 24, 36 and 48 h after oral administration. The three phenylpropanoids in rat plasma were quantified using an ultra-performance liquid-chromatography with tandem mass spectrometry (UPLC-MS/MS) method for pharmacokinetic study. Results In GZMH group, the area under the curve (AUC), mean retention time (MRT) of cinnamic acid and coumarin were increased significantly ( P <0.01, respectively), and biological half-life (t 1/2z ) was obviously shorter ( P <0.05) compared with the GZ group. There were no significant differences in the mean retention time from 0 to ∞ (MRT 0-∞ ), the peak concentration (C max ), the time to peak (T max ) and t 1/2z , except for AUC and MRT 0-t (the mean retention time from 0 to t) of cinnamic alcohol in the GZMH group by comparison to the GZ group ( P <0.01, respectively). The AUC, MRT (both P <0.01) and t 1/2z ( P <0.05) of coumarin were increased significantly, while C max , and T max were decreased slightly by comparison to the GZ group ( P >0.05). Conclusions There were statistically significant differences in some pharmacokinetic parameters of the three compounds between GZ and GZMH groups, which meant that MH could affect the absorption and elimination of the three compounds.

Effects of different selenium fertilizer types and dosages on non-volatile organic acids and aroma substances of flue-cured tobacco

Wanhui Jiang,Zijun Sun,Huaiyuan Li,Yaoxing Liang,Qihang Yang,Xin Yang,Lanjun Shao,Yuanyuan Wang,Jianjun Chen,Shiyuan Deng

Tobacco (Nicotiana tabacum L.) is well-known as an economic crop whose quality is evaluated according to its aroma quality. Researchers have found that selenium application can increase the aroma quality of tobacco, but until now, its mechanism is still unclear. So a field experiment was conducted to investigate the regulatory effect of the trace element selenium on enhancing aroma substances in flue-cured tobacco leaves. We compared selenium content, aroma precursors, non-volatile organic acids (NVOAs) content, and neutral aroma substances in tobacco upper leaves that applying three selenium fertilizer types (sodium selenite, organic selenium and nano-selenium) with three levels of pure selenium application rates (5.24 g·hm-2, 10.48 g·hm-2, and 15.72 g·hm-2) through foliar spraying. The results showed that the neutral aroma substances and NVOAs content were significantly increased. Metabolomics analysis of NVOAs showed that biosynthesis of phenylpropanoids pathway was significantly enriched. This indicates that selenium application may be able to regulate the accumulation of phenylpropane degradation products by regulating these NVOAs involved in the biosynthesis of phenylpropanoids pathway. Selenium content was significantly positively correlated with phenylpropane degradation products, NVOAs content, and phenolic compounds, while pyruvate was significantly down-regulated in each selenium application treatment. Therefore, it was speculated that selenium application could enhance the conversion of pyruvate into the biosynthesis of phenylpropanoids pathway. In summary, spraying organic selenium at 5.24–15.72 g·hm-2 or nano-selenium at 5.24–10.48 g·hm-2 could provide a suitable strategy for improving tobacco aroma quality.

Effect of Methyl Jasmonate on Phenolic Accumulation in Wounded Broccoli

Yuge Guan,Wenzhong Hu,Aili Jiang,Yongping Xu,Rengaowa Sa,Ke Feng,Manru Zhao,Jiaoxue Yu,Yaru Ji,Mengyang Hou,Xiaozhe Yang

In order to find an efficient way for broccoli to increase the phenolic content, this study intended primarily to elucidate the effect of methyl jasmonate (MeJA) treatment on the phenolic accumulation in broccoli. The optimum concentration of MeJA was studied first, and 10 μM MeJA was chosen as the most effective concentration to improve the phenolic content in wounded broccoli. Furthermore, in order to elucidate the effect of methyl jasmonate (MeJA) treatment on phenolic biosynthesis in broccoli, the key enzyme activities of phenylpropanoid metabolism, the total phenolic content (TPC), individual phenolic compounds (PC), antioxidant activity (AOX) and antioxidant metabolism-associated enzyme activities were investigated. Results show that MeJA treatment stimulated phenylalanine ammonia-lyase (PAL), cinnamate 4-hydroxylase (C4H), and 4-coumarin coenzyme A ligase (4CL) enzymes activities in phenylpropanoid metabolism, and inhibited the activity of polyphenol oxidase (PPO), and further accelerated the accumulation of the wound-induced rutin, caffeic acid, and cinnamic acid accumulation, which contributed to the result of the total phenolic content increasing by 34.8% and ferric reducing antioxidant power increasing by 154.9% in broccoli. These results demonstrate that MeJA in combination with wounding stress can induce phenylpropanoid metabolism for the wound-induced phenolic accumulation in broccoli. Keywords:wounded broccoli;methyl jasmonate;phenolic accumulation;phenylpropanoid metabolism;antioxidant capacity

Association of enriched metabolites profile with the corresponding volatile characteristics induced by rice yellowing process

Jinguang Liu,Yuqian Liu,Meng Jia,Xuedong Kang,Saimin Wang,Hui Sun,Meng Liu,Anqi Wang,Padraig Strappe,Zhongkai Zhou

Change in metabolites and volatiles during yellowing process in six rice cultivars was analyzed. Based on the yellowness, the study indicated Japonica was more prone to yellowing than Indica rice. Metabonomics analysis showed most differential metabolites were up-regulated, in which pathways of flavone and flavonol biosynthesis were significantly enriched following the yellowing process. Meanwhile, 54 differential metabolites were overlapped in six comparative groups, which is characterized by commonly-shared metabolic regulation pathway in each rice. Phenylalanine content was increased, followed by the enhanced phenylpropanoids formation, showing transformation between primary and secondary metabolites during yellowing process. Furthermore, 43 volatile compounds were identified, and the yellowed rice had more volatiles, including ketones, alcohols, esters and hydrocarbons, suggesting a positive correlation with the yellowing. Compounds 6-methyl-5-hepten-2-one and 6,10,14-trimethyl-2-pentadecanone were increased steadily during yellowing process, which may be applied for monitoring rice yellowing progress. This investigation provides further insight for revealing rice yellowing mechanism.

Bioconversion of Pinoresinol Diglucoside and Pinoresinol from Substrates in the Phenylpropanoid Pathway by Resting Cells of Phomopsis sp.XP-8

Yan Zhang,Junling Shi,Laping Liu,Zhenhong Gao,Jinxin Che,Dongyan Shao,Yanlin Liu

Pinoresinol diglucoside (PDG) and pinoresinol (Pin) are normally produced by plant cells via the phenylpropanoid pathway. This study reveals the existence of a related pathway in Phomopsis sp. XP-8, a PDG-producing fungal strain isolated from the bark of the Tu-chung tree ( Eucommiaulmoides Oliv.). After addition of 0.15 g/L glucose to Phomopsis sp. XP-8, PDG and Pin formed when phenylalanine, tyrosine, leucine, cinnamic acid, and p -coumaric acid were used as the substrates respectively. No PDG formed in the absence of glucose, but Pin was detected after addition of all these substrates except leucine. In all systems in the presence of glucose, production of PDG and/or Pin and the accumulation of phenylalanine, cinnamic acid, or p -coumaric acid correlated directly with added substrate in a time- and substrate concentration- dependent manner. After analysis of products produced after addition of each substrate, the mass flow sequence for PDG and Pin biosynthesis was defined as: glucose to phenylalanine, phenylalanine to cinnamic acid, then to p -coumaric acid, and finally to Pin or PDG. During the bioconversion, the activities of four key enzymes in the phenylpropanoid pathway were also determined and correlated with accumulation of their corresponding products. PDG production by Phomopsis sp. exhibits greater efficiency and cost effectiveness than the currently-used plant-based system and will pave the way for large scale production of PDG and/or Pin for medical applications.

Qingbo Liu,Jie Wang,Bin Lin,Zhuo-Yang Cheng,Ming Bai,Shaochun Shi,Xiao-Xiao Huang,Shao-Jiang Song

Alzheimer’s disease (AD) is characterized by the progressive accumulation of extracellular β -amyloid (A β ) aggregates. Recently, lignans and phenylpropanoids are attracting increasing attention to discovery useful agents of inhibition on A β aggregation. In the present study, to develop potential agents for slowing the progression of AD, Prunus tomentosa seeds were selected as a raw material for bioactive compounds, which led to the separation of two pairs of new enantiomeric lignans and phenylpropanoids using chiral HPLC. The planar structures of these compounds were elucidated by spectroscopic data analyses. And their absolute configurations were determined by comparing of experimental and calculated electronic circular dichroism (ECD). The biosynthesis pathway was also discussed. Additionally, the inhibitory activity on A β aggregation of all optical pure compounds was tested by thioflavin T (ThT) assay. The isolates ( 1a , 1b , 2a and 2b ) showed more potent inhibitory activity than positive control curcumin with inhibitory rate of 73.89 ± 3.41% 78.69 ± 1.50%, 63.25 ± 2.68%, and 67.13 ± 0.90% at 20 μM, respectively. More importantly, the inhibition profiles were explained by molecular dynamics and docking simulation studies.

Protective Effects of Hippophae rhamnoides L. Phenylpropanoids on Doxorubicin-Induced Cardiotoxicity in Zebrafish

Gang Li,Ming Chu,Yingying Tong,Yuexin Liang,Shenghui Wang,Chengjun Ma,Zhenhua Wang,Wenna Zhou

Hippophae rhamnoidesL. is a deciduous shrub that contains many unique bioactive substances. This sea buckthorn possesses anticancer, antioxidant, anti-inflammatory, and cardiovascular protective properties. Herein, the effects of phenylpropyl compounds extracted fromH. rhamnoidesL. on doxorubicin (Dox)-induced cardiotoxicity were evaluated in zebrafish. Cardiac injury in zebrafish was induced using 35 μM Dox for 96 h, and 30 μM phenylpropanoid compounds were used as the protective treatment. The cardioprotective effects and mechanisms of the four phenylpropanoids were investigated using microscopy, behavioral analysis, acridine orange staining, western blotting, flow cytometry, and real-time quantitative polymerase chain reaction. The extracted phenylpropanoids could significantly relieve Dox-induced cardiac injury in zebrafish and inhibit cardiomyocyte apoptosis. The mechanisms of action were mainly related to the stability of mitochondrial biogenesis and function maintained by phenylpropanoids in zebrafish. To our knowledge, this is the first report on the protective effect of sea buckthorn against myocardial injury in zebrafish. Our findings provide support for the further research and development of sea buckthorn and its components. Keywords:sea buckthorn;zebrafish;doxorubicin;cardiotoxicity;mitochondria

Changes in phenolic content, composition, and antioxidant activity of blood oranges during cold and on-tree storage

Ji-chun ZHAO,Miao AO,Xiao-qin HE,Wei-zhou LI,Li-li DENG,Kai-fang ZENG,Jian MING

Citrus fruits are rich in phenolic compounds that possess several health benefits. However, few studies have focused on the changes in phenolic compounds in citrus fruits during postharvest storage. This study dynamically monitored the phenolic content, components and antioxidant activity of ‘Tarocco’ blood oranges during a period of 12-week cold storage and on-tree storage, respectively. We investigated the alteration mechanism of phenolic compounds in blood oranges by evaluating phenylpropanoid pathway-related enzyme activities and gene expression. Results showed that flavanones were the main phenolic compounds in blood oranges. Both storage methods mainly stimulated the accumulation of phenolic acids to improve total phenolic content, which reached the maximum at week 12. Nonetheless, blood oranges had a higher phenolic content and antioxidant activity under on-tree storage than cold storage. Furthermore, the enzyme activities and gene expression of the phenylpropanoid pathway demonstrated that the accumulation of phenolics in blood oranges during storage was highly related to the activation of the phenylpropanoid pathway. These results demonstrate that on-tree storage is a potential approach for extending the supply period of blood orange from the perspective of phenolic compounds.

Evaluation of antioxidant, antidiabetic and antiobesity potential of phenylpropanoids (PPs): Structure-activity relationship and insight into action mechanisms against dual digestive enzymes by comprehensive technologies

Jiao Li,Chao-Feng Qin,Nai-Dong Chen

Phenylpropanoids (PPs), a group of natural compounds characterized by one or more C6-C3 units, have exhibited considerable potential in addressing metabolic disease. However, the comprehensive investigation on the relationship of compound structures and involved activity, along with the action mechanisms on the drug target is absent. This study aimed to evaluate the antioxidant and inhibitory activities of 16 PPs against two digestive enzymes, including α-glucosidase and pancreatic lipase, explore the structure–activity relationships and elucidate the mechanisms underlying enzyme inhibition. The findings revealed the similarities in the rules governing antioxidant and enzyme inhibitory activities of PPs. Specifically, the introduction of hydroxyl groups generally exerted positive effects on the activities, while the further methoxylation and glycosylation were observed to be unfavorable. Among the studied PPs, esculetin exhibited the most potent antioxidant activity and dual enzymes inhibition potential, displaying IC 50 values of 0.017 and 0.0428 mM for DPPH and ABTS radicals scavenging, as well as 1.36 and 6.67 mM for α-glucosidase and lipase inhibition, respectively. Quantification analysis indicated esculetin bound on both α-glucosidase and lipase successfully by a mixed-type mode. Further analyses by UV–Vis, FT-IR, fluorescence spectra, surface hydrophobicity, SEM, and molecular docking elucidated that esculetin could bind on the catalytic or non-catalytic sites of enzymes to form complex, impacting the normal spatial conformation for hydrolyzing the substrate, thus exhibiting the weakened activity. These results may shed light on the utilization value of natural PPs for the management of hyperglycemia and hyperlipemia, and afford the theoretical basis for designing drugs with stronger inhibition against the dual digestive enzymes based on esculetin.
