In this study, chitosan and sugarcane cellulose were used as film-forming materials, while the inorganic agent zinc oxide (ZnO) and natural compound phenyllactic acid (PA) were used as the main bacteriostatic components to fabricate biodegradable antimicrobial composite membranes. The water absorption and antimicrobial properties were investigated by adjusting the concentration of PA. The scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) results demonstrated that the components of the composite membrane were successfully integrated. The addition of ZnO improved the mechanical and antimicrobial properties of the composite membrane, while the addition of PA with high crystallinity significantly reduced the water absorption and swelling. Moreover, the addition of 0.5% PA greatly improved the water absorption of the composite membrane. The results of antimicrobial experiments showed that PA improved the antimicrobial activity of the composite membrane againstStaphylococcus aureus,Escherichia coli,Aspergillus nigerandPenicillium rubens. Among them, 0.3% PA had the best antimicrobial effect againstS. aureus,E. coliandA. niger, while 0.7% PA had the best antimicrobial effect againstP. rubens. Keywords:composite membrane;phenyllactic acid;antimicrobial activity

Objectives To achieve high production of rosmarinic acid and derivatives in Escherichia coli which are important phenolic acids found in plants, and display diverse biological activities. Results The synthesis of rosmarinic acid was achieved by feeding caffeic acid and constructing an artificial pathway for 3,4-dihydroxyphenyllactic acid. Genes encoding the following enzymes: rosmarinic acid synthase from Coleus blumei , 4-coumarate: CoA ligase from Arabidopsis thaliana , 4-hydroxyphenyllactate 3-hydroxylase from E. coli and d -lactate dehydrogenase from Lactobacillus pentosus , were overexpressed in an l -tyrosine over-producing E. coli strain. The yield of rosmarinic acid reached ~130 mg l −1 in the recombinant strain. In addition, a new intermediate, caffeoyl-phenyllactate (~55 mg l −1 ), was also produced by the engineered E. coli strain. Conclusion This work not only leads to high yield production of rosmarinic acid and analogues, but also sheds new light on the construction of the pathway of rosmarinic acid in E. coli .

Alicyclobacillus spp. are one of the major harmful microorganisms that can cause fruit juice spoilage. In this study, the inactivation activity and mechanism of phenyllactic acid (PLA) and glycerol monolaurate (GML) against Alicyclobacillus spp. were evaluated, and their effects on fruit juice quality were also investigated. The results displayed that the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of GML were 0.01–0.02 mg/mL and 0.04–0.60 mg/mL, while those of PLA were in the range of 0.25–0.50 mg/mL and 0.5–16.0 mg/mL, respectively. All of these antibacterial agents disrupted the permeability of cell membranes, destroyed the morphology of bacteria, and led to the leakage of cell contents. GML could stimulate bacteria to release a large number of reactive oxygen species, causing some oxidative damage and destroying bacteria. Treatment with L-PLA could reduce the level of adenosine triphosphate (APT) in cells by inactivating ATP synthase. In addition, the MBC of GML and L-PLA in apple juice were 0.02–0.6 and 0.25–16 mg/mL, respectively, which mainly depended on the strain difference. The treatment of L-PLA and GML had no significant adverse effect on the quality of apple juice, except that GML had some effect on color and organic acids, and L-PLA increased the total acid content. Sensory evaluation revealed that these treatments did not affect the organoleptic properties of apple juice. This study provides preliminary theoretical and data support for the application of GML and L-PLA to control Alicyclobacillus spp. contamination in apple juice.

Respiration and energy metabolism are intricately linked to the quality and senescence of fruit after harvest. This investigation was carried out to examine alternations in quality parameters, respiration, and energy metabolism of ‘Zaosu’ pears subsequent to phenyllactic acid immersion. Results demonstrated that phenyllactic acid treatment led to a reduction in L * , a * , b * values, ΔE, and mass loss. Additionally, it maintained ascorbic acid, reducing sugar, soluble sugar, soluble solids, and chlorophyll contents; delayed the decrease in flesh firmness and titratable acidity of pears. The activities and gene expressions of glucose-6-phosphate dehydrogenase, glucose phosphate isomerase, hexokinase, nicotinamide adenine dinucleotide kinase, cytochrome C oxidase, and succinate dehydrogenasepears in pears were enhanced by phenyllactic acid. Moreover, phenyllactic acid promoted PbPK , PbAOX , PbH + -ATPase , and PbCa 2+ -ATPase expressions, while inhibiting malate dehydrogenase activity and the expressions of PbPFK and PbMDH in pears. Phenyllactic acid also led to a decrease in the levels of nicotinamide adenine dinucleotide hydrogen and nicotinamide adenine dinucleotide, while causing an increase in the levels of nicotinamide adenine dinucleotide phosphate hydrogen and nicotinamide adenine dinucleotide phosphate within the fruit. Collectively, postharvest phenyllactic acid treatment can effectively preserve the storage quality of ‘Zaosu’ pears by regulating the activity and gene expression of the key enzymes involved in respiration and energy metabolism.

Black spot rot, caused by the pathogen Alternaria alternata , is a prevalent disease in apple cultivation. The present study investigated the effects of phenyllactic acid (PLA) immersion on black spot rot in apples and elucidated the potential mechanisms underlying its action. Results revealed that 1.00 g L -1 PLA inhibited the lesion development in A. alternata- inoculated apples. PLA resulted in a decrease in malondialdehyde (MDA) content, while simultaneously increasing the levels of H 2 O 2 , ascorbic acid (AsA), and reduced glutathione (GSH) in apples. PLA also induced the increase in gene expressions and enzymatic activities of superoxide dismutase (SOD), catalase (CAT), monodehydroascorbate reductase (MDHAR), dehydroascorbic acid reductase (DHAR), ascorbate peroxidase (APX), glutathione reductase (GR), peroxidase (POD), polyphenol oxidase (PPO), cinnamate 4-hydroxylase (C4H), 4-coumarate/coenzyme A ligase (4CL), and phenylalanine ammonia lyase (PAL) in apples. Moreover, inoculation of both PLA-treated and control fruit with A. alternata led to a further increase in the levels of H 2 O 2 , GSH, and AsA, along with enhanced gene expressions and enzymatic activities of CAT, SOD, DHAR, GR, APX , MDHAR, POD, PPO, C4H, 4CL and PAL, compared to un-inoculated fruit. Additionally, PLA dipping increased total phenols and flavonoids contents in apples, and inoculation with A. alternata further augmented the accumulation of these bioactive compounds. Furthermore, PLA-treated fruit displayed a more rapid response to A. alternata inoculation compared to control fruit, characterized by heightened enzymatic activity, gene expression, and metabolite content. Collectively, PLA has the potential to stimulate disease resistance against A. alternata in apples through regulating major enzyme activities, gene expressions, and metabolites contents involved in reactive oxygen species and phenylpropane metabolisms.

Abstract Phenyllactic acid (PLA) generally recognized as a natural organic acid shows against Vibrio parahaemolyticus activity. In this study, V. parahaemolyticus ATCC17802 ( Vp 17802) was cultured under the stress of 1/2MIC PLA, and then the antibacterial mechanisms were explored via transcriptomics. The minimum inhibitory concentration (MIC) of PLA against Vp 17802 was 3.2 mg/mL, and the time-kill analysis resulted that Vp 17802 was inhibited. PLA was able to destroy the bacterial membrane, leading to the leakage of intracellular substances and decline of ATP levels. The RNA-sequencing analysis results indicated that 1616 significantly differentially expressed genes were identified, among which 190 were up-regulated and 1426 were down-regulated. Down-regulation of the icd2 gene in the TCA cycle mediates blockage of tyrosine metabolic, arginine biosynthesis, and oxidative phosphorylation, causing insufficient energy supply of Vp 17802. Moreover, PLA could cause amino acids, metal ions, and phosphate transporters to be blocked, affecting the acquisition of nutrients. The treatment by PLA altered the expression of genes encoding functions involved in quorum sensing, flagellar assembly, and cell chemotaxis pathway, which may be interfering with the biofilm formation in Vp 17802, reducing cell motility. Overall, 1.6 mg/mL PLA inhibited the growth of Vp 17802 by disrupting to uptake of nutrients, cell metabolism, and the formation of biofilms. The results suggested a new direction for exploring the activity of PLA against Vp 17802 and provided a theoretical basis for bacterial pathogen control in the food industry. Key points •RNA sequencing was carried out to indicate the antibacterial mechanism of Vp17802. •The icd2 gene in the TCA cycle mediates blockage of metabolic of Vp17802. •The biofilm formation has interfered with 1.6 mg/mL PLA, which could reduce cell motility and virulence. Acidity plays a crucial role in determining the sensory attributes of the majority of fresh fruit and vegetables. This study investigated the variations of organic acids metabolism and salicylic acid biosynthesis in Zaosu pears after postharvest phenyllactic acid dipping. Results elucidated that phenyllactic acid dipping improved the contents of malic, succinic, citric, and salicylic acid in Zaosu pears. Phenyllactic acid also improved the enzymatic activities and gene transcription levels of phosphoenolpyruvate carboxylase, mitochondrial citrate synthase, mitochondrial aconitase, and NADP-isocitrate dehydrogenase, whereas declined NADP-dependent malate (NADP-ME) and cytoplasmic aconitase activities as well as PbNADP-ME transcription level in pears. Moreover, the enzymatic activities and gene transcription levels of phenylalanine ammonia lyase and shikimate dehydrogenase as well as the gene transcription levels of 3-dehydroquinate synthase, d -arabino-heptulosonate-7-phosphate synthase, chorismate synthase, shikimate kinase, isochorismate synthase, 5-enolppyruvylshikimate-3-phosphate synthetase, avrPphB susceptible 3, enhanced pseudomonas susceptibility 1, enhanced disease susceptibility 5, and chorismate mutase in Zaosu pears were promoted by phenyllactic acid treatment. Taken together, phenyllactic acid can regulate the pivotal enzyme activity and gene transcription in organic acids metabolism and salicylic acid biosynthesis to sustain the flavor and freshness of Zaosu pears.

It is well known that cofactors play a key role in the production of different compounds in bioconversion processes, while the high cost of cofactors limits their usage in industrial applications. In the present study, a NADH regeneration system was successfully developed in Lactobacillus plantarum by expressing the fdh gene coding for formate dehydrogenase (FDH) from Candida boidinii. Results indicated that the FDH was expressed with the highest activity of 0.82 U/mg of protein when cells entered early stationary phase. In addition, the expression of FDH increased the intracellular level of NADH and NADH/NAD + ratio in L. plantarum , and therefore, enhanced the NADH-dependent production of 3-phenyllactic acid (PLA) in repeated and fed-batch bioconversions. In brief, the results demonstrate that the NADH regeneration by expressing FDH is a promising strategy for producing NADH-dependent microbial metabolites in L. plantarum .

Polyphenol oxidase is the popular enzyme involved in fruit-vegetable browning and melanin synthesis. In the present paper, L-3-phenyllactic acid, a natural bacteriostatic substance, was investigated as an inhibitor of polyphenol oxidase. The results were demonstrated that the residual enzyme activity of polyphenol oxidase decreased gradually with the increase of the concentration of L-3-phenyllactic acid. The L and ΔE values of the reaction system increased gradually. The reversible mixed-type inhibition mode of L-3-phenyllactic acid was determined by Lineweaver Burk plot. At the same time, the results of fluorescence quenching demonstrated that L-3-phenyllactic acid was a quencher of polyphenol oxidase, and the molecular docking study provided the binding mode of L-3-phenyllactic acid and polyphenol oxidase at the molecular level. L-3-phenyllactic acid decreased the activity of polyphenol oxidase and browning of fresh-cut Agaricus Bosporus. This research first studied the inhibitory effect of L-3-phenyllactic acid on the activity of polyphenol oxidase, and would provide a theoretical foundation for the use of L-3-phenyllactic acid as anti-polyphenol oxidase agents.

Preservatives and sweeteners are always added in the food production process, which can extend the life and flavor of food. Benzoic acid and saccharin sodium are commonly used as preservatives and sweeteners respectively. Phenyllactic acid as a natural preservative, mainly produced by phenylalanine fermentation, may exist in fermented food. The objective of the current study is to develop a reverse-phase HPLC-UV method for simultaneous determination of benzoic acid, sorbic acid, phenyllactic, phenylalanine, and saccharin sodium in vinegar. They were separated on Sapphiresil TM (C 18 ) column (250 × 4.6 mm, 5 μm) with a mobile phase consisting of methyl alcohol:ammonium acetate buffer (pH 6.44; 0.02 M) (20:80, v/v) ran in isocratic mode at a flow rate of 1 mL/min. Complete baseline separation of five analytes was achieved in < 12 min, the linear range was 0.25–50 μg/mL with a correlation coefficient R 2 value > 0.9975, the method was validated with recoveries in the 96.93 to 118.42% range, and RSD was 0.39–6.21%. The detection limits of phenylalanine, benzoic acid, saccharin sodium, sorbic acid, and phenyllactic acid were 0.25, 0.01, 0.01, 0.025, and 0.05 mg/L. This method could be used for determining of benzoic acid, sorbic acid, phenyllactic acid, phenylalanine, and saccharin sodium in vinegar.

Phenyllactic acid (PLA) has gained a lot of attention due to its broad antimicrobial activity, but the mechanism of its antifungal action has been barely reported until now. Herein, the inhibitory activity of PLA against Aspergillus flavus spore germination and its mechanism were preliminarily investigated. Results indicated that PLA had a strong antifungal activity against A. flavus with the minimal inhibitory concentration (MIC) and minimal fungicidal concentration (MFC) of 6 and 12 mg/ml, respectively. As observed by scanning electron microscopy (SEM), the A. flavus spores displayed wrinkled and shrunken appearance after treatment with PLA. In addition, the permeability and integrity of A. flavus cell membrane were changed obviously after PLA treatment as indicated by the propidium iodide (PI) staining results, which was further confirmed by a rise in electric conductivity and increased leakage of intracellular protein and nucleic acid. Furthermore, reduced activities of mitochondrial ATPase and dehydrogenases caused by PLA were also observed in A. flavus spores, with a result of remarkable decrease in ATP synthesis. Therefore, it could be concluded that PLA was effective in inhibiting spore germination of A. flavus mainly by disrupting cell membrane and interfering with mitochondrial energy metabolism.

African swine fever (ASF) is a highly contagious and hemorrhagic disease caused by infection with the African swine fever virus (ASFV), resulting in a mortality rate of up to 100%. Currently, there are no effective treatments and commercially available vaccines for ASF. Therefore, it is crucial to identify biochemicals derived from host cells that can impede ASFV replication, with the aim of preventing and controlling ASF. The ASFV is an acellular organism that promotes self-replication by hijacking the metabolic machinery and biochemical resources of host cells. ASFV specifically alters the utilization of glucose and glutamine, which are the primary metabolic sources in mammalian cells. This study aimed to investigate the impact of glucose and glutamine metabolic dynamics on the rate of ASFV replication. Our findings demonstrate that ASFV infection favors using glutamine as a metabolic fuel to facilitate self-replication. ASFV replication can be substantially inhibited by blocking glutamine metabolism. The metabolomics analysis of the host cell after late-stage ASFV infection revealed a significant disruption of normal glutamine metabolic pathways due to the abundant expression of PLA (phenyllactic acid). Pretreatment with PLA also inhibited ASFV proliferation and glutamine consumption following infection. The metabolomic analysis also showed that PLA pretreatment greatly slowed down the metabolism of amino acids and nucleotides that depend on glutamine. The depletion of these building blocks directly hindered the replication of ASFV by decreasing the biosynthetic precursors produced during the replication of ASFV’s progeny virus. These findings provide valuable insight into the possibility of pursuing the development of antiviral drugs against ASFV that selectively target metabolic pathways.
