The unsatisfactory bioavailability of polyphenols caused by poor solubility, instability and low absorption rate limits their application in functional food field. This study fabricated the nanoparticles based on chitosan and sodium alginate for encapsulation of the polyphenols isolated from Malus baccata (Linn.) (MBP-NPs) and investigated their gastrointestinal digestive and absorption characteristics. MBP-NPs with uniform sphere morphology had the average particle size of 611.3 nm and non-crystalline properties. During simulated gastrointestinal digestion in vitro , MBP-NPs exhibited the sustain release characteristics. Through administration of MBP-NPs to mice , the phenolic composition of MBP-NPs after gastrointestinal digestion was similar to that of undigested MBP, which suggested that the degradation of polyphenols during gastrointestinal digestion was successfully prevented. After small intestinal absorption , more polyphenols were detected in liver and spleen via participating in systemic circulation , thereby improving the bioavailability of MBP. MBP-NPs also promoted the production of short chain fatty acids for maintainance of intestinal health. Collectedly, these findings proved that the nanoparticles prepared based on CS and SA could be served as an effective strategy for encapsulation of MBP to potentially ameliorate their bioavailability.

To effectively harness and valorize the often-overlooked pruning waste from Malus plants, an eco-friendly and efficient strategy for the simultaneous extraction and in situ de-glycosylation to prepare phloretin was introduced for the first time. By designing an acidic deep eutectic solvent (DES) system and adjusting the polarity and hydrophilicity of the non-acidic component as well as the acidity of the acidic component, a significant enhancement in the extraction yield and de-glycosylation efficiency of phloretin and its glycoside derivatives was achieved. The highest phloretin content was attained using ethylene glycol (EG) - p-toluenesulfonic acid (PTSA) (2:1), which consisted of ethylene glycol and p -toluenesulfonic acid with a molar ratio of 2:1 and 30 % water content. After treatment condition optimization, a peak phloretin content of 139.9380 ± 0.2191 μmol/g was achieved, which notably surpassed traditional organic solvent extraction methods. Furthermore, the phloretin was successfully recovered from the EG-PTSA (2:1) solution using macroporous resin adsorption method, with SP825 resin showing a recovery rate of 85.87 %. Applicability analysis confirmed that the EG-PTSA (2:1) treatment method is suitable for various Malus plant pruning wastes. As a result, this study not only offered a justification for improving the extraction and de-glycosylation efficiency through multi-dimensional modification of the acidic DES but also presented an innovative approach for utilizing Malus plant pruning waste.

Flower color is the most important ornamental character trait of Malus species. The study aimed to clarify the roles of phenolic compounds and compositions in the petal color change of Malus spp. cultivars at various flowering stages. Meanwhile, the antioxidant activity in the petals of Malus spp. was detected to estimate the potential of market application. Eight Malus spp. cultivars were divided into four systems according to the flower color, namely dark-red (‘Royalty’ and ‘Perfect Purple’), rose-red (‘Kelsey’ and ‘Sparkler’), pink-red (‘Strawberry Parfait’ and ‘Pink Spires’), and white (‘Jewelberry’ and ‘Snow Drift’). Total phenolic content (TPC), total flavonoid content (TFC), total anthocyanin content (TAC), and antioxidant activity of the petals were measured. The results revealed that high levels of TPC, TFC, TAC, and antioxidant activity in petals significantly decreased during flower development. These indicators were significantly and positively related to each other. Abundant phenolic metabolites in petals were determined by HPLC-DAD, and these compounds included anthocyanins, flavonols, flavanols, flavonones, phenolic acids, and dihydrochalcones. Anthocyanin concentrations and ratios in petals were notably different among the four color systems, with the highest levels noted in dark-red colored flowers, followed by rose-red, pink-red, and white flowers. Cyanidin-3- O -galactoside was the major anthocyanin noted. Its concentration reduced significantly with flower development, which is described as the primary cause of petal color fading. In addition, the petals contained abundance quercetin-3- O -glucuronide, hyperoside, taxifolin, chlorogenic acid, ferulic acid, phloridzin and other substances known for their antioxidant activities. Given these findings, we propose that the flowers of Malus spp. hold good utilization value as new sources of bioaccessible polyphenols and strong potential for development. Especially, the flowers at early stages will be supplements to the natural colorant market.

The study aims to analyze the phenolic compounds in Malus spp. and evaluate their antioxidant and pro-apoptotic effects in BGC-803 gastric cancer cells. The results showed that cyanidin-3-galactoside was the main polyphenol in Malus “Royalty” (MR), while catechin, epicatechin, and proanthocyanidin B1 and B2 contents were higher in Malus “Cinnabar” (MC) and Malus micromalus (MM) than in MR fruits. The total polyphenol content, total flavonoid content, and antioxidative properties of Malus spp. fruits followed an order of MR > MC > MM. Fruit extracts could inhibit BGC-803 cells growth and induce apoptosis, with IC 50 values of 0.47, 0.36, and 0.31 mg/ml for MR, MC, and MM, respectively. Furthermore, fruit extracts induced cell apoptosis through increasing Bcl-2 and Bcl-xl (pro-apoptosis) expression and inhibiting Bax and Bak (anti-apoptosis) expression, thereby accelerating cell apoptosis. Taken together, these results demonstrate that red fruits (i.e., MR and MC) were more effective against cancer cells than green fruits (i.e., MM). Practical applications Fruits of Malus spp. are ≤5 cm in size, considerably smaller than apples. They are rich in various natural bioactive compounds that are often consumed as a dietary supplement or used as natural raw materials for function food. In the current study, it is comprehensively characterized profile and bioactivity of phenolic metabolites in Malus spp. fruits of different colors, and found that red fruits are more effective in reducing the free radicals and inducing cancer cells apoptosis than green fruits. These findings are valuable for food technologists and food manufacturers, especially those who produce crabapple supplement. The study investigated the molecular mechanism of how Malus spp. fruits exert anti-cancer functions. This lays a theoretical foundation for future research on developing anti-cancer function food and provides helpful guidance for fruit market management and fruit processing industry.
