Effectiveness of 12 Insecticides to a Laboratory Population of Cydia pomonella (Lepidoptera: Tortricidae) Newly Established in China

Wu Zheng-Wei,Zhang Ya-Lin,Shang Su-Qin

The codling moth Cydia pomonella (L.) is an economically important fruit pest that has spread rapidly from its original site in Xinjiang to other northwestern regions of China. Insecticides are widely used to control this pest but its invasion has never been completely stopped. The aim of this study was to establish a laboratory population of the codling moth occurring in China, to investigate the effectiveness of 12 conventional insecticides to this laboratory population, and to recommend the discriminating doses for use in resistance monitoring. The laboratory population was generally similar to other laboratory strains although parameters such as survival rate and larval duration were low when compared with field populations. Toxicity varied among the insecticides tested with LC 50 values ranging from 0.016 mg/l for emamectin benzoate to 55.77 mg/l for chlorbenzuron. Discriminating dose levels were determined from dose-mortality reference curves for the detection of resistance in field populations. Effectiveness of 12 insecticides to a laboratory population of codling moth in China was evaluated for the first time. This can be integrated into resistance management strategies, especially in orchards with a history of frequent insecticides applications, in order to monitor or decrease insecticide resistance in the future.

Functional characterization of a novel λ-cyhalothrin metabolizing glutathione S-transferase, CpGSTe3, from the codling moth Cydia pomonella

Chao Hu,Wei Wang,Di Ju,Gao-Man Chen,Xiao-Ling Tan,David Mota-Sanchez,Xue-Qing Yang

BACKGROUND Recent work has shown that two codling moth ( Cydia pomonella ) glutathione S -transferase genes (GSTs), CpGSTd1 and CpGSTd3 , can metabolize λ-cyhalothrin, one of the recommended insecticides for C. pomonella control worldwide. However, systematical characterization of delta and epsilon GSTs, especially their potential contributions in the metabolism of λ-cyhalothrin, is currently still lacking in C. pomonella . RESULTS In this study, a total of nine cDNA sequences were identified in C. pomonella , including four in the delta and five in the epsilon subclasses. RT-qPCR showed that seven GSTs were ubiquitously expressed at all developmental stages, and CpGSTe2 , CpGSTe3 , and CpGSTe4 were mainly expressed in larvae. The mRNA levels of CpGSTd2 , CpGSTd4 , and CpGSTe5 were significantly higher in male than in female adults. Tissue-specific expression analysis revealed that the CpGSTe2 , CpGSTe3 , and CpGSTe4 were highly expressed in the midgut while CpGSTd2 and CpGSTd4 were predominantly expressed in the Malpighian tubules. The transcripts of these GSTs (except CpGSTe1 ) were co-expressed following exposure to LD 10 of λ-cyhalothrin for 3 h. Recombinant CpGSTd4, CpGSTe2, and CpGSTe3 proteins expressed in Escherichia coli displayed glutathione-conjugating activity toward 1-chloro-2,4-dinitrobenzene. In addition, λ-cyhalothrin could inhibit the activity of recombinant CpGSTd4, CpGSTe2, and CpGSTe3 enzymes, but only recombinant CpGSTe3 showed λ-cyhalothrin metabolic capacity, with 21.88 ± 1.09% of parental compound being depleted within 1 h. CONCLUSION These data show that CpGSTe3 is a third GST gene, encoding an enzyme that metabolizes λ-cyhalothrin in C. pomonella . © 2019 Society of Chemical Industry

Identification of G Protein-Coupled Receptors (GPCRs) Associated with Lambda-Cyhalothrin Detoxification in Cydia pomonella

Yu-Xi Liu,Chao Hu,Yu-Ting Li,Ping Gao,Xue-Qing Yang

While previous studies have reported G protein-coupled receptor (GPCR)-mediated insecticide resistance in various arthropods, the understanding of GPCR-associated resistance mechanisms in Cydia pomonella remains limited. In this study, a total of 95 CpGPCR genes categorized into four families were identified in C. pomonella. Results revealed high expression levels of the majority of the CpGPCRs during the first larval stage and in the head of C. pomonella. Exposure to lambda-cyhalothrin significantly increased the expression of 15 CpGPCRs, including CpGPCR70, which is highly expressed in all larval stages and shows the highest expression in the midgut. RNA interference (RNAi) demonstrated that downregulation of CpGPCR70 leads to reduced expression of key resistance-related genes and a decreased tolerance of larvae to lambda-cyhalothrin. These findings indicate that CpGPCR70 plays a crucial role in regulating the expression of detoxifying genes involved in lambda-cyhalothrin resistance, offering valuable insights for the development of more effective pest control strategies.

Characterization of Multiple Heat-Shock Protein Transcripts from Cydia pomonella: Their Response to Extreme Temperature and Insecticide Exposure

Xue-Qing Yang,Ya-Lin Zhang,Xiao-Qi Wang,Hui Dong,Ping Gao,Ling-Yi Jia

The economically important fruit pest Cydia pomonella (L.) exhibits a strong adaptability and stress tolerance to environmental stresses. Heat-shock proteins (HSPs) play key roles in insects in coping with environmental stresses. However, little is known about the spatiotemporal expression patterns of HSPs and their response to stresses in C. pomonella. In this study, a thermal treatment–recovery test was performed, and the expression profiles of a novel isolated HSP, named CpHSP40, and six CpHSPs were determined. Third-instar larvae were able to recover from cold shock (0 °C) and heat shock (40 °C). Escherichia coli BL21 (DE3) cells harboring recombinant pET-28a (+)-CpHSP40 plasmid showed significant temperature tolerance. CpHSPs were developmentally and tissue-specifically expressed. The responses of CpHSPs to 0 and 40 °C (with or without recovery) and insecticide exposure were varied. All of these indicated that the expression of HSPs plays a role in the development and in environmental adaptation in C. pomonella.

Stability, Inheritance, Cross-Resistance, and Fitness Cost of Resistance to λ-Cyhalothrin in Cydia pomonella

Chao Hu,Cong Zhang,Yi-Fan Tang,Yu-Xi Liu,Zi-Nan Xia,Yan Wang,Wei-Tu Li,Ping Gao,Yu-Ting Li,Yun-Tong Lv,Xue-Qing Yang

Insecticides are commonly utilized in agriculture and forestry for pest control, but their dispersal can pose hazards to humans and environment. Understanding resistance, inheritance patterns, and fitness costs can help manage resistance. A λ-cyhalothrin-resistant population (LCR) of Cydia pomonella, a global pest of pome fruits and walnuts, was obtained through selective insecticide breeding for 15 generations, showing stable moderate resistance (23.85-fold). This population was cross-resistant to deltamethrin (4.26-fold) but not to β-cypermethrin, chlorantraniliprole, chlorpyrifos, and avermectin. Genetic analysis revealed the resistance was autosomal, incompletely dominant, and controlled by multiple genes. Increased activity of glutathione S-transferases and cytochrome P450 monooxygenases (P450s) played a primary role in resistance, with specific genes up-regulated in LCR, and exhibited significant expression in midgut. LCR also exhibited fitness costs, including delays in development, reduced fecundity, and slower population growth. These findings contribute to understanding λ-cyhalothrin resistance in C. pomonella and can guide resistance management strategies.

Establishing a fluorescence-based technique for ABC transporters functional analysis in metabolism of insecticides in a Lepidopteron

Di Ju,Chao Hu,Yuntong Lv,Yuting Li,Ping Gao,Xueqing Yang

The insecticide resistance development poses challenges to the pest control effectiveness. As phase III detoxification enzyme, ATP-binding cassette (ABC) transporters play a crucial role in conferring resistance by facilitating the elimination of insecticides. However, the precise involvement of ABCs in insecticides metabolism remains unclear. In this study, a competitive assay utilizing microinjection of the ABC substrate Texas Red (TR) was established in Lepidoptera for the first time to investigate the contribution of ABCs to the detoxification process of Cydia pomonella towards lambda -cyhalothrin (LCT) and abamectin (ABM). Results revealed that the elimination of TR was hindered by ABC inhibitor verapamil, LCT, or a combination of both, implying a shared subsets of ABCs involved in the transportation of TR and LCT. Moreover, the application of ABM alone led to marked inhibition of TR elimination, with verapamil not further exacerbating this inhibitory effect, indicating that ABM exhibited a highly competitive ABC substrate characteristic and has a strong interaction with them. Furthermore, silencing key resistance associated ABC genes through RNAi resulted in increased mortality of larvae under LCT or ABM exposure, affecting TR elimination and ATP hydrolysis. These findings imply that ABC genes are critical for the active transport function of ABCs energized by ATP, ultimately reducing larvae tolerance to insecticides. In conclusion, our findings not only validated the feasibility of a competitive bioassay using fluorescent ABC substrate in lepidopteran larvae but also highlighted that ABCs in enhancing the efflux capacity of C. pomonella larvae, facilitating the binding and transportation of insecticides and their metabolites.

Characterization of a lambda-cyhalothrin metabolizing glutathione S-transferase CpGSTd1 from Cydia pomonella (L.)

Liu Jiyuan,Yang Xueqing,Zhang Yalin

In insects, glutathione S -transferases (GSTs) are enzymes involved in detoxification of insecticides. However, few data are available for the codling moth, Cydia pomonella (L.). In this study, we cloned a delta class GST gene CpGSTd1 from C. pomonella . Real-time quantitative PCR shows that CpGSTd1 was up-regulated with aging, and the mRNA level of CpGSTd1 was higher in the fat body and silk glands than in other tissues. The expression level of CpGSTd1 exposure to insecticide suggests that CpGSTd1 is up-regulated after chlorpyrifos-methyl and lambda-cyhalothrin treatments. Both lambda-cyhalothrin and chlorpyrifos-methyl altered GST activity in vivo. The purified CpGSTd1 protein exhibits a high catalytic efficiency with CDNB and was inhibited by lambda-cyhalothrin and chlorpyrifos-methyl in vitro. Metabolism assays indicate that lambda-cyhalothrin was significantly metabolized while chlorpyrifos-methyl was not metabolized by CpGSTd1. Binding free energy analysis suggests that CpGSTd1 binding is tighter with lambda-cyhalothrin than with chlorpyrifos-methyl. Our study suggests that CpGSTd1 plays a key role in the metabolism of insecticides in C. pomonella .

Different detoxification strategies of fruit borers adaptation to quercetin

Bing Bai,Huan-Juan Zhao,Bo-Kun Wang,Yu-Ting Li,Ping Gao,Yun-Tong Lv,Xue-Qing Yang

During long-term plant-insect interactions, plants have evolved diverse plant secondary metabolites (PSMs) as defenses against herbivory, while insects have developed sophisticated detoxification mechanisms to neutralize these compounds. Invasive and native herbivorous species may employ distinct metabolic strategies to cope with PSMs, a phenomenon that remains incompletely characterized. Here, we report the differential detoxification strategies of two closely related fruit borers, the invasive Cydia pomonella and the native Grapholita molesta , which exhibit interspecific competition, in response to quercetin stress—a flavonoid present in their host fruits. C. pomonella requires more generations to adapt 70 μg/g quercetin, indicating differential tolerance to flavonoid toxicity. Metabolic profiling shows G. molesta primarily detoxifies quercetin mainly via synergistic cytochrome P450 (P450) and transmembrane transport, whereas C. pomonella shifts from transport to a predominant role of oxidation over time. Given that C. pomonella was not pre-adapted to high quercetin stress in its native habitat, its constitutive defense system was likely suppressed under quercetin exposure. This species compensated by upregulating ATP-binding cassette (ABC) transporter-mediated excretion, as evidenced by HPLC quantification which indicated that the fecal quercetin content exceeded 50 % of the administered dose. Correlation analysis suggests that C. pomonella employs a biphasic detoxification: initial short-term dominated by ABC transporter activity, followed by a long-term reliance on P450-mediated metabolism. Conversely, G.molesta primarily utilizes a synergistic enzymatic detoxification system involving multiple enzymes. These findings elucidate divergent adaptive mechanisms to quercetin stress in the two fruit borers and enhance understanding of herbivorous insect-plant co-evolution and the interspecific interactions between invasive and native pest species.

Structural insights into Cydia pomonella pheromone binding protein 2 mediated prediction of potentially active semiochemicals

Tian Zhen,Liu Jiyuan,Zhang Yalin

Given the advantages of behavioral disruption application in pest control and the damage of Cydia pomonella , due progresses have not been made in searching active semiochemicals for codling moth. In this research, 31 candidate semiochemicals were ranked for their binding potential to Cydia pomonella pheromone binding protein 2 (CpomPBP2) by simulated docking and this sorted result was confirmed by competitive binding assay. This high predicting accuracy of virtual screening led to the construction of a rapid and viable method for semiochemicals searching. By reference to binding mode analyses, hydrogen bond and hydrophobic interaction were suggested to be two key factors in determining ligand affinity, so is the length of molecule chain. So it is concluded that semiochemicals of appropriate chain length with hydroxyl group or carbonyl group at one head tended to be favored by CpomPBP2. Residues involved in binding with each ligand were pointed out as well, which were verified by computational alanine scanning mutagenesis. Progress made in the present study helps establish an efficient method for predicting potentially active compounds and prepares for the application of high-throughput virtual screening in searching semiochemicals by taking insights into binding mode analyses.

Overexpression of CpGPCRs contributing to lambda-cyhalothrin resistance in Cydia pomonella

Yu-Xi Liu,Chao Hu,Zi-Nan Xia,Yan Wang,Yu-Ting Li,Ping Gao,Yun-Tong Lv,Jia Li,Xue-Qing Yang

G protein-coupled receptors (GPCRs) constitute a diverse and crucial family of membrane receptors, regulating a wide array of physiological processes. Although the involvement of GPCR signaling pathways in modulating key genes associated with insecticide resistance has been documented in various insect species, the molecular mechanisms underlying GPCR-mediated resistance in Cydia pomonella remain largely unknown. To elucidate the molecular basis of lambda-cyhalothrin (LCT) resistance in C. pomonella , we performed comparative transcriptome sequencing on whole-body, head, and midgut tissues of larvae from a susceptible population (SS) and a laboratory-selected LCT-resistant population (LCR) derived from SS. Five GPCR candidates ( CpGPCRs ) exhibiting overexpression in LCR were identified, including two members of Family A ( CpGPCR4 and CpGPCR55 ), two of Family B ( CpGPCR68 and CpGPCR76 ), and one of Family F ( CpGPCR94 ). Spatiotemporal expression profiling revealed that these genes were predominantly expressed during the first-instar larval and pupal-adult stages, with notably high expression levels in the larval heads. Notably, all genes exhibited significantly elevated expression in the midgut of LCR larvae compared to SS larvae. RNA interference (RNAi)-mediated knockdown of CpGPCR4 , CpGPCR55 , and CpGPCR68 increased larval susceptibility to LCT, while knockdown of CpGPCR4 and CpGPCR55 significantly reduced larval survival rates. Furthermore, larvae treated with dsGPCR55 exhibited molting defects and a decline in 20-hydroxyecdysone (20E) titers. These findings demonstrate that CpGPCRs play essential roles in the survival and development of C. pomonella and are critically involved in mediating resistance to LCT. This study enhances our understanding of the molecular mechanisms driving insecticide resistance and identifies potential targets for the development of novel pest management strategies against C. pomonella .
