Ectropis obliqua-Induced Secondary Metabolites Are Regulated by Methyl Jasmonate in a Threshold-Dependent Manner

Yongchen Yu,Xiaona Qian,Xiwang Li,Zhichao Chai,Dejiang Ni,Xiaoling Sun

The jasmonic acid (JA) signaling pathway has been demonstrated to play a crucial role in plant defense against herbivorous insects. However, the relationship betweenEctropis obliqua-induced defensive metabolites and the JA signaling pathway in tea plants remains poorly understood. In this study, we investigated seven key special metabolites, includingp-coumaroylputrescine, feruloylputrescine, prunin, naringenin, and three monolignols, to address this knowledge gap. Epicatechin was selected as a positive control based on its well-documented regulation through the JA signaling pathway. Notably, the content of all selected compounds was significantly increased byE. obliquainfestation. Furthermore, exogenous application of high-dose methyl jasmonate (MeJA) induced the accumulation of six of the eight compounds, excludingp-coumaryl alcohol and sinapyl alcohol, whereas low-dose MeJA failed to elicit their accumulation. To confirm the results, we screened two bioactive molecules, D-allose and L-theanine, which significantly increased the endogenous JA levels at low concentrations. Interestingly, neither D-allose nor L-theanine triggered the biosynthesis of these defensive compounds. Additionally, D-allose-treated tea leaves had no significant effect on the performance ofE. obliqualarvae. These findings demonstrate that the metabolic accumulation induced byE. obliquais mediated through a high-threshold JA signaling cascade. This study provides novel insights into the relationship between plant resistance and JA signaling pathway, advancing our understanding of special metabolites mediated plant-insect interactions.

Molecular and Biochemical Characterization of Jasmonic Acid Carboxyl Methyltransferase Involved in Aroma Compound Production of Methyl Jasmonate during Black Tea Processing

Jing Zhang,Huiyan Jia,Biying Zhu,Junyao Li,Tianyuan Yang,Zheng-Zhu Zhang,Wei-Wei Deng

Methyl jasmonate (MeJA), a volatile organic compound, is a principal flowery aromatic compound in tea. During the processing of black tea, MeJA is produced by jasmonic acid carboxyl methyltransferase (JMT) of the jasmonic acid (JA) substrate, forming a specific floral fragrance. CsJMT was cloned from tea leaves; the three-dimensional structure of CsJMT was predicted. Enzyme activity was identified, and protein purification was investigated. Site-directed deletions revealed that N-10, S-22, and Q-25 residues in the beginning amino acids played a key functional role in enzyme activity. The expression patterns of CsJMT in tea organs differed; the highest expression of CsJMT was observed in the fermentation process of black tea. These results aid in further understanding the synthesis of MeJA during black tea processing, which is crucial for improving black tea quality using specific fragrances and could be applied to the aromatic compound regulation and tea breeding improvement in further studies.

GLR36T807I Mutation of Casuarina equisetifoli Is Associated With a Decreased JA Response to Insect Feeding by Lymantria xylina

Huan Chang,Chengli Li,Tengfei Zhu,Shouping Cai,Jie Chen,Fangfang Zhan,Liqiong Zeng,Yu Fang,Gongfu Ye,Jian Li,Jun Su

Lymantria xylina is the most important defoliator, damaging the effective coastal windbreak tree species Casuarina equisetifolia . However, the underlying genetic mechanisms through which C . equisetifolia responds to L . xylina attacks remain unknown. Here, we compared the transcriptional, phytohormone and metabolic differences between susceptible (S) and resistant (R) C . equisetifolia cultivars in response to L . xylina feeding. The main L . xylina -induced resistance in C . equisetifolia was a jasmonate (JA) response and JA synthesis was highly induced by L . xylina feeding at both the transcriptional and metabolic levels, thus promoting flavonoid accumulation. The JA response was highly activated by L . xylina feeding on the R but not in the S cultivar, although the JA signalling pathway was intact in both cultivars. We found a single amino acid mutation in the homologues of glutamate receptor-like protein 3.6 (CeGLR3.6 T807I ) in the S cultivar. Compared with the GLR3.6 homologues in the R cultivar, phosphorylation of CeGLR3.6 T807I was not induced by insect feeding, leading to a decreased JA response in the S cultivar. Collectively, this study provides new insights into the function of CeGLR3.6 in regulating the JA response of C . equisetifolia to L . xylina feeding.

GhPLP2 Positively Regulates Cotton Resistance to Verticillium Wilt by Modulating Fatty Acid Accumulation and Jasmonic Acid Signaling Pathway

Zhu Yutao,Hu Xiaoqian,Wang Ping,Gao Linying,Pei Yakun,Ge Zhaoyue,Ge Xiaoyang,Li Fuguang,Hou Yuxia

Patatin-like proteins (PLPs) have non-specific lipid acyl hydrolysis (LAH) activity, which can hydrolyze membrane lipids into fatty acids and lysophospholipids. The vital role of PLPs in plant growth and abiotic stress has been well documented. However, the function of PLPs in plant defense responses against pathogens is still poorly understood. Here, we isolated and identified a novel cotton (Gossypium hirsutum) PLP gene GhPLP2. The expression of GhPLP2 was induced upon treatment with Verticillium dahliae, the signaling molecules jasmonic acid (JA) and ethylene (ETH) in cotton plants. Subcellular localization revealed that GhPLP2 was localized to the plasma membrane. GhPLP2-silenced cotton plants were more susceptible to infection by V. dahliae, while the overexpression of GhPLP2 in Arabidopsis enhanced its resistance to V. dahliae, which was apparent as mild symptoms, and a decrease in the disease index and fungal biomass. The hypersensitive response, deposition of callose, and H2O2 accumulation triggered by V. dahliae elicitor were reduced in GhPLP2-silenced cotton plants. The overexpression of GhPLP2 in Arabidopsis resulted in the accumulation of linoleic acid (LA, 18:2) and α-linolenic acid (ALA, 18:3) and facilitated the biosynthesis of JA and JA-mediated defensive responses. GhPLP2 silencing in cotton plants consistently reduced the accumulation of linoleic acid (LA, 18:2) and α-linolenic acid (ALA, 18:3) and suppressed the biosynthesis of JA and the defensive responses mediated by JA. These results indicate that GhPLP2 is involved in the resistance of cotton to V. dahliae by maintaining fatty acid metabolism pools for JA biosynthesis and activating the JA signaling pathway.

Tea Aphid-Induced β-Elemene Biosynthesis by CsELE Enhances JA-Dependent Herbivore Resistance in Tea Plants

Lanxin Luo,Ting Gao,Yanni Deng,Mengyao Chai,Bo Li,Hao Ni,Kai Wang,Mengting Zhang,Yaojia Liu,Hao Jiang,Chuankui Song,Tingting Jing

Herbivore-induced plant volatiles play a crucial role in enhancing plant resistance and adaptability. However, the key volatiles triggering this response and their activation mechanisms remain largely unknown. Although tea aphids ( Toxoptera aurantii ) are common tea plantation pests, the underlying biochemical pathways by which aphid-induced volatiles in Camellia sinensis contribute to aphid resistance are not well understood. In this study, we measured jasmonic acid (JA) accumulation induced by aphid feeding using LC-MS and DESI-IMS analysis. We also identified a diverse array of aphid-induced volatiles through GC-MS. Notably, tea aphids stimulated β-Elemene accumulation, enhancing ‘Shuchazao’ resistance to tea aphids. We also evaluated the insecticidal activity of β-Elemene through contact toxicity and repellent bioassays. For the first time, we identified aphid-induced CsELE as a terpene synthase enzyme catalyzing the final step in β-Elemene biosynthesis from farnesyl pyrophosphate. Transient overexpression and gene silencing of CsELE affected β-Elemene accumulation, thereby influencing tea plant resistance to aphids. Furthermore, Dual-Luciferase and Y1H assays revealed that MYC2a, a core transcription factor in the JA signaling pathway, positively regulates CsELE expression. Overall, this study advances our understanding of the transcriptional regulation of tea volatiles in response to pest attacks, providing new insights into the ecological significance of terpene volatiles under biotic stress.

Ethanol induced jasmonate pathway promotes astaxanthin hyperaccumulation in Haematococcus pluvialis

Yu-Hong Liu,Adili Alimujiang,Xiang Wang,Shan-Wei Luo,Srinivasan Balamurugan,Wei-Dong Yang,Jie-Sheng Liu,Lin Zhang,Hong-Ye Li

Haematococcus pluvialis is a main biological resource for the antioxidant astaxanthin production, however, potential modulators and molecular mechanisms underpinning astaxanthin accumulation remain largely obscured. We discovered that provision of ethanol (0.4%) significantly triggered the cellular astaxanthin content up to 3.85% on the 4th day of treatment. Amongst, 95% of the accumulated astaxanthin was esterified, particularly enriched with monoesters. Ultrastructural analysis revealed that ethanol altered cell wall structure and physiological properties. Antioxidant analyses revealed that astaxanthin accumulation offset the ethanol induced oxidative stress. Ethanol treatment reduced carbohydrates while increased lipids and jasmonic acid production. Transcriptomic analysis uncovered that ethanol orchestrated the expression of crucial genes involved in carotenogenesis, e.g. PSY, BKT and CRTR-b were significantly upregulated. Moreover, methyl jasmonic acid synthesis was induced and played a major role in regulating the carotenogenic genes. The findings uncovered the novel viewpoint in the intricate transcriptional regulatory mechanisms of astaxanthin biosynthesis.

Methyl jasmonate induction of tanshinone biosynthesis in Salvia miltiorrhiza hairy roots is mediated by JASMONATE ZIM-DOMAIN repressor proteins

Shi Min,Zhou Wei,Zhang Jianlin,Huang Shengxiong,Wang Huizhong,Kai Guoyin

Jasmonic acid (JA) is an important plant hormone involved in regulation of many aspects of plant growth and development including secondary metabolism and JASMONATE ZIM-DOMAIN (JAZ) proteins are key components in JA signal processes. In this study, two new JAZ genes named SmJAZ3 and SmJAZ9 were cloned from S. miltiorrhiza hairy roots and characterized. Expression profiles under methyl jasmonate (MJ) treatment revealed that SmJAZ3 and SmJAZ9 were both MJ-responsive. Subcellular localization assay showed that SmJAZ3 was located in nucleus while SmJAZ9 was preferentially in nucleus. Expression of SmJAZ3 and SmJAZ9 in S. miltiorrhiza hairy roots differently affected the production of tanshinone. Over-expression of SmJAZ3 or SmJAZ9 in hairy roots produced lower level of tanshinone compared with the control, tanshinone production was as low as 0.077 mg/g DW in line SmJAZ3-3 and 0.266 mg/g DW in line SmJAZ9-22 . Whereas, down-regulation of SmJAZs enhanced tanshione production, the content of tanshinone increased to 2.48 fold in anti - SmJAZ3-3 line and 1.35-fold in anti-SmJAZ9-23 line. Our work indicated that SmJAZ3 and SmJAZ9 are involved in regulation of tanshinone biosynthesis and act as repressive transcriptional regulators in the JA signaling pathway, which paves the way to further dissect molecular mechanism in details in the future.

Effects of Secondary Metabolites of Rice on Brown Planthopper and Its Symbionts

Ziyuan Deng,Chengling Lai,Jun Zhang,Fan Sun,Danting Li,Peiying Hao,Xuping Shentu,Kun Pang,Xiaoping Yu

The brown planthopperNilaparvata lugens(Stål) (BPH) is a main rice pest in China and many other Asian countries. In the control of BPH, the application of insect-resistant rice has proven to be quite effective. Secondary metabolites are essential weapons in plants’ defense against phytophagous insects. Studies have found that differences in the content of secondary metabolites play a crucial role in determining whether rice exhibits resistance or susceptibility to BPH. Simultaneously, symbionts are essential to the BPH. Nevertheless, there is limited research on the impact of secondary metabolites on the symbionts within BPH. Therefore, investigating the influence of secondary metabolites on both BPH and their symbionts is significant for the control of BPH. In this experiment, newly emerged female adults of BPH were fed artificial diets containing 10 different secondary metabolites. The results indicated that methyl jasmonate had inhibitory effects on the survival rate, weight gain, and reproductive capacity of BPH. Using qPCR methods, it was discovered that the number of symbiotic fungi (Ascomycetessymbionts) within BPH significantly decreased under methyl jasmonate stress. In conclusion, this experiment has preliminarily revealed the inhibitory effects of methyl jasmonate on BPH and its symbionts, demonstrating its potential for controlling BPH.Keywords:brown planthopper;secondary metabolites;symbionts;methyl jasmonate

Synergistic interaction between brassinosteroid and jasmonate pathways in rice response to cadmium toxicity

Zaoli Zhang,Tingting Zhang,Long Lu,Shunjiao Qiu,Zecong Huang,Yuan Wang,Xinyu Chen,Long Li,Yanyan Sun,Rongjun Zhang,Pinghua Chen,Yuanyuan Song,Rensen Zeng

Brassinosteroids (BRs) and jasmonic acid (JA) are known to be involved in regulating plant responses to cadmium (Cd) stress. However, their specific roles and interaction in this process remain unclear. In this study, we discovered that exogenous BR alleviated Cd-mediated growth inhibition of rice seedlings. Enhanced Cd tolerance was also observed in m107 , a BR-overproduction mutant. Phenotypic analysis of genetic materials involved in BR signaling confirmed the positive role of BR in regulating rice response to Cd toxicity. OsDLT, a key component in the BR signaling pathway, was found to be crucial for BR-mediated Cd tolerance. Further analysis demonstrated that activation of the BR pathway reduced the accumulation of Cd and reactive oxygen species (ROS) by modulating the expression of genes associated with Cd transport and ROS scavenging. Interestingly, transcriptome analysis indicated that the JA pathway was enriched in OsDLT-regulated differently expressed genes (DEGs). Gene expression and hormone assays showed that BR promoted the expression of JA pathway genes and JA levels in plants. Moreover, BR-induced tolerance was compromised in the JA signaling-deficient mutant osmyc2 , suggesting that BR-mediated Cd resistance depends on the activation of the JA signaling pathway. Overall, our study revealed the synergistic interaction between BR and JA pathways in rice response to Cd stress, providing insights into the complex hormonal interplay in plant tolerance to heavy metals.

Differential induction of JA/SA determines plant defense against successive leaf-chewing and phloem-feeding insects

Liu Yu-Xiao,Han Wen-Hao,Wang Jun-Xia,Zhang Feng-Bin,Ji Shun-Xia,Zhong Yu-Wei,Liu Shu-Sheng,Wang Xiao-Wei

Plants face an array of insect herbivores and have evolved complex defense approaches against various insect feeding strategies. However, little is known about how plants respond to successive attacks by herbivores with different feeding modes and coordinate their diverse defense mechanisms. In this study, we unveil that inducible jasmonic acid (JA) accumulation in response to leaf-chewing insects augments plant resistance and repellence to sequential leaf-chewing insect (caterpillar) and phloem-feeding insect (whitefly) infestations. Conversely, constitutive and whitefly inducible salicylic acid (SA) accumulation exclusively bolsters defense against later-stage whitefly invasion. Through assessments of herbivore performance and preferences on wild-type, JA-deficient, and SA-deficient plants, we show that JA/SA levels regulate plant resistance to both initial and sequential herbivores. Notably, JA or SA accumulation due to caterpillar or whitefly attacks does not substantially affect constitutive levels of the other compound, despite their antagonistic crosstalk. Furthermore, exogenous JA application in tobacco elicits efficient defense against successive caterpillar and whitefly assaults, surpassing SA's efficacy, albeit with associated growth penalties. Our discoveries demonstrate that plants can tailor their defense strategies against initial and sequential insects with different feeding modes. This customized defense is facilitated by JA/SA responses and their intricate cross-talk while taking account of the growth-defense trade-off.

In situ fluorescence labelling of jasmonic acid binding sites in plant tissues with cadmium-free quantum dots

Qiumei Liao,Ying Yu,Yujuan Cao,Bixia Lin,Jingjing Wei

The fluorescence labelling of plant hormone binding sites is an important analytical technique in research on the molecular mechanisms of plant hormone activities. The authors synthesised a jasmonic acid (JA)-conjugated ZnS:Mn quantum dot (QD) probe, with a cubic structure and average hydrodynamic sizes of about 17.0 nm. The maximum fluorescence emission of the probe was recorded at about 585 nm. The probe was used for fluorescence labelling of JA binding sites in mung bean seedling tissues. Analysis revealed that the probe exhibited high selectivity to JA binding sites and good performance in eliminating interference from background fluorescence in plant tissues. In addition, the probe did not exhibit any apparent biotoxicity, and is much more suitable than probes constructed from CdTe QDs for the analysis of biological samples.

Electrochemically deposited conductive composite sorbent for highly efficient online solid-phase microextraction of jasmonates in plant samples

Xu Ling,Zilin Chen

Conductive composite films composed of poly (3, 4-ethylenedioxythiophene) (PEDOT) and sulfonated graphene were electrochemically deposited on carbon fiber bundle in aqueous solutions for solid phase microextraction (SPME). During the electro-polymerization process, negatively charged sulfonated graphene were doped in PEDOT layer and uniformly dispersed in the composite. The modified carbon fiber bundle worked as sorbent was then successfully applied to online SPME-HPLC-UV analysis of jasmonates in wintersweet flowers. High extraction efficiency (over 420-fold), low limits of detection (0.1   ng   mL −1 for jasmonic acid and 0.01   ng   mL −1 for methyl jasmonate) and wide linear ranges (0.5–5000 and 0.05–5000   ng   mL −1 , R>0.9997) have been obtained. It has been demonstrated that this method can be well used for jasmonates analysis in fresh wintersweet flowers with good recoveries (92.4–98.9%). The results were confirmed by HPLC-MS method.
