ObjectivesThe current work aimed to isolate and identify new interesting small-molecule natural products from the leaves of a well-known medicinal plant Morus alba.MethodsThe ethanol extract of M. alba leaves was partitioned between EtOAc and water, and the EtOAc soluble part was fractionated by macroporous resin column chromatograph (CC) eluted with gradient EtOH. The 80% EtOH eluate was subject to silica gel, reversed phase C18, MCI gel and Sephadex LH-20 CC and finally purified by HPLC to afford four minor compounds 1–4, and the racemic 1 and 2 were further separated by chiral columns. Structural elucidation of these isolates was achieved via spectroscopic means especially HR-ESIMS and NMR, with the absolute configurations of new compounds assigned by ECD analyses. The cytotoxic screening was performed by MTT assay.ResultsA pair of new quinone enantiomers (1a/1b), a pair of new stilbenoid enantiomers (2a/2b), a known prenyl stilbenoid (3) and a known dihydrochalcone (4) were isolated and structurally characterized from the leaves of M. alba. They did not show significant cytotoxicity against MDA-MB231, A549, Hela and 4T1 tumor cell lines.ConclusionsThe M. alba leaves represent a rich source of new natural products, and the currently obtained constituents did not exert obvious growth inhibitory activity against the tested tumor cell lines. The bioactivities of the new isolates deserve further exploration in the future.

Two new stilbenoids, cajanstilbenoid C (1) and cajanstilbenoid D (2), together with eight other known stilbenoids (3-10) and seventeen known flavonoids (11-27), were isolated from the petroleum ether and ethyl acetate portions of the 95% ethanol extract of leaves ofCajanus cajan(L.) Millsp. The planar structures of the new compounds were elucidated by NMR and high-resolution mass spectrometry, and their absolute configurations were determined by comparison of their experimental and calculated electronic circular dichroism (ECD) values. All the compounds were assayed for their inhibitory activities against yeastα-glucosidase. The results demonstrated that compounds3,8-9,11,13,19-21,and24-26had strong inhibitory activities againstα-glucosidase, with compound11(IC50= 0.87 ± 0.05 μM) exhibiting the strongest activity. The structure–activity relationships were preliminarily summarized. Moreover, enzyme kinetics showed that compound8was a noncompetitive inhibitor, compounds11,24-26were anticompetitive, and compounds9and13were mixed-competitive.Keywords:Cajanus cajan;stilbenoid;flavonoid;α-glucosidase inhibitory activity

Background Paeonia suffruticosa is an important traditional Chinese herb used to treat osteoarthritis (OA) and oligostilbenes are the main active ingredient of the seeds of P. suffruticosa. The monomer trans-resveratrol of this species was demonstrated to have chondroprotective effects as a lead compound for the treatment of osteoarthritis, but it has not been applied due to its low efficacy. Methods Oligostilbenes were isolated by chromatography and were identified by NMR and HPLC. A rabbit osteoarthritis chondrocyte model was induced by interleukin-1β and was treated with individual drugs to systematically evaluate their effects. Cell Counting Kit 8 was used to test their effects on cell viability, calculate EC50 and plot a dose–response curve.Their effects on apoptosis were analyzed by Annexin V and PI staining, and the expression of chondrocyte-specific genes COL2A1, MMP13 and SOX9 was evaluated by real-time PCR.Results Paeonia suffruticosa seed extract could promote the cell viability of rabbit OA chondrocytes at low concentration and then ten oligostilbenes were isolated from it. Trans-oligostilbenes were better than their cis-forms, trimers and dimers were better than monomers for promoting the cell viability of rabbit osteoarthritis chondrocytes. None of the oligostilbenes was more effective than seed extract at the appropriate concentration; 1 μM oligostilbenes all showed various anti-apoptotic effects. Trans-gnetin H showed the best effect on proliferation and inhibition of MMP13 expression on OA chondrocytes, while trans-viniferin was most effective in promoting the expression of COL2A1 and SOX9.Conclusion sTen oligostilbenes from P. suffruticosa seed all have certain protective effects on OA chondrocytes at low concentration. The trans-viniferin and some trimers have the potential to be further developed for the treatment of osteoarthritis because they were more effective than resveratrol and diacerein. The synergistic effect that may exist between oligostilbenes also warrants further research.

Dried roots and rhizomes of rhubarb are widely used both as a vegetable and a medicinal product around the world. To date, most studies on the drying process of rhubarb have focused primarily on comparing different drying techniques and assessing whether the content of functional compounds meets standard requirements. However, few studies have explored the dynamic changes and underlying mechanisms of these functional ingredients during drying process. In this study, HPLC-DAD-based dynamic monitoring of anthraquinones, combined with widely targeted metabolomics, was employed to investigate the impact of moisture loss on the quality of rhubarb and to elucidate potential molecular mechanisms during the drying process. The results indicated that the contents of total and free anthraquinones reached their highest levels when the moisture content was 40 %, whereas other moisture levels generally led to a decrease. In contrast to anthraquinones, flavonoids, phenolic acids, and stilbenoids were identified as the most significantly changed functional components throughout the drying process. Further analysis revealed that the key metabolic pathways influencing these functional ingredients were glycolysis, TCA cycle, and pentose phosphate pathway, which are associated with respiratory metabolism. Phosphoenolpyruvate and D-erythrose-4-phosphate were the key precursors of shikimate pathway for the biosynthesis and interconversion between phenolic acids, flavonoids, stilbenoids and anthraquinones in rhubarb. This study provides a comprehensive understanding of how the drying process influences the functional components of rhubarb and offers valuable insights for determining the optimal moisture content in industrial drying processes.

Fifteen stilbenoid derivatives, including five previously undescribed ones (albaphenols A–E, 1–5) with diverse scaffolds, were obtained from the well-known agricultural economic tree Morus alba. Their structures, including absolute stereochemistries, were fully characterized by detailed interpretation of spectroscopic data and quantum chemical computational analyses of nuclear magnetic resonance (NMR) and electric circular dichroism (ECD). Albaphenol A (1) features an unprecedented rearranged carbon skeleton incorporating a novel 2-oxaspiro[bicyclo[3.2.1]octane-6,3′-furan] motif; albaphenol C (3) is likely derived from a cometabolite through an interesting intramolecular transesterification reaction; and albaphenol E (5) bears a cleavage-reconnection scaffold via a dioxane ring. All of the compounds exhibited significant inhibition against the diabetic target α-glucosidase, with low to submicromole IC50 values (0.70–8.27 μM), and the binding modes of selected molecules with the enzyme were further investigated by fluorescence quenching, kinetics, and molecular docking experiments. The antidiabetic effect of the most active and abundant mulberrofuran G (6) was further assessed in vivo in diabetic mice, revealing potent antihyperglycemic activity and comparable antidiabetic efficacy to the clinical drug acarbose.
