The fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), is a pest of many important crops globally. Effective control is challenging, with the pest exhibiting resistance to different synthetic pesticides across various groups. However, the mechanisms employed by resistant insects for overexpression of relevant detoxification genes remain unclear. The activity of detoxification enzymes was investigated in this study. Additionally, using RNA interference (RNAi), a functional analysis was completed of two P450s genes in an indoxacarb resistant population of fall armyworms. Elevated resistance levels (resistance ratio = 31.37-fold) in indoxacarb-selected populations of FAW were observed after 14 generations. The qRT-PCR showed higher expression of two cytochrome P450 genes, CYP321A7 and CYP6AE43, in this selected population compared to the control population. RNAi was applied to knock down the P450 dsCYP321A7 and dsCYP6AE43 genes in the FAW larvae. Droplet feeding of the dsRNAs (CYP321A7 and CYP6AE43) via an artificial diet significantly increased mortality rates in the indoxacarb treated population. A shorter larval developmental time of FAW was detected in all dsRNAs-fed larvae. Correspondingly, larval mass was reduced by dsRNAs in indoxacarb resistant populations of fall armyworm. Larval feeding assays demonstrate that dsRNAs targeting, specifically of CYP321A7 and CYP6AE43 enzymes, could be a beneficial technique in the management of indoxacarb resistant populations. Further study on the potential use of dsRNA and its application should be conducted in efforts to counter the development of resistance in FAW against various insecticides in the field.

Traditional pesticide formulations show poor utilization and environmental safety due to their low foliage adhesion and large auxiliaries. In this study, a novel and environment-friendly indoxacarb formulation was prepared to improve the pesticide’s utilization rate, target control characteristics and ecological security. Indoxacarb-loaded waterborne polyurethane–sodium alginate (PU/SA) nanoemulsions with film forming properties, alkaline responsive release, high effectiveness againstSpodoptera litura, and reduced acute contact toxicity for nontarget organisms were successfully prepared. The colloidal properties, swelling and release behaviors, leaf adhesion, degradation dynamics and bioactivity assay of the indoxacarb-loaded PU/SA nanoemulsions were determined. Results showed that the obtained indoxacarb-loaded microcapsule particulates were approximately 57 nm in diameter, electronegative −45.9 mV, and uniformly dispersed in the nanoemulsions. The dried latex films of PU/SA in the alkaline environment revealed better responsive swelling and release characteristics than those in acidic and neutral conditions. Compared with a commercial emulsifiable concentrate, the indoxacarb-loaded PU/SA nanoemulsions were useful for the targeted control ofS. litura, which have alkaline gut and showed reduced acute contact toxicity toHarmonia axyridia. Furthermore, the PU/SA formulation had better foliage adhesion and indicated the property of controlled-release and a persistent effect. Keywords:indoxacarb;anionic polyurethane;sodium alginate;nanoemulsions;bioactivity

Simple SummaryThe fall armyworm (FAW),Spodoptera frugiperda(J.E. Smith), is a voracious insect pest native to the Western Hemisphere, particularly in South America. The polyphagous fall armyworm feeds on more than 350 plants in such families, including Poaceae, Asteraceae, and Fabaceae. Transgenic plants that expressBacillus thuringiensisproteins (Bt plants) and synthetic insecticides are the main tactics to controlSpodoptera frugiperda, although widespread usage of synthetic chemicals has resulted in the emergence of resistance. We assessed cross-resistance, resistance mechanism, and fitness costs based on the life history traits ofSpodoptera frugiperda. Results indicated that after 24 generations of selection, the resistance to indoxacarb was increased by 472.67-fold as compared to the Ind-UNSEL. Significantly longer developmental time of larvae extended pupal duration, shorter adult longevity, and lower fecundity were observed in Ind-SEL as compared with the Ind-UNSEL population. Butoxide synergist increased susceptibility to indoxacarb, indicating that P450 enzymes may be involved in indoxacarb resistance. Therefore, it is crucial that we comprehend how insecticides work and how resistance develops in order to develop techniques for managing resistance. These data are valuable to understand the indoxacarb resistance mechanism and provides vital information for scientific-based guidance of pest management decisions.AbstractThe fall armyworm,Spodoptera frugiperda(J.E. Smith), is a voracious insect pest that is difficult to control due to resistance to insecticides and Bt proteins. We assessed cross-resistance, resistance mechanism, and fitness costs based on the life history traits ofS. frugiperda. We established anS. frugiperdastrain selected for resistance to indoxacarb (Ind-SEL) from a field-collected population and an unselected strain, Ind-UNSEL. Results indicated that after 24 generations of selection, the resistance to indoxacarb was increased by 472.67-fold as compared to the Ind-UNSEL. There was high cross-resistance to deltamethrin (31.23-fold) with very low or negligible cross-resistance to chlorantraniliprole, emamectin benzoate, and/or methoxyfenozide in the Ind-SEL population. Butoxide synergist increased susceptibility to indoxacarb, indicating that P450 enzymes may be involved in indoxacarb resistance. Significantly longer developmental time of larvae extended pupal duration, shorter adult longevity, and lower fecundity were observed in Ind-SEL as compared with the Ind-UNSEL population. The Net reproductive rate (R0) was the only growth parameter that differs between crosses of Ind-SEL♂ × Ind-UNSEL♀ (176 ± 46) and Ind-SEL♀ × Ind-UNSEL♂ (328 ± 57). On the other hand, all population growth parameters differ between Ind-SEL and Ind-UNSEL strains. Our work contributes to the growing body of research that demonstrates the importance of strain genetics in fitness cost experiments and helps resistance management programs make decisions. Keywords:Spodoptera frugiperda;indoxacarb selection;cross-resistance;inheritance of resistance;fitness cost

BACKGROUND Insecticide resistance continuously poses a threat to agricultural production. Chemosensory protein-mediated resistance is a new mechanism of insecticide resistance discovered in recent years. In-depth research on resistance mediated by chemosensory proteins (CSPs) provides new insight into aid insecticide resistance management. RESULTS Chemosensory protein 1 in Plutella xylostella ( PxCSP1 ) was overexpressed in the two indoxacarb-resistant field populations and PxCSP1 has a high affinity with indoxacarb. PxCSP1 was upregulated when exposed to indoxacarb and the knockdown of this gene elevated sensitivity to indoxacarb, which demonstrate that PxCSP1 is involved in the indoxacarb resistance. Considering that CSPs may confer resistance in insects via binding or sequestering, we explored the binding mechanism of indoxacarb in PxCSP1 -mediated resistance. Using molecular dynamics simulations and site-directed mutation, we found that indoxacarb forms a solid complex with PxCSP1 mainly through van der Waals interactions and electrostatic interactions. Between these, the electrostatic interaction provided by the Lys100 side chain in PxCSP1 , and especially the hydrogen bonding between the NZ atom and the O of the carbamoyl carbonyl group of indoxacarb, are the key factors for the high affinity of PxCSP1 to indoxacarb. CONCLUSIONS The overexpression of PxCPS1 and its high affinity to indoxacarb is partially responsible for indoxacarb resistance in P. xylostella . Modification of indoxacarb's carbamoyl group has the potential to alleviate indoxacarb resistance in P. xylostella . These findings will contribute to solving chemosensory protein-mediated indoxacarb resistance and provide a better understanding of the insecticide resistance mechanism. © 2023 Society of Chemical Industry.

The invasion area of red imported fire ants (RIFAs) in China continues to expand due to factors such as ecological changes and human activities, and the resulting damage is becoming increasingly severe. The development of smart nanopesticides that respond to environmental stimuli response with precisely controlled release has great potential in improving insecticidal effects. In this work, a smart pH and α-amylase dual stimuli-responsive pesticide delivery system was constructed by combining mono-(6-amino-6-deoxy)-β-cyclodextrin (β-CD-NH 2 ) with zeolitic imidazole skeleton-90 (ZIF-90) nanoparticles ( [email protected] ) loaded with indoxacarb. The average particle size of the prepared [email protected] was 249 nm, and the loading efficiency was 18.43%. The β-CD-NH 2 coating on the surface of nanoparticles reduced the photodegradation of indoxacarb and prevented the premature release of the active ingredient. In vitro release kinetics showed that [email protected] rapidly released IDC under acidic and amylase conditions, consistent with the oral and intestinal environment of RIFAs. The results of bioactivity tests showed that [email protected] enhanced the toxicity of IDC toward RIFAs by increasing feeding and damaging intestinal cells. The mortality rate of the [email protected] group was significantly higher than that of the IDC EC-treated group at 72 h. The nontargeted metabolomic results showed that [email protected] affected the metabolism of RIFAs by interfering with the metabolic pathways of six amino acids and one amino acid transport carrier, whereas IDC EC interfered with only two of these amino acid metabolic pathways. Overall, this work provides insight into the effective control of RIFA damage for sustainable pest management.

Control release of pesticides based on target plant characteristics is highly desirable for improving the effective utilization rate and reducing non-target organism risks. Here, the controlled release formulations (Cu-CS@IN@MIL-100 (Fe)) were successfully prepared by employing MIL-100 (Fe) as the carrier material, indoxacarb (IN) as the model pesticide, and chitosan (CS) as a coating to chelate copper ions by chemical crosslinking. The prepared Cu-CS@IN@MIL-100 (Fe) had an acceptable loading content (36.25 %). The introduction of CS and copper ions endowed it with sustained release properties under weak acidity conditions like cotton leaves. The laboratory bioassay experiments showed that Cu-CS@IN@MIL-100 (Fe) had practical good insecticidal activity (LC 50 = 4.936 mg/L) in controlling Helicoverpa armigera larvae. For long-term control efficacy experiments, after 14 days of application, the Cu-CS@IN@MIL-100 (Fe) treatment (73 %) mortality of Helicoverpa armigera larvae was significantly higher than the 15 % indoxacarb suspension concentrates (SC) treatment (27 %). Moreover, Cu-CS@IN@MIL-100 (Fe) had no adverse effect on cottonseed germination and the acute toxicity to Chrysopa pallens larvae was significantly lower than IN SC. Therefore, MIL-100 (Fe) could be used as an ecological safety carrier for pesticide delivery to improve IN effective utilization and reduce non-target organism risks.

Concerns over health risks associated with mixed pesticide residues have led to a demand for rapid analytical methods. We present a portable surface-enhanced Raman spectroscopy (SERS) device specifically designed for real-time monitoring of indoxacarb (INDX) and chlorfenapyr (CFP) in vegetable juice. This point-of-use sensor integrates silver nanoparticles with a metal-organic framework (Ag/MOF-808(Zr)) and employs a 19-well quartz plate alongside a portable Raman spectroscopy instrument, facilitating simultaneous adsorption, separation, and detection. Detection limits for INDX and CFP in ginger and spinach juices were 0.26–0.76 ppb. Notably, results closely matched those from liquid chromatography, and in-situ detection was achieved within 2 min without pre-treatment, significantly enhancing efficiency over conventional chromatographic methods. This innovative approach demonstrates substantial potential for detecting pesticide mixtures in non-laboratory settings, presenting a valuable option for point-of-use applications.

Spodoptera litura (Fabricius) is a widely distributed worldwide insect pest , particularly of vegetable crops . To determine the sensitivity of S. litura to insecticides , the toxicities of beta-cypermethrin, chlorpyrifos , emamectin benzoate, methoxyfenozide, and indoxacarb were tested from 2014 to 2016 using a diet incorporation assay on populations of S. litura collected from different districts in Sichuan Province and a susceptible strain (Lab-HN). Compared to the Lab-HN strain, field populations showed high levels of resistance to beta-cypermethrin (267.3- to 1789.2-fold). Field populations exhibited low or moderate resistance to chlorpyrifos in 2014 (8.9- to 24.4-fold) and high levels in 2016 (440.5- to 950.5-fold). All populations collected in 2014 and 2016 were susceptible or had a low or moderate level of resistance to indoxacarb (2.2- to 31.0-fold). No field populations exhibited resistance to emamectin benzoate or methoxyfenozide (0.7- to 1.5-fold and 0.9- to 1.4- fold) in 2014, whereas the resistance in 2016 increased to 29.8- to 55.2-fold and 38.1- to 59.0- fold, respectively. A synergist experiment indicated that the Lab-HN strain and field-collected population from Shuangliu in 2016 (SL16 population) to beta-cypermethrin may be related to cytochrome P450 monooxygenases (P450s), with the highest synergism ratios (SR) of piperonyl butoxide (PBO) of 34.4- and 73.3-fold, but not to carboxylesterase (CarE) or glutathione-S- transferase (GST). We detected the activities of detoxification enzymes and found that the ethoxycoumarin O -deethylase activities were also the strongest in the Lab-HN strain and the SL16 population with the highest SR values of 2.6- and 4.1-fold, respectively. Non-denaturing polyacrylamide gel electrophoresis (native PAGE) indicated that the inhibition of in vivo P450 activity caused by PBO was due to the binding of the synergist to the E5 or E6 isozyme , which were absent in the Lab-HN strain.

Pesticides are chemicals widely utilized in various industries, particularly agriculture, to manage pests and enhance crop yields. However, the excessive use of pesticides, such as phoxim, poses threats to the environment, food safety, and human health. Thus, it is crucial to use pesticides judiciously, limiting their application to what is necessary. Fluorescent probes have garnered attention for their cost-effectiveness, straightforward preparation, and high sensitivity, positioning them as promising candidates for detecting phoxim. In this context, orange ratiometric fluorescent carbon dots were synthesized using rhodamine B and NaOH as precursors through a hydrothermal method. The synthesized Rh B Na CDs were then purified via dialysis, thin-layer chromatography, and chromatographic columns to develop an accurate and precise ratiometric fluorescent probe. This nanoprobe demonstrated excellent linear ranges for detecting phoxim levels in various food samples. It exhibited a low detection limit of 4.486 nM for phoxim concentrations ranging from 0 to 25 μM and can accurately detect concentrations between 25 and 250 μM. Moreover, it proved sensitive in identifying phoxim content in samples of red grapes, green grapes, tomatoes, and pitayas.

A colorimetric sensor for the pesticide pymetrozine (PYM) based on analyte-induced aggregation of melamine-modified gold nanoparticles (M-Au NPs) has been developed with high selectivity and sensitivity with a detection limit of 80 nM by the naked eye and 10 nM by UV-Vis spectroscopy. This colorimetric sensing design exhibits lower detection limits, compared with limits of 46–148 nM for existing instrumental methods. The new aggregation-induced nanoparticle sensor is applicable for rapid colorimetric detection of PYM in real water environments (tap water and lake water) and food samples (green tea and apple juice). The technical approach for the colorimetric sensor is easier and simpler than detection of PYM by, for example, HPLC whilst giving equivalent results within the error of the measurements.

Paraquat (PQ) is a kind of herbicide, which is highly toxic to human body. In this work, we proposed a fast and simple detection method for PQ in aqueous samples based on electrostatic interactions between modified gold nanoparticles (Au NPs) and PQ. Au NPs were first modified with sodium 3-mercapto-1-propanesulfonate (Au NPs-3MPS). 3MPS with negative charge can interact with nitrogen atoms of PQ with positive charge via electrostatic force. As a result, Au NPs-3MPS aggregated together. Accordingly, the color of Au NPs-3MPS colloid changed from red to blue-gray, and the UV–vis absorption spectrum changed accordingly. The detection limit of PQ is 0.1 mg/L by naked eyes, and 0.001 mg/L by UV–vis spectroscopy. Moreover, this detection method has good specific selectivity and anti-interference performance, and it has been successfully used for detecting PQ in real environmental samples and fruit juice samples.

In this work, a barley husk biomaterial was successfully carbonized by hydrothermal method. The carbon had a high specific surface area and good stability. It was coated onto a stainless steel wire through sol-gel technique to prepare a solid-phase microextraction fiber for the extraction of trace levels of twelve pesticides (tsumacide, fenobucarb, indoxacarb, diethofencarb, thimet, terbufos, malathion, thiamethoxam, imidacloprid, buprofezin, acetamiprid, thiamethoxam) from vegetable samples prior to gas chromatography–mass spectrometric (GC–MS) detection. The main experimental parameters that could influence the extraction efficiency such as extraction time, extraction temperature, sample pH, sample salinity, stirring rate, desorption temperature and desorption time, were investigated. Under the optimized conditions, the linearity was observed in the range of 0.2–75.0 μg kg −1 for tomato samples, and 0.3–60.0 μg kg −1 for cucumber samples, with the correlation coefficients ( r ) ranging from 0.9959 to 0.9983. The limits of detection of the method were 0.01–0.05 μg kg −1 for tomato samples, and 0.03–0.10 μg kg −1 for cucumber samples. The recoveries of the analytes for the method from spiked samples were in the range of 76%–104%, and the precision, expressed as the relative standard deviations, was less than 12%.
