Antidepressant and immunosuppressive activities of two polysaccharides from Poria cocos (Schw.) Wolf

Wuxia Zhang,Lu Chen,Peng Li,Jinzhong Zhao,Jinyou Duan

Pursuit of novel effective antidepressants is an urgent task. Poria cocos ( Schw .) Wolf (PCW) is a traditional herb with antidepressant effects . Polysaccharides designated PCWPW and PCWPS were purified from PCW by DEAE-52 cellulose chromatography. Their structures were characterized using physicochemical and spectral methods. Chemical analysis indicated that PCWPW (37,154 Da) and PCWPS (186,209 Da) were mainly composed of mannose , glucose, galactose and fucose . Their antidepressant activities were evaluated by tail suspension test (TST), forced swimming test (FST) with chronic unpredictable mild stress (CUMS) model mice. To investigate the antidepressant mechanism, the neuroprotective and immunomodulation effects were assessed by MTT method. Results showed that PCWPW and PCWPS possess obvious antidepressant effects and can suppress ConA-stimulated T cell proliferation in a dose-dependent manner. In addition, PCWPS could protect the PC12 cells from H 2 O 2 -induced damage and suppress LPS-induced B cell proliferation . These findings implied that PCWPW and PCWPS might be a candidate for developing antidepressant or immunosuppressive agents in food and pharmaceutical industries.

Poria cocos-derived carbon dots for cellular imaging, free radical scavenging and pH sensing

Song Yuan,Yuanping Luo,Yuxiang Jiang,Wenjun Xu,Beijun Cheng,Zhili Peng

Green, economical preparation of carbon dots (CDs) from traditional Chinese herbal medicine Poria cocos via a one-step hydrothermal treatment was reported. The CDs exhibited excellent aqueous dispersibility , superior photoluminescence (PL) property and low cytotoxicity. Taking advantage of these properties, CDs were demonstrated as excellent multicolor bioimaging agents for the cellular imaging of MDA-MB-435S cells, with main distributions in the cell membranes and cytoplasm. Moreover, the CDs also exhibited good radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH) thanks to their inheritance of characteristic functionalities from Poria cocos. Lastly, our study also shown that the PL of CDs was reversibly sensitive to pH in the range of 4.0 and 9.0, and a highly sensitive pH-sensing assay was developed exploiting this phenomenon. In summary, the study opened up new development paths for the modernization and vitalization of traditional Chinese medicine in biomedical research.

Green fabrication of antimicrobial film for food preservation by polysaccharides from Poria Cocos

Mingjie Hu,Qian Liu,Lijuan Zhang,Deyu Yang,Guozhen Wang,Qi Wang,Cong Chang,Xinyan Chen,Zhangwei Yang,Guohua Zheng,Zhaohua Shi,Yan Meng

Recently, polysaccharide-based film materials have attracting widespread attentions due to their safety, biodegradability, and film-forming properties. However, the limited UV resistance, antibacterial and mechanical properties of polysaccharides severely restrict their application in food packaging. This work developed a biocomposite film (PGF@Ag) consited of a natural β -glucan from Poria Cocos (PCPA), in-situ synthesized AgNPs and glycerol, showing good antibacterial and antioxidant properties as well as biocompatibility and mechanical properties. Poria Cocos is an edible Chinese medicine, and its residue is rich in polysaccharide. In this system, the large amont of hydrogen groups on PCPA help fast reduce Ag + to Ag 0 in alkalescent solution and the triple-helix conformation of PCPA hinder the aggregation of AgNPs, leading the ultra small size of 9.0–12.3 nm and better ABTS radical scavenging rates of 93.30 ± 0.13 %. Additionally, the hydrogen bonding interactions between glycerol and polysaccharides significantly enhance the mechanical properties of the PGF@Ag films. The tensile strength is measured at 5.94 ± 0.5 MPa, while the elongation at break reaches 128.77 ± 12.45 %. Furthermore, the Young's modulus is recorded at 15.75 ± 1.91 MPa. Finally, the resulting films (PGF@Ag) exhibited excellent preservation effect on grapes, as reduced weight loss reduction of 14 %, hardness reduction of approximately 5.2 % over 12 days. This work provided a new strategy of sustainable constructing antibacterial films, showing great potentials for food preservation and packaging applications.

Monosaccharide analysis and fingerprinting identification of polysaccharides from Poria cocos and Polyporus umbellatus by HPLC combined with chemometrics methods

Jie Liu,Jing Zhou,Qian-qian Zhang,Min-hang Zhu,Mo-li Hua,Yun-hui Xu

Objective Poria cocos and Polyporus umbellatus are similar medicinal fungi in traditional Chinese medicines . A method for fingerprint analysis of monosaccharide composition of polysaccharides by HPLC combined with chemometrics methods has been developed for characterization and discrimination of them in this research. Methods The polysaccharides were extracted by decocting in water, and then completely hydrolyzed with hydrochloride. Monosaccharides in the hydrolyzates were derivatized with 1-phenyl-3-methyl-5-pyrazolone (PMP) for HPLC analysis. More than 20 batches of P. cocos and P. umbellatus from different regions were analyzed. Results The fingerprints of P. cocos showed five common characteristic peaks, which were identified by comparing with the reference substances. The five peaks corresponded to the derivatives of mannose , ribose , glucose, galactose , and fucose . At the same time, the fingerprints of P. umbellatus showed eight common characteristic peaks, of which seven were identified as the derivatives of mannose, ribose, rhamnose , glucose, galactose, xylose , and fucose. Moreover, the similarity of their fingerprints was respectively calculated by the Similarity Evaluation System for Chromatographic Fingerprint of TCM published by China Pharmacopoeia Committee (Version 2004A). And the data were further processed by hierarchical cluster analysis (HCA) and principal component analysis (PCA). The similarity evaluation and HCA indicated that there were no significant difference in P. cocos or P. umbellatus samples from different geographical regions, but PCA was performed to characterize the difference in monosaccharide constituents between P. cocos and P. umbellatus , and linear discriminant analysis (LDA) showed the overall correct classification rate was 100%. Conclusion The fingerprint analysis method of monosaccharide composition of water-soluble polysaccharides can distinguish P. cocos and P. umbellatus , and can be applied for the authentication or quality control for P. cocos and P. umbellatus.

Untargeted Metabolomics and Targeted Quantitative Analysis of Temporal and Spatial Variations in Specialized Metabolites Accumulation in Poria cocos (Schw.) Wolf (Fushen)

Yang Mei,Zhao Yujiao,Qin Yuejian,Xu Rui,Yang Zhengyang,Peng Huasheng

Poria cocos (Schw.) Wolf is a saprophytic fungus that grows around the roots of old, dead pine trees. Fushen, derived from the sclerotium of P. cocos but also containing a young host pine root, has been widely used as a medicine and food in China, Japan, Korea, Southeast Asian countries, and some European countries. However, the compound variations at the different growth periods and in the different parts of Fushen have not previously been investigated. In this study, an untargeted metabolomics approach based on ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q/TOF-MS) and targeted quantitative analysis was utilized to characterize the temporal and spatial variations in the accumulation of specialized metabolites in Fushen. There were 119 specialized metabolites tentatively identified using the UPLC-Q/TOF-MS. The nine growth periods of Fushen were divided into four groups using partial least squares discrimination analysis (PLS-DA). Four different parts of the Fushen [fulingpi (FP), the outside of baifuling (BO), the inside of baifuling (BI), and fushenmu (FM)] were clearly discriminated using a PLS-DA and orthogonal partial least squares discrimination analysis (OPLS-DA). Markers for the different growth periods and parts of Fushen were also screened. In addition, the quantitative method was successfully applied to simultaneously determine 13 major triterpenoid acids in the nine growth periods and four parts. The quantitative results indicated that the samples in January, March, and April, i.e., the late growth period, had the highest content levels for the 13 triterpenoid acids. The pachymic acid, dehydropachymic acid, and dehydrotumulosic acid contents in the FM were higher than those in other three parts in March, whereas the poricoic acid B, poricoic acid A, polyporenic acid C, dehydrotratrametenolic acid, dehydroeburicoic acid, and eburicoic acid in FP were higher beginning in October. These findings reveal characteristics in temporal and spatial distribution of specialized metabolites in Fushen and provide guidance for the identification of harvesting times and for further quality evaluations.

Imidazole derivative-based fluorescent arrays combined with multi-way pattern recognition for Poria cocos origin identification

Xia Hua,Chunling Yin,Leqian Hu,Liuchuang Zhao,Zhimin Liu,Peng Li

s Selective probes exclusively interact with a single metal ion species. In contrast, weakly selective probes can simultaneously coordinate with multiple metal ions, generating distinct fluorescence spectral signatures. This unique attribute enables them to provide more abundant chemical information. In this study, two weakly selective fluorescent probes of imidazole derivatives were designed and synthesized. We investigated the viability of utilizing a fluorescent array to trace the origin of Poria cocos based on the variations in fluorescence intensity and position of the distinctive fluorescence peaks created by their interaction with various metal ions. Subsequently, we collected three-dimensional fluorescence spectra of the interactions between the fluorescent array sensors and Poria cocos extracts from different geographical origins. These array data were analyzed using multi-way pattern recognition methods. The U-PLS-DA model exhibited excellent performance, with a prediction accuracy of 97.56 % that exceeded that of the N-PLS-DA model. This study demonstrates that the weakly selective probe array approach integrated with chemometric analysis provides a novel strategy for the traceability of non-fluorescent foods and traditional Chinese medicines.

Carbon dots derived from Poria cocos polysaccharide as an effective “on-off” fluorescence sensor for chromium (VI) detection

Qianqian Huang,Qianqian Bao,Chengyuan Wu,Mengru Hu,Yunna Chen,Lei Wang,Weidong Chen

Chromium is a harmful contaminant showing mutagenicity and carcinogenicity. Therefore, detection of chromium requires the development of low-cost and high-sensitivity sensors. Herein, blue-fluorescent carbon quantum dots were synthesized by one-step hydrothermal method from alkali-soluble Poria cocos polysaccharide, which is green source, cheap and easy to obtain, and has no pharmacological activity due to low water solubility. These carbon quantum dots exhibit good fluorescence stability, water solubility, anti-interference and low cytotoxicity, and can be specifically combined with the detection of Cr(VI) to form a non-fluorescent complex that causes fluorescence quenching, so they can be used as a label-free nanosensor. High-sensitivity detection of Cr(VI) was achieved through internal filtering and static quenching effects. The fluorescence quenching degree of carbon dots fluorescent probe showed a good linear relationship with Cr(VI) concentration in the range of 1–100 μM. The linear equation was F 0 / F  = 0.9942 + 0.01472 [Cr(VI)] ( R 2  = 0.9922), and the detection limit can be as low as 0.25 μM (S/N = 3), which has been successfully applied to Cr(VI) detection in actual water samples herein.

Metabolomics Study of Guizhi Fuling Capsules in Rats With Cold Coagulation Dysmenorrhea

Zhang Yu,Su Na,Liu Weiyi,Wang Qingqing,Sun Jianguo,Peng Ying

Dysmenorrhea refers to a kind of uterine cramping pain that occurs in women during the period of menstrual. Guizhi Fuling Capsules are mainly used for the treatment of various pain syndromes and especially effective in treating primary dysmenorrhea. However, the research on its modern pharmacology and mechanism of action have not been thoroughly carried out. It is not clear about the main active ingredients, potential targets and metabolic pathways involved in its efficacy. Therefore, this research project employed estradiol benzoate sensitization combined with oxytocin pain to construct the cold coagulation syndrome dysmenorrhea model, observed the anti-dysmenorrhea effect of Guizhi Fuling Capsules, and used the metabolomics to explore its mechanism. The results showed that Guizhi Fuling Capsules could considerably reduce the number of writhing times in dysmenorrhea rats, increasing the level of PGE2 and β-EP and reducing the contents of PGF2α in rat serum. Pathological sections of uterus and ovaries also showed that Guizhi Fuling Capsules could significantly relieve endometrial hyperplasia and improve ovarian function. The LC/MS-based metabolomics of rat uterine indicated that the model group has a great deviation from the control group. Compared with the model group, the Guizhi Fuling Capsules group had a tendency to shift to the control group, and the main metabolic changes was mainly concentrated on saturated and unsaturated fatty acids. Among them, arachidonic acid is in a pivotal position, and the expression of its rate-limiting enzyme (COX-2) involved in its cyclooxygenase metabolic pathway was significantly up-regulated in the model group, but significantly decreased after the intervention of Guizhi Fuling Capsules. In conclusion, Guizhi Fuling Capsules can effectively relieve primary dysmenorrhea, and this effect may be attributed to the regulation effects of Guizhi Fuling Capsules on endogenous metabolism, such as inhibiting arachidonic acid converted to prostaglandins through downregulate the expression of COX-2, which plays an anti-inflammatory effect.

Glycyrrhizic acid modified Poria cocos polyscaccharide carbon dots dissolving microneedles for methotrexate delivery to treat rheumatoid arthritis

Chen Qi,Wu Chengyuan,Wang Siwei,Wang Qiang,Wu Peiyun,Wang Lei,Yan Peiyu,Xie Ying

Introduction: Rheumatoid arthritis is an autoimmune disease characterized by chronic joint inflammation. Methotrexate is one of the most effective drugs for rheumatoid arthritis, but the adverse reactions caused by oral methotrexate greatly limit its clinical application. Transdermal drug delivery system is an ideal alternative to oral methotrexate by absorbing drugs into the human body through the skin. However, methotrexate in the existing methotrexate microneedles is mostly used alone, and there are few reports of combined use with other anti-inflammatory drugs. Methods: In this study, glycyrrhizic acid was first modified onto carbon dots, and then methotrexate was loaded to construct a nano-drug delivery system with fluorescence and dual anti-inflammatory effects. Then hyaluronic acid was combined with nano-drug delivery system to prepare biodegradable soluble microneedles for transdermal drug delivery of rheumatoid arthritis. The prepared nano-drug delivery system was characterized by transmission electron microscopy, fluorescence spectroscopy, laser nanoparticle size analyzer, ultraviolet-visible absorption spectroscopy, Fourier transform infrared spectroscopy, differential scanning calorimeter and nuclear magnetic resonance spectrometer. The results showed that glycyrrhizic acid and methotrexate were successfully loaded on carbon dots, and the drug loading of methotrexate was 49.09%. The inflammatory cell model was constructed by lipopolysaccharide-induced RAW264.7 cells. In vitro cell experiments were used to explore the inhibitory effect of the constructed nano-drug delivery system on the secretion of inflammatory factors by macrophages and the cell imaging ability. The drug loading, skin penetration ability, in vitro transdermal delivery and in vivo dissolution characteristics of the prepared microneedles were investigated. The rat model of rheumatoid arthritis was induced by Freund's complete adjuvant. Results: The results of in vivo animal experiments showed that the soluble microneedles of the nano drug delivery system designed and prepared in this study could significantly inhibit the secretion of pro-inflammatory cytokines and had a significant therapeutic effect on arthritis.Discussion: The prepared glycyrrhizic acid-carbon dots-methotrexate soluble microneedle provides a feasible solution for the treatment of Rheumatoid arthritis.

Protective mechanism of traditional Chinese medicine guizhi fuling pills against carbon tetrachloride-induced kidney damage is through inhibiting oxidative stress, inflammation and regulating the intestinal flora

Baogui Xu,Jiawen Zheng,Xiaoxiao Tian,Falei Yuan,zhongliang Liu,Yafeng Zhou,Zuisu Yang,Xianjun Ding

Background Chemical or drug-induced kidney damage has been recognized as a critical cause of kidney failure. The oxidative stress, inflammation, and imbalance of intestinal flora caused by carbon tetrachloride (CCl 4 ) play a fundamental role in chronic kidney damage. Guizhi Fuling pills (GZFL) is a traditional formula consisting of five traditional Chinese medicinal herbs, which can promote blood circulation and improve kidney function. The underlying mechanisms of GZFL improving kidney damage are not fully understood yet. Aim The current study aimed to explore the effects of GZFL on CCl 4 -induced kidney damage and intestinal microbiota in mice. Methods Male ICR mice were intraperitoneally administered with 20% CCl 4 (mixed in a ratio of 1:4 in soybean oil) twice a week, for 4 weeks to induce kidney damage. Creatinine (CRE), urea nitrogen, antioxidant enzymes, and inflammatory cytokines were measured and the histology of the kidney, jejunum, and colon examination to assess kidney and intestinal damage. The expressions of nuclear factor-erythroid 2-related factor 2 (Nrf2) family members, nucleotide-binding domain and leucine-rich repeat protein 3 (NLRP3) inflammasome in kidney tissues, and the tight junction proteins in colonic tissues were detected by Western blot. The gut microbiota was analyzed through 16S rRNA gene sequencing. Results GZFL treatment decreased the serum CRE and urea nitrogen levels. Moreover, GZFL reduced the levels of pro-inflammatory cytokines and increased antioxidant enzyme activities in kidney and colonic tissues. GZFL improved the kidney, jejunum, and colon histology. Furthermore, GZFL inhibited the expressions of NLRP3, ASC, and cleaved-Caspase-1, while Nrf2, HO-1, NQO1, GCLM, and tight junction proteins were increased. The dysbiosis of intestinal microbiota improved after GZFL treatment. Conclusions This study showed that GZFL could improve kidney damage, which might be mainly via the integrated regulations of the Nrf2 pathway, NLRP3 inflammasome, and composition of intestinal microbiota.

Guizhi Fuling Wan reduces autophagy of granulosa cell in rats with polycystic ovary syndrome via restoring the PI3K/AKT/mTOR signaling pathway

Min Liu,Hongqiu Zhu,Ying Zhu,Xiaodan Hu

Ethnopharmacological relevance Guizhi Fuling Wan (GFW) is a traditional Chinese medicine used to remove blood stasis and dissipate phlegm for treating gynecological diseases that was invented by Zhang Zhongjing in the Eastern Han dynasty. In recent years, GFW has been widely used to treat patients with polycystic ovary syndrome (PCOS). Clinical and animal studies have shown that it is effective in the treatment of PCOS, but its mechanism is unknown. Generally, it works by regulating autophagy via the PI3K/AKT/mTOR signaling pathway. Aim of the study This study investigated the effects and mechanism of GFW in PCOS rats with insulin resistance (IR) in order to provide better understanding of its observed clinical effects and a theoretical basis for the study of traditional Chinese medicine. Materials and methods Eighty-four female Sprague–Dawley rats were randomly divided into seven groups (n = 12 per group): 1) control, 2) PCOS model, 3) low-dose GFW, 4) medium-dose GFW, 5) high-dose GFW, 6) metformin , and 7) medium-dose GFW plus LY294002 . In all non-control groups, we induced PCOS through daily letrozole combined with intragastric high-fat emulsion for 21 days. After treatment, rats were sacrificed and serum follicle-stimulating hormone (FSH), testosterone (T), progesterone , luteinizing hormone (LH), 17β-estradiol, fasting insulin (FINS), and fasting plasma glucose levels were measured by enzyme-linked immunosorbent assay (ELISA). The LH/FSH ratios and HOMA-IR values were calculated. Ovarian morphology was observed by hematoxylin and eosin staining, and all follicles were counted under a microscope. MDC-positive vesicles were used as markers to detect autophagy, and the expression levels of p62, Beclin1 , and LC3-II were examined by immunostaining. Western blotting was used to measure PI3K/AKT/mTOR pathway activation, granulosa cell apoptosis , and autophagy. Results Compared with the PCOS model group, GFW-treated rats had less atretic and cystic follicles, and more mature follicles and corpus lutea. The GFW-treated rats had lower serum T, LH, and FINS levels than the PCOS model group, as well as lower LH/FSH ratios and HOMA-IR values. GFW treatment resulted in significantly reduced levels of cleaved-Caspase-3, cleaved-Caspase-9, BAX, Beclin1, Atg5, and LC3-II. Phosphorylation of PI3K, AKT, and mTOR was significantly higher in GFW-treated rats compared with the PCOS model group. The phosphorylation of PI3K, AKT, and mTOR was decreased with the use of a PI3K antagonist. Conclusions Our results indicate that GFW inhibited granulosa cell autophagy and promoted follicular development to attenuate ovulation disorder in PCOS-IR rats. This was associated with activation of the PI3K/AKT/mTOR signaling pathway.

Multiple-fingerprint analysis of Poria cocos polysaccharide by HPLC combined with chemometrics methods

Yilin Zhang,Mengqi Wu,Jingrui Xi,Chen Pan,Zhizhen Xu,Wei Xia,Wenqing Zhang

In this study, the multiple fingerprints, which were integrated with HPGFC-ELSD (high performance gel filtration chromatography - evaporative light scattering detector) fingerprint, PMP-HPLC-DAD (1-phenyl-3-methyl-5-pyrazolone-high performance liquid chromatography - diode array detector) fingerprint of complete acid hydrolysates and HILIC-HPLC-ELSD (hydrophilic interaction - high performance liquid chromatography - evaporative light scattering detector) fingerprint of enzyme hydrolysates, were established to evaluate the quality of polysaccharides from Poria cocos (PCPs). The similarity evaluation showed that 16 batches of PCPs from different origins had high similarity in structural characteristics based on the multiple fingerprints. The chromatographic data of multiple fingerprints of PCPs were fused, processed and analyzed by chemometric methods including HCA (hierarchical cluster analysis), PCA (principal component analysis) and PLS-DA (partial least squares discriminant analysis). The 16 batches of PCPs were divided into 3 categories in PCA, indicating a certain relationship between the structural characteristics and the origins. PLS-DA analysis indicated that Man, Glc, Gal, Fuc, the components with m/z of 2.22 × 10 4 ∼1.53 × 10 4 Da and 3.46 × 10 3 ∼2.69 × 10 3 Da, oligosaccharides with DPs of 6 and 7, respectively, could be regarded as potential chemical markers for the classification of PCPs from different origins. According to the multiple fingerprints and chemometric analysis, the two commercial samples were proved to be adulterants.
