Functional Characterization of an Antenna-Biased Odorant Receptor AaOr96 Involved in Tea Tree Oil Repellency Against Aedes aegypti

Mengli Chen,Zhanyi Xu,Guoxing Chen,Peitong Chen,Chunxia Tian,Jiali Qian,Tiefeng Song,Yongfeng Jin,Guonian Zhu,Ru Yan

Numerous essential oils have been well acknowledged as eco-friendly alternatives to combat insect pests due to synthetic insecticide-induced pest resistance and environment pollution. As a highly commercial essential oil, tea tree oil exhibits excellent insecticidal and repellent activities. However, the molecular mechanism of the olfactory system mediating the tea tree oil-induced repellency against insect pests remains unknown. In our study, mosquito was used as a suitable model to examine the molecular mechanism of tea tree oil-induced repellency against insect pests. The results showed that tea tree oil exhibited excellent spatial and oviposition repellency against Aedes aegypti adults and outstanding repellency against larvae, which were conferred by the main constituent terpinen-4-ol. The reduced repellency in the Orco–/– mutant strain revealed that tea tree oil-induced repellency against mosquitoes was dependent on odorant receptor(s). Moreover, we identified one antenna-biased odorant receptor, AaOr96, that was involved in detecting constituents of tea tree oil to elicit repellency, and the predicted protein–ligand complex indicated that AaOr96 interacted with terpinen-4-ol via van der Waals forces from five key residues. Finally, knocking out AaOr96 resulted in a reduced spatial repellency against A. aegypti by tea tree oil and terpinen-4-ol, and a reduced oviposition repellency by terpinen-4-ol, but not by tea tree oil. Our study not only reveals that tea tree oil has great potential in pest management but also provides more insights into the molecular basis of repellency of essential oils.

Essential oil-loaded chitosan/zinc (II) montmorillonite synergistic sustained-release system as antibacterial material

Jinghui Zhan,Huayao Chen,Hongjun Zhou,Li Hao,Hua Xu,Xinhua Zhou

Tea tree oil (TTO) is a broad-spectrum natural antimicrobial agent. Its application is limited due to its volatile property. To reduce its volatility and improve its sustained-release performance, a synergistic sustained-release system was prepared. Montmorillonite (Mt) was ion-exchanged with sodium ions (Na+) and zinc ions (Zn2+), then organically modified with cetyl trimethyl ammonium bromide (CTAB) and chitosan (CS) to improve its sustained release performance. The results indicated that Mt was exfoliated, but still maintained its ordered layer structure. The surface of CS-Zn-OMt was rough, because some pores and surfaces were filled. The addition of Zn2+ increased the loading of CS (37.8 − 40.8%) and induced more uniform distribution of CS on Mt. Also, CS-Zn-OMt achieved a high loading capacity for TTO and the loading value was 222.0 mg*g−1. Furthermore, the release studies showed that CS-Zn-OMt exhibited sustained-release behavior due to the release from the outer layer of CS and the Mt channel simultaneously. The results of antibacterial test showed that the addition of CS inhibited the drift of Mt and showed good antibacterial performance because of strong effective drug-carrier interaction. The developed system can be potentially applied for the delivery of volatile essential oils for antibacterial infection.

Purification of crude tea tree oil through enrichment of terpinen-4-ol by in-situ formation of deep eutectic solvent

Lijiao Yu,Lihong Deng,Jieyu Wu,Subhan Mahmood,Ke Yuan,Haoran Wu,Xiaodong Wang,Jing Zhang,Shun Yao

As a key component in tea tree oil (TTO), terpinen-4-ol exhibits ideal physiological activities. However, the high content of terpenes in crude TTO makes it challenging to enrich terpineol-4-ol. In this study, molecular simulations combined with experimental methods were used to select the optimal hydrogen bond acceptor (HBA), tetrabutylammonium chloride (TBAC), which forms a deep eutectic solvent (DES) with terpinen-4-ol in situ. Combined with necessary post-treatment process, it provided a feasible way for purification of TTO. Related DESs were prepared and characterized, and their main properties were also investigated. When the amount of TBAC was three times that of terpinen-4-ol, the formation of hydrogen bonds became more pronounced. At this point, the DES achieved its highest distribution coefficient for terpinen-4-ol (D t  = 15.02), as well as superior selectivity for terpinen-4-ol (S t/c  = 24.59, S t/γ  = 57.69). Water as the back-extractant and n-hexane as the auxiliary extractant facilitated the deep purification of terpinen-4-ol. Response surface analysis and experimental results confirmed that the optimal enrichment conditions were 30 min of enrichment time, an enrichment temperature of 60℃, and 0.5 g/g of n-hexane dosage. The back-enrichment and recovery of TBAC also proved to be satisfactory, and when applied to real tea oil, the developed method achieved over 90 % enrichment efficiency for terpinen-4-ol.

Long-lasting anti-bacterial activity and bacteriostatic mechanism of tea tree oil adsorbed on the amino-functionalized mesoporous silica-coated by PAA

Fan Gao,Hongjun Zhou,Zhichuan Shen,Guowei Zhu,Li Hao,Huayao Chen,Hua Xu,Xinhua Zhou

Tea tree oil (TTO) is an efficient natural antibacterial agent. However, the bacteriostatic effect of TTO does not prevail for a long period because of the volatile nature of the oil. Therefore, a novel sustained-release formulation of TTO should be developed for improving the applicability of TTO. Herein, the mesoporous silica was selected for constructing a carrier for TTO. Mesoporous silica is non-toxic, easy to modify and exhibited an adjustable pore size. First, the mesoporous silica was modified by an aminated silane coupling agent (NH 2 -MCM-41). Then, the polyacrylic acid (PAA) was bonded by electrostatic bonds (PAA-NH 2 -MCM-41), which imparted the sustained-release effect in the TTO, supported in the mesoporous silica channel (TTO/PAA-NH 2 -MCM-41). The prepared bacteriostatic agent can achieve long-term sustained-release properties. At room temperature (26 ℃), the release rate of TTO after 11 h release reached 50 %. However, the release rate of TTO from TTO/PAA-NH 2 -MCM-8 reached only 42 % after 24 h. Furthermore, the sustained release behavior of TTO/PAA-NH 2 -MCM-41 was consistent with the Korsmeyer-Peppas kinetic model. Compared to TTO, TTO/PAA-NH 2 -MCM-41 exhibited a stable and sustained bacteriostatic effect even after 50 days in a natural environment. The minimum inhibitory concentration (MIC) value of the TTO/PAA-NH 2 -MCM-41 against Escherichia coli ( E. coli ) was 0.37∼0.44 mg/mL. TTO altered the cell morphology of E. coli and broke the integrity of the cell membrane, leading to cell death.

Fabrication and Biological Activities of All-in-One Composite Nanoemulsion Based on Blumea balsamifera Oil-Tea Tree Oil

Yue Zhu,Teng Chen,Tingting Feng,Jiaojiao Zhang,Zejing Meng,Ning Zhang,Gang Luo,Zuhua Wang,Yuxin Pang,Ying Zhou

Nanoemulsion is a new multi-component drug delivery system; the selection of different oil phases can give it special physiological activity, and play the role of “medicine and pharmaceutical excipients all-in-one”. In this paper, we used glycyrrhizic acid as the natural surfactant, andBlumea balsamiferaoil (BB) and tea tree oil (TTO) as the mixed oil phase, to obtain a new green functional composite nanoemulsion. Using the average particle size and polydispersion index (PDI) as the evaluation criteria, the effects of the oil ratio, oil content, glycyrrhizic acid concentration, and ultrasonic time on the nanoemulsion were systematically investigated. The stability and physicochemical properties and biological activities of BB-TTO NEs prepared via the optimum formulation were characterized. The optimal prescription was BB: TTO = 1:1, 5% oil phase, 0.7% glycyrrhizic acid, and 5 min ultrasonication time. The mean particle size, PDI, and zeta potential were 160.01 nm, 0.125, and −50.94 mV, respectively. The nanoemulsion showed non-significant changes in stability after centrifugation, dilution, and 120 days storage. These nanoemulsions were found to exhibit potential antibacterial and anti-inflammatory activities. The minimal inhibitory concentration (MIC) of BB-TTO NEs againstEscherichia coli,Staphylococcus aureus, andPseudomonas aeruginosais 2975 μg/mL, 2975 μg/mL, and 5950 μg/mL, respectively. A lower level of inflammatory cell infiltration and proportion of fibrosis were found in the synovial tissue of AIA rats treated with BB-TTO NEs. These findings demonstrate that the BB-TTO NEs produced in this study have significant potential for usage in antibacterial and anti-inflammatory areas. Keywords:nanoemulsion;Blumea balsamiferaoil;tea tree oil;glycyrrhizic acid;anti-inflammatory;antibacterial

Electricallymodified bacterial cellulose tailored with plant based green materials for infected wound healing applications

Manjila Adhikari,Bianza Moise Bakadia,Li Wang,Ying Li,Zhijun Shi,Guang Yang

Effective treatment of infected wounds remains a challenge due to the rise of antibiotic-resistant microorganisms. The development of advanced materials with strong antimicrobial properties is necessary to address this issue. In this study, a unique composite of electrically modified bacterial cellulose (EBC) with allantoin (ABC) and zein was developed by dipping diffusion method. Morphological structural analysis revealed a uniform distribution of zein and aligned fibers, confirming the synthesis of the ABC-Zein composite. The formation of ABC-Zein was further confirmed by attenuated total reflection-Fourier transform infrared (ATR-FTIR), which displayed additional peaks corresponding to EBC, indicating the incorporation of zein into ABC. X-ray diffraction (XRD) analysis of ABC-Zein demonstrated a similar crystalline structure with EBC. The ABC-Zein showed mechanical integrity (tensile strength: 1.15 ± 0.21 MPa), thermal stability (degradation temperature: 290 °C), porous structure (porosity: 40.23 ± 0.21 %), and hydrophilic (water contact angle: 53.3 ± 5.3°) properties. Furthermore, the antimicrobial agent terpinen-4-ol (T4O), derived from tea tree oil, was incorporated into the ABC-Zein composite. Biological studies confirmed the antimicrobial efficacy ( Staphylococcus aureus inhibition: 88.5 ± 7.19 %) and biocompatible (cell viability: 84.95 ± 5.6 %, hemolysis: 4.479 ± 0.39 %) nature of the T4O-ABC-Zein composite. The combined effects of the aligned fiber structure, zein protein, and antimicrobial T4O significantly enhanced infected wound healing by day 7, promoting inflammatory response, granular tissue formation, cell proliferation, and angiogenesis. By day 14, T4O-ABC-Zein facilitated complete wound healing, with reepithelization, collagen I deposition, and downregulation of CD 31, Ki67, and α-SMA. Overall, the innovative T4O-ABC-Zein composite, with an aligned fiber structure, improved biocompatibility, and antimicrobial properties, holds significant potential for the treatment of infected wounds.

Electrospun chitosan-based nanofibers loading tea tree oil for fresh salmon fillet shelf-life extension

Yuhua Xia,Xu Tang,Xudong Wu,Yizhen Li,Changan Xu,Shaohua Chen,Peng Wu

Bioactive packaging can improve the shelf-life of food products and enhance consumer health. It can also alleviate environmental stress on the planet by reducing food waste. Here, the electrospinning of tea tree oil-loaded 2-hydroxypropyltrimethyl ammonium chloride chitosan nanofibers was investigated. The fabricated nanofiber films were characterized by scanning electron microscopy, thermal gravimetric analysis, Fourier transform infrared spectroscopy, and contact angle meter analysis. The prepared nanofibers have a well-defined diameter of about 200 nm and a smooth shape. They have good antibacterial properties against Staphylococcus aureus and Escherichia coli in vitro. Tea tree oil-loaded chitosan-based nanofibers were found to be effective in delaying spoilage and extending the shelf life of salmon by sensory evaluation, texture analysis, color, total viable counts, thiobarbituric acid, and total volatile basic nitrogen during storage in the freshness experiments, thus indicating their health benefits in bioactive packaging.

PVA@tea tree oil-emulsion core–shell microfiber membranes for extending the shelf life of fruits

Tianliang Dai,Ya Liu,Lina Wang,Juming Yao,Guocheng Zhu,Baochun Guo,Jiri Militk,Dana Kremenakova,Ming Zhang

In this work, a temperature-regulating and antimicrobial–antioxidant fiber membrane was fabricated using coaxial electrospinning technology. The tea tree oil (TTO) emulsion used as the core solution, polyvinyl alcohol (PVA) as the shell solution for coaxial electrospinning, has been demonstrated. Compared with the original PVA fibers, the average diameter of the fibers is increased after coaxial electrospinning, and the diameter increases with the core flow rate. The diameters corresponding to core flow rates of 0.05, 0.1, and 0.15 mL/h are 561, 597, and 849 nm, respectively. Via in vitro release experiment demonstrated that the quantity of TTO released from the composite membrane increased with the elevation of temperature. The inclusions TTO and polyethylene glycol give the film excellent antibacterial–antioxidant properties and a delayed thermal response. More intriguingly, core–shell fiber membranes containing TTO in the core part are effective in slowing down the formation of oxidation spots on the surface of banana compared with pure PVA membranes. Seven days after wrapping bananas with different membranes, the surface of banana in pure PVA film was full of oxidized spots, whereas banana in PVA core–shell fibers showed only one obvious spot.

β-Cyclodextrin/Tea Tree Oil Employed Thermoset with Controlled Hydrolysis and Synergistic Antibacterial Properties

Jin Han,Fang Chen,Weipeng Qin,Lei Wang,Dan Wu,Kun Shen,Xiaoxue Yin,Qingquan Xue

The practical application of tea tree oil (TTO) as a natural broad-spectrum antimicrobial agent is limited by high volatility, chemical instability, and compatibility challenges with conventional polymer matrices. In this study, an antimicrobial thermosetting system with controlled hydrolysis was constructed. The N,N,N′,N″,N″-pentamethyldipropylenetriamine (PMD) cured samples exhibited superior mechanical and thermal properties in comparison to the N,N,N′,N′-tetramethyl-1,6-hexanediamine (TMHDA) or N,N,N′,N′-tetramethyl-1,3-propanediamine (TMPDA) cured samples. The TTO-loaded β-cyclodextrin (β-CD) was cross-linked by a rapid quaternization reaction to give the thermoset with quaternary ammonium contact-mode antimicrobial and TTO pharmaceutical antimicrobial properties. The remaining tertiary amine groups in the system provide a weakly alkaline environment, while the quaternary ammonium ester (Quat Ester) groups undergo weak base accelerated hydrolysis. The tensile strength of the cured system reached 23.69 MPa, and the antimicrobial activity against Escherichia coli and Staphylococcus aureus reached 95.27 and 95.63%, respectively; the antimicrobial performance could be further enhanced when long-chain cetyl chloroacetate (CCA) was involved in the curing (the antimicrobial activity reached 98.33 and 98.07%, respectively). The antimicrobial and pH-responsive degradation properties provide ideas for the development of degradable and antimicrobial materials for three-dimensional (3D) printing, solid disinfectants and so on.

Antifungal modes of action of tea tree oil and its two characteristic components against Botrytis cinerea

Yu D.,Wang J.,Shao X.,Xu F.,Wang H.

AimsThe essential oil of Melaleuca alternifolia (tea tree) has been evaluated as a potential eco‐friendly antifungal agent against Botrytis cinerea. In this study, we investigated the antifungal activity and mode of action of tea tree oil (TTO) and its components against B. cinerea. Methods and ResultsOf the components we tested in contact phase, terpinen‐4‐ol had the highest antifungal activity, followed by TTO, α‐terpineol, terpinolene, then 1,8‐cineole. As one of characteristic components of TTO, terpinen‐4‐ol treatment led to pronounced alterations in mycelial morphology, cellular ultrastructure, membrane permeability under scanning electron microscope, transmission electron microscope and fluorescent microscope, and also reduced the ergosterol content of fungi. As another characteristic component, 1,8‐cineole caused serious intracellular damage but only slightly affected B. cinerea otherwise. When terpinen‐4‐ol and 1,8‐cineole were used together, the synergistic antifungal activity was significantly higher than either component by itself.ConclusionsThe results of our study confirmed that terpinen‐4‐ol and 1,8‐cineole act mainly on the cell membranes and organelles of B. cinerea, respectively, and when combined are similar to TTO in antifungal activity due to their differences.Significance and Impact of the StudyUnderstanding the mechanism of terpinen‐4‐ol and 1,8‐cineole antifungal action to B. cinerea is helpful for investigation on their synergistic effect and explaining antifungal action modes of TTO.

High-adhesion antimicrobial composite coating incorporating quaternary chitosan and tea tree oil for enhanced preservation of fruits and vegetables

Ying Liu,Yi Ding,Chao Wang,Jian Luo,Huanhuan Yao,Huili Zhang,Long Xu,Junfan Niu

This study developed high-adhesion quaternary chitosan coatings using amphiphilic HACC-oleic acid conjugates (HACC-OA) to enhance fruit and vegetable preservation. HACC-OA demonstrated improved viscosity and adhesion, providing a stable carrier for tea tree oil (TTO). The resulting TTO@HACC-OA coatings exhibited significantly reduced contact angle (43.7°) and increased retention amount on strawberry surfaces. Furthermore, TTO@HACC-OA demonstrated a lower volatilization rate of TTO (27.8 %) compared to pure TTO (49.5 %) and an enhanced synergistic antimicrobial activity (EC 50  = 1.51 mg/mL) against Botrytis cinerea Pers compared to HACC alone (EC 50  = 1.58 mg/mL). Preservation experiments revealed that TTO@HACC-OA effectively maintained color and firmness, reduced decay index and weight loss, delayed decline in vitamin C content of strawberries, postponed increase in total phenolic content, and mitigated malondialdehyde accumulation. Therefore, the prepared TTO@HACC-OA composite coating with excellent adhesion ability and preservation effect holds great potential for applications in fruit and vegetable preservation.

Sprayed Pickering emulsion with high antibacterial activity for wound healing

Xieru Bao,Jie Wu,Guanghui Ma

Effective treatment of wound injuries can prevent infections and promote healing. In this work, a sprayed Pickering emulsion composed of chitosan nanoparticles, tea tree oil and curcumin was formulated. The shear thinning behaviour allowed this Pickering emulsion to spray even with a high viscosity provided by the aqueous phase containing chitosan nanoparticles. Moreover, the Pickering emulsion could recover to the high viscosity after spraying onto the wounds, leading to the long residence time and intimate contact. In addition, the Pickering emulsion provided a carrier to encapsulate the tea tree oil, which caused sustained-release effects and further strengthened tea tree oil's antibacterial activity towards the Gram-negative Escherichia coli ( E. coli ), Gram-positive Staphylococcus aureus ( S. aureus ) and Gram-positive methicillin-resistant Staphylococcus aureus (MRSA). Besides, this multi-phase system offered a platform to load with hydrophobic curcumin, leading to better synergistic healing effects. Meanwhile, the Pickering emulsion avoided the irritation and toxicity related to surfactants, which was far safer than the classical emulsion especially when used in the defective skin. All these excellent properties endowed this sprayed Pickering emulsion with promising prospects for wound healing.
