Residue and dietary risk assessment of glyphosate, glufosinate-ammonium, and their metabolites in maize and soybean

Kuan Wang,Bin Jiao,Haixiang Gao,Xinglu Pan,Xiaohu Wu,Jun Xu,Fengshou Dong,Yongquan Zheng

Persistent weed growing in herbicide-resistant maize and soybean fields is controlled increasingly by glyphosate (GLY) and glufosinate-ammonium (GLU). There is increasing concern that consumers may be exposed to herbicide residues . A Total of 224 samples involved supervised trials and market sampling were carried out. And the levels of GLY and GLU were determined in maize and soybean samples by UPLC−MS/MS method. The residues of GLY and GLU were at a low level in all samples and ranged from <0.04 to 0.29 mg/kg and <0.04 to 0.91 mg/kg, respectively. Deterministic and probabilistic models were used to assess the dietary risk, and the highest risk quotient (RQ) values of GLY and GLU were 2.24 × 10 −4 and 3.27 × 10 −2 , respectively. The RQs of GLU are significantly higher than that of GLY. Risk assessment results showed that GLY and GLU residues should not pose serious health risks to Chinese consumers.

Simultaneous determination of glyphosate, glufosinate ammonium and their metabolites in maize and soybean by ultra-performance liquid chromatography with tandem mass spectrometry

Kuan Wang,Yue Geng,Zhe Dong,Xinglu Pan,Xiaohu Wu,Jun Xu,Haixiang Gao,Fengshou Dong,Yongquan Zheng

Glyphosate and glufosinate ammonium are non-selective, broad spectrum herbicides for controlling grasses in agriculture and forestry. Rapid and sensitive analytical methods for effective monitoring and subsequent risk control of glyphosate and glufosinate ammonium and their metabolites in crops are not available. In this paper, an analytical method for the simultaneous determination of glyphosate, glufosinate ammonium, and their metabolites in maize and soybean by ultra-performance liquid chromatography with tandem mass spectrometry was established. The seven compounds were well separated on an anion exchange analytical column within 10 min, with the mean recoveries of the target compounds ranging from 78.2 to 110.9%. The method showed good linearity (R2 ≥ 0.9819) with a limit of quantification for glyphosate, glufosinate ammonium, and their metabolites of 0.01 mg kg−1 in both maize and soybean. The method simplifies and expedites the sample preparation procedures, overcomes the traditional disadvantages including derivatization, weak retention, and low sensitivity, and has been successfully validated in actual maize and soybean samples.

Titanium dioxide-coated core-shell silica microspheres-based solid-phase extraction combined with sheathless capillary electrophoresis-mass spectrometry for analysis of glyphosate, glufosinate and their metabolites in baby foods

Jianing Liu,Wenxia Feng,Miaomiao Tian,Lihuan Hu,Qishu Qu,Li Yang

Because of their extremely low amount in complex samples, it is quite challenging to accurate determine residues of phosphorus-containing amino-acid-like herbicides (PAAHs) in food products. Here we develop novel core-shell mesoporous silica (CSMS) microspheres coated by titanium dioxide (CSMS@TiO 2 ) for extraction and enrichment of PAAHs in baby foods. After the dispersive solid phase extraction (d-SPE), sheathless capillary electrophoresis-mass spectrometry (sheathless CE-MS) is utilized to achieve efficient separation and sensitive detection. The synthesized CSMS@TiO 2 composites are characterized by various spectroscopic techniques, proving TiO 2 is uniformly distributed onto the channel surface of CSMS. The composites have essential features that are favorable for adsorption of the analytes on the material for d-SPE, including uniform diameter (1.0 μm with a shell thickness of 133 nm), large perpendicular mesopores (15.6 nm), high surface area (101.1 m 2 /g) and large pore volume (0.4 cm 3 /g). Taking glyphosate, glufosinate and their main metabolites (aminomethylphosphonic acid and 3-methylphosphinicopropionic acid) as analytes, selective and efficient enrichment is achieved by CSMS@TiO 2 -based d-SPE through the affinity interaction between titanium dioxide and phosphate groups. Sensitive detection of target compounds is achieved with low limits of quantitation (LOQs) between 0.3–1.6 ng/mL and excellent inter/intra-day repeatability. The compounds in nine different commercial baby foods from local markets are analyzed using the proposed method. Good recoveries of 82.3–102.6% are achieved with low RSDs ( n  = 5) of 2.1–8.3%. Our study indicates that the proposed CSMS@TiO 2 -based d-SPE combined with sheathless CE-MS is an accurate and reliable approach for sensitive determination of trace-amount PAAHs and their metabolites in complex samples.

Physiological and transcriptomic analyses reveal the threat of herbicides glufosinate and glyphosate to the scleractinian coral Pocillopora damicornis

Zhi Zhou,Xingzhen Ni,Zhongjie Wu,Jia Tang

The amino acid metabolism-related herbicides glufosinate and glyphosate are used worldwide and have flowed into the oceans, threatening the marine organisms. In the present study, physiological activities and transcriptomic profiles of the scleractinian coral Pocillopora damicornis and symbiotic Symbiodiniaceae were determined during a 48 h-exposure to the two herbicides with the final concentration of 10 μmol L −1 . Coral samples were collected at 0, 12, 24, and 48 h after exposure to determine symbiont density, chlorophyll content, as well as activities of superoxide dismutase (SOD), catalase (CAT), nitric oxide synthetase (NOS) and phenoloxidase (PO), and the caspase-3 levels, and the samples collected at 24 h were employed in the transcriptomic analysis. Specifically, the symbiont densities did not change significantly in response to the two herbicides, while the chlorophyll content increased significantly at 24 h post glufosinate exposure. SOD and CAT activities in the coral host increased significantly at 12 h after glufosinate and glyphosate exposure, while the activity of NOS in symbionts decreased significantly at 48 h after glufosinate exposure. Caspase-3 levels in the coral host declined significantly at 24 h after exposure to the two herbicides. In the transcriptomic analysis, glufosinate triggered the expression of genes related to the response to stimuli and immunoregulation in the coral host, and suppressed the expression of genes related to coral nitrogen-related metabolism, symbiont cell cycle, and response to nutrient levels. Furthermore, glyphosate activated the expression of genes involved in coral calcification and symbiont nutrient export and suppressed the expression of genes involved in coral meiosis and symbiont cell communication. These results suggest that although the coral-Symbiodiniaceae symbiosis is not disrupted, short-term glufosinate and glyphosate exposures alter several essential physiological processes including metabolism, calcification, and meiosis in the coral host, as well as the cell cycle and nutrient export in the symbiont. Summary Glufosinate and glyphosate herbicide exposures can disturb several essential physiological processes, including metabolism, calcification, and meiosis in the coral host as well as the cell cycle and nutrient export in the symbiont, threating the survival of scleractinian corals.
