According to the Chinese Pharmacopoeia 2015, only processed Aconitum tubers can be clinically applied, and the effect of processing is unclear. This research aimed to explore the effect of processing on cardiac efficacy of alkaloids in Aconitum tubers. First, the chemical ingredients in unprocessed and processed Aconitum tubers were identified and compared by using high performance liquid chromatography time-of-flight mass spectrometry (HPLC-TOF/MS) and multivariate pattern recognition methods. Then the representative alkaloids in Aconitum tubers, aconitine, benzoylaconine, and aconine, which belong to diester-diterpenoid alkaloids, monoester-diterpenoid alkaloids, and amine-diterpenoid alkaloids, respectively, were selected for further validation of attenuated mechanism. Subsequent pharmacological experiments with aconitine, benzoylaconine, and aconine in SD rats were used for validate the effect of processing on cardiac functions. After processing the Aconitum tubers, it was found that the contents of diester-diterpenoid alkaloids were reduced, and those of monoester-diterpenoid alkaloids and amine-diterpenoid alkaloids were increased, suggesting that diester-diterpenoid alkaloids were transformed into monoester-diterpenoid alkaloids and amine-diterpenoid alkaloids. Through further decocting the aconitine in boiling water, it was confirmed that the three alkaloids could be progressively transformed. Pharmacological experiments with aconitine, benzoylaconine, and aconine in SD rats showed that aconitine at a dose of 0.01 mg/kg and aconine at a dose of 10 mg/kg enhanced the cardiac function, while benzoylaconine at a dose of 2 mg/kg weakened the cardiac function. The effect of processing is attributed to the transformation of the most toxic diester-diterpenoid alkaloids into less toxic monoester-diterpenoid alkaloids and amine-diterpenoid alkaloids.

Aconiti Lateralis Radix Praeparata is one of the most famous traditional Chinese medicines possessing a variety of pharmacological activities on top of the toxicities. Due to the heterogeneity and non-standardization of the processing procedures, the subtypes and contents of the differential compounds between different processed products still remained indistinct, causing great risk in their proper use. In order to achieve the comparison and quality evaluation of different processed products of Aconiti Lateralis Radix Praeparata and develop new processed products with less toxicity, a quantification and pseudotargeted metabolomics method was developed based on the dynamic MRM mode of triple quadrupole (QqQ) mass spectrometry, and multivariate statistical analysis methods were applied to compare different processed products. Method validation results indicated good specificity, linearity, repeatability, precision, stability and recovery of the established quantification method and good linearity, precision and stability of the pseudotargeted metabolomics method. Differential compounds of different processed products were screened out and further confirmed by the quantification results. At last, the processing procedures were optimized to obtain new processed products of “Heishunpian” (black slices) with less toxicity, in which the contents of the toxic diester-type diterpenoid alkaloids were reduced from 106.98 μg/g to 0.85–12.96 μg/g. This study provided a valuable reference for the establishment of comprehensive quality evaluation methods of herbal medicines and a scientific basis for the optimization of processing procedures of Aconiti Lateralis Radix Praeparata.

A green and effective extraction system based on the use of an ultrasound-assisted ionic liquid aqueous two-phase system (IL-ATPS) was developed for the determination of aconitum alkaloids in lateral roots of Aconitum carmichaeli Debx (Fuzi in Chinese). IL-ATPS parameters, including the type and amount of ionic liquid, amount of salts, and pH value, as well as extraction time, were investigated. Under the optimized conditions, the recoveries of the analytes varied from 80.37 to 107.18%. The detection limits and quantification limits for the analytes were achieved within the ranges of 0.048–0.082 μg g−1 and 0.16–0.30 μg g−1, respectively. After extraction, the recovery of ionic liquids without the use of organic solvents was also confirmed. The hydrophilic ionic liquids (1-hexyl-3-methylimidazolium bromide) were converted into hydrophobic ionic liquids (1-hexyl-3-methylimidazolium hexafluorophosphate) by a metathesis reaction, and in situ liquid–liquid microextraction was triggered simultaneously. Finally, 0.1 mol L−1 HCl aqueous solution (containing 5% Tween-20) was used as the back-extraction agent to recover the ionic liquids.

The conversion of diester-diterpenoid alkaloids (DAs) into monoester-diterpenoid alkaloids (MAs) is an effective method to reduce the toxicity of Aconiti Lateralis Radix Praeparata (Fuzi in Chinese, ALRP), a popular traditional Chinese medicine. In the present study, deep eutectic solvents (DESs) were first utilized to simultaneously detoxify ALRP and extract aconitum alkaloids by gas bath thermostatic oscillator-assisted extraction. The effects of extraction parameters, including the extraction time, water content of DESs, temperature, liquid/solid ratio, and pH, on the extraction of MAs were evaluated by single-factor experiments, and the optimal condition was further optimized using response surface methodology. The results indicated that betaine-urea (Bet-Ure, 1:2 mol/mol) was the most appropriate DES to hydrolyze highly toxic DAs into less toxic MAs and improve the extraction efficiency of aconitum alkaloids in one step. The detoxification mechanism study showed that alkaline DESs had an excellent efficiency for DA hydrolysis and MA extraction, and the MA-extraction efficiency enhanced with the increase in pH. Compared with the conventional extraction, the newly developed detoxification process maximized the hydrolysis of DAs and exhibited a higher MA-extraction efficiency (1.913 mg/g vs 0.155 mg/g). The DES-based method offered an ecofriendly, simple, and feasible detoxication strategy for ensuring the safe and effective use of ALRP.

Parkinson's disease (PD), with widespread aggregation of α-synuclein in the form of Lewy bodies as a neuropathological hallmark, is a rising threat for ageing society. Glycyrrhiza uralensis is an herbal medicine and sweets used for centuries in Asia and Europe, its major pharmacologically active ingredients include isoliquiritigenin (ILG), liquiritin (LT), liquiritigenin (LG) and glycyrrhizic acid (GA). Here, we investigated the effects of Glycyrrhiza uralensis ethanol extract (GUE) and the above four compounds, on the aggregation of α-synuclein in vitro and in a transgenic Caenorhabditis elegans PD model NL5901. In vitro, ILG, LT and GUE inhibit the amyloid formation of α-synuclein and alleviate the toxicity caused by aggregates; moreover, ILG could disaggregate preformed fibrils. In vivo, ILG, LT and GUE not only reduce amyloid formation in C. elegans NL5901, but also extend its life span. Together, these data suggest that ILG and LT may be further considered as candidates for PD treatment.
