Metabolic pathways determine cellular fate and function; however, the exact roles of metabolites in host defence against influenza virus remain undefined. Here we employed pharmacological inhibition and metabolomics analysis to show that the metabolic pathways of oxaloacetate (OAA) are integrated with antiviral responses to influenza virus. Cytosolic malate dehydrogenase 1 senses intracellular OAA to undergo dimerization and functions as a scaffold to recruit the transcription factor ETS2 for phosphorylation by the kinase TAOK1 at serine 313. The phosphorylated ETS2 translocates into the nucleus and supports optimal expression of TBK1, an indispensable activator of type I interferon responses. OAA supplementation provides a broad-spectrum antiviral ability, and OAA deficiency caused by Acly genetic ablation decreases antiviral immunity and renders mice more susceptible to lethal H1N1 virus infection. Our results uncover a signalling pathway through cellular OAA sensing that links metabolism and innate immunity to coordinate defence against viral challenge.

Graphical The composition of chromogenic impurities in the glyoxylic acid prepared by electroreduction of oxalic acid is studied. The chromogenic impurities are mainly composed of glyceric acid and pyruvic acid. 92.97 % of the chromatic substances can be removed from glyoxylic acid via electrodialysis. Oxalic acid is also separated from glyoxylic acid by electrodialysis method. Electroreduction of oxalic acid (OA) is an effective process to produce glyoxylic acid (GA). However, electrochemically generated GA solution is yellow, which lowers the quality of GA and affects the application in producing the downstream products. It is urgent to investigate the composition of chromogenic substances (CSs) and separate them from the GA solution. In this work, the presence of tartaric acid, glyceric acid, and pyruvic acid in the electrochemically generated GA solution is successfully confirmed by high-performance liquid chromatography-electrospray-mass spectrometry. Tartaric acid is originally generated from GA and glycolic acid, and then further reduced to obtain chromogenic glyceric acid and pyruvic acid. 92.97 % of CSs could be successfully removed from the GA solution by electrodialysis at 240.64 A/m 2 and 25 °C. A separation of OA and GA is also achieved by electrodialysis. This study makes a significant contribution to the industrialization of electroreduction process to prepare GA.

A desirable MOF-on-MOF-based ratiometric fluorescent probe (denoted as HPU-26@ZIF-8@Cit@Eu) was designed through the seed-mediated method for classifying phenylglyoxylic acid (PGA, a real internal exposure level of styrene) and 2,6-dipicolinic acid (DPA, a unique bacterial endospore biomarker) with high selectivity and sensitivity. Characterization techniques were carried out with the PXRD pattern, XPS spectra, UV–vis absorption spectra, and DFT-calculated HOMO and LUMO energies for proven mechanisms. The results indicated that PGA could affect the LMCT-ET process in HPU-26 and be binded to Eu3+ in ZIF-8, thus resulting in a significant increase both in the blue fluorescence emission peak at 476 nm and the red fluorescence emission peak at 620 nm. According to the fluorescence intensity ratio (I620/I476), the limit of detection (LOD) reached as low as 0.63 μM within a wide concentration range from 0 to 100 μM, and a noticeable color change from blue to red could be observed. However, upon exposure to DPA, the energy transfer between Zn2+ and the ligand was influenced sharply, without prejudice to the fluorescent intensity of Eu3+ in ZIF-8. These phenomena made this probe with a stable reference signal have an ultralow LOD of 0.26 μM for DPA in the range of 0–175 μM, converting the fluorescence color from dark blue to bright blue. Meanwhile, the developed probe was applied to the construction of logic gates and hydrogel-based film for the determination of PGA and DPA in real samples with the help of a smartphone.

The enormous potential of carbon dots (CDs) in agriculture has been widely reported, whereas their accurate distribution, transformation, and metabolic fate and potential soil health effects are not clearly understood. Herein, 13C-labeled CDs (13C-CDs) were sprayed on maize leaf, accumulated in all tissues, and promoted photosynthesis. Specifically, 13C-CDs were internalized to participate in the synthesis of glucose, sucrose, citric acid, glyoxylate, and chlorogenic acid, promoting tricarboxylic acid cycle (TCA) and phenylalanine metabolism. Additionally, the catabolism of 13C-CDs in vivo was mainly mediated by O2•– produced by oxidative stress. 13C-CDs did not have an obvious impact on the soil environment at the overall level. The detection of 13C signals in soil fauna suggested 13C-CDs in soil food chain transmission. This study systematically described the exact fate of CDs in plants and potential soil ecological risks and provided a more comprehensive analysis and support for the potential advantages of CDs in agricultural application.

Pharmacologic intervention in chronic heart failure (HF) with renal insufficiency is one of the clinical challenges due to the fact that the mechanisms of cardio-renal interactions in chronic heart failure (CHF) progressing have not been fully revealed. In this paper, C57BL/6 mice were applied thoracic aortic narrowing surgery to establish pressure overload CHF model. Cardiac function, serum markers, renal pathologic changes and kidney metabolism were analyzed at 4th, 8th, 12th, and 16th week after surgery respectively to evaluate the heart-Kidney pathologic overlap. Kidney proteomic analysis was performed at 16th week after operation. As a result, renal hypofiltration and exacerbation of pathological damage was observed accompanying cardiac function deterioration after 12th week. 66 differentially expressed proteins and 13 differential metabolites were found to be involved in the cardio-renal pathological overlap. Joint proteomic and metabolomic analysis revealed that signal pathways like Phosphatidylinositol signaling system, Glucagon signaling pathway, the Glyoxylate and dicarboxylate metabolism; DEPs of Pten, Mtmr4, PLC and CPT1, differential metabolites like aspartic acid and isocitrate deserve further investigation.

Summary The role of neurotransmitters in suppressing anti-tumor immunity has garnered increasing attention. While glutamate has been extensively studied in neurological diseases, its potential role in regulating anti-tumor immunity, particularly in the context of neoadjuvant immunochemotherapy, remains underexplored. In this study, we find that glutamate levels are elevated in the plasma of head and neck squamous cell carcinoma (HNSCC) patients. The METTL3/m6A/CD98 axis in cancer-associated fibroblasts (CAFs) is a key driver of glutamate secretion. Glutamate induces CD8 + T cell exhaustion through SLC1A3 and impairs the formation of immune memory in secondary lymphoid structures. Additionally, glutamate promotes ferroptosis resistance in HNSCC. Notably, glutamate depletion enhances the efficacy of neoadjuvant immunochemotherapy. Our findings provide insights into how the METTL3/m6A/CD98 axis-mediated regulation of glutamate efflux may sensitize HNSCC to neoadjuvant immunochemotherapy.

Simple SummaryEdwardsiella tardais a bacterium that causes severe disease in fish, leading to major economic losses for aquaculture worldwide. Understanding how this bacterium causes infection is essential for developing effective prevention strategies. In this study, we investigated a key bacterial enzyme called phosphoenolpyruvate carboxykinase (PEPCK), which helps control the bacterium’s energy metabolism. When we removed the gene responsible for producing this enzyme, the bacteria became less harmful to fish, grew faster, and showed increased movement. Importantly, this modified bacterium triggered a strong immune response in fish and protected them against subsequent infection with the deadly form of the bacteria. Fish that received the modified bacteria survived at much higher rates compared to unprotected fish. These findings reveal that this enzyme plays a critical role in controlling bacterial disease-causing ability through its effects on cellular metabolism. The modified bacteria show promise as a safe and effective vaccine candidate to protect farmed fish againstEdwardsiellainfections, potentially reducing disease outbreaks and economic losses in aquaculture while decreasing the need for antibiotic treatments.Understanding the metabolic basis of bacterial virulence is essential for developing anti-infective strategies. Phosphoenolpyruvate carboxykinase (PEPCK), encoded bypckA, is a key enzyme at the metabolic junction between the pyruvate cycle and the TCA cycle, but its role in virulence regulation remains largely unexplored. In this study, we generated apckAdeletion mutant,ΔpckA, inEdwardsiella tardaand investigated its impact on virulence, metabolism, and vaccine potential. TheΔpckAmutant exhibited increased antibiotic sensitivity, faster growth, reduced autoaggregation, and significantly attenuated virulence in a tilapia infection model. Quantitative proteomics revealed global downregulation of type III and type VI secretion systems (T3SS and T6SS) and upregulation of flagellar proteins. Mechanistically, accumulation of oxaloacetate (OAA) in the mutant suppressed T3SS/T6SS expression without affecting flagellar motility. Importantly,ΔpckAinduced a robust immune response in zebrafish and conferred strong protection against lethal challenge, with relative percent survival values of 64% and 81.82% in zebrafish and tilapia, respectively. Taken together, our findings identify PEPCK as a critical metabolic regulator ofE. tardavirulence and highlightΔpckAas a promising live attenuated vaccine candidate.

Crosslinking is one of the most effective ways to enhance the performance of bio-based films, and suitable crosslinking agents are crucial for the enhancement. In this study, four α-ketoacids, namely glyoxylate, pyruvate, oxaloacetate, and α-ketoglutarate were used to crosslink chitosan at room temperature. The effects of crosslinking on the structure and properties of chitosan films were studied, and the reaction mechanism was explored. Fourier Transform Infrared spectroscopy and X-ray Photoelectron Spectroscopy indicated that ion attraction and Schiff base reactions occurred between keto acids and chitosan. Glyoxylate developed the most effective covalent crosslinking with chitosan, whereas α-ketoglutarate had the highest ionic crosslinking ratio. Keto acid crosslinking reduced the orderliness of chitosan, improved the uniformity of the film matrix and increased its UV-blocking capacity. Glyoxylate-crosslinked chitosan film demonstrated excellent tensile strength (160 MPa), water stability (water solubility about 11.71 %), and extremely low oxygen permeability (2.65 × 10 −16 cm 3 ·cm/cm −2 ·s −1 · Pa −1 ). Despite the weakened thermal stability and water barrier property, glyoxylate crosslinking shows great potential for the preparation of high-strength and high‑oxygen-resistance chitosan films. Furthermore, the glyoxylate-crosslinked chitosan film could be produced by hot pressing and performed satisfactorily.

Hydroxylamine (NH2OH) is an important nitrogenous feedstock in the production of numerous valuable chemicals. The selective electroreduction of nitrate (NO3–) into NH2OH paves a promising avenue for the synthesis of NH2OH. Bi-based catalysts are regarded as the most promising candidates owing to the weak adsorption for NH2OH, preventing the further reduction of NH2OH. It is worth noting that Bi-based catalysts exhibited a dramatic activity gap in the acid and neutral/alkaline electrolytes toward the synthesis of NH2OH. Unfortunately, the mechanistic origin of pH-dependent NH2OH production has rarely been investigated, lacking an insightful interpretation on the atomic and molecular scale. Herein, taking Bi nanosheets (NSs) as a model catalyst, we investigated the underlying origin of the pH-dependent selectivity in NO3– electroreduction toward NH2OH production. The Faradaic efficiency (FE) for NH2OH of Bi NSs dramatically decreased from 86.6 to 12.0%, with the pH value ranging from 0 to 3. Combining the in situ Fourier transform infrared spectroscopy and theoretical calculations, we revealed that the dominant 4-coordinated hydrogen-bonded water in acid media favored the involvement of *H in the hydrogenation of *NO to *NHO, thereby facilitating the synthesis of NH2OH. Moreover, 10 kinds of amino acids were successfully synthesized by Bi NSs, demonstrating the wide universality of Bi nanosheets for the synthesis of amino acids.

BACKGROUND Sclerotinia stem rot (SSR) caused by Sclerotinia sclerotiorum threatens oilseed rape cultivation, and the emergence of fungicide-resistant strains has led to control failures worldwide. Identifying novel chemical alternatives with different modes of action and high antifungal activities is thus crucial. Herein we evaluated the antifungal effects of 3-(2-pyridyl)methyl-2-(4-chlorphenyl)imino- thiazolidine (PMAS) on S. sclerotiorum to determine its efficacy for SSR management. RESULTS PMAS had an inhibitory effect on mycelial growth; the EC 50 values were 17.83 and 21.15 μg mL −1 for the carbendazim-susceptible strain Ss01 and carbendazim-resistant strain Hm25, respectively. PMAS treatment changed the color of inhibited mycelia to green, and the hyphae were sustained in the undifferentiated stage. Cysteine supplementation made this green color disappear, whereas methionine enhanced the color. Moreover, PMAS treatment markedly inhibited oxalic acid biogenesis, increased free thiol content in mycelia, and weakened the activities of oxaloacetase and malate dehydrogenase, but had little effect on the activity of glyoxylate dehydrogenase. Cysteine could reverse the inhibitory effects of PMAS on mycelial morphogenesis and biochemical constituents, except thiol production. In the pot-culture experiment, PMAS showed a good protective effect, with the control efficacy being >91% on SSR. CONCLUSION PMAS appears to be an effective fungicide for SSR management. © 2020 Society of Chemical Industry

Metabolites in the body fluid are becoming a rich source of disease biomarkers. Developing an effective and high throughput detection and analysis platform of metabolites is of great importance for potential biomarker discovery and validation. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) has been successfully applied in rapid biomolecules detection in large scale. However, non-negligible background interference in low molecule-weight region still constitutes a main challenge even though various nanomaterials have been developed as an alternative to traditional organic matrix. In this work, a novel composite chip, silicon nanowires loaded with fluorinated ethylene propylene (FEP@SiNWs) was fabricated. It can serve as an excellent substrate for nanostructure-initiator mass spectrometry (NIMS) detection with ultra-low background noise in low molecular weight region (<500 Da). Ion desorption efficiency and internal energy transfer of FEP@SiNWs were studied using benzylpyridinium salt and tetraphenylboron salt as thermometer chemicals. The results indicated that a non-thermal desorption mechanism might be involved in the LDI process on FEP@SiNWs. Owing to the higher LDI efficiency and low background interference of this novel substrate, the metabolic fingerprint of complex bio-fluids, such as human saliva, can be sensitively and stably acquired. As a proof of concept, FEP@SiNWs chip was successfully used in the detection of salivary metabolites. With the assistance of multivariate analysis, 22 metabolic candidates (p < 0.05) which can discriminate type 2 diabetes mellitus (2-DM) and healthy volunteers were found and identified. The role of these feature metabolites in the metabolic pathway involved in 2-DM was confirmed by literature mining. This work demonstrates that FEP@SiNWs-based NIMS might be served as an efficient and high throughput platform for metabolic biomarker exploration and clinical diagnosis.

Toxic heavy metals have been considered to be harmful environmental contaminations. The molecular mechanisms of heavy-metals-induced cytotoxicity and carcinogenicity are still not well elucidated. Previous reports showed exposures to toxic heavy metals can cause a change of DNA cytosine methylation (5-methylcytosine, 5-mC). However, it is still not clear whether heavy metals have effects on the recently identified new epigenetic marks in both DNA and RNA, i.e., 5-hydroxymethylcytosine (5-hmC), 5-formylcytosine (5-foC), and 5-carboxylcytosine (5-caC). Here, we established a chemical labeling strategy in combination with liquid chromatography–electrospray ionization–mass spectrometry (LC-ESI-MS/MS) analysis for highly sensitive detection of eight modified cytidines in DNA and RNA. The developed method allowed simultaneous detection of all eight modified cytidines with improved detection sensitivities of 128–443-fold. Using this method, we demonstrated that the levels of 5-hmC, 5-foC, and 5-caC significantly decreased in both the DNA and RNA of mouse embryonic stem (ES) cells while exposed to arsenic (As), cadmium (Cd), chromium (Cr), and antimony (Sb). In addition, we found that treatments by heavy metals induced a decrease of the activities of 10–11 translocation (Tet) proteins. Furthermore, we revealed that a content change of metabolites occurring in the tricarboxylic acid cycle may be responsible for the decline of the derivatives of 5-mC. Our study shed light on the epigenetic effects of heavy metals, especially for the induced decline of the derivatives of 5-mC in both DNA and RNA.
