Bletilla striata polysaccharide (BSP) and chitosan (CS) were chemically cross-linked using oxalyl chloride to prepare a composite hemostatic sponge (BSP-CS), and the process parameters were optimized using the Box–Behnken design (BBD) with response surface methodology. To optimize the performance of the hemostatic sponge, we adjusted the ratio of independent variables, the amount of oxalyl chloride added, and the freeze-dried volume. A series of evaluations were conducted on the hemostatic applicability of BSP-CS. The characterization results revealed that BSP-CS had a stable bacteriostatic effect on Escherichia coli , Staphylococcus aureus , and Pseudomonas aeruginosa within 72 h, and the bacteriostatic rate was above 30%. The CCK-8 cytotoxicity test demonstrated that BSP–CS had a certain effect on promoting cell proliferation of L929 cells. In the mouse tail-cutting experiment, the hemostasis time of BSP–CS was 463.0 ± 38.16 s, shortened by 91.3 s on average compared with 554.3 ± 34.67 s of the gauze group. The blood loss of the BSP–CS group was 28.47 ± 3.74 mg, which was 34.7% lower than that of the control gauze group ( 43.6 ± 3.83 mg). In the in vitro coagulation experiment, the in vitro coagulation index of the BSP–CS group was 97.29 % ± 1.8 % , which was reduced to 8.6% of the control group. The CT value of the BSP–CS group was 240 ± 15 s, which was 155 s lower than that of the gauze group ( 355 ± 31.22 s). All characterization results indicate that BSP–CS is an excellent hemostatic material.

Oil-water separation is a vital process across various industries, where membrane separation stands out as a promising, energy-efficient solution. Despite its potential, the widespread application is hindered by the limited durability of existing membrane surface engineering effects. This study introduces an innovative fabrication strategy to create a superhydrophobic and durable nanofibrous membrane for efficient oil-water separation. Utilizing electrospinning and a two-step sequential covalent-bonding dip-coating process, the membrane was chemically decorated with vinyl trichlorosilane, followed by a thiol-ene “Click” reaction with 1H, 1H, 2H, 2H-perfluorodecanethiol, resulting in a stable superhydrophobic and oleophilic surface. The membrane exhibited high permeation fluxes for various organic solvents, achieving separation efficiencies exceeding 99.5 % for non-emulsified mixtures (oil:water = 1:1) and over 99.7 % for hydrocarbon oil/water emulsions (oil:water = 99:1). Furthermore, the membrane demonstrated remarkable stability, enduring multiple filtration cycles and harsh chemical environments, including extended exposure to NaOH solutions. To validate its industrial applicability, the nanofibrous membrane was employed in a membrane extraction process post amide synthesis, successfully separating dichloromethane and water, thereby streamlining the extraction process. This work showcases a robust membrane surface engineering strategy for efficient oil-water separation.

Phase change materials (PCMs) can store thermal energy in the form of latent heat. However, traditional PCMs suffer from spontaneous solidification and uncontrolled heat release in the absence of effective insulation or continuous energy input, severely limiting their practical application. To address this, optically-controlled PCMs have emerged as a novel solution, developed by integrating molecular photoswitches into traditional PCM matrices. Solar energy can be converted and stored in the form of photochemical enthalpy and phase change enthalpy, and the thermophysical properties of the materials can be changed through photoisomerization. In this paper, 4-octanoyloxyazobenzene (4-OcOAzo), an azophenyl photo controlled PCM, was prepared experimentally. The phase transition temperatures for the cis and trans isomers were determined to be 294.37 K and 335.76 K, respectively. The material exhibits an energy storage density of 139.356 kJ/kg, and the thermal conductivity was 0.292 W/(m·K) at room temperature. Subsequently, a microscopic model of 4-OcOAzo was established, and its thermophysical properties were analyzed based on molecular dynamics. The calculated results show that the phase transition temperature of the trans sample is 333 K, the phase transition temperature of the cis sample is 296.41 K, and the thermal conductivity at room temperature is 0.282 ± 0.017 W/(m·K), which is highly consistent with the experimental results. In addition, through the analysis of microstructure parameters, the mechanism of optically controlled phase transition can also be clarified.

Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation (LPO), has emerged as a promising strategy for cancer therapy but remains limited by insufficient hydrogen peroxide (H₂O₂), restricted catalytic iron availability, and the extremely short lifetime of hydroxyl radicals (·OH) in the tumor microenvironment. Here, we report a self-assembled nano-prodrug designed to amplify oxidative stress through coordinated reactive oxygen species (ROS)-reactive nitrogen species (RNS) cascades. Transcriptomic analyses of melanoma cohorts revealed that the redox enzyme NAD(P)H: quinone oxidoreductase-1 (NQO1) is frequently upregulated and associated with oxidative stress and ferroptosis-related transcriptional programs, suggesting a potential redox vulnerability that can be therapeutically exploited. Guided by this observation, a glutathione-responsive nanoplatform (HFSAL) was constructed from a disulfide-bridged ferrocene–arginine scaffold (Fc-SS-Arg) with β-lapachone (β-lapa) co-loaded and hyaluronic acid coating for tumor-targeted delivery. Upon intracellular activation, β-lapa undergoes NQO1-mediated redox cycling to generate H₂O₂, which is further converted into ·OH through ferrocene-catalyzed Fenton reactions. Meanwhile, arginine-derived nitric oxide (NO) reacts with ROS intermediates to form highly reactive peroxynitrite (ONOO⁻), thereby amplifying oxidative damage and extending the effective range of redox stress. In addition, NO-mediated vascular remodeling alleviates tumor hypoxia and enhances intratumoral drug accumulation. Consequently, the HFSAL nano-prodrug effectively induces synergistic ferroptosis and apoptosis, leading to significant tumor growth inhibition and metastasis suppression in vivo with favorable biosafety. This work presents a redox-amplifying nano-prodrug strategy that exploits NQO1-associated metabolic vulnerability to enhance ferroptosis-based cancer therapy. Graphical

Combretastatin A4 (CA4) is a potent microtubule inhibitor, but its phenolic hydroxyl group is prone to oxidation into toxic quinone products, leading to poor metabolic stability and dose-limiting toxicity that restricts its clinical application. To overcome this limitation, we propose a novel dimer prodrug strategy that covalently links two CA4 molecules via an enzyme-cleavable linker, achieving temporary masking of the phenolic hydroxyl group. The resulting CA4-dimer1 and CA4-dimer2 exhibited potent in vitro antiproliferative activity. Of the two dimers evaluated, CA4-dimer1 demonstrated superior in vitro potency. Crucially, CA4-dimer1 achieved significant tumor suppression rates in patient-derived xenograft (PDX) liver cancer models without notable toxicity. CA4-dimer2 releases active CA4 through gradual hydrolysis but showed slightly lower in vivo release efficiency compared to CA4-dimer1. Flow cytometry and Western blot analysis revealed that this prodrug exerts antitumor activity through the same mechanism as the parent compound CA4. Collectively, the reported CA4-dimer prodrug strategy offers a highly promising approach for treating liver cancer and enhancing the therapeutic index of phenolic drugs, while also providing a potential strategy for structural optimization of drugs containing phenolic hydroxyl groups

Light-excited dual-mode imaging that integrates multispectral optoacoustic tomography (MSOT) and near-infrared region II fluorescence (NIR-II FL) imaging allows complementary deep-tissue visualization with high anatomical resolution and molecular sensitivity, thereby enhancing the accuracy of biomedical diagnostics. Activatable probes further advance this approach by producing signals upon recognition of disease-related biomarkers, which reduces background interference and improves imaging specificity. BTPE-NO 2 @F127, a benzothiadiazole-based nanoprobe, exhibits selective activation of both optoacoustic and NIR-II FL signals in response to hydrogen peroxide (H 2 O 2 ), a crucial early stage biomarker of liver injury, thus permitting cross-validated detection of hepatic damage in vivo with excellent signal-to-background ratio contrast. Here we present detailed procedures for preparation of the BTPE-NO 2 @F127 nanoprobe and its applications in MSOT/NIR-II FL dual-mode imaging of trazodone- or ischemia–reperfusion-induced liver injury in mice. Compared to conventional liver injury diagnostic methods, such as invasive tissue biopsy, ex vivo blood analysis and previously reported fluorescent/optoacoustic probes, BTPE-NO 2 @F127 offers real-time, in situ monitoring with high sensitivity and signal-to-background ratio, as well as mutually corroborating signals for increased reliability. The fabrication of BTPE-NO 2 @F127, including the chemical synthesis and characterization, requires ~17 d, while the in vitro validation of its H 2 O 2 responsiveness takes ~5 d. Notably, the complete workflow of data acquisition and analysis for MSOT/NIR-II FL dual-mode imaging of liver injury in mice using BTPE-NO 2 @F127 can be accomplished within 10 h. The protocol is easy to follow and suitable for clinicians and researchers with a basic understanding of chemistry and bioimaging techniques.

Due to its widespread usage but high toxicity, the detection of cyanide has drawn special attention. However, in many works usually only one colorimetric or fluorescent detector was put forward for chemical sensing. Here in this contribution, two kinds of polydiacetylene (PDA) derivatives substituted by either o -hydroxybenzaldehyde or dicyanovinyl group were designed and explored for the optical sensing of cyanide. Firstly, these two PDA derivatives were polymerized in the form of vesicle individually and showed chromatic transition from blue to purple or red after the addition of cyanide into the aqueous solution. Then polyvinylidene fluoride membrane was chosen to incorporate the PDAs to form strip-based solid sensors, which could exhibit obvious blue-to-red colorimetric response upon exposure to cyanide aqueous solution with different concentrations in an orthogonal way. Interestingly, these two PDA derivatives remained blue phase in the presence of many common anions, thus demonstrating excellent selectivity for the distinguishment of cyanide. Besides the experimental characterization methods, such as UV–visible absorption and Raman spectroscopy, to study the underlying chemical mechanism, the principal component analysis method was also applied for the multivariate statistical analysis in combination with RGB (Red, Green, Blue) values to obtain a deep insight into the chromatic sensing of PDAs.

Polyurea emerges as a promising material for acid nanofiltration membranes due to its high hydrogen-bonding capacity and low electron density from carbonyl groups. However, conventional polyurea-based composite membranes suffer from limited separation performance and acid stability due to insufficient hydrophilicity and conjugated structures. In this study, a novel monomer, 3,5-diisocyanate benzoyl chloride, was synthesized and employed as the organic-phase precursor in interfacial polymerization to fabricate thin-film composite (TFC) membranes with polyurea-amide active layers. During polymerization, the acyl chloride groups undergo partial hydrolysis, introducing carboxylate anions into the active layer. The NF membrane shows over 98.6% rejection of MgSO 4 at pH 7, and membrane water permeability can reach 3.41 L m −2 h −1 bar −1 . Additionally, the aqueous phase monomer 2,4-diaminobenzenesulfonic acid was incorporated to construct a fully aromatic active layer, which strengthens intermolecular conjugation and acid resistance. Comparative analysis demonstrated that the synthesized membranes outperform traditional polyurea-based membranes (prepared via toluene diisocyanate and fatty amines) in acid stability: after soaking in 20 wt% H 2 SO 4 for 100 days, the MgSO 4 rejection remained nearly unchanged. This study establishes a molecular design strategy integrating hydrophilic functionalization and conjugated aromatic structures to address critical limitations in polyurea membranes, offering a pathway toward robust nanofiltration systems for acid-resistant applications.

The growing environmental crisis caused by petroleum-based polymers has intensified the development of sustainable alternatives, with many biomass-derived polymers demonstrating potential for degradability, renewability, and low carbon footprints, though these properties can vary depending on structure and processing. However, traditional bio-based systems often lack tunability in mechanical properties, making it challenging to achieve both high strength and ductility. Herein, we report a high-performance, recyclable bio-based film (CAF-L) constructed via Diels-Alder dynamic covalent chemistry between furfuryl-functionalized cellulose acetate and maleimide-modified lignin. Thermally responsive dynamic Diels-Alder bonds, activated through heat-treatment, enable programmable network crosslinking that allows a smooth transition between strength- and ductility-dominated regimes, while maintaining high mechanical performance (tensile strength up to 52.3 MPa and elongation at break up to 545%). Structural characterization and molecular simulations reveal that Diels-Alder bond dynamics drive thermally induced structural reorganization of the polymer network, imparting rare adaptivity to biomass-based systems. In addition, CAF-L films exhibit outstanding UV shielding, oxygen barrier properties, and dual-mode recyclability through solvent dissolution and hot pressing. This work provides a scalable platform for constructing mechanically tunable, structurally reconfigurable, and environmentally resilient cellulosic bioplastic for sustainable packaging and circular material systems.

Natural biological tissues exhibit different mechanical and surface properties. These disparate features make their connections with engineering materials quite difficult due to the lack of universal methods for tuning the interfacial bonding over a wide range. However, the precise control of interfacial properties, including modulus and adhesion on diverse biological tissues, requires overcoming multiple inherent and external barriers. Here we propose an interface-enhanced strategy by spatial and temporal anchoring of magnetic nanostickers for controlled bioadhesive properties. Fully exploiting the interactions from nanostickers by remote control enables the attached patch to achieve extremely high adhesion energy ( ~ 1250 J m -2 ) and interfacial fatigue resistance with a threshold of ~50 J m -2 , at a very low area density of nanostickers (4 μg/mm 2 ). The controlled interfacial properties as well as space and time for anchoring, lead to comprehensively tunable bioadhesion on diverse tissues such as skin, intestine, liver, and kidney, which are strongly desired in biomedical applications. Integration with fragile tissues in female Sprague-Dawley rats for 10 days further demonstrates that the anchored biointerface can adapt to the in vivo environment and promote postoperative recovery. The biointerface bridged by intelligent nanostickers prompts the methodology for bioadhesion towards controllable orientation.

Despite the development of numerous prodrugs for tumor therapy, their clinical application remains limited due to issues such as non-targeted delivery, uncontrolled release, and excessively rapid drug release rates. In this study, we synthesized a dual-functional prodrug, glycyrrhizic acid-polyethylene glycol-disulfide curcumin (GA-PEG-SS-CUR), by conjugating glycyrrhizic acid (GA), which possesses liver-targeting capabilities, with the anticancer agent curcumin (CUR). GA-PEG-SS-CUR can self-assemble in aqueous solutions to form micelles with an approximate diameter of ∼140 nm. In vitro drug release studies demonstrated that GA-PEG-SS-CUR exhibits a sensitive glutathione (GSH) response and sustained release characteristics; after 72 hours, the drug release rate was 30.5% at low GSH concentrations and 46.7% at high GSH concentrations. Due to GA receptor-mediated targeting, the uptake of GA-PEG-SS-CUR in HepG2 cells was significantly higher than that in L02 cells, and the internalization of GA-PEG-SS-CUR in HepG2 cells was markedly greater compared to unmodified PEG-SS-CUR. Cytotoxicity assays indicated that GA-PEG-SS-CUR enhanced the cytotoxicity against tumor cells compared to free CUR, while exhibiting lower toxicity towards normal L02 cells. Pharmacokinetic studies revealed that the half-life of CUR in GA-PEG-SS-CUR was 8.3 times longer than that of free CUR, extending the in vivo circulation time, with CUR accumulation at the tumor site being 6 times greater than that of free CUR, indicating significant tumor-homing ability. Therefore, the developed GA-PEG-SS-CUR represents an ideal delivery system for curcumin in tumor therapy.

Background/Objectives:Ulcerative colitis (UC) involves chronic colon inflammation and oxidative stress. Treating UC is challenging due to systemic drug side effects and poor targeted delivery. Nanocarriers responsive to the UC microenvironment, particularly elevated reactive oxygen species (ROS), could overcome these limitations. This study developed an oral delivery system for ROS-triggered drug release and active targeting. Using ferulic acid (FER), a system was designed to enhance site-specific accumulation and therapeutic efficacy against colitis. Methods:A ROS-sensitive covalent organic framework (COF) was synthesized from γ-cyclodextrin and functionalized with folic acid (FA) to create a carrier (COF-FA) designed for potential active targeting. This carrier was loaded with FER to form FER@COF-FA. The system was characterized (SEM, FTIR, TGA), and its ROS scavenging and sustained drug release profiles were confirmed in vitro. Biocompatibility was evaluated in cell lines, and therapeutic efficacy was tested in a DSS-induced murine colitis model. Results:The synthesized FER@COF-FA demonstrated high drug loading, potent ROS-scavenging capability, and a sustained drug release profile. It showed excellent biocompatibility and, in the murine model, significantly outperformed free FER. Treatment alleviated disease severity, prevented colon shortening, restored healthy tissue histology, and rebalanced pro- and anti-inflammatory cytokines. Conclusions:The FER@COF-FA system represents a highly promising therapeutic strategy for UC. Its superior efficacy is attributed to a synergistic multi-mechanism approach, combining sustained release, ROS-responsive drug delivery, intrinsic antioxidant activity, and potential folate receptor-mediated targeting, which collectively enhance site-specific accumulation and therapeutic outcomes in the inflammatory colon microenvironment.
