A rapid and sensitive liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed to simultaneously determine berberine, palmatine, coptisine, epiberberine and jatrorrhizine in rat plasma, using tetrahydropalmatine as an internal standard. The separation of the five compounds was carried out on a C18 column using a mobile phase consisting of acetonitrile and water (containing 5 mmol ammonium acetate adjusted to pH 5.0 with formic acid). The detection was performed in the multiple reaction monitoring (MRM) mode via the electrospray ionization (ESI) source operating in the positive ionization mode. Linear calibration curves were obtained over the concentration range of 0.20–200.0 ng ml−1 for berberine, and 0.20–100.0 ng ml−1 for palmatine, coptisine, epiberberine and jatrorrhizine. The accuracy, precision and extraction recovery of the five analytes were all satisfactory. The validated method was successfully applied to study the pharmacokinetic differences of the five protoberberine-type alkaloids after the oral administration of Rhizoma coptidis (RC) and Jiao-Tai-Wan (JTW) extract. It was found that the Tmax values of the five ingredients in the JTW extract were significantly increased (P < 0.05) when compared with those in RC. In addition, the AUC0–t of palmatine, coptisine and epiberberine were significantly increased (P < 0.05).

The determination of isoquinoline alkaloids (IQAs) is crucial for maximizing the pharmacological benefits of Rhizoma coptidis (RC). In this study, the dendritic silica sphere (dSiO 2 ) was covalently modified with cyanuric chloride (CNCl) to synthesize a porous dNSiO 2 -CNCl NP. The fluorescence of berberine (Ber), palmatine (Pal), coptisine (Cop), and jatrorrhizine (Jat) was remarkably enhanced by dNSiO 2 -CNCl NP for at least 38-fold, inspiring the accurate determination of the isoquinoline total alkaloids (IQTAs) in RC using fluorometry. The method exhibited good linearity within the concentration range of 0.005 to 1 μM, with a limit of detection (LOD) and quantitation (LOQ) of 0.001 and 0.004 μM. Specially, Jat was discovered to form a stable covalent linkage with dNSiO 2 -CNCl, showing an innovative and enduring fluorescent nanoparticle derived from a natural product. Furthermore, using dNSiO 2 -CNCl as a substrate for fluorescence enhancement and a dispersive solid-phase extraction (d-SPE) sorbent, a fluorescence-mediated HPLV-UV method was developed to monitor Ber, Pal, and Cop in serum. Rapid qualitative analysis using fluorescence was initially performed. In chromatography analysis, the linear range for Cop was 0.20 to 10 μM, with LOD and LOQ at 0.035 and 0.110 μM, while Pal and Ber showed good linearity in the range of 0.05 to 10 μM, with LODs and LOQs at 0.013 and 0.040 μM. The spiked recovery rates ranged from 90.4% to 99.8% for the three IQAs, with intra-day precision below 7.9% and inter-day precision below 9.9%. The dual-functional dNSiO 2 -CNCl NP exhibited promising prospect in fluorescence sensing and separation applications for detecting drugs.

Rhizoma coptidis has been clinically used for a long time for the treatment of various diseases in China, such as hypertension, diabetes, and inflammation. Previous studies have shown that alkaloid components of Rhizoma coptidis extract could be extensively metabolized and the metabolites were also considered to be the therapeutic material basis. However, until now, pharmacokinetic studies of the in vivo metabolites have not been revealed yet. The aim of the present study was to characterize the pharmacokinetics and excretions of five main alkaloids (berberine, jatrorrhizine, palmatine, epiberberine, and coptisine) and their seven metabolites (berberrubine, demethyleneberberine, jatrorrhizine-3-O-β-D-glucuronide, thalifendine-10-O-β-D-glucuronide, berberrubine-9-O-β-D-glucuronide, demethyleneberberine-2-O-sulfate, and demethyleneberberine-2-O-β-D-glucuronide) in rats after oral administration of Rhizoma coptidis extract. Meanwhile, comparative pharmacokinetics and excretions of these analytes in diabetic model rats were also investigated, since Rhizoma coptidis is widely used for the treatment of diabetes. Our results showed that the in vivo existing forms of alkaloid components were phase II metabolites, highlighting the glucuronidation metabolic pathway. In diabetic model rats, the utilization of Rhizoma coptidis alkaloids was significantly increased and the biotransformation of berberine into berberrubine was significantly inhibited.

Ethnopharmacological relevance Jiao-Tai-Wan (JTW), an important herbal formula consists of Rhizoma coptidis and Cortex cinnamomi powder, is a famous prescription which has been used for centuries to treat insomnia in Traditional Chinese Medicine . The purpose of this study is to compare the pharmacokinetic properties of five protoberberine-type alkaloids (i.e. berberine , palmatine , coptisine , epiberberine and jatrorrhizine), the main bioactive constituents in JTW, between normal and insomnic rats. We also investigate the differences between single-dose and multiple-dose pharmacokinetics of five protoberberine-type alkaloids. Materials and methods The insomnic rat models were induced by intraperitoneal injection of one-dose para-chlorophenylalanine acid (PCPA). Quantification of five protoberberine-type alkaloids in rat plasma was achieved by using a rapid LC–MS/MS method. Plasma samples were collected at different time points to construct pharmacokinetic profiles by plotting drug concentration versus time and estimate pharmacokinetic parameters . An unpaired Student׳s t test was used for comparisons with SPSS 17.0. Results The five protoberberine-type alkaloids of single-dose normal groups had slow absorption and low bioavailability, as well as a delay of peak time. In the single-dose oral administration, the C max and T max of five ingredients in insomnic rats had significant differences compared with those of normal rats. In the multiple-dose oral administration, the pharmacokinetic parameters of five protoberberine-type alkaloids varied greatly in insomnic rats. In the normal rats, there were significant differences ( P <0.05) in the principal pharmacokinetic parameters such as C max and T max between single-dose and multiple-dose oral administration. In the insomnic rats, the five ingredients of multiple-dose groups showed better absorption than the single-dose groups. Particularly, three peaks were observed in multiple-dose model group of plasma–concentration curves. Conclusions The pharmacokinetic behavior of five protoberberine-type alkaloids was described in this paper. In both normal groups and model groups, the pharmacokinetic behavior of multiple-dose had significant differences comparing with the single-dose; either single-dose or multiple-dose, the pharmacokinetic behavior of insomnic rats had significant differences comparing the normal rats. Multiple dosing may improve the absorption of JTW in insomnic rats, which will increase the bioavailability and bring into active role in therapeutical effect.

Coptis chinensis Franch. and Sophora flavescens Ait. is a herbal pair frequently used in treating ulcerative colitis. However, the bio-disposition profile of the major components in the inflamed gut remains unclear, which is essential to understand the pharmacological material basis of this herb pair. Here we established an integral quantitative and chemometric method to deduce the colonic metabolism differences of this herbal pair in normal and colitis mice. With this LC-MS method, a total of 41 components have been found in the Coptis chinensis Franch. and Sophora flavescens Ait. extract, and 28 metabolites were found in the colon after oral administration. Alkaloid and its phase I metabolites were the main components in the colon of normal and colitis mice. The results of principal component analysis at 6 h after oral administration showed significant colonic metabolism differences between normal and colitis mice. Heamap results showed that colitis induced significant changes in the colonic bio-disposition of this herbal pair extract. In particular, in the context of colitis, the phase I metabolism of berberine, coptisine, jatrorrhizine, palmatine, and epiberberine has been inhibited. These results may provide a basis for understanding the pharmacological material basis of Coptis chinensis Franch. and Sophora flavescens Ait. in treating ulcerative colitis.

Berberine (BBR) has seen growing application in ophthalmology, yet the ocular toxicity of BBR and its metabolites remains poorly understood. This study aimed to evaluate the ocular toxicity of BBR and its major metabolite M1 and unravel their underlying mechanisms. Ocular toxicity was evaluated in human corneal epithelial cells and wild-type AB zebrafish. Mechanistic studies utilized fluorescence imaging, biochemical quantitative assays, and qPCR analyses in AB zebrafish and transgenic mitochondrial fluorescent zebrafish (strainTg(Xla.Eef1a1:mlsEGFP)). Both BBR and M1 induced significant ocular toxicity across models, with BBR showing higher toxicity than M1. Mechanistic analyses revealed their toxicity stemmed from photoreceptor cell damage and Sirtuin 3 (SIRT3) inhibition, triggering a cascade of pathological events: mitochondrial dysfunction, oxidative stress, autophagic dysfunction, apoptosis, and inflammation. This study provides a reference for individualized risk assessment and clinical management of BBR-based therapies and paves the way for developing BBR derivatives with reduced ocular toxicity.

Protoberberine alkaloids are an important class of isoquinoline alkaloids with diverse potent pharmacological activities. The rapid, label-free, and sensitive detection of these chemical compounds is of great significance for quality control, pharmacological research, and clinic medical assays. However, common detection methods such as high-performance liquid chromatography have the shortcomings of time-consuming and strict operation. Here, a rapid and label-free method was developed for sensitive detection of protoberberine alkaloids. Raman spectra of four protoberberine alkaloids were recorded. Three of them exhibited strong fluorescence backgrounds, and the results revealed that the near-infrared laser could effectively reduce the fluorescence signals. The four protoberberine alkaloids showed distinct Raman peaks and intensity ratios, which could be used to identify them. Furthermore, a plasmonic sensor was prepared for detection of berberine with 0.1-mM sensitivity. The relative standard deviations of the SERS intensity were small, indicating the favorable spectral reproducibility. Besides, the SERS intensities measured in the first and seventh week were comparable, demonstrating the excellent stability of the plasmonic sensor, which may be attributed to the stable gold nanoparticles and film used. This rapid, fingerprint, and sensitive method provides a powerful new tool for protoberberine alkaloids detection.

A capillary-electrophoresis-based method for the screening of tyrosinase inhibitors in traditional Chinese medicines was developed. The method integrated electrophoretically mediated microanalysis with sandwich mode injection, partial filling, and rapid polarity switching techniques, and carried out on-column enzyme reaction and the separation of substrate and product. The conditions were optimized including the background electrolyte, mixing voltage, and the incubation time. Finally, the screening of nine standard natural compounds of traditional Chinese medicines was carried out. The inhibitors can be directly identified from the reduced peak area of the product compared to that obtained without any inhibitor. Chlorogenic acid (100 μM) showed inhibitory activity with the inhibitory percentage of 19.8%, while the other compounds showed no inhibitory activity. This method has great application potential in drug discovery from traditional Chinese medicines.

Cucurbit[7]uril (CB7), one host macromolecule, possesses a hydrophobic inner cavity and two identical hydrophilic carbonyl fringe portals, thus interacting with a variety of guest molecules through hydrophobic, hydrogen bonding and ion–dipole interactions. These characteristics make CB7 a good candidate as the stationary phase (SP) modifier in high performance liquid chromatography (HPLC). In the present work, CB7 was immobilized onto silica (CB7-SiO 2 ) by a sol–gel approach. The obtained CB7-SiO 2 was characterized by Fourier transform infrared spectroscopy, thermogravimetric analysis and elemental analysis. The chromatographic performance of CB7-SiO 2 was then studied in detail, in both reversed phase (RP) and hydrophilic interaction liquid chromatographic (HILIC) modes. For probe aromatic compounds and dyes, the typical RP behavior was observed, mainly due to the predominant hydrophobic interaction between the analytes and CB7-SiO 2 . For probe alkaloids, the “U-shape” curves were obtained; in the methanol-rich part, HILIC performance was obvious, probably owing to hydrogen bonding and ion–dipole interactions. Compared to bare SiO 2 SP, the stronger retention and different selectivity were observed on CB7-SiO 2 . The CB7-SiO 2 SP remained stable within at least 11,000 column volumes of the MP, revealing the satisfactory stability of it. The proposed method integrated the advantages of CB7 and sol–gel method provided a feasible way for the wider application of CBs in separation science.

In the present study, one of the new generation of host molecules, cucurbit(6)uril (CB(6)), was immobilized onto silica (CB(6)/SiO 2 ) by a sol–gel approach. CB(6)/SiO 2 was characterized by NMR spectroscopy, Fourier transform infrared spectroscopy, thermogravimetric analysis, and elemental analysis. It was used as a high-performance liquid chromatographic stationary phase and its chromatographic performance was systematically investigated with different types of analytes as probes. The results revealed that the CB(6)/SiO 2 stationary phase exhibited weak hydrophobic and strong hydrophilic properties. Hence, the variables for hydrophilic interaction liquid chromatography, including components and pH of the mobile phase, were further investigated to explore the retention mechanism of this CB(6)/SiO 2 stationary phase. For less polar analytes, both hydrophobic and hydrophilic interactions could contribute to the retention, while for polar analytes, hydrophilic interaction may be predominant. Compared to the tetraethoxylsilane-coated SiO 2 stationary phases, the CB(6)/SiO 2 stationary phase exhibited a different retention behavior toward basic analytes with excellent stability. It is a novel promising hydrophilic interaction liquid chromatography stationary phase.

Exploiting the specific recognition probe for all of the biomolecules is difficult in “lock-and-key” biosensors. The cross-reaction or the semispecific probe in pattern recognition mode is an alternative strategy through extracting a multidimensional signal array from recognition elements. Here, we design a pattern recognition sensor array based on the alkaloid-inhibited catalytic activity of dopzymes for the discrimination and determination of six alkaloids. In this sensor array, three different G-rich sequences, i.e., G-triplex (G3), G-quadruplex (GQ1), and the G-quadruplex dimer (2GQ1) possessing various peroxidase activities, conjugated with a dopamine aptamer and the dopzymes (G3-d-apt, GQ1-d-apt, and 2GQ1-d-apt) were obtained with an enhanced catalytic performance for the substrate. Through the interactions between six target alkaloids and G3, GQ1, and 2GQ1 regions, the pattern signal (6 alkaloids × 3 dopzymes × 5 replicates) was obtained from the diverse inhibited effect for the dopzyme activity. In virtue of the statistical method principal component analysis (PCA), the data array was projected into a new dimensional space to acquire the three-dimensional (3D) canonical scores and grouped into their respective clusters. The sensor array exhibited an outstanding discrimination and classification capability for six alkaloids with different concentrations with 100% accuracy. In addition, the nonspecific recognition elements of the sensor array showed high selectivity even though other alkaloids with similar structures to targets existed in the samples. Importantly, the levels of the six targets can be analyzed by the most influential discrimination factor, which represented the vector with the highest variance, evidencing that the sensor array has potential in drug screening and clinical treatment.

In this work, a novel magnetic nanomaterial functionalized with a molecularly imprinted polymer was prepared for the extraction of protoberberine alkaloids. Molecularly imprinted polymers were made on the surface of Fe 3 O 4 nanoparticles by using berberine as template, acetonitrile/water as porogen, acrylamide as functional monomer and ethylene glycol dimethacrylate as cross-linker. The optimized molar ratio of template/functional monomer was 1:7. The polymeric magnetic nanoparticles were characterized by transmission electron microscopy and Fourier transform infrared spectroscopy. The stability and adsorption capacity of the molecularly imprinted polymers were investigated. The molecularly imprinted polymers were used as a selective sorbent for the magnetic molecularly imprinted solid-phase extraction and determination of jatrorrhizine, palmatine, and berberine. Extraction parameters were studied including loading pH, sample volume, stirring speed, and extraction time. Finally, a magnetic molecularly imprinted solid-phase extraction coupled to high-performance liquid chromatography method was developed. Under the optimized conditions, the method showed good linear range of 0.1–150 ng/mL for berberine and 0.1–100 ng/mL for jatrorrhizine and palmatine. The limit of detection was 0.01 ng/mL for berberine and 0.02 ng/mL for jatrorrhizine and palmatine. The proposed method has been applied to determine protoberberine alkaloids in Cortex phellodendri and rat plasma samples. The recoveries ranged from 87.33–102.43%, with relative standard deviation less than 4.54% in Cortex phellodendri and from 102.22–111.15% with relative standard deviation less than 4.59% in plasma.
