Sorafenib-mediated cleavage of p62 initiates cellular senescence as a mechanism to evade its anti-hepatocellular carcinoma efficacy

Du Jiaying,Bai Dongsheng,Gu Chunyang,Zhao Jiawei,Zhou Chen,Wang Yuxiang,Zhao Yue,Lu Na

Hepatocellular carcinoma (HCC) stands as one of the most aggressively advancing and lethal malignancies. Sorafenib is presently endorsed as a primary therapy for advanced liver cancer, but its resistance presents a formidable challenge. Previous studies have implicated a connection between post-sorafenib discontinuation rebound and the development of drug resistance, yet the underlying mechanism remains elusive. In this study, we discerned that Sorafenib induced a senescent phenotype in HCC cells and caused a cleavage of ubiquitin-binding protein p62. Mechanistic studies establish that truncated p62 drives cellular senescence by promoting proteasome-dependent degradation of 4EBP1. Furthermore, truncated p62 induced specific ubiquitination of 4EBP1. Crucially, virtual drug screening uncovered that dacinostat inhibited cellular senescence by blocking sorafenib-induced p62 cleavage. In summary, our findings imply that truncated p62 from sorafenib cleavage promotes senescence via 4EBP1 degradation. The prevention of p62 cleavage could emerge as a crucial strategy for impeding the sorafenib-induced cellular senescence.

Quantitative proteomics identifies FOLR1 to drive sorafenib resistance via activating autophagy in hepatocellular carcinoma cells

Chu Hongwei,Wu Changqing,Zhao Qun,Sun Rui,Yang Kuo,Zhao Baofeng,Liu Yang,Liang Zhen,Zhong Shijun,Zhang Lihua,Zhang Yukui

Sorafenib is commonly used to treat advanced human hepatocellular carcinoma (HCC). However, clinical efficacy has been limited by drug resistance. In this study, we used label-free quantitative proteomic analysis to systematically investigate the underlying mechanisms of sorafenib resistance in HCC cells. A total of 1709 proteins were confidently quantified. Among them, 89 were differentially expressed and highly enriched in the processes of cell–cell adhesion, negative regulation of apoptosis, response to drug and metabolic processes involving in sorafenib resistance. Notably, folate receptor α (FOLR1) was found to be significantly upregulated in resistant HCC cells. In addition, in vitro studies showed that overexpression of FOLR1 decreased the sensitivity of HCC cells to sorafenib, whereas siRNA-directed knockdown of FOLR1 increased the sensitivity of HCC cells to sorafenib. Immunoprecipitation-mass spectrometry analysis suggested a strong link between FOLR1 and autophagy-related proteins. Further biological experiments found that FOLR1-related sorafenib resistance was accompanied by the activation of autophagy, whereas inhibition of autophagy significantly reduced FOLR1-induced cell resistance. These results suggest the driving role of FOLR1 in HCC resistance to sorafenib, which may be exerted through FOLR1-induced autophagy. Therefore, this study may provide new insights into understanding the mechanism of sorafenib resistance.

IFI6 Downregulation Reverses Oxaliplatin Resistance of Colorectal Cancer Cells by Activating the ROS-Induced p38MAPK Signaling Pathway

Chen Huang,Tao Zhou,Lihua Ma,Shipeng Zhao

Oxaliplatin (OXA) is a usual chemotherapeutic agent applied in the colorectal cancer (CRC) clinical treatment. Interferon-alpha inducible protein 6 (IFI6) has been proved to promote proliferation and suppress apoptosis in several tumor cells, while the impacts of IFI6 on OXA resistance in CRC still need exploration. HCT116 and SW620 cells were used as the parental to obtain OXA-resistant cells. The influence of IFI6 on OXA sensitivity, cell proliferation and apoptosis were evaluated by overexpression or knockdown IFI6 in cells. In this work, we found that the level of IFI6 was significantly enhanced in HCT116/OXA and SW620/OXA cells as compared to the parental cells. Overexpression of IFI6 decreased the sensitivity of HCT116 and SW620 cells to OXA. However, knockdown of IFI6 enhanced the sensitivity of HCT116/OXA and SW620/OXA cells to OXA. And upregulated IFI6 promoted the proliferation and repressed apoptosis in HCT116 cells, while suppressed IFI6 markedly reduced proliferation and increased apoptosis in HCT116/OXA cells. Additionally, IFI6 suppressed the phosphorylation level of p38, and silenced IFI6 enhanced it. The addition of the p38 kinase inhibitor, SB203580, alleviated the decreased cell proliferation and increased apoptosis in HCT116/OXA cells. Suppressed IFI6 enhanced the reactive oxygen species (ROS) level in HCT116/OXA cells, and blockade of ROS with N-acetyl-L-cysteine (NAC) decreased the enhancement level of ROS and the phosphorylation level of the p38, which was induced by IFI6 down-regulation. We, therefore, implied that suppressed IFI6 reverses OXA-resistance of CRC cells via promoting the ROS-induced p38 mitogen-activated protein kinase (MAPK) signaling pathway.

Identification of Fasudil as a collaborator to promote the anti-tumor effect of lenvatinib in hepatocellular carcinoma by inhibiting GLI2-mediated hedgehog signaling pathway

Yilan Huang,Siwei Wang,Xiaojun Zhang,Chen Yang,Sikai Wang,Hongxia Cheng,Aiwu Ke,Chao Gao,Kun Guo

Lenvatinib is a frontline tyrosine kinase inhibitor for patients with advanced hepatocellular carcinoma (HCC). However, just 25% of patients benefit from the treatment, and acquired resistance always develops. To date, there are neither effective medications to combat lenvatinib resistance nor accurate markers that might predict how well a patient would respond to the lenvatinib treatment. Thus, novel strategies to recognize and deal with lenvatinib resistance are desperately needed. In the current study, a robust Lenvatinib Resistance index (LRi) model to predict lenvatinib response status in HCC was first established. Subsequently, five candidate drugs (Mercaptopurine, AACOCF3, NU1025, Fasudil, and Exisulind) that were capable of reversing lenvatinib resistance signature were initially selected by performing the connectivity map (CMap) analysis, and fasudil finally stood out by conducting a series of cellular functional assays in vitro and xenograft mouse model. Transcriptomics revealed that the co-administration of lenvatinib and fasudil overcame lenvatinib resistance by remodeling the hedgehog signaling pathway. Mechanistically, the feedback activation of EGFR by lenvatinib led to the activation of the GLI2-ABCC1 pathway, which supported the HCC cell's survival and proliferation. Notably, co-administration of lenvatinib and fasudil significantly inhibited IHH, the upstream switch of the hedgehog pathway, to counteract GLI2 activation and finally enhance the effectiveness of lenvatinib. These findings elucidated a novel EGFR-mediated mechanism of lenvatinib resistance and provided a practical approach to overcoming drug resistance in HCC through meaningful drug repurposing strategies.

High salt condition alters LPS synthesis and induces the emergence of drug resistance mutations in Helicobacter pylori

Hongming Huang,Huang Yang,Shunhang Feng,Xiaoyan Zhang,Chu Chen,Hongyu Yan,Rui Li,Mengxin Liu,Juan Lin,Yancheng Wen,Feifei She

The burgeoning emergence of drug-resistant Helicobacter pylori strains poses a significant challenge to the clinical success of eradication therapies and is primarily attributed to mutations within drug-targeting genes that lead to antibiotic resistance. This study investigated the effect of high salt conditions on the occurrence of drug-resistance mutations in H. pylori. We found that high salt condition significantly amplifies the frequency of drug resistance mutations in H. pylori. This can be chiefly attributed to our discovery indicating that high salt concentration results in elevated reactive oxygen species (ROS) levels, initiating DNA damage within H. pylori. Mechanistically, high salt condition suppresses lipopolysaccharide (LPS) synthesis gene expression, inducing alterations in the LPS structure and escalating outer membrane permeability. This disruption of LPS synthesis attenuates the expression and activity of SodB, facilitates increased ROS levels, and consequently increases the drug resistance mutation frequency. Impairing LPS synthesis engenders a reduction in intracellular iron levels, leading to diminished holo-Fur activity and increased apo-Fur activity, which represses the expression of SodB directly. Our findings suggest a correlation between high salt intake and the emergence of drug resistance in the human pathogen H. pylori, implying that dietary choices affect the risk of emergence of antimicrobial resistance.

Synthesis and Antifungal Activity of Novel Tetrahydrogeranyl Quaternary Ammonium Salts

Yun Peng,Jiayu Chang,Zhuangquan Xiao,Jiazong Huang,Ting Xu,Shangxing Chen,Guorong Fan,Shengliao Liao,Zongde Wang,Hai Luo

Due to the excessive use of antifungal agents, drug resistance and ecological problems are increasing. Some antifungal agents are difficult to degrade and have high toxicity and several side effects. In this study, 15 novel tetrahydrogeranyl quaternary ammonium salts (8a–8o) were synthesized from the natural compound citral. The structures of the quaternary ammonium salts were characterized by Fourier transform infrared, proton nuclear magnetic resonance, carbon-13 nuclear magnetic resonance spectroscopy, and mass spectrometry, and the antifungal activities of these compounds at a concentration of 0.25 mg/mL against 10 plant pathogenic fungi were tested. The results showed that compound 8i had the best antifungal activity, and its inhibition rates against Rhizoctonia solani, Phytophthora parasitica var. nicotianae, Sphaeropsis sapinea, Fusarium oxysporum f. sp. niveum, and Poria vaporaria reached 100%. For Fusarium verticillioides, the inhibition rate of compound 8i was 93.28%, which was higher than that of chlorothalonil. In addition, it was found that the inhibition rates of compounds with N,N-di-n-propyl group (8l, 8m) against R solani, F oxysporum f. sp. niveum, S sapinea, P parasitica var. nicotianae, F verticillioides, Colletotrichum acutatum, and Coriolus versicolor were higher than compounds with N,N-diethyl and N,N-dimethyl groups (8a, 8b, 8j, 8k). The inhibition rates of compounds with morpholine groups (8n, 8o) were generally low.

Clinical insight-driven micron-sized cholesterol oxidation platform for membrane lipid therapy of advanced ovarian cancer

Fei Weidong,Xin Yu,Qian Wenqiang,Liu Mingqi,Zheng Caihong,Liu Yunxi,Chen Danfei,Zhou Ying,Xu Shanshan,Wu Xiaodong,Zhao Mengdan

Due to the insidious onset of ovarian cancer, the majority of patients are diagnosed at advanced stages, often presenting with extensive pelvic and abdominal metastasis. A significant proportion of these patients exhibit primary or acquired drug resistance, limiting improvements in the five-year survival rate. In this study, an analysis of patient-derived samples demonstrated a strong correlation between cholesterol levels and ovarian cancer progression. Membrane lipid therapy, a strategy that targets the composition, structure, and function of the cell membrane, has demonstrated notable potential in tumor therapy. Subsequently, a novel cholesterol oxidation-mediated membrane lipid therapy employing PLGA microspheres co-loaded with miriplatin (MiR) and cholesterol oxidase (COD) was proposed for drug-resistant advanced ovarian cancer therapy. The microspheres had a particle size of approximately 2.58 μm, with encapsulation efficiencies of 82.98 ± 0.09% for MiR and 32.83 ± 4.30% for COD. In vitro experiments demonstrated that COD-induced cholesterol oxidation modulated the membrane rigidity and fluidity of SKOV3-TR cells, thereby compromising membrane structural integrity and attenuating tumor cell migration. Additionally, the reactive oxygen species generated during cholesterol oxidation disrupted mitochondrial membrane potential and adenosine triphosphate production. The resulting energy deficiency and compromised membrane integrity reduced the expression and function of drug resistance-associated proteins, thereby enhancing chemosensitivity. Moreover, the combined effects of reactive oxygen species and MiR drove resistant cells towards apoptosis. In vivo studies demonstrated that the large-particle PLGA formulation effectively resided in the peritoneal cavity, resulting in superior therapeutic outcomes against drug-resistant metastatic ovarian tumor, as evidenced by fewer peritoneal metastatic nodules and diaphragmatic colonization sites, more extensive tumor tissue destruction, and prolonged survival. More importantly, PLGA encapsulation significantly decreased the toxic side effects associated with continuous platinum-based chemotherapy. Overall, cholesterol oxidation-mediated membrane lipid therapy represented a promising approach for treating advanced ovarian cancer. Graphical abstract

Tetramethylpyrazine attenuates the cancer stem cell like-properties and doxorubicin resistance by targeting HMGCR in breast cancer

Xuan Jiang,Manli Wang,Guoliang Cui,Yuanyuan Wu,Zhonghong Wei,Suyun Yu,Aiyun Wang,Wei Zou,Yanhong Pan,Xiaoman Li,Yin Lu

Background Tetramethylpyrazine (TMP), a key bioactive constituent derived from Ligusticum wallichii Franchat , has demonstrated efficacy in mitigating multidrug resistance (MDR) in human breast cancer (BC) cells. However, the precise mechanisms underlying its action remain poorly understood. Purpose Cancer stem cells (CSCs) are widely recognized as the primary contributors to MDR. This investigation seeks to elucidate the role and mechanisms through which TMP counteracts MDR by attenuating CSC-like characteristics. Methods Various assays, including flow cytometry, sphere formation, and Western blotting, were employed to evaluate TMP's effects on breast cancer stem cell (BCSC)-like phenotypes in vitro. In vivo , extreme limiting dilution assays and immunohistochemistry (IHC) were executed to assess the impacts of TMP on BCSC frequency and the levels of stemness markers. Mechanistically, RNA sequencing was performed to uncover the key biological processes involved in TMP's effects on BCSCs. Further experiments, encompassing micro scale thermophoresis (MST), drug affinity responsive target stability (DARTS), cellular thermal shift assay (CETSA) and amino acid mutation analyses, were utilized to identify the essential targets and corresponding binding sites of TMP. Finally, the effects of TMP on BCSC-like phenotypes were confirmed using cells with mutated amino acid residues, which allowed us to investigate the specificity of TMP's binding sites. To further evaluate the impact of TMP on drug resistance, doxorubicin-resistant MCF7 (MCF-7 ADR ) cells, along with corresponding cell lines harboring mutated amino acid residues, were employed. Results TMP was found to inhibit BCSC-like properties both in vitro and in vivo , evidenced by a reduction in the CD44 + /CD24 - population, sphere formation capability, and expression of stemness markers. Mechanistic studies revealed that TMP targets 3‑hydroxy-3-methylglutaryl-CoA reductase (HMGCR), a rate-limiting enzyme in cholesterol biosynthesis. TMP binds to Asp-767 of HMGCR, thereby inhibiting its activity and reducing cholesterol synthesis. The influence of TMP on BCSC-like phenotypes was nullified by overexpression of wild-type HMGCR, while mutations in the binding site of HMGCR had no effect on TMP's inhibition of BCSC-like properties. Additionally, TMP mitigated MDR by targeting HMGCR. Conclusion These findings suggest that TMP alleviates MDR by reducing BCSC-like traits through targeting HMGCR and disruption of cholesterol biosynthesis in BC. This provides new insights into the mechanisms through which TMP alleviates MDR and offers new lead compound for exploring HMCGR antagonists.

Improved anti-malarial parasite efficacy with heparin-artemisinin nanoemulsions

Yuxin Zheng,Ning Jiang,Qilong Li,Ruiming Fan,Kexin Zheng,Ziwei Su,Ran Chen,Ying Feng,Yiwei Zhang,Xiaoyu Sang,Qijun Chen

Artemisinin and its derivatives have been widely applied as the most effective drugs in the treatment of malaria since their introduction in the last century. However, challenges such as short half-life, limited bioavailability, and increasing drug resistance have prompted researchers to explore new strategies in the development of antimalarial drugs. In this study, a nano-targeting drug delivery system (HEP@ART@NEs) is developed consisted of heparin as the outer shell and artemisinin in the inner core, utilizing a high-pressure homogenization approach. The specific targeting effect of HEP@ART@NEs is achieved through surface multivalent interactions between heparin with both Plasmodium falciparum merozoites as well as the surface of infected red blood cells (iRBCs). The physical and chemical properties of HEP@ART@NEs, as well as its anti-malarial activity both in vitro and in vivo , are systematically investigated. HEP@ART@NEs demonstrate enhanced antimalarial effects, delay malaria recrudescence, and improve survivability in a rodent model with P. berghei ANKA infection compared to the treatment with conventional artemisinin treatment. Overall, this study indicates that HEP@ART@NEs hold promise as a platform for a rational design of targeted delivery of antimalarial drugs.

Glucocorticoid resistance induced by ANXA5 overexpression in B-cell acute lymphoblastic leukemia

Ju Yang,Ping Liu,Dan Ma,Peng Zhao,Yan Zhang,Yinghao Lu,Yanju Li,Yi Huang,Ying Chen,Jishi Wang

Development of chemo‑resistance is ultimately responsible for treatment failure and relapse in B-cell acute lymphoblastic leukemia (B-ALL). However, the mechanism underlying glucocorticoid (GC) resistance remains unclear. This study was performed to identify GC resistance-related genes using the transcriptome chip from the GEO database, and preliminarily analyze drug resistance mechanism in B-ALL. Here, we found that ANXA5 expression was upregulated in B-ALL cells and high-level ANXA5 was associated with dexamethasone (DEX) resistance. Then, small interfering RNA (siRNA) was designed to silence ANXA5 expression in the B-ALL cell lines, and the apoptotic rate of cells treated with DEX was detected by flow cytometry. As a result, cell apoptosis was dramatically promoted in B-ALL cells following silencing of ANXA5 and DEX administration versus that in ANXA5-silenced alone or DEX-treated alone cells. It was further found that down-regulation of ANXA5 in B-ALL cells significantly increased the relative amount of cleaved Caspase 3 and Caspase 9 induced by DEX. Collectively, inhibition of ANXA5 gene expression may represent a novel method to restore the sensitivity of treatment-resistant B-ALL tumors to GC-induced cell death, which is of important clinical significance to overcome drug resistance associated with B-ALL.

CD44-targeted hyaluronic acid–curcumin reverses chemotherapeutics resistance by inhibiting P-gp and anti-apoptotic pathways

Lu Diao,Ao Shen,Yunxu Yang,Jin Tao,Ying Hu

Chemotherapeutic drug resistance poses a great challenge in cancer therapy. Drug efflux and anti-apoptotic processes are the two most common mechanisms leading to chemotherapy resistance. In this study, we focused on the applicability of curcumin (CUR) as a sensitizer for chemotherapeutics (doxorubicin [DOX] as the model drug) modified with hyaluronic acid (HA) as an effective therapeutic strategy against multidrug resistance (MDR) in cancer cells. We constructed an HA–CUR/DOX delivery system measuring approximately 180 nm with superior encapsulation efficacy and serum stabilities. In vitro, we found that HA modification could facilitate the efficient delivery of chemotherapeutics through CD44 receptor-mediated targeted delivery. MTT assay results confirmed that the combination of CUR and DOX/paclitaxel (PTX) had a significant synergistic effect and significantly reversed MDR. Further experiments including real-time polymerase chain reaction and western blotting proved that the main mechanisms by which CUR reversed MDR in tumor cells were inhibiting the expression and activity of P-glycoprotein (P-gp) and inducing apoptosis through mitochondrial pathway. Taken together, our new engineered tumor-targeting nanoparticle delivery system may have the potential for overcoming MDR in cancer.

Combined Inhibitions of Glycolysis and AKT/autophagy Can Overcome Resistance to EGFR-targeted Therapy of Lung Cancer

Mingtong Ye,Sufan Wang,Ting Wan,Rui Jiang,Yun Qiu,Lei Pei,Nengzhi Pang,Yuanling Huang,Yufeng Huang,Zhenfeng Zhang,Lili Yang

Efficacy of EGFR-targeted tyrosine kinase inhibitors (TKIs), such as erlotinib, to treat human non-small cell lung cancers (NSCLCs) with activating mutations in EGFR is not persistent due to drug resistance. Reprogramming in energy (especially glucose) metabolism plays an important role in development and progression of acquired resistance in cancer cells. We hypothesize that glucose metabolism in EGFR-TKI sensitive HCC827 cells and erlotinib-resistant sub-line of HCC827 (which we name it as erlotinib-resistant 6, ER6 cells in this study) is different and targeting glucose metabolism might be a treatment strategy for erlotinib-resistant NSCLCs. In this study, we found increased glucose uptakes, significant increase in glycolysis rate and overexpression of glucose transporter 1 in ER6 cells compared to its parental cells HCC827. We also found AKT and autophagy of ER6 cells were more activated than HCC827 cells after glucose starvation. Combining glucose deprivation and AKT or autophagy inhibitor could synergize and overcome the acquired resistance against EGFR-targeted therapy for NSCLCs. Our data suggest that the combinations of inhibitors of AKT or autophagy together with glucose deprivation could be novel treatment strategies for NSCLC with acquired resistance to targeted therapy.
