Unsaturated fatty acid promotes the production of triterpenoids in submerged fermentation of Sanghuangporus baumii

Jing Huang,Kun Wang,Song Zuo,Lei Chen,Zhongyang Ding,Mohamed El-shazly,Bo-Bo Zhang

Sanghuangporus species are precious edible and medicinal fungi that are traditionally used in Asian countries. Triterpenoids, the major bioactive constituents of Sanghuangporus spp. are currently receiving a lot of attention from the scientific community due to their potent pharmacological activities. However, very few reports have been conducted about the possibility of adjusting the fermentation conditions to increase the triterpenoids yield from certain fungi such as Sanghuangporus baumii . In this study, we examined the effects of four different extracts from mulberry shavings on the production of triterpenoids and traced the major components from the effective extracts by GC-MS to provide a reference for further exploration of the triterpenoids synthetic mechanism. It was found that unsaturated fatty acids significantly promoted the production of triterpenoids. The results suggested a direct correlation between the degree of unsaturation of fatty acid and the triterpenoids yield. The best stimulatory additive was linolenic acid . It increased the triterpenoids yield to 573 mg/L, which was 2.53 times higher than the control. This study not only showed that addition of mulberry shavings extract was an efficient fermentation protocol to improve the triterpenoids yield of S. baumii , but it also highlighted the importance of fatty acid content in the production of triterpenoids.

Organic pollutants removal by phosphoric acid modified biochar from residue of Inonotus obliquus

Caili Guo,Fangcong Hua,Peiying Chen,Xiaoyin Wang,Xiaohong Hou,Jianhua Qu,Qi Hu

Herbal medicine is used widely, and the residues increase rapidly. Biochar preparation from the residues is a potential way to treat wastes with wastes. Inonotus obliquus (IO) is a natural porous fungus and is famous for its hypoglycemic effect. The residues of Inonotus obliquus (IOR) are potential raw materials for biochar because of their porous structure. P-IORBC and Zn-IORBC, activated by H 3 PO 4 and ZnCl 2 respectively, were prepared from IOR. The distinct properties of P-IORBC and Zn-IORBC were shown in the study. P-IORBC was optimized by response surface method (RSM), and the optimized parameters of P-IORBC preparation were 166 min (pyrolysis time), 374 °C (pyrolysis temperature), and 3.91 (the mass ratio of H 3 PO 4 to IOR). The actual methylene blue (MB) adsorption (951.56 mg/g) was near the predicted value (971.00 mg/g), indicating the reliability of the response surface model. Zn-IORBC has a larger pore volume and pore size, while P-IORBC has a larger specific surface area. The specific surface areas of Zn-IORBC and P-IORBC were 1676.78 and 2014.51 m 2 /g, respectively. Compared to Zn-IORBC, the pyrolysis temperature of P-IORBC was reduced from 547 °C to 374 °C. The tetracycline (TC) adsorption for P-IORBC and Zn-IORBC was 1515.46 mg/g and 1033.66 mg/g. P-IORBC was more effective at the adsorption of cationic dyes than Zn-IORBC. In general, P-IORBC had more potential due to its high yield and environmentally friendly characteristics. The data provides a reference for biochar preparation from residues of herbal medicine.

Ultraviolet radiation promotes the production of hispidin polyphenols by medicinal mushroom Inonotus obliquus

Rong Zhou,Hanbing Yang,Ting Lu,Yanxia Zhao,Weifa Zheng

Production of hispidin polyphenols in Inonotus obliquus is a stress-induced response triggered by environmental factors. As one of the important environmental factors, ultraviolet (UV) radiation plays regulatory roles in fungal growth and development . However, whether UV radiation regulates the formation of hispidin polyphenols remains to be established. In this study, we cultivated I. obliquus on solid medium and imposed intermittent UV radiation. We showed that UV exposure inhibited the growth of mycelia but increased the production of polyphenols. Further bioassays revealed that UV radiation also increased the catalytic activities of phenylalanine ammonia-lyase (PAL) and chalcone isomerase (CHI), up-regulated expression of genes related to redox, transcriptional regulation, and metabolism. In addition, the total extracts from the UV-irradiated group were more capable of scavenging DPPH and ABTS + free radicals, especially at the later stage of culture. Thus, UV radiation, acting as one of the environmental factors, stimulated the accumulation of polyphenols in I. obliquus by regulating the activities of enzymes and the expression of genes related to growth and metabolism, and can be tentatively used as a feasible strategy to enhance the production of polyphenols in I. obliquus .

Alterations in Functional Constituents and Bioactivities of Tartary Buckwheat via Solid-State Fermentation with Three Edible-Medicinal Fungi: A Metabolomics-Based Analysis

Qi Li,Jiaying Zhu,Xiangying Lv,Jin Liu,Hong Liu,Qingyuan Wang,Yunlong Li

To functionally enhance Tartary buckwheat and elucidate the underlying mechanisms of change, solid-state fermentation (SSF) was conducted using three edible-medicinal fungi—Auricularia auricula(A. auricula),Ganoderma lucidum(G. lucidum), andHericium erinaceus(H. erinaceus). The in vitro antioxidant (DPPH/ABTS) and α-glucosidase inhibitory activities were quantitatively evaluated. Notably, SSF withH. erinaceusspecifically elevated α-glucosidase inhibitory activity by 50% under the tested conditions. Non-targeted metabolomics further profiled metabolite alterations to identify key up-regulated bioactive compounds. Epicatechin gallate (ECG) was significantly up-regulated in all three samples, and the fold change in quercetin 3′-O-sulfate in GFTB was significantly higher than that in the other two samples. Metabolic pathway analysis identified the biosynthesis of secondary metabolites and the metabolism of terpenoids and polyketides as the most prominently affected pathways. This study demonstrates that SSF with edible-medicinal fungi is an effective bioprocessing strategy to boost the bioactivity and value of Tartary buckwheat.

Metabolomics analysis of rice fermented by medicinal fungi providing insights into the preparation of functional food

Xing Luo,Ji-Hang Jiang,Shi-Liang Liu,Jian-Yun Gao,Li-Wei Zhou

Rice, a primary staple food, may be improved in value via fermentation. Here, ten medicinal basidiomycetous fungi were separately applied for rice fermentation. After preliminary screening, Ganoderma boninense , Phylloporia pulla , Sanghuangporus sanghuang and Sanghuangporus weigelae were selected for further LC-MS based determination of the changes in metabolic profile after their fermentation with rice, and a total of 261, 296, 312, and 355 differential compounds were identified, respectively. Most of these compounds were up-regulated and involved in the metabolic pathways of amino acid metabolism, lipid metabolism, carbohydrate metabolism and the biosynthesis of other secondary metabolites. Sanghuangporus weigelae endowed the rice with the highest nutritional and bioactive values. The metabolic network of the identified differential compounds in rice fermented by S. weigelae illustrated their close relationships. In summary, this study provides insights into the preparation and application of potential functional food via the fermentation of rice with medicinal fungi.

Metabolomics-based analysis of changes in functional constituents and activities of Tartary buckwheat by solid-state fermentation with three kinds of edible-medicinal fungi

Jiaying Zhu,Ting Zhang,Linhua Cui,Hong Liu,Qingyuan Wang,Yunlong Li,Qi Li

Solid-state fermentation of Tartary buckwheat using Auricularia auricula , Ganoderma lucidum , and Hericium erinaceus enhanced its functional components, increasing levels of phenols, polysaccharides, proteins, and amino acids, alongside improved antioxidant activity. The fermentation notably elevated flavonoid levels, particularly Quercetin-3-O-sulfate, which increased tenfold with Ganoderma lucidum . Analysis revealed 3078 metabolites, with downregulated metabolites predominantly being amino acids, carbohydrates, flavonoids, and phenolic compounds. Key metabolic pathways identified included amino sugar metabolism and biosynthesis of plant secondary metabolites, as per KEGG enrichment analysis. This research highlights solid-state fermentation as a promising method to enhance the functional properties of Tartary buckwheat, paving the way for developing functional foods.
