Development of a Captiva EMR-lipid clean-up and LC-MS/MS methodology for detecting H2 receptor antagonists in functional foods and food-medicine homology materials

Guodong Shan,Xiuqin Fan,Qingcheng Wang,Li Zhang,Lijun Ge,Weibo Lu,Jing Xue,Kang Chen,Zejun Wang,Qing Shen

The inappropriate use of H2 receptor antagonists (H2RAs) in functional foods (FFs) and food-medicine homology materials (FMHs) designed to enhance gastrointestinal function (IGF) presents health risks due to potential adverse effects. In this study, an analytical method that integrates Captiva EMR-Lipid (CEL) solid-phase extraction (SPE) with liquid chromatography tandem mass spectrometry (LC-MS/MS) has been developed for the detection of H2RAs (nizatidine, famotidine, cimetidine, ranitidine, and lafutidine) in these products. Through the optimization of CEL-SPE parameters, such as using 200 mg of sorbent and eluting with ACN/water (80/20, v/v), the recovery rates of H2RAs in IGF-FF and IGF-FMH ranged from 87.4 % to 94.6 % and 90.2 % to 97.1 %, respectively. The assessment of matrix effects revealed that the five H2RAs exhibit matrix suppression, necessitating the construction of matrix-matched standard curves with correlation coefficients (r 2 ) exceeding 0.9991. The detection limits ranged from 0.4 to 0.8 μg·kg −1 and the quantification limits ranged from 1.3 to 2.7 μg·kg −1 , with recovery rates spanning from 85.4 % to 97.1 % and relative standard deviations ≤ 15 %. The successful development of this technique not only helps safeguard consumer health but also provides scientific basis and technical support for market supervision, thereby preventing and addressing food safety risks.

Development and validation of an ultra-performance liquid chromatography tandem mass spectrometry for target determination of bioactive compounds in Dendrobium spp.

Jingjing Liang,Ruyan Chen,Wenping Zhang,Lei Luo,Yun Wang,Qing Shen

As a medicine-food homology herb, Dendrobium spp. has versatile applications in modern medicine and food industry. Herein, an ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) based method was established for simultaneous quantification of six active components, including gigantol, erianin, naringenin, quercetin, rutin, and p -coumaric acid in Dendrobium spp., on the basis of optimized sample preparation, mass spectrometry conditions, and chromatography conditions. Sample extraction was carried out using methanol at a temperature of 60 °C, followed by separation on a T3 C18 column utilizing a gradient eluting program. The results demonstrated excellent linearity (r > 0.999) for the six active components within a specified concentration range. The average recovery rates ranged from 84.7 % to 106.9 %, and the precision (RSD) was within 7.4 %. The detection and quantification limits of this method ranged from 0.34 to 4.17 ng mL −1 and 1.12–13.91 ng mL −1 , respectively. The established method demonstrates high accuracy and reliability and is applicable in practical sample detection. Different Dendrobium spp. exhibit specific variations in compound composition, with D . fimbriatum Hook. having a higher content of benzyl compounds and D . crystallinum . Rchb. f. having a higher content of flavonoids. This study provides experimental evidence for the quality and safety regulation of Dendrobium spp.

Occurrence, transfer rule, and dietary risk assessment of aflatoxins in medicinal and edible seeds

Jianxin Lv,Huiru Zhang,Xuwen Wang,Jiaoyang Luo,Yudan Wang,Dandan Kong,Chuang Wang,Mengyue Guo,Meihua Yang

The last three decades have seen a rise in the use of herbal remedies and supplements, with more than 80% of the world's population relying on them for primary care. However, aflatoxin contamination in herbal remedies may constitute a risk to human health. In this study, a simple and robust method for the simultaneous detection of aflatoxins in four types of medicinal and edible seeds was developed by using UFLC-MS/MS combined with QuECHERS pretreatment. Meanwhile, the transfer rule of the aflatoxins in medicinal and edible seeds during the decoction process was explored, followed by dietary risk assessment based on Monte Carlo simulation. Results showed that aflatoxin contamination was found in 80.6% of the samples (62/77), with the contamination levels of 0.04–258.72 μg/kg. Notably, high levels of aflatoxins were found in Ziziphi Spinosae Semen and Platycladi Semen, while low aflatoxin transfer rates (1.13%–11.86%) were observed during the decoction process. The estimated daily intake (EDI) of AFB 1 could reach 15.314 and 0.333 ng/kg bw/day by ingesting aflatoxin-susceptible Ziziphi Spinosae Semen and Platycladi Semen, respectively. The population cancer risks ranged from 3.34 × 10 −8 to 2.86 × 10 −4 hepatocellular carcinoma cases per 10 5 persons per year. The probabilistic risk assessment indicated a short-term and long-term health concern of intaking these two medicinal and edible seeds contaminated by aflatoxins. This study revealed the occurrence and transfer pattern of aflatoxins in the medicinal and edible seeds and evaluated the risks of their dietary exposure, providing a basis for their safe use.

Modification of Kudzu Starch with Food-Medicine Homology Extracts Alters Starch Digestion

Yueyuan Chen,Ningtao Zhao,Jingru Song,Xiaojie Yan,Xiaohua Jiang,Fenglai Lu,Min Zhang,Dianpeng Li

Starch-guest complexes are gaining popularity due to their low digestibility and unique physicochemical properties. In this study, kudzu starch (KS) was modified by complexing it with four food-medicine homology extracts (FMH) to form starch-guest complexes (KS-FMH). The assembly process involved pre-gelatinization, autoclaving, and ultrasonic treatment. Digestion and structural characterization of KS-FMH were investigated to assess their potential as low glycemic index (GI) foods. Following the complexation treatment, the KS-FMH exhibited a more favorable hydrolysis pattern compared to native KS, showing a lower hydrolysis rate and equilibrium concentration ( C ∞ ) ( p < 0.05). Moreover, the addition of FMH affected the structural properties of KS, possibly due to non-covalent interactions between KS and FMH. The morphology and distribution of phenolic guests in KS-FMH were determined using scanning electron microscopy, confocal laser scanning microscopy, and high-performance liquid chromatography. These analyses demonstrated the selective inclusion characteristics of FMH in complexation, confirming the formation of KS-FMH complexes. This study underscores the potential of FMH as functional additives for creating low-GI diets with KS, thus promoting their practical application in the food industry.

Determination of pyrethroid pesticides in medicine–food homology tea via homogeneous liquid–liquid microextraction coupled with high-performance liquid chromatography

Xiaojiao Zheng,Yulin Wang,Hongli Zhang,Qingrong Peng,Xianzhao Zhang,Xu Jing,Fanrong Zhao,Jiajun Han

A green, rapid, and efficient homogeneous liquid–liquid microextraction technique involving the use of bio-based solvent 2-methyltetrahydrofuran was developed to extract deltamethrin, beta-cypermethrin, and bifenthrin from medicine–food homology tea samples, including mint-, ginger-, cinnamon- and chrysanthemum-based tea. Various solvents were screened via a conductor-like screening model, and three superior solvents were optimised and validated. 2-Methyltetrahydrofuran was selected as the extractant and added to the sample, and phase separation was induced with ammonium sulfate, achieving extraction in under 1 min without centrifugation. Mechanistic studies using density functional theory calculations and molecular docking revealed that van der Waals forces and hydrogen bonds dominate the interactions. The method exhibited excellent linearity within the range of 0.005–0.5 μg mL −1 and recoveries between 80.6 % and 109.8 %. The environmental sustainability of the entire process was verified. These results emphasise the methold's potential for applications in the trace analysis of pesticides in diverse food matrices .

Rapid Construction of Recombinant PDCoV Expressing an Enhanced Green Fluorescent Protein for the Antiviral Screening Assay Based on Transformation-Associated Recombination Cloning in Yeast

Yucan Zhang,Chenxi Shi,Jin Yuan,Yue Zhang,Xiaohui Jin,Shaopo Zu,Honglei Zhang,Hui Hu

Porcine deltacoronavirus (PDCoV) is an emerging enteropathogenic coronavirus that mainly causes diarrhea and death in suckling piglets and also has the potential for cross-species transmission, threatening public health. However, there is still no effective vaccine or drug to prevent PDCoV infection. In order to accelerate the development of antiviral drugs, we established a high-throughput screening platform using a novel genome editing technology called transformation-associated recombination cloning in yeast. The recombinant PDCoV and PDCoV reporter virus expressing enhanced green fluorescent protein were both rapidly rescued with stable genealogical characteristics during passage. Further study demonstrated that the reporter virus can be used for high-throughput screening of antiviral drugs with a Z-factor of 0.821–0.826. Then, a medicine food homology compound library was applied, and we found that three compounds were potential antiviral reagents. In summary, we have established a fast and efficient reverse genetic system of PDCoV, providing a powerful platform for the research of antiviral drugs.

A medicine and food homology formula prevents cognitive deficits by inhibiting neuroinflammation and oxidative stress via activating AEA–Trpv1–Nrf2 pathway

Li Ming-Jie,Xu Jing-Yi,Zhang Hua-Yue,Guo Min,Lan Meng-Ning,Kong Jie,Liu Shi-Wei,Zheng Hua-Jun

Alzheimer’s disease (AD) is a neurodegenerative disorder frequently accompanied by neuroinflammation and oxidative stress. The medicine and food homology (MFH) has shown potential for treating neuroinflammation and oxidative stress. This study aimed to provide a safe and efficient therapy for AD based on MFH. In this study, we develop a MFH formula consisting of egg yolk oil, perilla seed oil, raphani seed oil, cinnamon oil, and noni puree (EPRCN). To evaluate the ameliorative effects of EPRCN on AD-related symptoms, a mouse model of AD was constructed using intraperitoneal injection of scopolamine in ICR mice. Experimental results demonstrated that EPRCN supplement restored behavioral deficits and suppressed neuroinflammation and oxidative stress in the hippocampus of scopolamine-induced mice. An in vitro study was then performed using induction of Aβ (25–35) in glial (BV–2 and SW–1783) and neuron (SH–SY5Y) cell lines to examine the improvement mechanism of EPRCN on cognitive deficits. Multi-omics and in vitro studies demonstrated that these changes were driven by the anandamide (AEA)-Trpv1-Nrf2 pathway, which was inhibited by AM404 (an AEA inhibitor), AMG9810 (a Trpv1 inhibitor), and BT (an Nrf2 inhibitor). Consequently, EPRCN is an effective therapy on preventing cognitive deficits in mouse models of AD. In contrast to donepezil, EPRCN exhibits a novel modes action for ameliorating neuroinflammation. The mechanism of EPRCN on preventing cognitive deficits is mediated by improving neuroinflammation and oxidative stress via activating the AEA–Trpv1–Nrf2 pathway.
