In this work, the interaction mechanism, structure and foaming properties between three whey proteins (α-lactalbumin (ALa), β-lactoglobulin (BLg) and bovine serum albumin (BSA)) and Mogroside V (Mog) at pH 7.0 were systematically formulated via multi-spectroscopic analysis and silico methods. Mog quenched the fluorescence of three whey proteins in a static mode. Mog had greater binding affinity for ALa than that for BLg or BSA. Hydrogen bonds, van der Waals force and hydrophobic interaction acted as the primary driven forces in ALa/BLg/BSA-Mog system. Moreover, content of α-helix in ALa and β-sheet in BLg decreased after Mog addition, whereas there was no significant change in the secondary structure of BSA. In addition, hydrophobic crevice in ALa, “bottom of β-barrel” in BLg and interfacial region between two domains in BSA were the binding areas for Mog using molecular docking. Molecular dynamics (MD) simulation further demonstrated that the complex of ALa and Mog possessed the lowest binding free energy and most stable state. Furthermore, compared with single proteins, the foaming ability of protein/Mog complexes were improved. Especially, Ala-Mog attained the highest increase rate. Bubble morphology presented that three whey proteins containing Mog possessed compacter, smaller and even bubbles. This research could establish theoretical basis for the binding mechanism between different food proteins and low weight surfactants. Besides, whey protein-saponin complexes could be served as the novel food-grade foaming agents applied in food industry.

Morroniside is a biologically active polyphenol found in Cornus officinalis Sieb . et Zucc (CO) that exhibits a broad spectrum of pharmacological activities, such as protecting nerves, and preventing diabetic liver damage and renal damage. However, little data are available regarding the mechanism of its intestinal absorption. Here, an in vitro human intestinal epithelial cell model of cultured Caco-2 cells was applied to study the absorption and transport of morroniside. The effects of donor concentration, pH and inhibitors were investigated. The bidirectional permeability of morroniside from the apical (AP) to the basolateral (BL) side and in the reverse direction was studied. When administered at three tested concentrations (5, 25 and 100 μM), the apparent permeability coefficient (P app ) values in the AP-to-BL direction ranged from 1.59 × 10 −6 to 2.66 × 10 −6 cm/s. In the reverse direction, BL-to-AP, the value was ranged from 2.67 × 10 −6 to 4.10 × 10 −6 cm/s. The data indicated that morroniside transport was pH-dependent. The permeability of morroniside was affected by treatment with various inhibitors, such as multidrug resistance protein inhibitors MK571 and indomethacin, as well as the breast cancer resistance protein inhibitor apigenin. The mechanisms of the intestinal absorption of morroniside may involve multiple transport pathways, such as the passive diffusion and efflux protein-mediated active transport especially involving multidrug resistance protein 2 and breast cancer resistance protein. After the addition of CO, the P app values in the AP-to-BL direction increased significantly, therefore, it can be assumed that some ingredients in the CO promote morroniside absorption in the small intestine.

Chronic fructose consumption is becoming one leading risk factor for NAFLD due to its hazard on cellular lipid metabolism, which plays an essential role in the pathogenesis of NAFLD. Morroniside, an iridoid glycoside from Cornus officinalis, has been suggested to have potent in regulating cellular glucolipid metabolism. However, the effect and mechanism of morroniside on improving fructose-induced hepatic steatosis and retarding NAFLD progression remain ambiguous. In this study, we illustrate the efficacy of morroniside in alleviating fructose-triggered hepatic steatosis in mice. Mechanically, morroniside suppressed de novo lipogenesis and promoted β-oxidation by inhibiting the activation of PGC1β. Additionally, morroniside was found to work in attenuating hepatic inflammation in response to long-term fructose intake. Taken together, the current study reveals that morroniside is a promising food and medicinal therapy for NAFLD treatment and is effective in delaying the progression of NAFLD to NASH, especially in subtypes caused by excessive carbohydrate intake.
